Studies on the Ginseng Saponins(I) On the Determination of the Ginseng Saponins in Ginseng Tea and Extract (인삼 사포닌에 관한 연구 (I) 인삼 사포닌의 분별 정량에 대하여)

  • A determinatien of the saponins in ginseng tea and extract was carried out by using the quantitative TLC autodetector equipped with a hydrogen flame ionization detector. In order to apply to the Quality central of the ginseng tea and extract. the optimum condition and recovery percentage for the quantitative determination of saponins in these products duo studied. The results obtained were as follows: The method was adequate to estimate whether the ginseng extract used for the Products and She raw ginseng extract were the same quality or net. Most of the individual peak area was increased with the concentration of the total saponin. But some of the peak areas were net increased quantitatively in the ease of the sample containing high concentration ginseng extract. To deternine the saponins in ginseng tea correctly high volume low concentration was better than the low volume high concentration. Optimum concentration of ginseng extract in sample to determine the individual saponins was in the range of 0.5∼1.5g. The recovery percentage of the total saponin was 99.5% on the average.

  • Saponins, the major component of ginseng root, mediate the pharmacological action of the ginseng. It has been reported that ginseng roots have protective effect against various toxins. In this study, the effects of ginseng total saponin (GTS) on tumor necrosis factor-alpha (TNF-$\alpha$) production induced by bacterial toxin was investigated. TNF-$\alpha$ level in lipopolysaccharides (LPS)-activated serum was remarkably reduced by intraperitoneal administration (50 mg/kg)of ginseng total saponin (GTS) into mice. The inhibitory effect against TNF-$\alpha$ production was not significant when GTS was given after the LPS injection, and by oral administration. These results suggested that ginseng root may have protective activity against liver damage accompanying the overproduction of TNF-$\alpha$ and GTS is the active component of ginseng.

Potential Effects of Ginseng Saponin Fractions on Macrophage Chemotaxis and Intracellular Calcium and Actin Mobilization (대식세포의 화학주성과 세포내 칼슘과 Actin의 증가에 미치는 인삼사포닌 성분의 영향)

  • In the present study, We have tested the potential effects of ginseng saponin fractions on macrophage chemotaxis and intracellular calcium and F-actin mobilization. Peritoneal macrophages treated with various ginseng saponin fractions showed 28.4% to 71% of increasement of chemotaxis as compared with untreated cells. The activity of intracelluar calcium mobilization was increased up to 65% by treatment with saponins, and F-actin content also increased 10% in the cells loaded with NBD-phallacidin. When the cells were activated with calcium of PMA and treated with saponin fractions, the intracelluar F-actin content increased significantly and prolonged for 2 minutes. These results suggest that ginseng saponin fractions might be a chemoattractants.

Effects of Media and Growth Regulators on the Growth and Saponin Production of Ginseng Root (인삼 Root의 생장 및 사포닌 생성에 미치는 배지와 생장조절물질의 영향)

  • Effects of media and growth regulators on the growth and saponin accumulation of ginseng root were investigated to develop the ginseng root culture system. When Panax ginseng C. A. Meyer roots were induced and cultured in various liquid media, the maximum root growth and saponin production were obtained in SH medium and an initial doubleing time of ginseng root was approximately 10 days. The patterns and contents of ginsenosides of cultured ginseng root in various media were different from each other. SH and White media resulted in higher total ginsenosides contents than the other media. Among different combinations and concentrations of growth regulators, SH medium containing 4.0mg/ L NAA gave best growth of ginseng roots, while saponin content was highest in SH medium containing 0.5mg/L BAP. These results suggested that the rapid production of ginseng saponin is possible by root culture of Panax ginseng.

Changes of saponin Contents in Panax ginseng Leaves by Different Harvesting Months (인삼엽의 채엽시기에 따른 사포닌 성분의 함량 및 조성)

  • To study of production of Panax ginseng leaf tea, after harvested the leaves in July, August, and September as ripening season, the content and composition of ginseng saponin were investigated. 1. Crude saponin contents in the leaves were a about 16.5%, and they were found to be lower in the leaf harvested in September than those harvested in July or August. 2. As similar patterns were observed with month to month in ginsenoside, sum of major ginsenosides of -Re, -Rd and -Rg1 was fixed about 70% of saponin at harvested in each month. And minor components were ginsenoside -Rb1, -Rb2 and -Rc as in order. 3. The ratio of protopanaxadiol(PD)/protopanaxatriol(PT) was revealed reduction of 1.13 of harvested in July to 0.85 of those in September gradually. The contents of protopanaxadiol were high in the leaves of August and protopanaxatriol was high in those September.

  • Preventive effect of the saponin fraction extracted from Panax ginseng C.A. Meyer against ethanol intoxication of the liver has been investigated biochemically and morphologically. Previous work in this laboratory showed that the moderate amounts of ginseng sponins stimulated several enzymes including mitochondrial dehydrogenases and transaminases so far examined in vitro. It was also realized that the half life of the saponin in the liver was estimated approximately five hours and the saponin concentration in the liver was around $10^{-5}\%$ level at two hours after the saponin (1mg) administration orally. In this study, it was confirmed that ginseng saponins stimulated alcohol dehydrogenase, aldehyde dehydrogenase and microsomal ethanol oxidizing system in vivo as well as in vitro. It seemed likely that toxic aldehyde formed during ethanol oxidation in the body might be removed relatively quickly from the liver and the excess hydrogen was used for the biosynthetic work in the presence of the saponin, resulting in the liver protection from alcohol intoxication. Electron microscopic observation demonstrated that the hepatocytes of rats doses with $12\%$ ethanol instead of water for six days were found severely damaged while those of the ginseng saponin administered rats were not impaired suggesting that the sapcnin protected the liver against ethanol intoxication.

An Rapid Extraction of Ginseng Saponin Compounds (인삼사포닌 화합물의 신속한 추출)

  • A new rapid saponin extraction method was developed with using of organic solvent and waring blonder. There was a good correlation between previous distillation method and this method in f major ginsenosides ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rg1) contents. When the ratio of methanol and chloroform was 7:3, this method showed similar saponin contents (total major. ginsenosides contents) comparing with distillation method. Contents of total major ginsenosides were 2.41% in this method and 2.54% in distillation method. However, crude saponin content of this method was higher than that of distillation method.

  • The effect of the saponin fraction extracted from Panax ginseng C.A. Meyer on the antioxidant activity of ${\alpha}-tocopherol$ was investigated in vitro as well as in vivo. Microsomal preparation of albino rat (Sprague-Dawley, 180-200g) was incubated in the mixture containing NADPH, $Fe^{3+},$ ATP, ${\alpha}-tocopherol$ with and/or without ginseng saponin fraction for 30 minutes and the malondialdehyde formed was assayed and found that the saponin fraction stimulated the antioxidant activity of ${\alpha}-tocopherol$ cooperatively. It was also realized that the cooperative stimulation of the antioxidant activity of ${\alpha}-tocopherol$ was most eminent when the concentration of the saponin fraction was around $10^{-5}\%$ in the reaction mixture. Alcoholic suspension of ${\alpha}-tocopherol$ with and/or without ginseng saponin fraction was administered orally to rats in which the lipid peroxidation was induced by ethanol administration and the lipid peroxide contents of the liver were assayed at certain periods of time after ${\alpha}-tocopherol$ administration in this animal. It was reported that the saponin fraction stimulated the absorption of ${\alpha}-tocopherol$ in rats and this was confirmed again in the present work. From the previous work and present experimental results, it seemed that the saponin fraction accelerated the absorption of ${\alpha}-tocopherol$ and therefore stimulated the antioxidant activity of ${\alpha}-tocopherol$ more effectively in the animal body.

Saponins of Gynostemma pentaphyllum (돌외의 Saponin 성분에 관한 연구)

  • Saponins of Gynostemma pentaphyllum that collected from Geochang region(Korea) and Dokujima(Japan) were extracted by the method for ginseng saponin. Comparison by retention time in chromatogram(HPLC) of G. pentaphyllum to that of ginseng showed that it is hardly to find out a common saponin between penta-phyllum and ginseng saponins. Saponin content extracted from G. pentaphyllum growing in Geochang region was higher than that growing in Dokujima region.
