The study was performed to investigate the property of rhizosphere microorganisms, and community structure during GMO, and Non-GMO rice cultivation. In the dilution plate technique, there were no significant differences in microbial populations of rhizosplane with genetically modified, and non-genetically modified rice cultivation, and rhizosphere were also the same results. Dominant bacterial genera were Afipia 12.5%, Spingomonas 10.0%, Ramlibacter 10.0%, Mycobacterium 7.5%, and Tetrasphaera 7.5% in rhizosphere soil of genetically modified rice plant, while Afipia 7.3%, Spingomonas 12.2%, Ramlibacter 7.3%, Mycobacterium 17.1%, Tetrasphaera 14.6% in non-genetically modified cultivated at Suwon test fields in 2006. Majorgenera isolated from root surface cultivated in Yesan fields were Arthrobacter 12.7% in rhizoplane of genetically modified plant, and Burkholderia 22.2% of non-genetically modified plant in 2007, Paucimonas 26.6% of genetically modified plant, Chryseobacterium 15.4% of non-genetically modified plant in 2008. Also the microbial communities in rhizosphere soils of genetically modified, and non-genetically modified plants were characterized using phospholipid fatty acid, and denaturing gradient gel electrophoresis. The phospholipid fatty acid profiles of soils in this condition showed different pattern, but did not show significant differences between soils cultivated with genetically or non-genetically modified rice plants.

One of the primary concerns about the environmental risks of genetically modified (GM) crops is that they may have adverse effects on the local arthropod communities. In this study, we investigated whether the arthropod diversity and community structure in fields of GM rice tolerant to protoporphyrinogen oxidase (PPO)-inhibiting herbicides differ from those in non-GM (control) rice fields. The aim of this study was to assess the potential adverse effects of GM rice on the local arthropod communities. During the growing seasons in the study period, we collected arthropods from both fields by using yellow sticky traps and compared the diversity and community structure of arthropods from the two sites. Overall, the GM rice had no significant effect on the diversity of the local arthropod communities. In addition, multivariate analyses (permutational multivariate analysis of variance and nonmetric multidimensional scaling) showed that the structures of arthropod communities were not affected by the rice genotype (GM vs. non-GM), although these comparisons were made using data obtained at different sampling dates.

We investigated the systemic effects of leaf folder (Cnaphalocrocis medinalis) resistant rice (transgenic rice, Nakdongbyeb) in Sprague-Dawley rats for 13 weeks. Leaf folder resistant rice was added to the diet at percentage levels of 0, 5 and 20 percentage/feeder and was administered for 13 weeks. The results did not show any changes in food and water intake. There were also no biologically significant changes in both body and organ weights, hematological and blood biochemical parameters, autopsy and histopathology between the treatment and control groups. Based on these results, no observed adverse effect level (NOAEL) of transgenic rice was considered to be more than 10,000 mg/kg b.w. under the conditions of the present study.

This experiment was conducted to evaluate pollen viability of Nakdongbyeo, transgenic rice lines, an F$_1$ hybrid from a cross between Milyang weedy rice and ABC-promoter transgenic rice line containing basta-resistant (bar) gene and subsequent selfed progenies, F$_2$ and F$_3$. The reaction of pollen with 3-{4,5 dimethylthiazolyl-2}-2,5-diphenyl monotetrazolium bromide (MTT) as a staining chemical immediately after pollen shedding showed maximum pollen viability of 86% in Nakdongbeyo, 75% in ABC-promoter transgenic rice line, 62% in ubiquitin-promoter transgenic line, 68% in F$_1$, 79% in F$_2$ and 78% in F$_3$. Viability gradually declined during subsequent observations at 20-minute intervals. However, there was a drastic decline in pollen viability after 40 minutes of pollen shedding. The mean difference of pollen viability among rice lines and time was highly significant, indicating significantly different pollen viabilities at different time intervals. Maximum viability of 36.2% was observed in F$_3$ and minimum viability of 3.5% was found in F$_2$ at 90 min after pollen shedding. Results of this experiment on pollen viability and longevity elucidate potential risks of pollen-mediated flow of herbicide-resistant gene from transgenic rice lines and possible integration of it into the weedy rice population.

This study was conducted to analyze nutrient composition of a genetically modified $\beta$-carotene biofortified rice (GM rice), developed by the Rural Development Admistration in Korea. The nutritional constituents of GM rice were compared with those of the parental rice cultivar 'Nakdong' as a non-GM control to access nutritional equivalence. Proximate components (moisture, starch, protein, lipid, and ash) of the GM rice were similar to those of the conventional non-GM rice. $\beta$-Carotene contents of GM brown and milled rice were 2.35, 2.03 ${\mu}g/g$(d.b.), respectively. There were no significant differences between the GM and non-GM rice with respect to most of their nutrient composition, despite minor differences in most amino acids and minerals. This result demonstrated that the nutritional composition of this GM rice would be equivalent to that of the parental non-GM rice without major changes in its chemical contents.
Genetically modified (GM) crops have been developed worldwide through the recombinant DNA technology and commercialized by global agricultural companies. Until now, GM crops have not been cultivated commercially in Korea. Commercialization of GM crops requires a compulsory assessment of environmental risk associated with the release of GM crops. This study was conducted to evaluate the frequency of pollen mediated gene flow from Bt transgenic rice (Agb0101) to japonica non-GM rice (Nakdongbyeo), indica non-GM rice (IR36), and weedy rice (R55). A total of 729,917, 596,318 and 230,635 seeds were collected from Nakdongbyeo, IR36, and R55, respectively, which were planted around Agb0101. Selection of the hybrids was determined by repeated spraying of herbicide and Cry1Ac1 immunostrip assay. Finally, the hybrids were confirmed by PCR analysis using specific primer. The hybrids were found in all non-GM rice and out-crossing ranged from 0.0005% at IR36 to 0.0027% at Nakdongbyeo. All of hybrids were located within 1.2 m distance from the Agb0101 rice plot. The meteorological elements including rainfall and temperature during rice flowering time were found to be important factors to determine rice out-crossing rate. Consideration should be taken for many factors like the meteorological elements of field and physiological condition of crop to set up the safety management guideline to prevention of GM crops gene flow.

The cultivation of genetically modified (GM) crops has increased due to their economic and agronomic advantages. Before commercialization of GM crops, however, we must assess the potential risks of GM crops on human health and environment. The aim of this study was to investigate the possible impact of Bt rice on the soil microbial community. Microbial communities were isolated from the rhizosphere soil cultivated with Bt rice and Nakdong, parental cultivar and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional rice were not significantly different. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures during cultural periods were very similar each other. Analysis of dominant isolates in the rhizosphere cultivated with Bt and Nakdong rice showed that the dominant isolates from the soil of Bt rice and Nakdong belonged to the Proteobacteria, Cloroflexi, Actinobacteria, Firmicutes, and Acidobacteria. These results indicate that the Bt rice has no significant impact on the soil microbial communities during cultivation period. Further study remains to be investigated whether the residue of Bt rice effect on the soil environment.
Genetically modified (GM) rice Agb0101, which expresses the insecticidal toxin modified cry1Ac (mcry1Ac1) gene, was developed by the Rural Development Administration in Korea. To monitor the probable release of Agb0101 in the future, it is necessary to develop a reliable detection method. Here, we developed the PCR detection method for monitoring and tracing of GM rice. The primer pair (RBEgh-1/-2) from a starch branching enzyme (RBE4) gene was designed as an endogenous reference, giving rise to an expected PCR amplicon of 101 bp. For the qualitative PCR detection, construct- and event-specific primers were designed on the basis of integration sequence of T-DNA. Event-specific PCRs amplified specifically 5'- or 3'-junction region spanning the native genome DNA and the integrated gene construct, while none of amplified product was shown on crops, rice varieties, and other insect-resistant transgenic rice lines. The event-specific real-time PCR method was performed using TaqMan probe and plasmid pRBECrR containing both rice endogenous gene RBE4 sequence and 5'-junction sequence as the reference molecule. The absolute limit of quantification (LOQ) of real-time PCR was established with around 10 copies for one plasmid molecule pRBECrR. Thereafter, the different amounts of transgenic rice (1, 3, 5, and 10%, respectively) were quantified by using the established real-time PCR method, with a range below 19.55% of the accuracy expressed as bias, 0.06-0.40 of standard deviation (SD) and 3.80-7.01% of relative standard deviations (RSD), respectively. These results indicate that the qualitative and quantitative PCR methods could be used effectively to detect the event Agb0101 in monitoring and traceability.
The genetically modified leaffolder-resistant (Cry1Ac1) rice plant was evaluated for the changes of resistance by comparing the occurrence of major diseases with a japonica type Korean rice variety, Nakdong which was the mother plant of the transgenic rice event, in greenhouse and field conditions. There was no difference in the occurrence of sheath blight and Helminthosporium blight between the two varieties in the fields. We couldn't find any difference of resistance for fungal blast and bacterial leaf blight by artificial inoculation in greenhouse. There was also no difference in the susceptibility to sheath blight in artificial inoculation tests confirming the results in the fields. The possibility of gene transfer of Bar and Cry1Ac1 from the genetically modified rice plant to naturally infected pathogens such as Fusarium moniliforme and Pyricularia oryzae in the field conditions was tested by PCR. And the possible transfer of those genes by continuous inoculation of Xanthomonas oryzae pv. oryzae and Rhizoctonia solani was also tested. However, we couldn't find any possibility of transfer of the genes in natural and artificial conditions.

Anthranilate synthase (AS) is a key enzyme in the biosynthesis of tryptophan (Trp), which is the precursor of bioactive metabolites like indole-3-acetic acid and other indole alkaloids. Alpha anthranilate synthase 2 (OsASA2) plays a critical role in the feedback inhibition of tryptophan biosynthesis. In this study, two vectors with single (F124V) and double (S126F/L530D) point mutations of the OsASA2 gene for feedback-insensitive ${\alpha}$ subunit of rice anthranilate synthase were constructed and transformed into wildtype Dongjinbyeo by Agrobacterium-mediated transformation. Transgenic single and double mutant lines were selected as a single copy using TaqMan PCR utilized nos gene probe. To select intergenic lines, the flanking sequence of RB or LB was digested with a BfaI enzyme. Four intergenic lines were selected using a flanking sequence tagged (FST) analysis. Expression in rice (Oryza sativa L.) of the transgenes resulted in the accumulation of tryptophan (Trp), indole-3-acetonitrile (IAN), and indole-3-acetic acid (IAA) in leaves and tryptophan content as a free amino acid in seeds also increased up to 30 times relative to the wildtype. Two homozygous event lines, S-TG1 and D-TG1, were selected for characterization of agronomic traits and metabolite profiling of seeds. Differentially expressed genes (DEGs), related to ion transfer and nutrient supply, were upregulated and DEGs related to co-enzymes that work as functional genes were down regulated. These results suggest that two homozygous event lines may prove effective for the breeding of crops with an increased level of free tryptophan content.
