Rapid Detection of Lactobacillus Genus Inhabiting in Vagina of 20's Healthy Women Using Multiplex PCR (Multiplex PCR 분석을 통한 건강한 20대 여성의 질 내에 서식하는 Lactobacillus 속 유산균의 신속한 검출)

  • The purpose of this study is to simultaneously detect and identify specific genus of Lactobacillus distributed in vagina of healthy women in their twenties by using multiplex primer. Vaginal fluid samples were taken from 166 women who were healthy and did not have vaginosis symptoms caused by bacterial infection. Six species of lactic acid bacteria belong to Lactobacillus genus that are known to inhabit in healthy vagina were selected to make a species-specific multiplex primer. Multiplex primer I was specified to selectively detect L. iners, L. crispatus, L. gasseri and multiplex primer II was specified to selectively detect L. acidophilus, L. jensenii, L. fermentum. L. crispatus (77%) was most frequently detected, and L. acidophilus (57%) and L. jensenii (57%) were relatively higher than others. Although the proportion of L. iners (59%) was a little higher than those of L. acidophilus and L. jensenii, this should be further validated in healthy women's vagina to be sure since they often appear in women having bacterial vaginosis and/or during the antibiotic therapy. Conclusively, the multiplex PCR technique using the species-specific primer could a useful tool to predict variation of vaginal health condition and process of recovery from vaginal disease.

Bioconversion of nitrogen oxides and reduction of ferric ions by probiotic lactic acid bacteria (프로바이오틱스 유산균에 의한 질소 산화물 전환 및 철 이온 환원활성)

  • Many lactic acid bacteria (LAB) have probiotic properties that exert various health benefits. In this study, the reduction potential of nitrogen oxide compounds and ferric ions by six LAB, including Lactobacillus kimchicus, L. lactis, L. casei, L. plantarum, L. rhamnosus GG, and Leuconostoc mesenteroides were evaluated. The L. kimchicus strain produced a substantial amount of nitrite reduced from nitrate added to the media, whereas the other five LAB strains did not. L. kimchicus also showed the most potent reducing activity of ferric to ferrous ions. However, the reduction potential of the autoclaved L. kimchicus was little pronounced. The scavenging activities of viable LAB or their cell lysates against different radicals were not consistent with the potency of the LAB's reducing ability. The present results indicate that L. kimchicus has a strong reduction potential for nitrogen oxides in viable status, and that this ability can be used as a probiotic property for various health benefits.

Analysis of Marker Components of Fermented Opuntia ficus-indica var. saboten Stem Extracts (유산균 발효에 의한 손바닥선인장 줄기추출물의 지표물질 함량 변화 분석)

  • The fruit and stem of Opuntia ficus-indica var. aboten (OFS), a native plant of Jeju Island, are considered a safe food source. Moreover, stem extracts have been previously reported to possess a variety of biological effects (e.g. anti-inflammatory and anti-oxidant, including the ability to partially ameliorate cognitive impairment), suggesting that this plant may have utility as a functional food. The present study investigated whether fermentation by lactic acid bacteria enhances the biological effects of OFS extracts. The acetylcholinesterase (AChE) inhibitory activity of fermented or non-fermented OFS extracts was evaluated, and the content of marker components dihydrokaempferol (DHK) and quercetin-3-methyl ether (3-MeQ) was analyzed using high-performance liquid chromatography. Fermented (relative to non-fermented) OFS extracts exhibited improved AChE inhibitory activity ($IC_{50}=28.35 mg/mL$), with AChE inhibitory activity resulting from fermentation by L. plantarum ($IC_{50}=12.56mg/mL$) exceeding that resulting from fermentation by L. fermentum ($IC_{50}=17.71mg/mL$). Furthermore, fermented (relative to non-fermented) OFS extracts exhibited a 16.7 % increase in DHK content, and 3-MeQ content of OFS extracts fermented by L. plantarum and L. fermentum increased by 28.6 % and 21.4 %, respectively. Therefore, OFS stem extract AChE inhibitory activity, as well as DHK and 3-MeQ content, was enhanced by fermentation with Lactobacillus spp. This suggests that fermented OFS extracts may contribute to prevention or improvement of cognitive impairment. These data are anticipated to be useful in the development of enhanced-efficacy OFS products.

Chemical Characteristics in Fermented Cordycepin-enriched Cordyceps militaris (발효 동충하초의 이화학적 특성)

  • The comparative effects of fibrinolytic, antioxidative activity and electrophoretical protein patterns with Cordyceps militaris powder (CM) and cordycepin-enriched Cordyceps militaris JLM0636 powder (CCM) and fermented cordycepin-enriched Cordyceps militaris JLM0636 powder by several microscopic organisms were investigated. In addition, nutritional materials such as mineral, protein, and fatty acids were also measured. The protein concentration was higher in CCM than that in CM. The protein concentration in fermented CCM was the highest in CCM treated by Aspergillus kawachii among the various samples. When total protein patterns of CM, CCM, and both fermented CCMs were analyzed by native- and SDS-PAGE, there were slightly varietal differences in electrophoretical protein patterns. Major minerals were K, Ca, Mg, and Zn. Major fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid, and linolenic acid. Fibrinolytic activity was the highest in the fermented CCM by Bs treatment among the various samples. The ${\alpha},{\alpha}^{\prime}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radical scavenging activity was slightly stronger in the CCM treated with Aspergillus kawachii among the various samples; however, these samples all exhibited relatively low levels of activity compared with the butylated hydroxytoluene (BHT). These results may provide the basic data necessary to understand the biological activities and chemical characteristics of Cordyceps militaris JLM0636 powder fermented by several microscopic organisms to develop functional foods.

Comparison of quality characteristics of Platycodon grandiflorum according to steaming and fermentation (증숙 및 발효에 따른 도라지의 품질특성 비교)

  • This study was designed to improve the sensory characteristics and mitigate the bitter taste of Platycodon gradiflorum. It was aimed at investigating the qualitative properties of fermented P. gradiflorum after repeated steaming and drying treatments. P. gradiflorum was heated for 2 hr at $95^{\circ}C$, being the first and third treatments compared afterwards. Lactobacillus plantarum, at a concentration of 10%, was used as starter culture. As a result, the third steaming process and the addition of starter improved the physical and chemical properties of P. gradiflorum i.e., crude saponin and total polyphenol contents increased significantly. Moreover, P. gradiflorum steamed three times and fermented by L. plantarum showed the higher overall preference score. Our results indicated that the three-time steaming and drying was an effective manufacturing process for the production of high-quality fermented P. gradiflorum. Lactic acid-fermented P. gradiflorum also could have a potential use as a valuable resource for the development of functional products.

젓갈 유래 프로바이오틱스의 항산화 및 콜레스테롤 저하 효과

  • Lactococcus lactis NK24, Lactobacillus plantarum NK181, and strain HW161 were previously isolated from Jeot-gal. For antioxidative activity, the culture supernatants of there probiotic strains were shown to be 78, 77, and 81% by DPPH method. Also, the peroxidation of linoleic acid of them was investigated and they were about 3.5, 4.6, and 2.0%. And cholesterol-lowering activity of Jeot-gal probiotic strains were about 40, 33, and 60%, respectively.

식품 발효중 기능성 물질의 생전환 및 기능성 변화

  • 본 연구에서는 프로바이오틱스로 사용하는 유용 미생물과 식용으로 안전하게 사용 가능한 미생물 자원을 이용하여 배당체의 전환 연구를 실시하여 전환체의 새로운 생리활성 작용에 대하여 연구하였다. 연구에 사용한 주요 소재로는 인삼의 사포닌 배당체, 길경의 platycodin 배당체, 호로파의 사포닌 배당체, 칡과 두유의 이소플라본 배당체 등이었다. 사용 균주로는 bifidobacteria, lactobacilli, leuconostocs, yeasts, aspergilli 등이었다. 유산균 중에는 Bifidobacterium sp. Int-57의 전환능이 우수하였고 식용 곰팡이 중에는A. niger의 전환능이 우수하였다. 다양한 미생물 또는 효소를 이용한 전환체에 대하여 아질산 소거능, 항산화능, a-glucosidase와 a-amylase 저해능 등을 조사하였을 때 일반적으로 전환 후의 활성이 중가하였다. 길경 사포닌은 전환 후에 세포 독성이 줄어들었으며 관능성이 유의적으로 개선되었다. 결론적으로, 프로바이오틱스와 식용 미생물을 이용하여 천연물 중에 존재하는 배당체의 생리활성 및 관능성을 증가시킬 수 있어 새로운 기능성 식품의 개발에 활용할 수 있을 것이다.

Screening for Ginseng-Fermenting Microorganisms Capable of Biotransforming Ginsenosides (Ginsenoside 전환이 가능한 인삼 발효 미생물의 선별)

  • Panax ginseng has been drawing world-wide attention since it was used for medicinal purposes and its effects was discovered in scientific manners. However, it is necessary to develope new ginseng products as functional foods to compete with western ginseng. Fermented ginseng could be an excellent solution, where useful probiotics are provided and ginsenosides are specifically transformed to functional forms. In this study, we investigated the growth and ginsenoside biotransformation by 21 Bacillus strains isolated from Chongkukjang and 12 lactic acid bacteria. 2.5% (w/v) and 1% (w/v) of ginseng were used in culture media containing only ginseng powder as a sole nutrient source, and their biotransformation abilities were tested after the growths were checked. All used Bacillus strains and lactic acid bacteria were able to grow well in ginseng powder media at higher levels than $10^{7}\;CFU/ml$. Most of Bacillus strains displayed ginsenoside transformation in a strain-specific manner. Therefore, the results of this study demonstrated that the strains tested in this study could be used as potential starters for the ginseng fermentation.

Protection of Infection and Eradication Activity of Culture Product by Pediococcus pentosaceus CBT SL4 Showing Antimicrobial Activity against Helicobacter pylori (Helicobacter pylori에 대한 항균활성을 나타내는 Pediococcus pentosaceus CBT SL4 배양물의 감염방어 및 제균활성)

  • New food ingredient was developed to eradicate and protect against re-infection of Helicobacter pylori in fermentation broth of lactic acid bacteria (LAB) showing antimicrobial activity against pathogenic microorganisms such as H. pylori and Listeria monocytogenes. LAB strain CBT SL4 was identified as Pediococcus pentosaceus by 16S rDNA sequencing and its culture broth showed antimicrobial activity of 800 AU/mL against H. pylori in optimized fermentation process. Using thin layer concentration system and spray-typed fluid bed drier system, concentrated powder product showing activity of 12,800 AU/g was harvested. Product showed eradication and protection activities against H. pylori infection on feeding test (50 AU/day) using Mongolian gerbil infection model. After 4 weeks therapy of 8,000 AU/day, ${\Delta}13CO_2$ level (DOB30) decreased about 40% in urea breath test on patient with H. pylori infection. Result show concentrated culture product of P. pentosaceus CBT SL4 has eradicating and protecting activities against H. pylori infection and can be used as food-active ingredient for prevention of gastric and duodenum ulcer caused by H. pylori.

Effect of Effective Microorganism Applications on Growth, Yield and Fruit Nutrient Contents in Hot Pepper (유용미생물 처리가 고추의 생육 및 과실성분에 미치는 영향)

  • The aim of this study was to determine the effect of effective microorganisms (EM) on growth, yield and fruit nutrient contents of two cultivars ('Muhanjilju' and 'Daetong') of hot pepper. The number of injection of EM cultivated are 6 times from the pepper plant seedlings until harvested in both cultivars. Stem girth was highest in the treatment of Lactobacillus plantarum whereas lowest in no EM control. In 'Muhanjilju', the number of branches was highest (79.3) in the treatment of Bacillus subtilis whereas in 'Daetong' was highest (119.0) as treated with Lactobacillus plantarum. Yield of the red hot pepper fruits that sum of two varieties was highest in the treatment of Rhodopseudomonas capsulatas (53.1kg FW per 10a). Regardless of EM treatments, average yield was 8% higher in 'Daetong' than is 'Muhanjilju' (33.8kg vs 31.2kg per 10a). The incidence of antracnose was lowest by the Rhodopseudomonas capsulatas treatment, which led to the highest yield. As for the effect on fruit mineral nutrients, total N and phosphorus concentrations were highest in the treatment of Lactobacillus plantarum and Bacillus subtilis in both varieties, respectively. The highest content of total carotenoids was obtained from the treatment of Lactobacillus plantarum in both varieties.
