Antiviral Properties of Probiotic Mixtures against Rotavirus in the Rat (랫드에서 로타바이러스에 대한 유산균혼합물의 항 바이러스활성)

  • Rotavirus is a major cause of acute gastroenteritis in young children in developed and developing countries. The use of probiotics for the treatment of gastrointestinal diseases is both safe and easily accessible. In this study, we evaluated the anti-rotaviral activities of probiotic mixtures in a Sprague-Dawley rat. 24 litters with their dams were randomly assigned to four groups; placebo, phosphate buffered saline (PBS), and two probiotic mixture (PRO-1 and PRO-2) groups. All rats were inoculated with rotavirus at dose of 8 log plaque forming units per rat at 5 days old. Animals in the PRO-1 and PRO-2 groups were orally administered probiotic mixtures 1 or 2, respectively, at a dose of 8 log colony forming units daily during 4 days. For control purposes, placebo and PBS groups were orally administered the same amount of placebo (containing maltose and polydextrose) or PBS once daily for 4 days, respectively. Antiviral analysis was performed by real-time quantitative PCR (RT-qPCR) and observing intestinal villi. As a result, weights of small intestines were greater in the PRO-1, PRO-2 groups than in control groups. Villi were short and villous epithelial necrosis was exhibited in control groups, but these morphological changes were not observed in PRO-1, PRO-2 treated rats. RT-qPCR analysis showed that VP7 gene level of rotavirus in fecal samples and small intestinal epithelial cells were lower in the PRO-1 and PRO-2 groups. These findings suggest that probiotic mixtures may be useful probiotics for the treatment of or as alternative therapies for rotaviral gastroenteritis.

  • 발효는 어떤 식품에 유용한 미생물이 자라 큰 분자의 영양소를 분해하여 소화되기 쉬운 식품으로 변하게 한다. 그리고 음식속의 영양소를 이용하여 산성분, 맛성분, 생리활성 물질을 만들어내어 음식물의 보존기간을 연장할 뿐 아니라 균자체(유산균, 프로바이오틱 이라함)나 그 생성물들은 사람의 건강을 증진시키는 효과를 갖는다. 우리나라의 발효식품에는 김치, 장류(된장, 간장, 청국장, 고추장 등), 젓갈류, 막걸리가 대표적이다. 서양에서는 우유 발효식품으로 요구르트, 치즈, 채소발효식품으로는 사우어 크라우트, 피클, 올리브가 있고, 발효빵, 발효소세지, 술로는 포도주, 맥주 등이 있다. 발효 콩제품으로는 템페(인도네시아), 미소(일본된장), 쇼유, 낫또, 수후, 도우치 등 다양하다. 이에 우리나라에서 가장 많이 섭취하고 있는 김치, 된장 및 청국장의 암 예방효과에 대하여 알아보고자 한다.

Application of a Undaria pinnatifida for industrial cultivation of Lactobacillus (유산균의 산업적 배양을 위한 미역의 유용성 평가)

  • To optimize the medium composition of Undaria pinnatifida as a pH modulator the growth of Lactobacillus plantarum industrial scale, we analyzed the pH and growth characteristics of L. plantarum in basal medium. Subsequently, the medium compositions addition of carbon, nitrogen sources and buffering agents were optimized. When 0.5% yeast extract and 2% glucose, L. plantarum grew to maximum cell density in experimental condition. However, the growth of L. plantarum rapidly pH 4.0 in basal medium. A high alkali-ash value and low cost-effective utilization n the waste part as examined. ddition of U. pinnatifida extract alleviated the serious decrease. Among them, juice of U. pinnatifida was most helpful for the growth of L. plantarum ($36.3{\pm}1.810^8CFU/mL$). These results show that U. pinnatifida be large-scale cultivation of L. plantarum. This optimized U. pinnatifida medium can be used for safe and economical production of Lactobacillus.

Antioxidant and ACE Inhibiting Activities of Sugared-Buchu (Allium ampeloprasum L. var. porum J. Gay) Fermented with Lactic Acid Bacteria (부추 당침액의 유산균 발효에 따른 항산화 및 ACE저해활성)

  • In recent years, the growing interest in the health care benefits of sugared-plant fermented enzymes has led to increased consumption. This study investigated the fermentation of sugared-buchu (Leek:sugar, 1:3) by lactic acid bacteria (Lactobacillus acidophilus AML 0422, Lactobacillus brevis HLJ 59, Lactobacillus helveticus AML0410, Lactobacillus plantarium KCTC 13093) and the antibacterial activity, antioxidant activity, and functionality (e.g., anti-hypertensive activity) of the fermented product. The fermented sugared-buchu showed high antibacterial activity against Staphylococcus aureus KCTC 1916, at 31.43 mm, and its total polyphenols, total flavonoid content, and DPPH scavenging activity were 160.8-178 mg/ml, 100-108 mg/ml, and 51.4-58.1%, respectively. DPPH scavenging activity was to that of vitamin C (50 ppm). ACE inhibitory activity was 50.4-67%, depending on the strain of lactic acid bacteria, and the control of sugared-buchu activity was higher than 32.6%. These results suggest that sugared-buchu fermented with lactic acid bacteria has strong antibacterial, antioxidant, and ACE inhibitory activities.

  • Coffee is a commonly consumed beverage that contains anti-inflammatory compounds such as caffeine, chlorogenic acid, cafestol, trigonelline, and kahweol. Lactobacillus plantarum is a lactic acid bacterium most frequently used in the fermentation of food products of plant origin. L. plantarum is able to degrade some food phenolic compounds and provide high value-added compounds such as powerful antioxidants or food additives approved as flavouring agents. In this study, we investigated the anti-inflammatory effects of coffee extract fermented by L. plantarum on RAW264.7 macrophages. In lipopolysaccharide-stimulated RAW264.7 cells, these coffee extracts exhibited anti-inflammatory activities through the reduction of nitric oxide (NO) production and inducible NO synthase expression. Fermented coffee extracts significantly decreased the expression of inflammatory cytokines such as tumor necrosis factor ${\alpha}$, interleukin $1{\beta}$, interleukin 6, and interferon ${\gamma}$. Cyclooxygenase-2, which is one of the key biomarkers for inflammation, was significantly suppressed. These results might be helpful for understanding the anti-inflammatory mechanism of fermented coffee extract on immune cells and, moreover, suggest that fermented coffee extract may be a beneficial anti-inflammatory agent.

From food wastes into useful probiotics: Development of Lactic acid bacteria as useful probiotics for animal feed without antibiotics using food waste (음식물류폐기물의 자원화를 위한 항세균 활성 유산균의 개발)

  • For the conversion of food waste into a good recycling material such as animal feed without antibiotics, thirteen lactic acid bacteria, which can be used as good probiotics for animal feed, were isolated from the intestine and feces in pigs. All isolates showed strong tolerance to high salt (4% of NaCl), acid(pH4.0), and bile juice (0.8% of oxgall). The growth rate was best at $37^{\circ}C$ in all strains. Among the isolates, Lactobacillus plantarum CJY-22, L. brevis CJY-42, L. arizonensis CJY-3, and Pediococcus sp. CJY-41 showed higher and broader spectrum of antimicrobial activities against six different pathogens such as Salmonella, typhimurium. L. plantarum CJY-22 has also grown well at $25^{\circ}C$, making this strain as an appropriate candidate for the fermentation of food waste at room temperature, thus conducting the fermentation process cost-effectively.

Application of Lactic Acid Bacteria to Inhibit Fungal Contamination of Cured Cheeses (항곰팡이능 보유 유산균의 숙성치즈 적용 연구)

  • Lactic acid bacteria with antibacterial activity can be effectively used as probiotics to inhibit the growth of harmful bacteria that cause food spoilage or food poisoning. In this study, Pediococcus pentosaceus M132-2, isolated from soybean paste, was analyzed for its effects on three major contaminating fungi. M132-2 was confirmed to exert antifungal activity by inhibiting the growth of all three fungi tested. In addition, M132-2 displayed excellent salt resistance and low temperature tolerance. Thus M132-2 can survive at the salinity level in cheese and at the low temperatures used in the aging process. Finally, when supernatant from an M132-2 culture was applied to Gouda cheese, the growth of contaminating fungi was significantly inhibited. Consequently, M132-2 may be useful for the prevention of spoilage of various foods, including cheese.

  • 아로니아(Aronia czarna)는 anthocyanin, polyphenol, flavonoid, cathechine, chlorogenic acid와 같은 생리활성물질이 풍부하게 존재하며 항산화, 항암, 항균, 피부건강개선, 노화방지 등 다양한 생리활성에 대한 효능이 있는 것으로 알려져 있어 건강 및 기능성식품, 화장품 등의 원료 소재로 각광받고 있다. 생물전환(Bioconversion)은 미생물 또는 효소의 생물학적 촉매 반응을 활용하여 기존 소재의 성분을 변환시키는 기술이다, 최근 생물전환을 활용한 천연소재의 생리활성 물질 기능성, 생체이용률, 안전성을 증대시키기 위한 방안으로 많은 연구가 진행되고 있으며 식품, 의약품, 화장품 등 다양한 분야에서 활성화 되고 있다. 본 연구는 젓갈로부터 분리한 균주를 유전학적 특성을 확인하기 위하여 16S rDNA 염기서열을 분석한 뒤 그중 유산균을 발효공정에 활용하였다. 전북 순창에서 수확된 아로니아 분말과 발효공정을 수행하였으며 아로니아 최적 추출조건 선정, 발효공정 전 후 추출물의 기능성 평가를 진행하기 위하여 DPPH radical scavenging activity, Total polyphenol 함량을 확인하여 항산화 효능 및 유효성분 함량을 평가하였다. 또한 대식세포인 Raw 264.7을 사용하여 MTT assay, Nitric oxide (NO) 생성 억제 효능을 확인하여 세포독성 및 항염증 활성을 평가하였다. 실험결과, 젓갈류 발효물로부터 16종의 다양한 균주를 확보하였으며, 그중 L. rhamnosus, L. plantarum, P. pentosaceus 균을 발효 공정에 활용하여 유용 균주를 선정 결과 P. pentosaceus 종 유산균 처리군에서 무처리군 대비 DPPH radical 소거능 및 polyphenol 함량이 증가됨을 확인하였다. 발효공정 후 항산화 활성은 무처리군 대비 약 119%, polyphenol의 함량은 무처리군 대비 약 119%로 증가됨을 확인되었다. 또한 Raw 264.7 세포실험 결과 발효공정 후 독성활성이 감소되는 경향을 확인되었으며, 항염증 활성이 월등히 증가됨을 확인하였다.

Dietary effect of Lactobacillus plantarum CJLP55 isolated from kimchi on skin pH and its related biomarker levels in adult subjects (김치유산균 Lactobacillus plantarum CJLP55 섭취가 성인 남녀의 피부 산도 및 관련 구성 인자의 변화에 미치는 영향)

  • Purpose: The skin pH is maintained by epidermal lactate, free fatty acids (FFAs), and free amino acids (FAAs). As a significant determinant of skin health, the skin pH is increased (less acidic) under abnormal and aged skin conditions. In a search for dietary alternatives that would promote an acidic skin pH, this study investigated the dietary effects of Lactobacillus plantarum CJLP55 isolated from Korean kimchi on the skin pH, and epidermal levels of lactate, FFAs, and FAAs in adult subjects. Methods: Seventy eight subjects (mean age $24.9{\pm}0.5years$, range 19 ~ 37 years) were assigned randomly to ingest CJLP55, Lactobacillus strain from kimchi, (n = 39, CJLP group) or placebo supplements (n = 39, placebo group) for 12 weeks in a double-blind, placebo-controlled trial. Skin pH and epidermal levels of lactate, FFAs and FFAs were assessed at 0, 6 and 12 weeks. Results: Although significant decreases in skin pH were observed in both the CJLP and placebo groups at 6 weeks, the skin pH was decreased significantly only in the CJLP group at 12 weeks. In parallel, the epidermal level of lactate in the CJLP group was also increased by 25.6% at 12 weeks. On the other hand, the epidermal level of FAAs were not altered in the CJLP and placebo groups, but the epidermal level of total FFAs, including palmitic acid and stearic acid, was lower in the CJLP group than in the placebo group over 12 weeks. The changes in the other FFAs, such as palmitoleic acid and oleic acid, were similar in the CJLP and placebo groups over 12 weeks. Conclusion: Overall, a dietary supplement of CJLP55 promotes acidic skin pH with a selective increase in epidermal lactate in adult subjects.

Preparation and Quality Characteristics of the Fermentation product of Ginseng by Lactic Acid Bacteria (FGL) (유산균을 이용한 발효인삼 제조 및 품질 특성)

  • Ginseng as a raw material for production of probiotic ginseng product by lactic acid bacteria (LAB) was evaluated in this study. Either white ginseng (WG) or red ginseng (RG) (1% or 5%, w/v) were directly inoculated with a 24 hold seed culture of twenty seven substrains of four different LAB ($1.0{\times}10^6CFU/ml$); Lactobacillus spp., Streptococcus/Enterococcus spp., Leuconostoc/Lactococcus spp. and Bifidobacterium spp., and incubated at $37^{\circ}C$ for 24 or 48 h. Among 27 kinds of LAB, seven substrains of Lactobacillus (MG208, MG311, MG315, MG501, MG501C, MG505, MG590) and one Bifidobacterium (MG723) were selected based on their dose dependent stimulation of the growth of LAB in the presence of ginseng and changes in pH, acidity and viable cell counts during fermentation were examined. Lactobacillus MG208 specifically was found to show the best growth on 5% RG and reached nearly $14.0{\times}10^8CFU/ml$ after 48 h of fermentation and produced the titratable acidity as $0.84{\pm}0.02%$, whereas the pH was significantly lowered from $6.80{\pm}0.01\;to\;3.42{\pm}0.02$. These results indicated that ginseng can be an appropriate material to prepare the fermentation product by several strains of LAB. Therefore we should further check whether probiotic ginseng product may have synergistic health benefits of both probiotics and ginseng to serve for vegetarians and lactose-allergic consumers.
