Analysis of Physiological Activity and Cytotoxicity of Residue Extracts after Pepper Harvest (고추 수확 후 잔재물 추출물의 생리활성과 세포독성 분석)

  • The purpose of this study was to investigate the value of residue discarded after harvesting a species of pepper (Capsicum annuum L.). This study analyzed the physiological activity and cytotoxicity of a fermented extract and a hot water extract gathered from the residues of the Cheongyang pepper and the Nokkwang pepper. The total nitrogen, phosphoric acid, potassium, calcium, and magnesium contents of the fermented extract of the Nokkwang pepper were approximately twice as high as those of the Cheongyang pepper, while the hot water extracts had similar levels of these components across both peppers. Among the microelements, only boric acid, iron, and silicon components were detected, whereas zinc, manganese, molybdenum, and copper components were not detected in either extract. The total polyphenol and flavonoid contents of the fermented extracts were more than two times higher than those of the hot water extracts for both peppers. The DPPH radical scavenging ability (indicating antioxidant activity) of the fermented extract was higher than that of the hot water extract, while the ABTS radical scavenging ability (indicating antioxidant activity) of the hot water extract was higher than that of the fermented extract. A cytotoxicity test was conducted on the extracts using an MTT assay. Both extracts exhibited weak cytotoxicity at all concentrations for both peppers. These results suggest that the extract of residue discarded after harvesting these peppers could be suitable for functional feed or agricultural materials.

Quality characteristics and antioxidative activity of fermented milk added with hot water extract of Cordyceps militaris grown upon Tenebrio molitor (갈색거저리 유래 밀리타리스 동충하초 열수 추출물을 첨가한 발효유의 품질 특성과 항산화 활성)

  • This study was carried out to investigate the physicochemical and antioxidant properties of fermented milk with addition of hot water extract of Cordyceps militaris grown upon Tenebrio molitor. The general components of Cordyceps militaris grown upon Tenebrio molitor are moisture 10.85%, crude protein 18.44%, crude fat 2.07%, crude ash 5.46%. The DPPH radical scavenging activities of different solvents were the highest (74.81 EDA%) with hot water extract sample. The acidity of fermented milk was high with increasing amount of extracts. The pH of fermented milk reached 4.60-4.66 after 4 h of fermentation, and the number of lactic acid bacteria was highest (11.70 log CFU/mL) with 1% fermented milk. The moisture content of fermented milk showed no significant difference. In addition, contents of crude protein were not significantly different according to addition amount. Regarding Hunter's color values, L value decreased as the amount of extract increased, whereas a value and b value increased. The content of free amino acid increased with increasing amount of extract. The DPPH free radical scavenging ability and ABTS free radical scavenging ability of fermented milk were significantly different as the addition amount of extract increased. In the sensory evaluation, fermented milk containing 1% of extract showed the highest preference.

Anti-Diabetic, Alcohol-Metabolizing, and Hepatoprotective Activities of Moringa (Moringa oleifera Lam.) Leaf Extracts (모링가 잎 추출물의 항당뇨, 알코올 대사 및 간 보호 활성)

  • This study was carried out to investigate anti-inflammatory, anti-diabetic, alcohol metabolizing, and hepatoprotective effects of hot water (MOW) and 80% ethanol (MOE) extracts from moringa (Moringa oleifera Lam.) leaf. The total phenol content of MOW and MOE were 45.49 and 63.06 mg tannic acid equivalents/g, respectively. 1,1-Diphenyl-2-picrylhydrazyl radical scavenging activities of MOW and MOE were remarkably elevated in a dose-dependent manner, and about 60.8% and 71.3% at 1 mg/mL, respectively (P<0.01). Superoxide dismutase-like activities of MOW and MOE were 2.8% and 7.4% at 5 mg/mL, respectively (P<0.05). ${\alpha}-Glucosidase$ inhibitory activity also increased in a dose-dependent manner in both extracts, and MOE was higher about two times than MOW at 5 mg/mL (P<0.001). The effects of MOW and MOE on alcohol metabolizing activity were determined by measuring generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). ADH and ALDH activities significantly increased upon addition of MOW and MOE (P<0.05). Anti-inflammatory activity was examined in lipopolysaccharide-stimulated RAW 264.7 cells. Nitric oxide production was reduced to 32.1% and 81.2% by addition of MOW and MOE at 1 mg/mL, respectively (P<0.05). MOW and MOE showed significant protective effects against tacrine-induced cytotoxicity in Hep3B cells at $100{\mu}g/mL$. These results suggest that moringa leaf extracts have great potential as natural health products.

Antioxidative Activity of Hot Water and Ethanol Extracts of Lespedeza cuneata Seeds (야관문(Lespedeza cuneata) 종자의 열수 및 Ethanol 추출물의 항산화 효과)

  • Hot water and 95%(v/v) ethanol extracts were prepared from dried Lespedeza cuneata seeds and antioxidant compounds were isolated by solvent fractionation, silica gel adslorption chromatography, Sephadex LH-20 column chromatography, and preparative HPLC. Antioxidant activity was measured using DPPH radical scavenging activity. The 80%(v/v)ethanol and ethylacetate fraction of Lespedeza cuneata seed extracts had stronger antioxidant effects than did the n-hexane fraction. The active antioxidant compounds obtained from hot water and 95%(v/v) ethanol extracts may be identical, based on analysis by Sephadex LH-20 column chromatography and preparative HPLC.

Inhibitory Effect of Semen Sinapis Albae on Immediate Hypersensitivity Reaction (백개자의 즉시형 과민 반응에 대한 억제 효과)

  • 목적 : 본 연구에서는 백개자 열수추출물이 활성화된 대식세포 및 사람 비만세포주, HMC-1에서 염증 반응을 효과적으로 억제하는가를 관찰하고자 하였다. 방법 : 대식세포에 여러 농도의 백개자 열수추출물을 가한 뒤 LPS로 염증을 유도하여 NO 생산, iNOS와 COX-2 단백질 발현을 관찰하였으며 HMC-1에도 여러 농도의 백개자 열수추출물을 가한 후 PMACI로 염증을 유도하여 histamine 분비와 NF-${\kappa}B$ 활성 및 $I{\kappa}B$-${\alpha}$의 인산화, MAPKs pathway에 대한 저해효과를 관찰하였다. 결과 : 백개자 열수추출물은 대식세포에서 LPS로 유도된 NO 생성 및 INOS, COX-2 단백질 발현을 농도 의존적으로 저해하였으며 HMC-1에서 PMACI로 유도된 histamine의 분비와 p38 MAPK, ERK, JNK의 인산화 반응 및 $I{\kappa}B$-${\alpha}$의 인산화와 NF-${\kappa}B$의 활성을 저해하였다. 결론 : 백개자 열수추출물은 대식세포 및 비만세포의 활성을 저해함으로써 알레르기 질환의 치료에 사용될 잠재성이 크다고 사료된다.

Antimicrobial Effect of Forsythiae Fructus Extracts on Several Food-Borne Pathogens (연교 추출물의 항균활성 검색 및 일부 식중독균의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effect of the Forsythiae Fructus extracts against food-borne pathogens. First, Forsythiae Fructus was extracted with methanol at room temperature and the methanol extracts were fractionated by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Forsythiae Fructus extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Forsythiae Fructus showed the highest antimicrobial activity against Salmonella paratryphimurium and Salmonella typhimurium. A synergistic effect in inhibition was observed when Forsythiae Fructus extract was mixed with Ulmus davidiana Japonica extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Forsythiae Fructus against Shigella flexneri and Salmonella paratyphimurium. The aqueous extract of Forsythiae Fructus had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 10,000 ppm. At this concentration, the growth of Shigella fexneri was retarded for more than 24 hours and for up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Forsythiae Fructus efficiently inhibited Staphylococcus epidermidis and Shigella flexneri.

Comparative Study of the Antioxidant Activity Effects of Gami-Sumiwon Extract using Water and 80% Ethanol (가미수미원의 열수 추출물과 80% 에탄올 추출물의 항산화 효능 비교)

  • In this study, Gami-Sumiwon (GS; Cynanchi wilfordii R., Angelica gigantis R., Lycium chinense M., Betula platyphylla S., Cryptotympana atrata F. and Carthamus tinctorius L.) and/or Sinomenium acutum R. (SC) was extracted with 70% ethanol or water. And We investigated the antioxidant activity effect of GS±SC. The following experimental techniques were used to evaluate the antioxidant efficacy of GS±SC. HPLC chromatogram, heavy metal content, ABTS/DPPH radical scavenging analysis, SOD-like activity assay, FACS, and NO assay. As a result of the experiment, the sinomenine content was found to be higher in DW extracts, and decursin was found to be higher in 80% ethanol extracts. And, the amount of heavy metals in all extracts was below the standard value. ABTS, and DPPH radical scavenging activity was identified that GS±SC(EtOH) was found to have a higher scavenging activity than GS±SC(DW). But, SOD showed the opposite result. No cytotoxicity of GS was observed on Raw 264.7 cells at concentration of 1~100 ㎍/㎖. The ROS production was significantly decreased that GS±SC(DW) was found to have a higher scavenging activity than GS±SC(EtOH). However, NO production showed the opposite result. Looking at the results of SOD and ROS analysis, SC does not seem to have a function of prevention. SC is thought to have an effect on the removal of free radicals generated after oxidative stress. This result objectively confirmed the antioxidant effect of GS±SCs. We will continue to conduct in-depth research. Therefore, it is believed that the possibility of using GS±SCs as a functional material can be established. The more diverse the objectives, the higher the value of GS utilization is thought to be.

Antioxidant Activities and Biological Properties of Phellinus linteus Extracts according to Different Extraction Methods (추출방법에 따른 상황버섯 추출물의 항산화활성 및 생물학적 특성)

  • Antioxidant activities and biological properties such as antimutagenic and cytotoxic effect of Phellinus linteus extracts from different extraction conditions were measured against Salmonella typhimurium and human cancer cell lines. DPPH free radical scavenging activities of the extracts were higher in the solutions extracted with ethanol (17.14) and ethanol after water (17.79), respectively. In the Ames test, ethanol extract of P. linteus alone did not exhibit any mutagenicity but showed substantial inhibitory effect against mutations induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), 4-nitroquinoline-1-oxide (4NQO), and benzo $({\alpha})$ pyrene $(B({\alpha})P)$. The extracts of ethanol and ethanol after water of P. linteus $(200\;{\mu}g/plate)$ had the highest inhibitory effect of 61.5 and 60.9%, respectively, on the mutagenesis on S. typhimurium TA98 strain induced by $B({\alpha})P$. Extracted solutions of ethanol and ethanol after water of P. linteus showed high antimutagenic effect against MNNG, 4NQO, Trp-P-1 and $B({\alpha})P$, causing mutations in S. typhimurium TA100 strain. The anticancer effects of P. linteus extracts were investigated against human fibrosarcoma HT-29 and human hepatocellular carcinoma HepG2. The treatment of 0.5 mg/ml of ethanol, ethanol after water and water extracts of P. linteus had the highest cytotoxicity of 59, 57, 54%, respectively against HT-27 cell line, whereas low cytotoxicity effects were observed against HepG2 cell line in the range of $10{\sim}30%$. The ethanol and water extracts of P. linteus also showed the nitrate scavenging ability at different pHs. The ethanol extract showed higher nitrate-scavenging ability compared to water extract of P. linteus.

ROS Scavenging Effect and Cell Viability of Opuntia humifusa Extract on Osteoblastic MC3T3-E1 Cells (천년초 추출물이 조골세포의 증식과 ROS소거능에 미치는 영향)

  • In this study, the effect of the Opuntiahumifusa extracts on proliferation, alkaline phosphatase (ALP) activity, collagen synthesis and ROS level of a cell was investigated using an osteoblast. Opuntiahumifusawas separated intoOpuntiahumifusapeel (OH-P), seed (OH-Se) and stem (OH-St).These were subjected to extraction by using hot water and ethanol. The proliferation of the MC3T3-E1 osteoblastic cells that were treated with OH-Se water extract were increased by approximately 120%. Regarding the effects of OH-Se on ALP activity, the $50{\mu}g/ml$ ethanol extract group showed the highest activity. The synthesis of collagen increased significantly in response to treatment with OH-Se water extract. The ROS scavenging effects of Opuntiahumifusawere investigated for involvement of oxidativedamage, cell culture and staining. Also, when OH-Se water extract $100{\mu}g/ml$ was added, the ROS level decreased by 54%. These results indicate that Opuntiahumifusa extracts have an anabolic effect on bone through the promotion of osteoblastic differentiation, suggesting that it could be used for the treatment of common metabolic bone diseases.

The research on antioxidative effect of Sasa quelpaertensis extractum and assessment of cytotoxicity (제주조릿대 잎 추출물의 항산화효능 및 세포독성 평가에 관한 연구)

  • This research analyzed cell survival rate according to antioxidative effect and cytotoxicity using Sasa quelpaertensis and Ethanol extracts. Sasa quelpaertensis used extracts of $95^{\circ}C$ boiled-water extract and 70% ethanol liquid each. For total polyphenol content, tannin content, and flavonoid content, polyphenol content of $95^{\circ}C$ boiled-water extract was 26.6 mg/g while that of 70% ethanol extract liquid was 22.3 mg/g, which means polyphenol content was higher in $95^{\circ}C$ boiled-water extract. Tannin content was 72.1 mg/g in $95^{\circ}C$ boiled-water extract which was higher than 61.2 mg/g in 70% ethanol extract liquid. Total flavonoid content was higher in 70% ethanol extract liquid (25.4 mg/g) than in $95^{\circ}C$ boiled-water extract (17.8 mg/g). Further, the researcher measured DPPH radical elimination and ABTS radical elimination. As a result, as the concentration of each extract increased, DPPH radical elimination and ABTS radical elimination increased. In the DPPH radical elimination, L-ascorbic acid was eliminated at 500 ppm and showed no change even though the concentration increased, whereas it increased in BHT and Sasa quelpaertensis leaf extracts according to concentration. ABTS radical elimination indicated the similar phenomenon, whereas BHT showed maximum elimination at 500 ppm and decreased gradually as the concentration increased. On the other hand, it gradually increased in Sasa quelpaertensis leaf extract and showed 90% elimination at 10,000 pm, and it increased in $95^{\circ}C$ boiled-water extract. Using human skin fibroblasts (3T3) to check the cell survival rate of Sasa quelpaertensis leaves showed that cell survival rate decreased between 85.6 and 66.6% in $95^{\circ}C$ boiled-water extract. The cell survival rate was between 95.4 and 92.1% when using 70% ethanol extracts. Sasa quelpaertensis has efficacy as a natural antioxidant as well as a material hyangjang.
