Effect of Substrate to Inoculum Ratio on Biochemical Methane Potential in the Thermal Pretreatment of Piggery Sludge (양돈분뇨의 열전처리에서 기질과 접종액의 비율이 메탄생산 퍼텐셜에 미치는 영향)

  • This study was carried out to investigate the effect of substrate to inoculum ratio on ultimate methane potential ($B_u$) from piggery wastes. BMP(Biochemical methane potential) assays were executed for the three samples that have different organic characteristics (Filtrate of pig slurry, LF; Thermal hydrolysate of piggery sludge cake, TH; Mixture of LF and TH at the ratio of 4 to 1, Mix), and $B_u$ values obtained from BMP assays were compared with the theoretical methane potential ($B_{th}$) of each samples. While $B_u$ values (0.27, 0.44, and $0.46Nm^3\;Kg^{-1}-VS_{added}$) of TH sample that was pretreated with thermal hydrolysis were below the $B_{th}$ at all S/I ratios (0.1, 0.3, and 0.5), and $B_u$ values of LF (0.64 and $0.53Nm^3\;Kg^{-1}-VS_{added}$ for the S/I ratios of 0.1 and 0.3, respectively) at the lower S/I ratios of 0.1 and 0.3 exceeded the $B_{th}$ values ($0.418Nm^3\;Kg^{-1}-VS_{added}$). And also biodegradability ($B_u/B_{th}$) of LF sample were obtained as 152.07%, 122.67%, and 95.71% at the S/I ratios of 0.1, 0.3, and 0.5, respectively, and unreasonable $B_u/B_{th}$ values were presented at lower S/I ratios of 0.1 and 0.3. $B_u$ and $B_u/B_{th}$ of Mix sample showed a similar tendency with those of LF sample. Therefore, TH sample by thermal hydrolysis pretreatment showed lower anaerobic biodegradability than those of other samples (LF and Mix) and ultimate methane potentials of LF and Mix samples were overestimated in the lower S/I ratio of 0.1 and 0.3.

  • Woodfiber insulation board can be considered as a one of the key material for low energy consumption, comfortable and safety construction of residential space because of its eco-friendly and high thermal insulation performance. This study was carried out to investigate the formaldehyde (HCHO) total volatile organic compounds (TVOC) emission properties and combustion shapes by flame test of low density fiberboards (LDFs) prepared with different adhesives. HCHO TVOC emission and combustion properties of LDFs prepared by melamine urea formaldehyde (MUF), phenol formaldehyde (PF), emulsified methylene diphenyl diisocyanate (eMDI) and latex resin adhesives were measured by desiccator method, 20 L chamber method, and flame test, respectively. As results, LDFs manufactured by MUF, eMDI and latex resin adhesives satisfied the Super $E_0$ grade of HCHO emission performance except PF resin. Furthermore, TVOC emission of all LDFs were satisfied the Korean indoor air quality standard (below $400{\mu}g/m^2{\cdot}h$). Especially, LDF with eMDI resin adhesive showed the lowest HCHO and TVOC emissivity, that $0.14mg/{\ell}$, $12{\mu}g/m^2{\cdot}h$, respectively. However, eMDI emitted the small amount ($3{\mu}g/m^2{\cdot}h$) of toluene in VOC components. In the flame test, LDF with MUF resin adhesives showed the most favorable shape after flame test compare to LDFs prepared other adhesives. Based on HCHO and TVOC emission, and combustion shapes, MUF resin adhesive may be recommended to prepare LDF for insulation purpose.

Expression of Nesfatin-1/NUCB2 and Its Binding Site in Mouse Ovary (생쥐 난소 내 Nesfatin-1/NUCB2 발현과 결합 부위 확인)

  • It was recently reported that nesfatin-1/NUCB2, which is secreted from the brain, controls appetite and energy metabolism. The purpose of this research was to confirm whether or not the protein and its binding site should have been expressed in the mouse reproductive organs and to know the possible effects of nesfatin-1 on the reproductive function. Using the ICR female mouse ovary and uterus, the expression of NUCB2 mRNA was confirmed with the conventional PCR and the relative amount of NUCB2 mRNA in the tissues was analyzed with real-time PCR. Immunohistochemical staining was performed using the nesfatin-1 antibody to investigate the nesfatin-1 protein expression and the biotin conjugated nesfatin-1 to confirm the binding site for nesfatin-1 in the ovary. Furthermore, in order to examine if the expression of NUCB2 mRNA in the ovary and uterus is affected by gonadotropin, its mRNA expression was analyzed after PMSG administration into mice. As a result, the expression level of NUCB2 mRNA in the ovary and the uterus was as much as the expression level in hypothalamus. As a result of the immunohistochemical staining, nesfatin-1 proteins were localized at the theca cells, the interstitial cells, and some of the luteal cells. However, the granulosa cells in the follicles did not stain. Interestingly, the oocytes in the some follicles were stained with nesfatin-1. On the other hand, nesfatin-1 protein binding sites were displayed at the theca cells and the interstitial cells near the tunica albuginea. After PMSG administration the expression level of NUCB2 mRNA was increased in the ovary and the uterus. These results demonstrate that for the first time the nesfatin-1 and its binding site were expressed in the ovary and NUCB2 mRNA expression was controlled by gonadotropin, suggesting an important role in the reproductive organs as a local regulator. Therefore, further study is needed to elucidate the functions of nesfatin-1 on the reproductive organs.

Alternative Concept to Enhance the Disposal Efficiency for CANDU Spent Fuel Disposal System (CANDU 사용후핵연료 처분시스템 효율향상 개념 도출)

  • There are two types of nuclear reactors in Korea and they are PWR type and CANDU type. The safe management of the spent fuels from these reactors is very important factor to maintain the sustainable energy supply with nuclear power plant. In Korea, a reference disposal system for the spent fuels has been developed through a study on the direct disposal of the PWR and CANDU spent fuel. Recently, the research on the demonstration and the efficiency analyses of the disposal system has been performed to make the disposal system safer and more economic. PWR spent fuels which include a lot of reusable material can be considered being recycled and a study on the disposal of HLW from this recycling process is being performed. CANDU spent fuels are considered being disposed of directly in deep geological formation, since they have little reusable material. In this study, based on the Korean Reference spent fuel disposal System (KRS) which was to dispose of both PWR type and CANDU type, the more effective CANDU spent fuel disposal systems were developed. To do this, the disposal canister for CANDU spent fuels was modified to hold the storage basket for 60 bundles which is used in nuclear power plant. With these modified disposal canister concepts, the disposal concepts to meet the thermal requirement that the temperature of the buffer materials should not be over $100^{\circ}C$ were developed. These disposal concepts were reviewed and analyzed in terms of disposal effective factors which were thermal effectiveness, U-density, disposal area, excavation volume, material volume etc. and the most effective concept was proposed. The results of this study will be used in the development of various wastes disposal system together with the HLW wastes from the PWR spent fuel recycling process.

High-Resolution Numerical Simulations with WRF/Noah-MP in Cheongmicheon Farmland in Korea During the 2014 Special Observation Period (2014년 특별관측 기간 동안 청미천 농경지에서의 WRF/Noah-MP 고해상도 수치모의)

  • In this paper, the high-resolution Weather Research and Forecasting/Noah-MultiParameterization (WRF/Noah-MP) modeling system is configured for the Cheongmicheon Farmland site in Korea (CFK), and its performance in land and atmospheric simulation is evaluated using the observed data at CFK during the 2014 special observation period (21 August-10 September). In order to explore the usefulness of turning on Noah-MP dynamic vegetation in midterm simulations of surface and atmospheric variables, two numerical experiments are conducted without dynamic vegetation and with dynamic vegetation (referred to as CTL and DVG experiments, respectively). The main results are as following. 1) CTL showed a tendency of overestimating daytime net shortwave radiation, thereby surface heat fluxes and Bowen ratio. The CTL experiment showed reasonable magnitudes and timing of air temperature at 2 m and 10 m; especially the small error in simulating minimum air temperature showed high potential for predicting frost and leaf wetness duration. The CTL experiment overestimated 10-m wind and precipitation, but the beginning and ending time of precipitation were well captured. 2) When the dynamic vegetation was turned on, the WRF/Noah-MP system showed more realistic values of leaf area index (LAI), net shortwave radiation, surface heat fluxes, Bowen ratio, air temperature, wind and precipitation. The DVG experiment, where LAI is a prognostic variable, produced larger LAI than CTL, and the larger LAI showed better agreement with the observed. The simulated Bowen ratio got closer to the observed ratio, indicating reasonable surface energy partition. The DVG experiment showed patterns similar to CTL, with differences for maximum air temperature. Both experiments showed faster rising of 10-m air temperature during the morning growth hours, presumably due to the rapid growth of daytime mixed layers in the Yonsei University (YSU) boundary layer scheme. The DVG experiment decreased errors in simulating 10-m wind and precipitation. 3) As horizontal resolution increases, the models did not show practical improvement in simulation performance for surface fluxes, air temperature, wind and precipitation, and required three-dimensional observation for more agricultural land spots as well as consistency in model topography and land cover data.

Effects of Maternal Genetic Potential and Parity with Pre- and Postpartum on Body Weights, Body Condition Score and Blood Metabolites in Hanwoo Cows (한우 암소의 유전능력과 산차에 따른 분만 전?후 체중, Body Condition Score 및 혈중 대사물질의 변화)

  • This study was conducted to investigate effects of maternal genetic potential and parity with pre- and postpartum periods on body weights, body condition score (BCS) and blood metabolites in relation to physiological stress and nutritional metabolism in Hanwoo cows. Also, this study was designed to develop effective husbandry technique for Hanwoo cows concerning of pre- and postpartum periods, and to get basic data for it. Forty five cows were allocated into two groups, 24 cows with high maternal genetic potentials and 21 cows with low maternal genetic potentials. The average parity of experimental cows with high and low maternal genetic potentials were 2.83±1.63 and 3.00±1.77, respectively. The growth performances such as body weights, average daily gain (ADG) and BCS were not different between two groups regardless of maternal genetic potential. However, pre- and postpartum periods had effects on the growth performances (p<0.05). Parity had no effects on ADG and BCS (p>0.05), but effect on body weight of cows (p<0.05). The metabolites of physiological stress such as neutrophil, hematocrit and cortisol, and nutritional metabolites such as albumin, blood urea nitrogen (BUN), triglyceride, and non-esterified fatty acid (NEFA) concentrations in blood of cows were affected by pre- and postpartum periods in a large scale, while those were partially affected by maternal genetic potential. However, among the metabolites in blood, only neutrophil and triglylceride concentrations were affected by different parity of cows. Therefore, the present study suggests that nutritional intake and digestion are affected by physiological stress due to the parturition, and it should need to consider different husbandry technique based on the maternal genetic potential, and pre- and postpartum periods of cows.

Review: Distribution, Lactose Malabsorption, and Alleviation Strategies of Lactose Intolerance (유당불내증(Lactose Intolerance)의 발생 원인과 경감 방안에 대한 고찰)

  • Milk is called an almost complete food in terms of nutrition, especially for the younger generations because it contains a number of nutrients required for growth and development. Lactose intolerance is defined as a malabsorption of lactose in the intestine with some typical symptoms of abdominal pains and bloating, and occurred at 75% of global populations, which hampers milk consumption worldwide. Lacks of milk consumption in the underdeveloped countries frequently lead to many nutrients deficiencies, so that diseases including osteoporosis, hypertension, and colon cancer are more prevalent in the recent days. Lactose in foods needs to be hydrolyzed prior to intestinal absorption. The hydrolytic enzyme responsible for splitting lactose into its monomeric forms, glucose and galactose, is called as lactase or $\beta$-galactosidase. The former is primarily used as blood sugar and energy source and the latter used in glycolipid synthesis of brain tissues in infants. Lactose is clinically diagnosed with the breath hydrogen production test as well as intestinal biopsy. Reportedly, symptoms of lactose intolerance are widely prevalent at 25% of Europeans, 50 to 80% of Hispanics, South Indians, Africans, and Jews, almost 100% of Asians and native Americans. For the adults, phenotype of lactase persistence, which is able to hydrolyse lactose, is more common in the northern Europeans, but in the other area lactase non-persistence or adult-type hypolactasia is dominant. Genetic analysis on human lactase gene continued that lactase persistence was closely related to the err site of 1390 single nucleotide polymorphism from the 5'-end. To alleviate severity of lactose intolerance symptoms, some eating patterns including drinking milk a single cup or less, consumption along with other foods, whole milk rather than skimmed milk, and drink with live yogurt cultures, are highly recommended for the lactose maldigesters. Also, delay of gastric emptying is effective to avoid the symptoms from lactose intolerance. Frequency of lactose intolerance with conventional diagnosis is thought overestimated mainly because the subjects are exposed to too much lactose of 50 g rather than a single serving amount. Thus simple and accurate diagnostic method for lactose intolerance need to be established. It is thought that fermented milk products and low- or free lactose milks help improve currently stagnant milk consumption due to lactose intolerance which contributes to major barrier in milk marketing especially in Asian countries.

Physiological responses and subjective sensation of human body wearing Cool Mapsi in air-conditioning environment (냉방환경에서 쿨맵시 착용에 따른 생리적 반응과 주관적 감각)

  • The purpose of this study is to test the performance of the recommended summer dressing for office man through the analysis of skin temperature changes by air-conditioning temperature. We tested two clothing combinations; formal wear with necktie and casual shirts without necktie as for Cool mapsi. 4 male subjects sat to stabilize for thirty minutes after entering artificial-climate chamber with both temperature of $25^{\circ}C$, $27^{\circ}C$ and $50{\pm}10%$ R.H. And during 60 minute experiments of simulating office work, the subjective feelings including thermal, humidity and comfort sensation, skin temperature, clothing humidity and sweat amount were measured at the equal intervals. The result is that formal wear of $25^{\circ}C$ and Cool mapsi of $27^{\circ}C$ show good values such as low skin temperature, low clothing humidity and neutral thermal sensation. And Cool mapsi of $25^{\circ}C$ shows the risk of low rectal temperature for long and static energy level of office work. Formal wear of $27^{\circ}C$ shows high values of mean skin temperature, clothing humidity and thermal sensation. Second experiment was to find the ambient temperature when the subject wearing formal wear shows the skin temperature corresponding to which he shows on Cool mapsi of $27^{\circ}C$. The air-conditioning temperature on wearing formal wear has to be $2^{\circ}C$ lower to produce the corresponding skin temperature to which shows on wearing Cool mapsi of $27^{\circ}C$. Therefore it is possible to increase room temperature to $27^{\circ}C$, when wear Cool mapsi for summer office, for skin temperature and thermal sensation are produced the same.

Solid Phase Synthesis of N-(3-hydroxysulfonyl)-L-homoserine Lactone Derivatives and their Inhibitory Effects on Quorum Sensing Regulation in Vibrio harveyi (고체상 합성법에 의해 합성된 N-(3-hydroxysulfonyl)-L-homoserine Lactone 유사체들의 Vibrio harveyi 쿼럼 센싱에 대한 저해 효과)

  • The inhibitors against Vibrio harveyi quorum sensing (QS) signaling were developed by modifying the molecular structure of the major signal, N-3-hydroxybutanoyl-L-homoserine lactone (3-OH-$C_4$-HSL). A series of structural derivatives, N-(3-hydroxysulfonyl)-L-homoserine lactones (HSHLs) were synthesized by the solid-phase organic synthesis method. The in vivo QS inhibition by these compounds was measured by a bioassay system using the V. harveyi bioluminescence, and all showed significant inhibitory effects. To analyze the interaction between these compounds and LuxN, a 3-OH-$C_4$-HSL receptor protein of V. harveyi, we tentatively determined the putative signal binding domain of LuxN based on the sequence homology with other acyl-HSL binding proteins, and predicted the partial 3-D structure of the putative signal binding domain of LuxN by using ORCHESTRA program, and further estimated the binding poses and energies (docking scores) of 3-OH-$C_4$-HSL and HSHLs within the domain. In comparison of the result from this modeling study with that of in vivo bioassay, we suggest that the in silica interpretation of the interaction between ligands and their receptor proteins can be a valuable way to develop better competitive inhibitors, especially in the case that the structural information of the protein is limited.

Estimation of Fetal Dose during Radiation Therapy of Pregnant Patient (임산부의 방사선치료 시 태아선량 평가)

  • Purpose: To evaluate the effectiveness of a simple and practical shielding device to reduce the fetal dose for a pregnant patient undergoing radiation therapy of brain metastasis. Materials and Methods: The dose to the fetus was evaluated by simulating the treatments using the anthropomorphic phantom. The prescription dose at mid-brain is $300cGy{\times}10$ fractions with 6 MV photon with $18{\times}22cm^2$ field size. The additional shielding devices to reduce the fetal dose are a shielding wall, cerrobend plates and lead (Pb) sheets over acrylic bridge. Various points of measurement with off-field distance were detected by using ion-chamber (30, 40, 50, and 60 cm) with and without the shielding devices and TLD (30, 40, 50, 60, and 70 cm) only with the shielding devices. Results: The doses to the fetus without shielding were 3.20, 3.21, 1.44, 0.90 cGy at the distances of 30, 40, 50, and 60 cm from the treatment field edge. With shielding, the doses were reduced to 0.88, 0.60, 0.35, 0.25 cGy, and the ratio of the shielding effect varied from 70% to 80%. TLD results were 1.8, 1.2, 0.8, 1.2, and 0.8 cGy (70 cm). The total dose to the fetus was expected to be under 1 cGy during the entire treatment. Conclusion: The essential point during radiation therapy of pregnant patient would be minimizing the fetal dose. 10 cGy to 20 cGy is the threshold dose for fetal radiation effects. Our newly developed device reduced the fetal dose far below the safe level. Therefore, our additional shielding devices are useful and effective to reduce the fetal dose.
