A novel acetylalginate esterase (AcAlgE) gene was previously cloned and characterized from Sphingomonas sp. MJ-3. In this study, the synergistic effects of MJ-3 AcAlgE, and KS-408 alginate lyase on the degradation of acetylalginate from Pseudomonas aeruginosa were investigated by using high-field 1H-NMR and an FPLC-equipped peptide column. The alginate lyase coupled assay of AcAlgE showed that degradation of high molecular weight acetylalginate was more difficult than degradation of acid hydrolyzed acetylalginate. The degradation of acetylalginate by alginate lyase was easier after AcAlgE was used to remove the acetyl group from acetylalginate. This result showed that the recombinant AcAlgE enhanced the degradation of acetylalginate by alginate lyase.
In order to expand the utility of alginic acid in the food industry, we have investigated to prepare low viscous alginic acid using a method for degradation of alginic acid with the enzyme system of Vibrio sp. AL-145. The enzyme showed maximum activity at pH 8.0 and $37^{\circ}C$, and was stable in the pH range 7.5 to 8.5 and at temperature up to $30^{\circ}C$, and 0.5M NaCl needed for the enzyme activity. The viscosity of alginic acid decreased with the reaction time courses regardless of alginic acid and enzyme concentration, and 90% of viscosity decreased with 60 min of reaction time, but the changes of reducing sugar not exhibited. The soluble concentration of partially degradated alginic acid(PDA) in water was about 10%(w/v), and adsorption capacity of $Ca^{2+}$ ion increased with increasing the concentration of PDA. The alcohol concentration on precipitation of PDA was higher than Na-alginic acid.

Uronate compositions and molecular weights of alginates from the four kinds of brown algae, sea mustard (Undaria pinnatifida), sea tangle (Laminaria japonica), gulf weed (Sargassum fulvellum), and seaweed fusiforme (Hizikia fusiforme), in regard with the harvesting season were investigated. Sea mustard contained the highest amount of alginates in the four kinds of brown algae. D-Mannuronic acid to L-guluronic acid (M/C) ratio of the alginates was high in order of seaweed fusiforme, gulf weed, sea mustard, and sea tangle, and especially in water-soluble alginate. Molecular weights of the alginates were greater with the growing period ranging in $4,500\~4,800\;kDa$ for sea tangle, $4,000\~4,200\;kDa$ for sea mustard, $3,300\~3,400\;kDa$ for seaweed fusiforme, and $3,000\~3,200\;kDa$ for gulfweed. In water-soluble alginate of sea mustard, M/G ratio was much higher in sporophyll than in midrib and blade.
A polyguluronate-specific lyase from Streptomyces sp. strain M3 has been previously cloned and characterized. In this study, the M3 alginate lyase gene in the pColdI vector was mutated by site-directed mutagenesis and random mutagenesis to enhance the alginate degrading activity. Six mutants were obtained: Ser25Arg, Phe99Leu, Asp142Asn, Val163Ala, Lys191Glu, and Gly194Cys. Phe99Leu and Lys191Glu mutants completely lost their alginate lyase activity, whereas the alginate degrading activity of Gly194Cys mutant increased by nearly 10 fold. The 3-D protein structure of M3 alginate lyase, which was constructed using the Swiss-Model automodeler, was also compared to the crystal structure of another alginate lyase. A mutated glycine residue was positioned between Gly193 and Tyr195 of the C-terminal conserved sequence, YFKAGXYXQ. A phenylalanine residue (at position 99) and a glycine residue (at position 194) mutated in this study were distant from the active site, but the degrading activity was strongly affected by their mutation.

Oligosaccharides production showed various biological activities in vivo like functional foods and industrial materials utilized available within many practical applications which have obtained from the degradation of alginate. Alginate is rich in the main component of seaweeds especially the brown algae. We investigated what degrading alginate from seaweeds to make alginate oligosaccharides can utilize in various fields using enzyme secreting Erwinia tasmaniensis. In this study, we observed an optimal culture condition of E. tasmaniensis, and characteristics of alginate lyase secreting E. tasmaniensis. These bacteria, E. tasmaniensis, were isolated from Nuruk. In this case, a suitable growth factor for E. tasmaniensis was culture it for 36 h in broth media on concentration of 1.0% (w/v) alginate. The enzyme showed the highest level of alginate lyase activity when cultured on broth media containing 1.0% (w/v) sodium alginate for 72 h. Optimal condition of pH, temperature and duration time for alginate lyase activity were found to be pH 6.0, $20^{\circ}C$ and 60 min, respectively.
This study examines the effects of alginic acid, a source of dietary fiber, in a glucose-derived media. In particular, we examined how the presence or absence of alginic acid affected the differentiation and triglyceride densities of 3T3-L1 cells. We established that the addition of insulin-like growth factor-I (IGE-I) to 3T3-L1 cells results in acceleration of differentiation. We sought to determine the role of alginic acid in the production of fat by adding alginic acid to 3T3-L1 cells and examining its ability to limit or potentiate this stimulatory effects of IGE-I and IGF binding proteins. We have determined that alginic acid restricts 3T3-L1 cell differentiation and the creation of triglycerides, effectively attenuating 3T3-L1 cell metablolism and growth.
Phase separation is the typical phenomenon in protein-polysaccharide mixtures because of thermodynamic incompatibility between two macromolecules. Phase separations of casein-alginate-water systems were investigated by using phase diagram under varying pH (6, 8 and 10) and NaCl concentrations (0, 0.25 and 0.5 M). Incompatibility decreased with increasing pH and decreasing NaCl concentration. Molecular weight of alginates did not significantly affect the phase diagram of casein-alginate-water systems. The results strongly suggested that compatibility of casein and alginate involved electrostatic interactions.
본 연구의 목적은 치아 회분말과 연석고 및 알긴산을 혼합하여 이식한 후 초기 골 형성을 알아보는 데 있다. 백서 30마리의 두개골에 8mm 직경의 골결손부를 형성한 후 인간의 치아 회분말과 연석고 및 알긴산을 혼합하여 이식한 후 대조군, 치아 회분말과 연석고 처치군, 그리고 치아 회분말과 연석고 및 알긴산 혼합 처치군에서 각각의 골 결손부 치유과정을 조직병리학적, 조직형태계측학적으로 비교 관찰하였다. 통계학적으로 주간 변화(4주, 8주)는 유의하지 않았고, 각 그룹 간에 있어도 치아 회분말과 연석고 처치군과 치아 회분말과 연석고 및 알긴산 혼합 처치군 간 모두(4주, 8주) 유의한 차이가 없었다. 그러나 이들 치아 회분말과 연석고 처치군과 치아 회분말과 연석고 및 알긴산 혼합 처치군은 대조군에 비해서는 유의하게 우수한 신생골 형성을 보였다. 비록 통계학적으로는 치아 회분말과 연석고 처치군과 치아 회분말과 연석고 및 알긴산 혼합 처치군이 유의한 차이를 보이지 않았지만 치아 회분말과 연석고 및 알긴산 혼합 처치군에서 보다 더 우수한 신생골 형성 경향을 보였다. 뿐만 아니라, 신생골의 내용면에서도 치아 회분말과 연석고 및 알긴산 혼합 처치군에서 좀더 많은 신생 직조골의 융합이나 골 소주형성이 관찰되어 신생골의 형성 및 성숙에 알긴산이 역할을 담당하는 것으로 사료되었다. 결론적으로 골결손부 치료 방법으로 치아 회분말과 연석고 처치군 또는 치아 회분말과 연석고 및 알긴산 혼합 처치로 결손부에 이식하는 것은 신생골 형성의 양적, 질적 개선에 통계학적으로 유의하며, 알긴산을 이식재와 함께 처리한 경우에 신생골 형성의 양적, 질적 개선에 도움을 줄 수 있을 것으로 사료된다.$1^{\circ}C$ 냉수로 세척하고 PETE tray로 포장하여 $4^{\circ}C$로 저장한 경우 깻잎 고유의 초록색과 향을 유지하고 있어 저온냉수 세척과 tray 포장이 세척 청경채의 선도 유지에 효과가 있는 것으로 나타났다.1%,\;pendimethalin\;1.3{\sim}2.9%$ 및 $3.8{\sim}10.8%,\;ethoprophos\;0.6{\sim}2.7%$ 및 $0.1{\sim}0.3%$이었다. 인공강우실험 후 공약의 토심별 분포를 살펴 본 결과 alachlor와 ethopropho는 토심 $10{\sim}15cm$까지 이동하였고, ethalfluralin과 pendimethalin는 대부분 토심 5 cm 이내에 잔류하였다. 경사도 30%의 경우가 10%에 비하여 각 농약의 유실량이 $0.2{\sim}1.9$ 배 증가하였는데 유출수에 의한 농약의 유실량 차이는 유출수 중 농도 차이로 판단되며, 유실토양에 의한 농약 유실량 차이는 토양 유실량과 관계되는 것으로 생각되었다. 농약의 강우에 의한 유실은 복잡하게 작용하는 많은 환경적 요인에 의하여 영향을 받지만 정교하게 구성된 환경 시나리오에 의하여 예측 가능할 것으로 판단되었다.고 도라지는 물에 우려 푹 삶았고, 감자, 송이 등은 잘게 썰어 쌀과 함께 밥을 조리하였다. 4. 약선 음식조리방법 약선음식의 재료는 평상시 식생활에 사용되고 있던 식품들의 기능성분과 약이성을 이용하여 만성적인 질병과 급성적인 복통 설사 등에 재료의 전처리를 통해 죽으로 많이 이용하였다. 특히 곡류 등은
Partial hydrolyzing condition for low-molecularized alginate and rheological properties such as viscosity, solubility, emulsitying ability, oil absorption capacity, bile acids binding and metal ion binding of the low-molecularized alginates from the sea mustard ( Undaria pinnatifida) and giant kelp (Macrocystis pyrifera) were investigated. Alginate from sea mustard was regularly hydrolyzed with the increase of HCl concentration in the range of 0.2 N to 0.5 N and with the prolonged reaction time at $100^{\circ}C$. The molecular weight of alginate was decreased to a part of 100 after hydrolysis for 50 min with 0.3 N HCl. The ratio of mannuronate to guluronate was increased with the acid hydrolysis and total yield was estimated to be $75\%\~80\%$. Low-molecularization of alginate was featured in the apparent decrease of viscosity, whereas solubility, emulsifying ability, and bile acids binding ability were increased with the low-molecularization. Oil absorption capacity of the acid$\cdot$alkali soluble alginate was slightly higher than that of the water soluble alginate. Metal ion binding capacity was the highest in acid$\cdot$alkali soluble alginate, and decreased with the low-molecularization.

This study investigated the effects of alginate oligosaccharide on lipid metabolism in mice fed a high-cholesterol diet for 6 weeks. Male apoE mice were assigned to four groups: normal diet group (N), high-cholesterol diet group (HC), HC with 5% alginate oligosaccharide group (HC-AOL), and 10% alginate oligosaccharide group (HC-AOH). Epididymal adipose tissue and kidney adipose tissue weights were significantly reduced in the HC-AOH group by 131.4% and 148.4%, respectively, as compared to the HC group. Serum total cholesterol (TC), triglyceride (TG), and LDL-cholesterol levels were also significantly reduced in the HC-AOH group by 57.5%, 51.4%, and 82.9%, respectively, as compared to the HC group. Hepatic TC and TG levels in the HC-AOH group were significantly reduced by 72.3% and 33.5%, respectively, as compared to the HC group. These results indicate that alginate oligosaccharide might improve lipid metabolism and reduce fat accumulation.
