Identification and Characterization of the Aquaporin Gene aqpA in a Filamentous Fungus Aspergillus nidulans (사상성 진균 Aspergillus nidulans에서 아쿠아포린 유전자 aqpA의 분리 및 분석)

  • Aquaporin is a water channel protein, which is classified as Major Intrinsic Protein (MIP), found in almost all organisms from bacteria to human. To date, more than 200 members of this family were identified. There are two major categories of MIP channels, orthodox aquaporins and aquaglyceroporins, which facilitate the diffusion across biological membranes of water or glycerol and other uncharged compounds, respectively. The full genome sequencing of various fungal species revealed 3 to 5 aquaporins in their genome. Although some functions of aquaporins found in yeast were characterized, however, no functional characteristics were studied so far in filamentous fungi, including Aspergillus sp. In this study, one orthodox aquaporin homolog gene, aqpA, and four aquaglyceroporin homologs, aqpB-E, in a model filamentous fungus Aspergillus nidulans were identified and the function of the aqpA gene was characterized. Knock-out of the aqpA gene didn't show any obvious phenotypic change under the osmotic stress, indicating that the function of the gene does not involved in the osmotic stress response or the function could be redundant. However, the mutant showed antifungal susceptibility resistance phenotype, suggesting that the function of the aqpA gene could be involved in sensing the antifungal substances rather than the osmotic stress response.

  • Salinity gradient power is a system which sustainably generates electricity for 24 hrs, if the system is constructed at a certain place where both seawater and river water are consistently pumped. Since power is critically determined by the water flux and the salt rejection, a membrane of water-semipermeable aquaporin protein in cell membranes was studied for pressure-retarded osmosis. NaCl was used as a salt, and $NaNO_3$ was used as a candidate to check the ion selectivity. The water flux of biomimetic aquaporin membranes was negligible at a concentration below 2M. Also, there is no remarkable dependence of water flux and ion selectivity on concentrations higher than 3M. Therefore, the biomimetic aquaporin membrane could not be applied into pressure-retarded osmosis; however, if a membrane could overcome the current limitations, the properties shown by natural cells could be accomplished.

  • Functional nanochannels were fabricated in order to control selective ion transportation with high permeability and low energy consumption. In this research, nanochannel platform fabrication process and surface functionalization process were developed. In addition, selective ion transportation and concentration measurement system was also set-up. By using fabricated multilayer metal membrane with electrical bias, 95% of ion ($Cl^-$) was blocked. This developed process is new-conceptional membrane fabrication technology and is expected to be applied to next-generation water purification/desalination, portable artifical kidney, and artificial sense organ.

  • Background : Aquaporins (AQPs) may play a role in the pathogenesis of pulmonary inflammation and edema. This study investigated the role ofAQPs in acute lung injury following bleomycin inhalation in rats. Methods : Sprague-Dawley rats were treated via inhalation with 10 U/kg bleomycin hydrochloride dissolved in 5 ml of normal saline. The control rats were treated with 5 ml normal saline. The animals (n = 6-8 rats per group) were sacrificed at 4, 7, and 14 d. The changes in AQP1, AQP4, and AQP5 expression levels over time were analyzed by Western blotting. The nitrate and nitrite concentrations in the bronchoalveolar lavage fluid (BALF) were measured using a modified Griess reaction. ELISA was used to check cytokines. Results : The respiration rates were significantly higher 4 and 7 days after the bleomycin treatment compared with those of the control rats. The tidal volume was lower in rats at 4 days after the bleomycin treatment, and the wet/dry weights of the lung were significantly higher than those of the control group. The nitrite and nitrate concentrations in the BALF from the rats at 4 days after exposure to bleomycin were greater than those from the saline-treated rats. Immunoblotting studies demonstrated that the AQP1 and AQP4 expression levels were lower in the rats at 4 days. However, the AQP4 expression level was higher at 7 days. The AQP5 expression level increased at 4, 7 and 14 days after the bleomycin treatment. Conclusion : This study demonstrates that AQPs are expressed differently in bleomycin-induced pulmonary edema.

Conflicting Physiological Characteristics and Aquaporin (JcPIP2) Expression of Jatropha (Jatropha curcas L.) as a Bio-energy Crop under Salt and Drought Stresses (바이오에너지 작물 소재로서 자트로파의 염과 가뭄 스트레스 하에서 상반되는 생리적 특성과 아쿠아포린(JcPIP2)의 발현)

  • This study was undertaken to collect basic knowledge of Jatropha which is one of bio-energy crops, based on the understanding of physiological and molecular aspects under salt and drought conditions. The treatments were followed as: 100, 200 and 300 mM NaCl for salt stress and 5, 10, 20 and 30% PEG for drought stress for 8 days, respectively. Leaf growth, stomatal conductance, chlorophyll fluorescence and gene expression of aquaporin (JcPIP2) of Jatropha were investigated. From 2 days after treatments, plants treated with higher than 100 mM NaCl and 10% PEG respectively were significantly suppressed in leaf length, width, and stomatal conductance, but 5% PEG treatment showed that plant growth was improved more than control plant. Semi-quantitative RT-PCR analyses revealed that the JcPIP2 gene was expressed in root, stem, cotyledon and leaves. It was not detected in leaves at 200 and 300 mM NaCl treatments. However, transcripts of JcPIP2 were induced in roots and stems under salt and drought conditions compared to those of healthy plants. Therefore, it was concluded that JcPIP2 plays an important role in improving drought tolerance.

Anti-aging & Skin Hydration Effects of Spore oil Extracted from Ganoderma lucidum (영지버섯에서 추출한 포자오일의 항노화 및 보습 효능)

  • This study evaluated the anti-aging activity with antioxidant, anti-inflammatory and moisture activity of Ganoderma lucidum spore oil(GLS). GLS increased DPPH radical scavenging activity in a dose-dependent manners. Anti-inflammatory assay measured the inhibitory effect of GLS on NO, TNF-α and IL-6 production in LPS-stimulated RAW264.7 cells. As a result GLS inhibited NO and pro-inflammatory cytokine, TNF-α, IL-6 production. Also using human fibroblast cell to the procollagen production analysis and COL1A1 mRNA expression level analysis for defining, and for AQP-3 mRNA expression level analysis, used human keratinocyte cell. GLS increased procollagen production and COL1A1, AQP-3 mRNA expression. Our results suggest that the GLS have potential anti-inflammatory and wrinkle improves, skin moisture effect.

  • Several studies have reported a possible link between anti-aquaporin-4 antibody and vitamin B12 deficiency in neuromyelitis optica spectrum disorder (NMOSD). Bilaterally symmetric hyperintense signals on magnetic resonance imaging (MRI) of the posterior columns, called the inverted V sign, are a characteristic feature of subacute combined degeneration associated with vitamin B12 deficiency. We report a patient with anti-aquaporin-4 antibody-positive NMOSD and an inverted V sign on MRI of the spinal cord and address the association between anti-aquaporin-4 antibody and functional vitamin B12 deficiency.

The Effect of Soil Amended with β-glucan under Drought Stress in Ipomoea batatas L. (𝛽-glucan 토양혼합에 따른 고구마의 가뭄피해 저감 효과 )

  • Biopolymer is a versatile material used in food processing, medicine, construction, and soil reinforcement. 𝛽-glucan is one of the biopolymers that improves the soil water content and ion adsorption in a drought or toxic metal contaminated land for plant survival. We analyzed drought stress damage reduction in sweet potatoes (Ipomoea batatas L. cv. Sodammi) by measuring the growth and major protein expression and activity under 𝛽-glucan soil amendment. The result showed that sweet potato leaf length and width were not affected by drought stress for 14 days, but sweet potatoes grown in 𝛽-glucan-amended soil showed an effect in preventing wilting caused by drought in phenotypic changes. Under drought stress, sweet potato leaves did not show any changes in electrolyte leakage, but the relative water content was higher in sweet potatoes grown in 𝛽-glucan-amended soil than in normal soil. 𝛽-glucan soil amendment increased the expression of plasma membrane (PM) H+-ATPase, but it decreased the aquaporin PIP2 (plasma membrane intrinsic protein 2) in sweet potatoes under drought stress. Moreover, water maintenance affected the PM H+-ATPase activity, which contributed to tolerance under drought stress. These results indicate that 𝛽-glucan soil amendment improves the soil water content during drought and affects the water supply in sweet potatoes. Consequently, 𝛽-glucan is a potential material for maintaining soil water contents, and analysis of the major PM proteins is one of the indicators for evaluating the biopolymer effect on plant survival under drought stress.
