Development and Characterization of EMS-induced Mutants with Enhanced Salt Tolerance in Silage Maize (EMS 유도 내염성 증진 사료용 옥수수 돌연변이체 선발 및 특성 분석)

  • Maize (Zea mays L.) is one of the most valuable agricultural crops and is grown under a wide spectrum of environmental conditions. However, maize is moderately sensitive to salt stress, and soil salinity is a serious threat to its production worldwide. In this study, we used ethyl methane sulfonate (EMS) to generate salt-tolerant silage maize mutants. We screened salt-tolerant lines from 203 M3 mutant populations by evaluating the morphological phenotype after salt stress treatment and selected the 140ES91 line. The 140ES91 mutant showed improved plant growth as well as higher proline content and leaf photosynthetic capacity compared with those of wild-type plants under salt stress conditions. Using whole-genome re-sequencing analysis, 1,103 single nucleotide polymorphisms and 71 insertions or deletions were identified as common variants between KS140 and 140ES91 in comparison with the reference genome B73. Furthermore, the expression patterns of three genes, which are involved in salt stress responses, were increased in the 140ES91 mutant under salt stress. Taken together, the mutant line identified in our study could be used as an improved breeding material for transferring salt tolerance traits in maize varieties.

Functional characterization of domestic and foreign green tea cultivars at different harvest periods (채취시기가 다른 국내외 녹차잎의 기능성분 함량, 뇌세포 생존 및 대사 효소 활성 조절 효과 조사)

  • This study was performed to compare nutritional compounds and physiological functions of five domestic and imported green tea cultivars at three time points. The five cultivars were compared for theanine, γ-aminobutyric acid, and catechin content by LC-MS/MS and HPLC. Furthermore, the five tea cultivars were functionally characterized with respect to antioxidant activity, brain cell protective effect, and inhibitions of α-glucosidase and HMG-CoA reductase activities. Among green tea cultivars, Chamnok had the highest content of catechins (198 mg/g DW), theanine (11.89 mg/g DW), and tannin (23.6 mg/g DW). Considering functional properties, Chamnok treatment resulted in the maximum viability of brain cells and reduced the cortisol content of SH-SY5Y cells. The inhibition of α-glucosidase and HMG-CoA reductase was the strongest following Chamnok treatment (72.9% and 69.8%, respectively). These results indicate that Chamnok could be optimal for consumption or favorable processing owing to its high nutritional compounds, such as theanine and catechin, and remarkable brain cell protective effects.

  • Recent developments and improvements of multiple technological elements including mass spectrometry (MS) instrument, multi-dimensional chromatographic separation, and software tools processing MS data resulted in benefits of large scale proteomics analysis. However, its throughput is limited by the speed and reproducibility of the protein digestion process. In this study, we demonstrated a new method for rapid proteolytic digestion of proteins using acoustic technology. Tryptic digests of BSA prepared at various conditions by super acoustic for optimization time and intensity were analyzed by LC-MS/MS showed higher sequence coverage in compared with traditional 16 hrs digestion method. The method was applied successfully for complex proteins of a breast cancer cells at 30 min of digestion at intensity 2. This new application reduces time-consuming of sample preparation with better efficiency, even with large amount of proteins, and increases high-throughput process in sample preparation state.

Physiological Responses of Rice(Oryza sativa L.) Varieties to Ozone (벼 품종의 오존에 대한 생리적 반응)

  • This experiment was conducted to select and utilize the tolerant rice varieties for ozone stress through comparing the physiological responses. In SOD activity, Backunchalbyeo which treated once by 0.15 ppm $0_3$, Ilpumbyeo and Inbujinado treated twice showed significantly different SOD activity compared with $non-O_3$ treatment. No significant difference of SOD activity in three-times $0_3$ treated Sangnambatbyeo and Hangangchalbyeo was found as compared to ($non-O_3$ treated)control, POD activity, when Sangnammbatbyeo, Hangsngchalbyeo, Ilpumbyeo and Inbujinado were sequently treated by third ozone, showed no significant difference between $O_3$ treatment and $non-O_3$. In the comparison of amino acid content in rice leaf blade between three times $O_3$ treatment and $non-0_3$ treatment in 3 varieties, phenylalanine content represent as increased quantities. Especially, in three times $0_3$ treatment of Sangnambatbyeo, phenylalanine content(O.66 mg/g) was significantly increased as compared with $non-O_3$. Visual-growing characters of Sangnembatbyeo were significantly different between $0_3$ treatment and non-treatment. However, grain-filling rate showed no difference. SOD and POD related to resistant mechanism were high activity. In addition, phenylalanine known as precursor which is synthesized resistant materials was increased by $0_3$ treatment. These results suggested that Sangnambatbyeo which increased SOD and POD activity, and phenylalanine content under ozone stress could be selected as a tolerant variety.

  • This experiment was carried out to study the practical utility of Mucor rennet in making Blue cheese and to investigate the changes in the level of various nitrogen compounds. 1. The Mucor rennet cheese, the calf rennet cheese and the mixed rennet cheese did not show any significant difference in their yields. 2. The dry matter contents of Blue cheese was increased during the ripening and the levels of Mucor rennet did not have any influence in these respect. 3. The water soluble nitrogen contents of Blue cheese increased during ripening. On 0 day of ripening the Mucor rennet cheese contained water soluble nitrogen than the calf rennet cheese. On 40 days of ripening the mixed cheese contained less water soluble nitrogen than the calf rennet cheese. 4. Non protein nitrogen, peptone amino nitrogen, water soluble protein nitrogen, proteose nitrogen and peptone nitrogen appeared to contain similar levels of water soluble nitrogen. 5. The results of electrophoresis indicated a greater degredation on as-casein and ${\beta}$-casein in the Blue cheese made with Mucor rennet than in those made with calf rennet. On 60 days of ripening the mixed cheese more casein than did the Mucor rennet cheese. 6. The free amino acid contents of the Mucor rennet cheese was higher than the calf rennet cheese at 60 days of ripening.

Antitumor and Immunomodulating Effects of Seaweeds toward Sarcoma-180cell (파래와 곤피에서 추출한 당단백질의 Sarcoma-180 cell에 대한 항암효과 및 면역활성)

  • This study was investigated on the antitumor effects of protein-polysaccharide fraction(PPF) extracted from seaweeds such as sea-lettuce and gonpi toward sarcoma-180 cells. In the PPF extracted from these seaweeds, the polysaccharide contents of sea-lettrce and gonpi were 52.20% and 48.16%, respectively. The highest levels of constituents monosaccharides found in seaweeds was fructose. The major amino acids were aspartic acid, glutamic acid, glycine and cystein. The solid tumor growth inhibition showed the highest level of 64.55% when 50mg/kg sea-lettuce was administerated. The life prolongation effect was 18.31% at 50mg/kg of gonpi. In the effects of immunologic activity, when 50mg/kg sea-lettuce was administrated, the number of circulating leucocyte showed the highest level (65.11%). The number of total peritoneal exudate cells of the sea-lettuce administerated group was increased significantly in comparison with the control group. The hematological analysis of the experimental group was similar with that of the control group.

Isolation of indigenous Lactobacillus plantarum for malolactic fermentation (말로락틱 발효에 적합한 토착 Lactobacillus plantarum 분리)

  • The malolactic fermentation (MLF), which is widely used in winemaking, is the conversion of malic acid to lactic acid conducted by the malolactic enzyme (Mle) of lactic acid bacteria. In order to select the strains with MLF among 54 lactic acid bacteria isolated from the traditionally fermented foods, we designed a primer set that specifically targets the conserved regions of the mle gene and then selected four strains that harbor the mle gene of Lactobacillus plantarum. All strains were identified as L. plantarum by analyzing the 16S rRNA sequences, biochemical properties, and the PCR products of the recA gene. From comparison of the mle gene sequences consisting of 1,644 bp, the nucleotide and amino acid sequence of strain JBE60 correspond to 96.7% and 99.5% with those of other three strains, respectively. The strain JBE60 showed the highest resistant against 10% (v/v) ethanol among the strains. The strains lowered the concentration of malic acid to average 43%. Considering the ethanol resistance and conversion of malic acid, the strain JBE60 is considered as a potential starter for the malolactic fermentation.

Studies on Antitumor Components of the Cultured Mycelia of Interspecific Protoplast Fusant F-2 of Ganoderma lucidum and Ganoderma applanatum (영지와 잔나비걸상버섯의 원형질체 융합균주의 항암 성분에 관한 연구)

  • On the five interspecific protoplast fusants of Ganoderma lucidum and G. applanatum was the antitumor test performed. The fusant F-2 was selected, to examine the cultured mycelia (protein bound polysaccharide) as antitumor components. When a dose of 20 mg/kg/day of each components purifed from F-2 fusant was, i.p., injected into ICR mice, the inhibition ratio of Fr. II against the solid form of sarcoma 180 increased to 1.5 times as compared with that of their parents. When Fr. II was examined for immunopotentiation activity, it increased the amount of the superoxide anion in activated macrophages to 1.2 times and the count of hemolytic plaque forming cells in the spleen to 4.3 times as compared with that of each control group. Its chemical analysis showed 85.2% polysaccharide which consisted of glucose, galactose, mannose, fucose and xylose, and 0.39% protein of 15 amino acids. The content of hexosamine was 0.39% and the molecular weight of Fr. V was $5.6{\times}10^4$ dalton.

Antitumor Effect and Immunology Activity of Seaweeds toward Sarcoma-180 (청각과 김에서 추출한 당단백질의 Sarcoma-180에 대한 항암효과 및 면역활성)

In the PPF extracted from these sewaweeds, the polysaccharide contents of sea-staghorn and laver were $62.26\%$ and $65.78\%$, respectively. The highest levels of polysaccharides found in seaweeds was fructose. The major amino acids were aspartic acid, glutamic acid, glycine and cystein. The solid tumor growth inhibition showed the highest level of $53.30\%$ when 50mg/kg sea-staghorn was administrated. The life prolongation effect was $17.35\%$ at 50 mg/kg of laver. In the effects of immunologic activity, when 100mg/kg sea-staghorn was administrated, the number of circulating leucocyte showed the highest level of $82.23\%$ but decreased leucocyte for prolonged times. The number of total peritoneal exudate cells of the sea-staghorn administerated group was increased significantly in comparison with the control group. The hematobiolgoical analysis of the experimental group was similar with that of the control group. This experiments indicated that hemeastasis still maintained nor-mal state and not showed any harmful effects in normal mice.

A Study on the Solubilization of $\alpha$-Chymotrypsin Using AOT Reverse Micelles; Effects of pH and salts (AOT 역미셀을 이용한 $\alpha$-chymotrypsin의 가용화에 대한 연구;pH와 염의 영향)

  • Micellar aggregates are known to be useful for the selective isolation of biologically active materials such as amino acids, proteins, and enzymes from crude mixtures sparsely dispersed in water. In this study, the effects of pH, salt type and its concentration on the solubilization of $\alpha$-chymotrypsin into the organic micellar phase, which consisted of AOT (sodium 야(2-ethylhexy)sulfosuccinate) and iso-octane, were comprehensively examined. It was found that maximum extraction efficiency was attained at a pH below the isoelectric point of $\alpha$-chymotrypsin; at pH=5.0 for NaCl and KCl, and at pH=7.0 for $CaCl_2$and $MgCl_2$. In order to avoid complications stemming from the precipitationof protein at low pH interfaces, the protein concentrations in the organic and aqueous phases were directly measured. The size of the micelle water pool was estimated by measuring the molar ratio of the surfactant to the water, W(sub)o. The resulting values of W(sub)o were nearly constant at 30 and 19 for NaCl and KCl, respectively, and were independent of pH. The addition of 1:2 salts like $MgCl_2$and $CaCl_2$ led to much lower, but a constant value of, W(sub)o than the 1:1 salts.
