Effect of Plant Growth Regulators on Regeneration from the Cotyledon Explants in Watermelon (Citrullus lanatus (thunb.) Matsum. & Nakai) (수박(Citrullus lanatus (thunb.) Matsum. & Nakai) 자엽 절편의 재분화에 미치는 생장조절물질의 영향)

  • In this study, we developed a high frequency watermelon regeneration system using three breeding lines ('B02', 'B05' and 'D04') of watermelon (Citrullus lanatus (thunb.) Matsum. & Nakai) which are differed in their fruits in shape, color of pericarp and flesh. The highest frequency of explants with callus was observed by using explants that consist of cotyledon proximal part end in all breeding lines, and the highest rate of callus induction was obtained on MS medium containing 1.0 mg/L BAP + 0.5 mg/L IAA for 'B02' (94%), 3.0 mg/L BAP for 'B05' (95%), 3.0 mg/L BAP + 0.1 mg/L IAA for 'D04' (90%). The highest shoot regeneration rates from derived callus were obtained on MS medium containing 1.0 mg/L BAP + 0.5 mg/L IAA for 'B05' (94%), and then a 'B02' (81%) with a same culture conditions, and the lowest regeneration was obtained on MS medium containing 1.0 mg/L BAP for 'D04' (56%). Regenerated plants showed the best rates of root formation on MS containing 0.1 mg/L IBA for 'B02' (67%), 0.1 mg/L NAA for 'B05' (87%), 0.5 mg/L IAA for 'D04' (88%). The regenerated plants showed a 100% survival rate in soil condition. The tissue culture and regeneration conditions obtained from this study will be useful for regenerating plants in breeding applications, and will be a useful tool for further genetic transformation studies on watermelons.

Bush Growth and Yield of Highbush Blueberry 'Duke' as Influenced by Different Pruning Times in Unheated Plastic House (블루베리 '듀크' 품종의 무가온 시설재배시 전정시기가 수체생장과 과실수량에 미치는 영향)

  • This experiment was conducted to compare effects of summer pruning at different times and dormant pruning on shoot growth and yield of northern highbush blueberry. Using 7 (2018) to 9 (2020) years old 'Duke' bushes grown in 180 L containers, summer pruning was treated on June 20 (around 30 days after harvest), July 20, and August 20 consecutively in both 2018 and 2019 removing 30% of the total woods, while dormant pruning (conventional) was on January 20 in both 2019 and 2020. Summer pruning reduced shoot growth the following year, especially when treated in late summer. Total shoot length per bush decreased to 47%, 37%, and 33% on October 15, 2020 in June, July, and August pruning, respectively, compared with that of dormant pruning. Summer pruning at different times in 2018 and 2019 did not affect berry characteristic in the following year. Yield per bush was not significantly changed in 2019, but it decreased by 21 to 38% in 2020 in the summer pruning treatments compared with 2.9 kg of the dormant pruning. It was concluded that consecutive summer pruning in 'Duke' under unheated plastic house could weaken the shoot growth with reducing yield.

In vitro shoot regeneration from leaf tissue of "Whangkeumbae" pear(Pyrus pyrifolia Nakai) (황금배(Pyrus pyrifolia Nakai) 잎 조직으로부터 기내 신초 재분화)

  • In order to establish an efficient adventitious shoot regeneration conditions from leaf explants for Asian pear 'Whangkeumbae', the effect of concentration and kinds of plant growth regulator and carbon source was investigated. Leaf explants of cultures grown on Murashige and Skoog (MS) medium containing 8 g/L plant agar were used. When the medium contained 0.25 mg/L thidiazuron (TDZ) and 0.3 mg/L indolebutyric acid (IBA), the adventitious shoot regeneration rate (ASRR) was greater as 61.1% than others treated and higher TDZ concentrations (2.5 and 5 mg/L) treatment significantly reduced the ASRR. As the effect of IBA and indoleacetic acid (IAA) concentration on the ASRR, 0.5 mg/L TDZ plus different concentration of IAA exhibited relatively high ASRR and 0.5 mg/L TDZ plus 0.3 mg/L IAA showed the highest ASRR of 76.7%. Also the effect of sucrose and sorbitol as carbon source on regeneration was examined. The highest ASRR and the most shoots per explants averaged 94.4% and 3.49 by treatment of 30 mg/L sorbitol, respectably. Sorbitol is considered better carbon source than sucrose for shoot regeneration of 'Whangkeumbae' pear.

Shoot Growth and Fruit Characteristics of 'Soomee' Peach according to Length of Fruit Bearing Branch (결과지 길이에 따른 복숭아 '수미'의 신초 생장 및 과실 특성)

  • We analyzed the correlation among fruit bearing branch (FBB) and shoot and fruit characteristics in order to select the length of FBB suitable for producing high-quality fruits of 'Soomee', a peach tree developed in Korea. The length and diameter of FBB were 26.1 cm and 6.1 mm, respectively, shoot and leaf number per FBB were 3.2 and 38.6, respectively. Of these, the coefficient of variation was very high in the shoots and leaf number. The average weight and soluble solid content (SSC) of fruit were 298.6 g and 12.2 Brix, respectively, and coefficient of variation of the fruit weight was 18.0 %, which was higher than that of SSC. As the FBB of 10-20 cm and 20-30 cm length per tree were 27.1 % and 25.4 %, respectively, the sum of short and middle FBB frequency per tree was more than 50 %. Fruits of 250-350 g and 11.0-13.0 Brix per tree were distributed in 68.6 % and 74.0 %, respectively. As a result of correlation analysis, fruit weight and shoot number were affected by the length of FBB. In particular, length of FBB showed the relation of fruit weight with $y=-0.0482x^2+2.4512x+277.36$. As a result, the length of FBB that can maximize fruit weight was analyzed as 25.4 cm. Therefore, in the filed, the suitable FBB for producing 'Soomee' peach is estimated to be about 20-30 cm.

Influence of Seasonal incidence and Defoliation Degree of Marssonina Blotch on Fruit Quality and Shoot Growth of 'Fuji'/M.9 Apple Tree (갈색무늬병의 시기별 이병 및 낙엽 정도가 '후지'/M.9 사과나무의 과실품질 및 신초생장에 미치는 영향)

  • This study investigated the influence of seasonal incidence and defoliation degree caused by Marssonina blotch (Diplocarpon mali Harada et Sawamura) on shoot growth and fruit quality of 'Fuji'/M.9 apple tree. The occurrence of marssonina blotch in Gunwi region was observed from the mid of July 2009, and percentage of defoliation was about 10% in the end of October. In 2010, that started in the early of June. By the end of September, percentage of defoliation was 20% and it reached 50% in late October. Secondary growth of terminal shoot in 2010 was three times as much as that in 2009. In 2009, the soluble solid contents and the red color (hunter a value) during fruit maturation increased to $13.8^{\circ}Brix$ and 16.2, respectively. In 2010, the soluble solid content during fruit maturation remained in the $12.1-12.6^{\circ}Brix$ range after early October, and the red color began to increase after the end of September when the percentage of defoliation was 20%. As for fruit quality by defoliation degree, the red color decreased when defoliation percentage of bourse shoot at the end of October was more than 30%. Fruit weight and soluble solids also decreased when defoliation percentage was more than 50%. Fruit length and diameter of over 30% defoliation treatments begun to decrease after the middle of August, compared with those of under 30% defoliation treatment. Photosynthetic rate of the leaves that was located at the secondary growth of bourse shoot was similar to that of leaves which was located at the middle of bourse shoot. However, the size and the soluble solid content of fruit were not affected by photosynthesis activities of bourse shoot leaves which was develop in the secondary shoot growth.

Factors Affected on Plant Regeneration of Phyllitis scolopendrium (L.) Newm. In vitro (기내에서 변산일엽의 식물체 재생에 영향을 미치는 요인들)

  • This study was conducted to develop the efficient propagation method of fern Phyllitis scolopendrium using In vitro culture. The influence of the origin of the donor explant sources (rhizome, stipe, three parts of blade) and the homogenization of explants was investigated. Rhizome and stipe explants showed the organogenic capacity among the five explant sources and plant regeneration was promoted by homogenization of culture material. Optimum condition for vigorous and excellent growth of multiple shoots was the half-strength MS medium with 1% sucrose concentration. Generally, addition of $NaH_2PO_4$ to media enhanced shoot multiplication. The highest rate of shoot proliferation was observed on the media containing $5{\mu}M$ NAA. Also, combination of activated charcoal $(0.1{\sim}0.2%)$ and growth regulators to growth medium prevented the formation of multiple bud primordia, 'nodule'-like bud clusters and improved the normal morphogenesis of sporopytes in P. scolopendrium.

Medium Composition Affecting In Vitro Plant Regeneration and Acclimation of Pteris cretica 'Wilsonii' (Pteris cretica 'Wilsonii'의 기내 식물체 재생에 미치는 배지구성물질의 영향 및 기외순화)

  • Adventitious shoots were induced from pinnae, petiole and rhizome in Pteris cretica 'Wilsonii' in order to develop the efficient mass propagation method, using in vitro culture. Only homogenized rhizome segments could regenerate young sporophytes. Efficient regeneration of multiple shoots was obtained on the one-eighth strength MS medium containing 1% sucrose, and $50mg{\cdot}L^{-1}$ $NaH_2PO_4$. To achieve higher rate of regeneration from rhizome segments, rhizome segments were exposed to growth regulators for 2 months and then subcultured on hormone-free medium. The greatest shoot regeneration was obtained by $1{\mu}M$ kinetin with $5{\mu}M$ NAA. BA was effective in formation of GGB (kind of meristems), but they showed low shoot regeneration rate. Plants obtained from present experiments were transplanted to examine good environmental conditions for acclimation. Juvenile plants obtained by the one-eighth strength MS medium showed highest survival rate and vigorous growth at the seedling stage.

Establishment of propagation system for Alstroemeria plants by using hormones and gelling agents (호르몬 및 배지 고형제를 이용한 알스트로메리아 식물체 대량증식 체계 확립)

  • Alstroemeria (Alstroemeriaceae) is one of the most important cut flowers in international market. But, its characteristics such as low multiplication rates, time consuming process and high risk of carrying viral disease, in vitro propagation techniques based on rhizome meristems culture have been developed. In this study, we conducted this experiment to investigate the effect of hormones and gelling agents on the growth for Alstroemeria in vitro tissue culture. The highest number of shoot and root formation were obtained from the growth medium contains BA 1.0 mg/L and NAA 0.1 mg/L. When 0.25% of gelrite was added up to 50% shoots and roots length were observed compared to other gelling agents. Using these results, it is expected to contribute to the establishment of mass production systems in Alstroemeria.

Trends of turfgrass withering and dying on the base of trees in golf courses (골프코스내 수목 근부 지역의 잔디고사 경향)

  • This study was conducted to find out withering and dying trends of turfgrasses on the base of tree, and to investigated how much area of turfgrasses withering and dying were progressed on angle of inclination at golf course. Turfgrasses withering and dying on base of trees on slope was extended more to the down-solpe direction than to the up-solpe direction. This trend was common in all experiment plot. Turfgrasses on the base of trees were withered and dead by competing with tree root fur nutrients and moisture in soil. But the trend of the withering and dying in some experiment plots occurred by soil erosion following turfgrass withering and dying occurring on upside ridge. This trend was generally appeared in region of tree community at upside ridge. The form of turfgrass withering and dying on the base of tree in some experiment plots was similar to that of the tree crown. This withering and dying from occurred at the $11^{th}$ experiment plot in Ji San Golf Course and the $6^{th}$ experiment plot in East valley Golf Course.

Effect of Head Rice Ratio on Rice Palatability (쌀의 완전미 비율 증가가 식미에 미치는 영향)

  • This experiment aimed to estimate the effect of imperfect rice on rice palatability. Rice cultivar, Ilpumbyeo, was cultivated by direct-seeding on flooded paddy surface with 11 kg/10a nitrogen application. Palatability of harvested rice was measured by NIR spectroscope. Brown rice was divided according to their appearance namely, perfect, discolored, green-kerneled, and immature opaque with a composition ratio of $75.7\%,\;11.0\%,\;8.0\%,\;and\;5.3\%$ respectively. When the perfect brown rice was milled, the grain were composed of head, cracked, and white core & belly, at $64.7\%,\;25.3\%\;and\;10.0\%$ respectively. The milled rice of discolored brown rice had similar composition with the perfect rice. The milled green-kerneled vice, on the other hand, had $36\%$ head rice and $64\%$ white core & belly rice. The immature opaque brown rice, when milled, had $25.3\%$ white core & belly and $74.7\%$ damage & opaque rice. With the respect to grain quality, the viscosity of white core at belly rice and damaged & opaque rice was lower than that of head rice. In contrast, their protein content was a little higher than that of head rice. The palatability value of pure imperfect rice was much lower than head rice. The palatability value of damaged & opaque rice was the lowest among the imperfect rices. When mixed with head rice, the damaged & opaque rice impacted on the deterioration of vice palatability. Mixing $1\%$ each of white core at belly rice and damaged h opaque rice decreased the palatability value by $5\%$ as compared with the head rice.
