Glucosinolate Content Varies and Transcriptome Analysis in Different Kale Cultivars (Brassica oleracea var. acephala) Grown in a Vertical Farm (수직농장에서 자란 케일(Brassica oleracea var. acephala) 품종에 따른 글루코시놀레이트 함량의 변화 및 전사체 분석)

  • Kale (Brassica oleracea var. acephala) is one of the most frequently consumed leafy vegetables globally, as it contains numerous nutrients; essential amino acids, phenolics, vitamins, and minerals, and is particularly rich in glucosinolates. However, the differences in the biosynthesis of glucosinolates and related gene expression among kale cultivars has been poorly reported. In this study, we investigated glucosinolates profile and content in three different kale cultivars, including green ('Man-Choo' and 'Mat-Jjang') and red kale ('Red-Curled') cultivars grown in a vertical farm, using transcriptomic and metabolomic analyses. The growth and development of the green kale cultivars were higher than those of the red kale cultivar at 6 weeks after cultivation. High-performance liquid chromatography (HPLC) analysis revealed five glucosinolates in the 'Man-Choo' cultivar, and four glucosinolates in the 'Mat-Jjang' and 'Red-Curled' cultivars. Glucobrassicin was the most predominant glucosinolate followed by gluconastrutiin in all the cultivars. In contrast, other glucosinolates were highly dependent to the genotypes. The highest total glucosinolates was found in the 'Red-Curled' cultivar, which followed by 'Man-Choo' and 'Mat-Jjang'. Based on transcriptome analysis, eight genes were involved in glucosinolate biosynthesis. The overall results suggest that the glucosinolate content and accumulation patterns differ according to the kale cultivar and differential expression of glucosinolate biosynthetic genes.

Morphological Characteristics Based on Phenotypes of Flower and Fruit in a New Variety of Anthoxylum schinifolium (산초나무(Zanthoxylum schinifolium) 신품종의 꽃과 열매에 기반한 형태적 특성 분석)

  • In this study, the morphological characteristics of flower and fruit in five varieties (Hancho 915ho, 930ho, 10ho, 1020ho, 1030ho) from Zanthoxylum schinifolium were investigated as new varieties with different fruiting seasons. Qualitative and quantitative characteristics were investigated, spectively. For quantitative characteristics can be measured numerically, such as size and number, the varieties were identified using ANOVA and Scheffe's multiple range test. In addition, the validity of whether the quantitative characteristics of five varieties can be explained by factor analysis was reviewed. Using UPGMA, the relationship between the five varieties was confirmed by dendrogram. As a result of ANOVA and Scheffe's multiple range test there was significant difference in inflorescence width and female flower length. It had a high contribution to distinguishing varieties, which was similar to the PCA results using factor analysis. As a result of dendrogram using cluster analysis, Hancho 915ho, 1020ho and 1030ho were similar, but there was a distinct difference in the characteristics of different flowering and fruiting periods, confirming that they were different varieties.

  • 영농형 태양광발전은 태양의 일사량을 전기발전과 영농에 공유하는(solar-sharing) 방식이다. 본 연구는 신재생에너지의 활용의 극대화를 위하여 추적식 영농형 태양광발전시스템을 구축하고 시설하부에서 일정 기간 재배중인 작물의 하부 환경과 생육을 조사하여 영농형태양광 하부작물개발을 위한 기초자료를 확보하고자 하였다. 구축한 추적식 영농형 태양광발전시스템은 4열 6단의 24장 모듈(8m × 6m)을 가지며, 발전시설 중심축 기둥 간 중심간격 14m로 단일지주식 스크루 공법으로 순천대학교 부속농장 답작포(순천시 죽평리)에 설치하여 하부 환경과 하부작물의 생육을 조사하였다. 태양광발전시설 하부작물의 생육을 조사하기 위하여 순천 농협육묘장에서 벼(신동진)를 육묘하여 2022년 6월 16일 이앙하였다. 태양광발전시스템 하부 지역을 4방위 방향에 따라 강음영(중심축으로부터 1~3m), 중음영(5m), 약음영(7~9m) 구역으로 설정하여 생육을 조사한 결과, 방위에 따른 초장은 남쪽에서 음영간 차이가 상대적으로 낮게 나타났으며, 1번기 태양광 발전시설에 의하여 음영이 중첩된 2번기 시설의 동쪽에서 대조구 대비 초장이 상대적으로 낮은 경향을 나타내었다. 음영강도에 따른 초장은 대체로 강음영구에서 낮게 나타났으며, 약음영구로 갈수록 높게 나타났다. 엽수는 방위에 따라서, 그리고 음영의 강도에 따른 차이가 초장에 비하여 작게 나타났다. 출수기의 경우 방위별로는 남쪽에서 음영별 차이가 작게 나타났으며, 음영강도에 따라서 차이를 보였다. 또한 태양광시설 하부에 데이터수집장치(Model 1650, Spctrum Technonogies, USA)를 설치하여 음영에 따른 토양전도도, 토양함수량, 토양온도, par light 등 생육환경을 조사, 비교하였다.

Website Monitoring on the Behavior of Consumers for Educational Pet Insects (애완학습곤충 소비자의 행동 모니터링)

  • As the market of educational pet insects is expanding, understanding the consumer needs became more crucial. To achieve the ideal analysis on the market, this research monitored the behavior of consumers. The posting on the blogs of consumers, who have visited insect museums and farms, or have bought insects were collected as data. Moreover, the informational contents, photographs and texts, were analyzed. The results showed that the family-unit visitors with elementary school lower graders were the main type of visitors for their children's education. The visiting areas were concentrated in Seoul and the Metropolitans of Gyeonggi province, and the visits were mostly occurred during their children's vacation period. The analysis of posted photographs showed the visitors' high interest in the hands-on program. According to the texts on visitors' blogs, especially, the largest number of visitors satisfied with the variety of program. It implies the necessity of development in diverse and differentiated hands-on program. Otherwise, the programs available to connect insects to other animals and plants should be introduced to reduce aversion against insects, which was reported as the strongest dissatisfaction. In conclusion, diversification on insect species and development in systematized hands-on program seem to be required for the continuous growth of educational pet insects market.

Effects of Harvesting Time on Seed Yield, Seed Germination and Seedling Growth of Rye (Secale cereale L.) (호밀 수확시기가 종실의 수량과 발아 및 유묘생장에 미치는 영향)

  • To know the optimum harvest time for seed yield and seed quality a local variety of rye 'Paldang-homil'was harvested at seven different harvest times from 25 to 55 day after heading (DAH) at five-day intervals in 1984 ani 1986. Seed development, seed germination and seedling growth were observed. The l000-grain weight increased as harvest time delayed until 50 DAH in both years. Although grain yield tended to increase with delay of harvest time, the yield differences between succeding harvest time was highest between 40 DAH and 45 DAH. Germination rate of seeds harvested before 30 DAH were lower than those after 35 DAH at 20 C, but at 10 and 30 C before 35 DAH were lower after 40 DAH. Plant height and dry weight of seedlings increased with delay of harvest time up to 45 DAH in pot. Heading stages were similar among the seeds harvested 40-55 DAH. Culm length was not different among the harvest times. The optimum harvest time for seed production of rye seems to be 45 DAH (38 days after flowering).

Development of Transgenic Crops and Research Projects for Biotechnology Application (유전자 전환작물 개발 연구 현황과 과제)

  • The main objective of this topic is to establish strategies and to plan biotechnology researches which are related to the agricultural improvements especially focusing on the crop breeding in Korea. From 1960's to 1980's government policy had been emphasized to develope high yielding cultivars for the self sufficient supply of the staple food crops. As a result, considerable increase of rice production has been made with accumulating technology and man's powers. Recently genetically modified crops harboring useful characteristics have been developed using biotechnology and released in the developed countries. National research institutes and private companies have been developed biotechnology researches to establish competitive capabilities, however they have not been successfully used in commercialization. Therefore it is necessary to promote the practical. application by connecting molecular technology with conventional breeding. Proposed research projects are; (1) basic researches including plant genome studies, (2) developing new cultivars through gene transformation, (3) screening and producing antioxidants, secondary metabolite substances and edible vaccines. To set a government policy, both domestic and international research trends were reviewed and possibility of success based on the economic view point were discussed. The intellectual property and preservation of environment play a key role to decide the research priority. It is also necessary for us to make one step system for the distribution of research resources such as microorganisms, genes cloned, plant seeds and research informations for promoting research activities.

Analyzing Self-Introduction Letter of Freshmen at Korea National College of Agricultural and Fisheries by Using Semantic Network Analysis : Based on TF-IDF Analysis (언어네트워크분석을 활용한 한국농수산대학 신입생 자기소개서 분석 - TF-IDF 분석을 기초로 -)

  • Based on the TF-IDF weighted value that evaluates the importance of words that play a key role, the semantic network analysis(SNA) was conducted on the self-introduction letter of freshman at Korea National College of Agriculture and Fisheries(KNCAF) in 2020. The top three words calculated by TF-IDF weights were agriculture, mathematics, study (Q. 1), clubs, plants, friends (Q. 2), friends, clubs, opinions, (Q. 3), mushrooms, insects, and fathers (Q. 4). In the relationship between words, the words with high betweenness centrality are reason, high school, attending (Q. 1), garbage, high school, school (Q. 2), importance, misunderstanding, completion (Q.3), processing, feed, and farmhouse (Q. 4). The words with high degree centrality are high school, inquiry, grades (Q. 1), garbage, cleanup, class time (Q. 2), opinion, meetings, volunteer activities (Q.3), processing, space, and practice (Q. 4). The combination of words with high frequency of simultaneous appearances, that is, high correlation, appeared as 'certification - acquisition', 'problem - solution', 'science - life', and 'misunderstanding - concession'. In cluster analysis, the number of clusters obtained by the height of cluster dendrogram was 2(Q.1), 4(Q.2, 4) and 5(Q. 3). At this time, the cohesion in Cluster was high and the heterogeneity between Clusters was clearly shown.

Analysis of Heating Effect of an Infrared Heating System in a Small Venlo-type Glasshouse (소형 벤로형 유리온실에서 적외선등 난방 시스템의 난방효과 분석)

  • An infrared heating system, installed in a small venlo-type glasshouse ($280m^2$) in Gyeongsang National University, Jinju, Korea, was used to investigate its heating effect with potted Phalaenopsis, Schefflera arboricola 'Hongkong', Ficus elastica 'Variegata', and Rosa hybrida 'Yellow King' as the test plants. Temperature changes in test plants with the system turned 'On' and 'Off' were measured by using an infrared camera and the consumption of electricity by this infrared heating system was measured and analyzed. In potted Phalaenopsis, when the set air temperature of the greenhouse was $18^{\circ}C$, temperature of leaves and the growing medium were $22.8{\sim}27^{\circ}C$ and $21.3{\sim}24.3^{\circ}C$, respectively. In such tall plants as Schefflera arboricola 'Hongkong' and Ficus elastica 'Variegata', the upper part showed the highest temperature of 24.0 and $26.9^{\circ}C$, respectively. From the results of temperature change measurements, the plant temperatures were near or above the set point temperatures with some fluctuations depending on the position or distance from the infrared heating system. When air temperature between night and dawn dropped sharply, plant temperatures were maintained close to the set temperature ($18^{\circ}C$). There was a significant difference between 'On' and 'Off' states of the infrared heating system in average temperatures of root zone and leaf: 21.8 and $17.8^{\circ}C$ with the system 'On' and 20.4 and $15.5^{\circ}C$ with the system 'Off', respectively, in a cut rose Rosa hybrida 'Yellow King'. The heating load was about $24,850{\sim}35,830kcal{\cdot}h^{-1}$, which comes to about 27,000~40,000 won in Korean currency when calculated in terms of the cost of heating by a hot water heating system heated by petroleum. The cost for heating by the infrared heating system was about 35% of that of a hot water heating system. With the infrared heating system, the air temperature during the night was maintained slightly lower than the set point air temperature, probably due to the lack of air tightness of the glasshouse. Therefore, glasshouses with an infrared heating system requires further investigation including the installation space of the heat-emitting units, temperature sensor positions, and convection.

Hazard Analysis for the Application of Good Agricultural Practices(GAP) on Paprika During Cultivation (파프리카의 농산물우수관리제도(GAP)적용을 위한 재배단계의 위해요소 분석)

  • This study established hazards which may cause risk to human at farm during cultivation stage of paprika. Samples of plants (paprika, leaf, stem), cultivation environments (water, soil), personal hygiene (hand, glove, clothes), work utensils (carpet, basket, box) and airborne bacteria were collected from three paprika farms (A, B, C) located in Western Gyeongnam, Korea. The collected samples were assessed for biological (sanitary indications and major foodborne pathogens), chemical (heavy metals, pesticide residues) and physical hazards. In biological hazards, total bacteria and coliform were detected at the levels of 1.9~6.6 and 0.0~4.610g CFU/g, leaf, mL, hand or 100 $cm^2$, while Escherichia coli was not detected in all samples. In major pathogens, only Bacillus cereus were detected at levels of ${\leq}$ 1.5 log CFU/g, mL, hand or 100 $cm^2$, while Staphylococuus aureus, Listeria monocytogenes, E. coli O157 and Salmonella spp. were not detected in all samples. Heavy metal and pesticide residue as chemical hazards were detected at levels below the regulation limit, physical hazard factors, such as insects, pieces of metal and glasses, were also found in paprika farms. Proper management is needed to prevent biological hazards due to cross-contamination while physical and chemical hazards were appropriate GAP criteria.

A Sustainable Operation Plan for School Gardens - Based on a Survey of Elementary School Gardens in Seoul (학교 텃밭의 지속적인 운영방안에 관한 연구 - 서울특별시 초등학교의 학교 텃밭 실태조사를 바탕으로 -)

  • This study surveyed 599 elementary schools in Seoul to provide measures for the quantitative expansion and sustainable operation of environmentally-friendly school garden. Of all schools, 161 schools had formed and were operating school gardens. The total area of school gardens was $166,901m^2$ and the mean area was $131.2m^2$ in elementary, junior high and high schools in Seoul. Meanwhile, the total area of school gardens was $65,493m^2$ and the mean area was $363m^2$ in 161 schools that participated in the survey, indicating $1.15m^2$ per student. Of these schools, 11.8% were operating gardens themselves, while 50.3% were operating gardens that had been newly renovated or environmentally improved by institutional support projects after initially managing gardens themselves. According to the locations of school gardens, mixed-type gardening (a combination of school gardening and container vegetable gardening) accounted for 34.8%, followed by school gardening at 32.9%, container vegetable gardening at 29.2%, and suburb community gardening at 3.1%. Those in charge of garden operations were teachers at 51.6%, comprising the largest percentage. Facilities built when forming the garden included storage facilities for small-scale greenhouses and farming equipment at 26.1%, accounting for the largest percentage. No additional facilities constructed accounted for 21.7%. The greatest difficulty in operating gardens was garden management at 34.2%. The most needed elements for the sustainable operation of gardens were improvement in physical environment and the need for hiring a paid garden, each accounting for 32%. The most important purpose for school gardening was creating educational environments (81.6%). The major source for gaining information on garden management was consultation from acquaintances (67.8%). Schools that utilize plant waste from gardens as natural fertilizers accounted for 45.8% of all schools. Responses to the impact of operating school gardens for educational purpose were positive in all schools as 'very effective' in 63.2% and 'effective' in 36.8%. This study was meaningful in that it intended to identify the current status of the operation of school gardens in elementary schools in Seoul, support the formation of school gardens appropriate for each school with sustainable operation measures, implement a high-quality education program, develop teaching materials, expand job training opportunities for teachers in charge, devise measures to support specialized instructors, and propose the need for a garden management organization.
