First Report of Fusarium Wilt Caused by Fusarium oxysporum on Kohlrabi in Korea (Fusarium oxysporum에 의한 콜라비 시들음병)

  • In April 2014, Fusarium wilt was found on kohlrabi seedlings in Iksan, Korea. Symptoms included wilting of foliage, drying and withering of older leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelium. Macroconidia were falcate to almost straight, thin walled and usually 3-septate. Microconidia were usually formed abundantly in false-heads on short monophialides on the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, $4-11{\times}2.5-5{\mu}m$. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers (internal transcribed spacer rDNA and translation elongation factor $1{\alpha}$), the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch's postulates. To our knowledge, this is the first report on the occurrence of Fusarium oxysporum on kohlrabi in Korea.

  • From 2014 to 2016, Fusarium wilt disease was found on fassionfruit in Iksan and Jeju, Korea. Symptoms included wilting of foliage, drying and withering of leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelia with 35 mm after one week. Macroconidia were falcate to almost straight, thin-walled and usually 2-3 septate. Microconidia were usually formed on monophialides of the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, $3-12{\times}2.5-6{\mu}m$. On the basis of the morphological characteristics and phylogenetic analyses of two molecular markers, internal transcribed spacer rDNA and translation elongation factor $1{\alpha}$, the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch's postulates. To our knowledge, this is the first report on the occurrence of F. oxysporum on fassionfruit in Korea.

Surveys on Disease Occurrence in Major Horticultural Crops in Kangwon Alpine Areas (강원도 고랭지 주요 원예작물의 병해 발생 상황)

  • These surveys were conducted to check the occurrence of disease in various horticultural crops in alpine areas, especially Daekwallyong areas, Pyongchang-Gun, and Hyeongseong-Gun in Kangwon province. TuMV on Chinese cabbage was one of the most serious diseases, especially in 1994 and 1997. The incidence of soft rot and clubroot has been increased gradually. Brittle root rot on Chinese cabbage was significantly decreased. Soft rot, gray mold, downy mildew, powdery mildew, bottom rot and Alternaria leaf spot were the common diseases on most vegetable crops. Gray mold (Botrytis cinerea) on celery, cercospora leaf spot (Cercospora sp.) on melon, powdery mildew (Erysiphe cichoracearum) on lettuce, and clubroot (Plasmodiophora brassicae) on parsley are newly found in Korea. The most common and predominant diseases were viruses, especially CMV, TMV, TuMV, BBMV, and gray mold, wilts, and cercospora leaf spot on many flowers in alpine areas. Fusarium oxysporum f. sp. eustomae causing wilting on lisianthus (Eustoma grandiflorum), Turnip mosaic virus causing mosaic and color breaking on stock, Cercospora spp. causing cercospora leaf spot on various wild lily, Cladosporum echinulatum causing leaf spot on carnation, and phytoplasma causing witches' broom on statice (Limonium sinuatum) and blazing star (Liatris spp.) were newly found during these surveys in Korea.

Effect of Chitin Compost on Biological control of Fusarium wilt in Tomato Field (키틴퇴비를 이용한 토마토의 Fusarium 시들음병의 생물학적 제어)

  • Biological control by chitinolytic microorganisms is being evaluated as management options for soilborne diseases. Forty kilograms of chitin compost (CTC) and control compost (CC) were amended on tomato plots ($15m{\times}0.5m$) 7 d before transplanting to evaluate enzymatic activities and the control of Fusarium wilt. Samples were taken on day 1, 3, 5, and 7, the day 1 corresponded to the 66 d after transplanting, the day on which the initial wilting symptoms occurred in plants of CC treated plots. The chitinase activity in soil of CTC was always higher compared to the control. Pathogenesis related (PR) protein (chitinase, ${\beta}$-1, 3-glucanase and peroxidase) activities in tomato roots in CC increased every day and showed marked differences compared to CTC. Wilting symptoms (96 d after transplanting) were reduced by 25% in CTC compared to the control. Protection of tomato plant may be correlated with the high levels of soil enzyme activities resulting from the chitin compost.

Occurrence of Fusarium Wilt on Cruciferous Vegetable Crops and Pathogenic Differentiation of the Causal Fungus (십자화과 채소작물에서의 후사리움 시들음병 발생과 그 원인균의 병원성 분화)

  • Occurrence of Fusarium wilt was surveyed in fields of cruciferous vegetable crops in Korea from 1996 to 1998. The disease severely occurred up to 40% in fields of Chinese cabbage and radish but slightly in Fields of cabbage. A total of 123 isolates was obtained from roots of the diseased plants and identified as Fusarium oxysporum based on the morphological and cultural characteristics. Pathogenicity of nine isolates selected from the isolates was tested by artificial inoculation to the hosts. All the isolates had similar virulence on Chinese cabbage and cabbage, although there were some differences in virulence on cultivars tested among the isolates. The isolates from radish were more virulent to radish than those from Chinese cabbage and cabbage. All isolates from the crucifers were not virulent to eight species of vegetable crops except the crucifers. The results of pathogenicity tests showed that the pathotype of Chinese cabbage-infecting isolates was identical to that of cabbage-infecting isolates, but somewhat different from that of radish-infecting isolates.

Investigation of Fungi in Pesticide Fumigated Oak Wilt-Diseased Logs (훈증방제 처리한 참나무시들음병 감염목의 사상균 조사)

  • Korean oak wilt disease caused by Raffaelea quercus-mongolicae is vectored by the ambrosia beetle Platypus koryoensis. To prevent the spread of the disease, the beetle infested oak tree had been cut into logs, covered with plastic vinyl, fumigated with a pesticide, and stored for three years on the site where the tree was cut. This study was carried out to get information on the fungi colonizing the fumigated oak wood. Wood disk samples collected from the fumigated oak logs at two locations in the Taejo Mountain, Cheonan city, were used for fungal isolation. A total of 99 filamentous fungal isolates were obtained from the wood disk samples. Hypocrea spp., Trichoderma spp. and Penicillium spp. were identified based on morphological characteristics and nucleotide sequence analysis of translation elongation factor 1-alpha gene and ITS rDNA region. Trichoderma was the major fungal group. R. quercus-mongolicae, and P. koryoensis were not detected from the fumigated oak wood. Our work provided evidence that after three years of storage, the fumigated oak wilt-diseased logs should be no longer harmful source of oak wilt disease transmission.

Selection and Antagonistic Mechanism of Bacillus thuringiensis BK4 against Fusarium Wilt Disease of Tomato (토마토시들음병의 생물학적 방제를 위한 토착길항세균 Bacillus thuringiensis BK4의 선발과 길항기작)

  • In oder to select an antifungal substance-producing antagonistic bacterium against Fusarium oxysporum casuing fusarium wilt on tomato, strain BK4 was isolated from local soil of Gyeoungbuk and was identified as Bacillus thuringiensis by 16s rDNA analysis, biochemical test, and Mcirolog TM 3.0 System. The antibiotic of B. thuringiensis BK 4 was highly produced at $30^{\circ}C$ in nutrient broth (pH 9.0). The crude antibiotic was even stable at $121^{\circ}C$ and more stable at slight alkalic condition than acid condition. It was also remained $50{\%}$ activity at pH 3.0. B. thuringiensis BK4 showed the inhibition of spore germination and the biocontrol ability against F. oxysporum causing fusarium wilt of tomato in vivo test. According to these results, B. thuringiensis BK4 was enough to use with a microbial agent for biocontrol against fusarium wilt.

  • 고추$\cdot$참깨등 경제작물에 발생하는 병해는 수십여종이 있으나 그중 국내에서 흔히 발생하며 피해가 큰 병해는 곰팡이병으로 노균병, 잿빛곰팡이병, 탄저병, 균핵병, 시들음병, 역병 등 6종, 세균병으로는 무름병, 풋마름병, 세균성점무늬병등 3종, 그리고 바이러스에 의한 병을 들수 있다. 이 병해들은 그 종류별로 병징, 병발생 양상, 전염방법 등에 특징이 있기 때문에 기생하고 있는 작물이 다를지라도 병의 구별이 가능하고 동일종류에서는 병의 방제방법도 비슷하다. 최근 문제되고 있는 병해를 중심으로 진단법과 방제법을 알아본다.
