In this study, we conducted the hot box tests to compare the changes in thermal insulation for the four types of multi-layer thermal screens by the used period after collecting them from the greenhouses in the field when they were replaced at the end of their usage. The main materials for these four types of multi-layer thermal screens were matt georgette, non-woven fabrics, polyethylene (PE) foam, chemical cotton, etc. These materials were differently combined for each multi-layer thermal screen. We built specimens ($70{\times}70cm$) for each of these multi-layer thermal screens and measured the temperature descending rate, heat transmission coefficient, and thermal resistance for each specimen through the hot box tests. With regard to the material combinations of multi-layer thermal screens, thermal insulation can be increased by applying a multi-layered PE foam. However, it is considered that the multi-layered PE foam significantly less contributes to heat-retaining than chemical wool that forms an air-insulating layer inside multi-layer thermal screens. For the suitable heat-retaining performance of multi-layer thermal screens, basically, materials with the function of forming an air-insulating layer such as chemical cotton should be contained in multi-layer thermal screens. The temperature descending rate, heat transmission coefficient, and thermal resistance of multi-layer thermal screens were appropriately measured through the hot box tests designed in this study. However, in this study, we took into consideration only the four kinds of multi-layer thermal screens due to difficulties in collecting used multi-layer thermal screens. This is the results obtained with relatively few examples and it is the limit of this study. In the future, more cases should be investigated and supplemented through related research.
The archaeal clusters of orthologous genes (arCOG) algorithm, which identifies common genes among archaebacterial genomes, was used to identify conservative genes among 168 archaebacterial strains. The numbers of conserved orthologs were 14, 10, 9, and 8 arCOGs in 168, 167, 166, and 165 strains, respectively. Among 41 conserved arCOGs, 13 were related to function J (translation, ribosomal structure, and biogenesis), and 10 were related to function L (replication, recombination, and repair). Among the 14 conserved arCOGs in all 168 strains, 6 arCOGs of tRNA synthetase comprised the highest proportion. Of the remaining 8 arCOGs, 2 are involved in reactions with ribosomes, 2 for tRNA synthesis, 2 for DNA replication, and 2 for transcription. These results showed the importance of protein expression in archaea. For the classes or orders having 3 or more members, genomic analysis was performed by averaging the distance values of the conservative arCOGs. Classes Archaeoglobi and Thermoplasmata of the phylum Euryarchaeota showed the lowest and the highest average of distance value, respectively. This study can provides data necessary for basic scientific research and the development of antibacterial agents and tumor control.
The YrdC superfamily is a group of proteins that are highly conserved in almost all organisms sequenced so far. YrdC in Escherichia coli was suggested to be involved in ribosome biogenesis, translation termination, cold adaptation, and threonylcarbamoyl adenosine formation in tRNA. In this study, to unambiguously demonstrate that yrdC is essential in E. coli, we constructed two yrdC mutant strains of E. coli and examined their phenotypes. In the temperature-sensitive yrdC mutant strain, cell growth stopped almost immediately under nonpermissive conditions and it appeared to accumulate 16S ribosomal RNA precursors without significant accumulation of 30S ribosomal subunits. We also cloned yeast and human homologs and demonstrated that they complement the E. coli yrdC-deletion strain. By mutational study, we demonstrated that the concave surface in the middle of the YrdC protein plays an important role in E. coli, yeast, and human versions. By comparison of two yrdC-deletion strains, we also unambiguously demonstrated that yrdC is essential for viability in E. coli and that the functions of its yeast and human homologs overlap with that of E. coli YrdC.
Identification of blood cells and their physological functions in the cultured scallop, Patinopecten yessoensis collected from Abashiri Bay, Hokkaido, Japan were studied by electron microscopic structure and histological observations. The physiological function of each blood cell type was studied on the basis of its cytological structure, the lysosome in the blood cell and its phagocytosis. The blood cells were classified as Type I, Type II and Type III. The morphological characteristics of each blood cell type are as follows ; Type I : The cell is oval shaped and its cytoplasm contains comparatively low electron dense materials. The oval nucleus is sometimes ramified into two nuclei. Lumps of tubular smooth endoplasmic reticula and vacuoles are distributed near the nucleus. Type II : The cell appears long and oval shaped, and its cytoplasm contains high electron dense materials. The oval nucleus does not ramify, and large numbers of sac-like smooth endoplasmic reticula and free ribosomes are developed around the nucleus. No vacuoles exist in the cytoplasm Type 1II : The cell is round in shape and the electron density of the cytoplasm is the highest among the three types of cells because of large quantities of roughsurfaced endoplasmic reticula and no vacuoles. Particularly, the nucleus reveals a wheel-like shape owing to lumps of tuberous chromatin. The cells of Type I and II seem to have the role of carrying out phagocytosis on either foreign materials such as bacteria or endogenous old cells and the transport of nutritive materials. The type III cell, which has not been found in any bivalve species of non Pectinidae, may be said to have the function of production and the secretion of protein related to some humoral defense materials.

This study was carried out to determine effects of bamboo species on the distribution of nutrients in aboveground biomass of Phyllostachys bambusoides and Phyllostachys nigra var. nenosis, Damyanggun, Korea. The study site was established around 40-year-ago to produce bamboo culm and edible shoot production. Total 28 bamboos (14 P. bambusoides and 14 P. nigra var. nenosis) were cut to measure nutrient concentration of each bamboo component, such as culm, branches and leaf. Magnesium concentration in each bamboo component was significantly higher in the P. bambusoides than in the P. nigra var. nenosis. Nutrient concentrations except for calcium were significantly higher in the current-year-old bamboos than in the > 1-year-old bamboos. The nutrient concentration in leaf was generally highest in carbon, followed by potassium or nitrogen, phosphorus, calcium or magnesium. Total nutrient content in each bamboo component was significantly higher in the P. bambusoides than in the P. nigra var. nenosis. The nutrient content of bamboo biomass was the highest in carbon, followed by potassium, nitrogen, phosphorus, magnesium and calcium. The results indicate that P. bambusoides uptakes more nutrients compared with P. nigra var. nenosis during growth development.
This study was accomplished to compare energy saving effects of several heat insulation materials in greenhouse and to develop new automatic opening and closing equipment which is suitable to the most effective heat insulation material. To find out more effective heat insulation material, the magnitude of heat transfer occurred through aluminum screen (ALS), non-woven fabric (NWF), double-layer aluminum screen with chemical cotton sheet (DAL), and multi-layer fabric screen material quilted with non-woven fabric, chemical cotton, poly foam, and polypropylene (MLF) were compared relatively. The results showed that the relative magnitude of heat transfer occurred through MLF was lower than DAL and ALS by 23.3% and 43.0% respectively. MLF screen material was the most effective compared with other heat insulation materials. But because of thickness, there was a need of new mechanism for automatic operation in greenhouse. Accordingly, new screen system using MLF-thick but profitable for keeping warm in greenhouse-was developed. Opening & closing equipment was designed to roll MLF with pipe axis during opening process and pull MLF with string during closing process with electric motors, clutches, drums, and so on. In hot pepper cultivation and energy saving test during winter time, the early stage yield of pepper under MLF screen system was higher than NWF by 27%, and gasoline consumption of MLF screen system was lower than NWF by 46%.
$H^+-ATPase$ located on plasma and vacuolar membranes play major roles in various cellular physiological processes. In order to investigate the physiological roles of $H^+-ATPase$, microsomes were prepared from tomato roots and the effects of various anions were measured on the activities of $H^+-ATPase$. $H^+-ATPase$ was inhibited by various anions. Citrate and phosphate were chosen to investigate detailed inhibitory mechanisms on $H^+-ATPase$ since they showed different levels of inhibition. Inhibitory effect of citrate was observed at the concentrations above 3 mM. When 20 mM citrate was added, the ATPase activity was decreased by 50-60%. However, the inhibitory effect of citrate was decreased by increasing the concentration of$Mg^{2+}$ The citrate-induced inhibited activity was recovered by the addition of $Mg^{2+}$ Addition of 7 mM $Mg^{2+}$ completely removed the inhibitory effect of citrate and the activity recovered to the level of the control experiment. These results imply that citrate chelates $Mg^{2+}$ and thus inhibits $H^+-ATPase$. Meanwhile, the inhibitory effect of phosphate was observed at the concentration above 3 mM and the activity was decreased by 50% in the presence of 30 mM phosphate. Further addition of $Mg^{2+}$ showed no recovery on the activity. These results imply that the inhibitory effect of phosphate is not dependent upon the concentration of $Mg^{2+}$.
Dandelion has been frequently used as a remedy for women's disease, inflammatory diseases and disorders of the liver and gallbladder. Dandelion extracts water extract, an herbal medication, may have an effect on the activity of hepatic antioxidant enzymes in diabetic rat. This study aims demonstrate the effect of dandelion extracts, one of the natural chelator, on the biochemical and enzyme activity changes in the mouse liver caused by $HgCl_2$. Mice approximately 30 gm in weight were grouped into the control, mercury chloride-treated, and the dandelion extracts-treated after mercury chloride groups. $HgCl_2$ (5 mg/kg) and dandelion extracts (3 g/kg) were delivered orally. Serum AST and ALT were measured, enzyme activity of liver were examined by spectrophotometer and ultrastructural alteration of liver were examined by light and electron microscopy. Dandelion extracts were decreased the increase of serum AST and ALT level induced by mercury. The catalase activity was decreased in the dandelion extracts group. The activity of SOD was dereased, but did not show significant differences. Mercury chloride-treated hepatic cell were irregular nucleus, enlarged and reduced number of mitochodria, enlarged rough endoplasmic reticulum, loss of ribosomes. Cells treated with dandelion extracts were similar to those of the control group. In conclusion, dandelion extracts may protect the mercury-induced toxicity on Liver.
In order to investigate the mechanism of growth inhibition by salt stress, lettuces were grown hydroponically in three different nutrient solutions, normal and 30 mM or 50 mM $KNO_3$-added nutrient solutions, and the electrical conductivities of these solutions were 1.0, 4.5, and 6.5 dS/m, respectively. The activities of plasma and vacuolar $H^+$-ATPases in the root tissue of lettuce were measured by specific inhibitors, 100 ${\mu}M$ vanadate and 50 mM $NO_3^-$, respectively. Microsomal ATPase activity of lettuce grown in the normal nutrient solution was $356\pm1.5$ nmol/min/mg protein. When lettuces were grown in 30 mM and 50 mM $KNO_3$-added nutrient solutions, total activities of microsomal ATPases were increased by 1.6 and 1.9 times, respectively, and the increases were mainly mediated by vacuolar $H^+$-ATPase. These results show that lettuces adapt themselves to salt-stressed condition by increasing the activities of $H^+$-ATPases. Effects of various heavy metal ions were investigated on the microsomal ATPases and various metal ions at 100 $\mu M$ inhibited the activities by 10$\sim$25%. $Cu^{2+}$ showed the highest inhibitory effect on the vacuolar $H^+$-ATPase. These results suggest that lettuce increases the activities of root ATPases, specially that of vacuolar $H^+$-ATPase, in salt-stressed growth conditions and $Cu^{2+}$ could be a useful tool to control the activity of vacuolar $H^+$-ATPase.
The purpose of this study was to evaluate the effect the lead toxicity of chitosan on the rats. A healthy male of Wistar rat was used for experiment. The lead acetate (30 mg/kg, Pb) was injected two times per a week into the abdomen. 0.1% chitosan solution (1 mg/ml) was supplied by feeding ad libitum. After 4 or 8 weeks, we was observed kidney by electron microscope. The experimental groups were divided into two groups. Group 1 was control group which was not treated with chitosan solution after contaminatation of lead acetate. Group 2 was experimental group which was treated with chitosan solution after contamination of lead acetate. All groups were used to 10 rats. The results were as follow: On the proximal convoluted tubules of the group with only Pb, the number of microvillis were decreased and the number of lysosomes were increased on the cytoplasm. The mitochondria and rough endoplasmic reticulum were extended and ribosomes dropped from the rough endoplasmic reticulum. On the proximal convoluted tubules of the group with Pb-chitosan, the shapes and number of microvilli were not changed and the number of lysosomes were decreased. The shapes of mitochondria and endoplasmic reticulum observed not changes. We concluded that chitosan has effect on reducing toxicity of lead in the rat kidney.
