This paper describes fabrication of a micro cell counter integrated with an oxygen micropump and counting experiment with Sephadex G-25 beads ($70{\sim}100\;{\mu}m$). The pumping part consisted of a microheater, catalyst (manganese dioxide) enveloped with paraffin, hydrogen peroxide, and microchannel, and the counting part consisted of collimated light, a microwindow, and a phototransistor including an external circuit. The micropump generated oxygen gas by decomposing hydrogen peroxide with manganese dioxide, which was initiated by melting the paraffin with the microheater, and pumped beads in the microchannel. When the beads passed the microwindow, they shaded the collimated light and changed the illumination on the phototransistor, which caused the current variation in the circuit. The signals, according to the bead size, reached up to 22 mV with noise level of 2 mV during 50 seconds and the numbers of peaks were analyzed by magnitude.

We present a novel flow-rate independent cell counter using a fixed control volume between double electrical sensing zones. The previous device based on the single electrical cell sensing in a given flow-rate requires an accurate fluid volume measurement or precision flow rate control. The present cell counter, however, offers the flow-rate independent method for the cell concentration measurement with counting cells in a fixed control volume of $22.9{\pm}0.98{\mu}{\ell}$. In the experimental study, using the RBC (Red Blood Cell), we have compared the measured RBC concentrations from the fabricated devices with those from Hemacytometer. The previous and present devices show the maximum errors of $20.3\%\;and\;16.1\%$, which are in the measurement error range of Hemacytometer (about $20\%$). The present device also shows the flow-rate independent performance at the constant flow-rates ($5{\mu}{\ell}/min$ and $10{\mu}{\ell}/min$) and the varying flow-rate (4, 2, and $4{\mu}{\ell}/min$). Therefore, we demonstrate that the present cell counter is a simple and automated method for the cell concentration measurement without requiring an accurate fluid measurement and precision flow-rate control.

세포막지질의 산화는 심각한 세포막의 기능저하를 유발하고 심하면 세포를 죽음에 이르게하여 생물학적으로 중요한 지표이다. 세포막지질의 산화는 간접적인 화학적 방법으로 측정하거나, 지질을 추출해내어 질량분석기나 핵자기공명분광기 같은 물리적 방법으로 분석한다. 우리는 이온유도 이차전자 방출계수(${\gamma}$) 변화를 측정하여 세포막지질의 산화를 지질추출 없이 측정할 수 있는지 조사해 보았다. 세포막분리가 쉬운 적혈구를 모델세포로 사용하였고, 다양한 라디칼을 발생시키는 대기압 공기 DBD플라즈마 장치를 이용하였다. 적혈구를 플라즈마에 노출하는 시간으로 산화의 정도에 차이를 만들어 측정값과 비교하였다. ${\gamma}$값은 Auger의 중화이론에 바탕을 둔 이온유도 이차전자 방출빔(${\gamma}$-FIB)장비를 이용하여 측정하였다. 측정결과 적혈구가 산화됨에 따라서 ${\gamma}$값이 증가함을 볼 수 있었고, 동시에 workfunction값이 변화함을 보았으며, 그 결과를 화학적 방법과 비교해 보았다.
River and stream are the important water supply source in our lives. Eutrophication causes excessive green algae growth including microcystis, which makes harmful to ecosystem and human health. Therefore, the water purification process to remove green algae is essential. In Korea, green algae alarm system exists depending on the concentration of green algae cells in river or stream. To maintain the growth amount under control, green algae monitoring system is being used. However, the unmanned, small and automatic monitoring system would be preferable. In this study, we developed the 3D printed device to measure the concentration of green algae cell using microfluidic droplet generator and deep learning. Deep learning network was trained by using transfer learning through pre-trained deep learning network. This newly developed microfluidic cell counter has sufficient accuracy to be possibly applicable to green algae alarm system.
The effect of microsparged aeration in mammalian cell bioreactor on the oxygen transfer rate and cell viability was studied. The microspargers with differ- ent micron-sized pores were used to supply oxygen to the medium. The oxygen transfer coefficients (k$_{L}$a) measured in the bioreactor were markedly increased, which is due to the increase of the contacting area between air bubbles and liquid medium when the pore size of microsparger decreases. When the impellers of two different types (square-pitch marine impeller and $45^{\circ}$ pitched flat blade impeller) were used for agitation, the k$_{L}$a values were slightly higher with the marine impeller than with the blade impeller. The detrimental effect of direct gas sparging with microsparger on mammalian cells was investigated in bubble columns with various air flow rates and different pore sized microspargers. The first-order cell death rate constant ($k_{d}$ /7) was shown to be directly proportional to the air flow rate and inversely proportional to the pore size. During the cultivation of hybridoma cells using microsparger with the pore size of $0.57\mu$m in the mammalian cell culture bioreactor, the continuous sparging caused the cell death and suppressed the cell growth. However, cells grew normally and cell viability was maintained above 90% in the logarithmic phase when the air was intermittently sparked in order to maintain the dissolved oxygen level above 20%.
In this paper, a new micro polymer chip platform and protocol were developed for rare cell sample preparation. The proposed platform and protocol overcome the current limitation of the dilution method which is based on statistics and the FACS method which expensive and requires fluorescence staining. It allows collecting exact number of target cells simply and selectively because the cells are visually confirmed during the collecting process. The collected cells can be transported or spiked into a desired locations, such as a microchamber, without cell loss. This research may applicable not only to a rare cell sample preparation for Lab on a Chip cancer diagnosis, but also to a single/double/multiple cell sample preparation for a cell analysis field. To verify this platform and protocol, five human breast cancer cells (MCF-7) were collected and transported into a hemocytometer chamber.
Purpose: The purpose of this study was to evaluate the relationship between radiation-induced activation of DNA repair genes and radiation induced apoptosis in A431 cell line. Materials and Methods: Five and 25 Gys of gamma radiation were given to A431 cells by a Cs-137 cell irradiator. Apoptosis was evaluated by flow cytometry using annexin V-fluorescein isothiocyanate and propidium iodide staining. The expression of DNA repair genes was evaluated by both Northern and Western blot analyses. Results: The number of apoptotic cells increased with the increased radiation dose. It increased most significantly at 12 hours after irradiation. Expression of p53, p21, and hRAD50 reached the highest level at 12 hours after 5 Gy irradiation. In response to 25 Gy irradiation, hRAD50 and p21 were expressed maximally at 12 hours, but p53 and GADD45 genes showed the highest expression level after 12 hours. Conclusion: Induction of apoptosis and DNA repair by ionizing radiation were closely correlated. The peak time of inducing apoptosis and DNA repair was 12 hours in this study model. hRAD50, a recently discovered DNA repair gene, was also associated with radiation-induced apoptosis.

Recovery from potentially lethal damage (PLDR) after irradiation was studied in plateau-phase culture of Vero cells in vitro. Unfed plateau-phase cells were irradiated with dose of 1 to 9Gy using Cs-137 irradiator. Cells then were incubated again and left in situ for 0, 1, 2, 3, 4, 5, 6, and 24 hours and then were trypsinized explanted, and subcultured in fresh RPMI-1640 media containing $0.33\%$ agar. Cell survival was measured by colony forming ability. An adequate number of heavily irradiated Vero cells were added as feeder cells to make the total cell number constant in every culture dish. As the postirradiation in situ incubation time increased, surviving fraction increased by PLDR. The rate of PLDR was so rapid that increased surviving fraction reached saturation level at 2 to 4 hours after in situ incubation. As the radiation dose increased, the rate of PLDR fastened and the magnitude of increased surviving fraction at saturation level by PLOR also increased. In analysis of cell survival curve fitted to the linear-quadratic model, the linear inactivation coefficient $(\alpha)$ decreased largely and reached nearly to zero but the quadratic inactivation coefficient $(\beta)$ increased minimally by increment of postirradiation in situ incubation time. So PLDR mainly affected the damage expressed as $\alpha$, In the multitarget model, significant change was not obtained in $D_0\;but\;in D_q$. Therefore, shoulder region in cell survival curve was mainly affected by PLDR and terminal slope was not influenced at all. And dose-modifying factor by PLDR was relatively higher in shoulder region, that is, in low dose area below 3 Gy.

본 고에서는 한국통신(Korea Telecom) IMT-2000 시험시스템(이하: Trial system 라고 함) 단말기용 전력증폭단으로 적용하기 위한 다단구동증폭기 및 전력증폭기를 GaAs MMIC로 설계 구현하는 기술을 제시하였다. 설계된 구동증폭기는 3단으로구성하여 RF(Radia Frequency) 송신신호(1955$\pm$70MHz)대역에서 2단 (중간단)의 이득 조정범위가 40 dB이상이 될 수 있도록 능동부품인 MESFET를 Cascade 형으로 구성하고 MESFET의 게이트(gate)에 조정전압을 인가하는 증폭기를 설계하여 GaAs MMIC화 1 침(크기4$\times$5 mm)으로 제작하였다. 아울러, 본 논문에서는 제시한 구동증폭기는 동작주파수 대역폭 범위기 3.5배이고 출력전력은 15dBmm 이며, 출력전력이득이 25~27dB이고 반사계수는 -15~20dB이며 이득평탄도 3dB(동작주파수 대역폭내)로써 Trial system용 단말기의 최종단인 전력증폭단의 출력단 특성을 효과적으로 나타내었다. 그리고, 전력 증폭기는 2개의 입력단에서 출력되는 신호를 분배하는 전력분배기와 병렬구조인 4개의 증폭단에서 출력되는 출력신호를 외부에서 접속하는 전력결합기를 접소하여 구성하였으며 RF(Radio Frequency) 주파수(1955 $\pm$70NHz)에서 대역폭을 4배로 설계하여 광대역인 대역폭을 구현하였고 출력전력은 570mW이며, 출력부가효율(PAE; Power Added Efficency)가 -15$\pm$20dB이고, 이득 평탄도(Gain flatness)는 동작주파수 대역내에서 0.5dB이며 입출력 전압정재파비(Input & Output VSWR)가 13이하인 고출력 전력증포기를 GaAs MMIC화 1칩 (크기; 3$\times$4mm)으로 제작하였다.의 다양성이나 편리성으로 변화하는 것이 국적을 바꾸는 것보다 어려운 시 대가 멀지 않은 미래에 도래할 것이다. 신세기 통신 과 SK 텔레콤에는 현재 1,300만명이 넘 는 고객이 있으며. 이들 고객은 어 이상 음성통화 중심의 이동전화 고객이 아니라 신세기 통신과 SK텔레콤이 함께 구축해 나갈 거대란 무선 네트워크 사회에서 정보화 시대를 살아 갈 회원들이다. Purpose: Radiation adaptive response in human peripheral lymphocytes and mouse bone marrow cells was investigated using both metaphase analysis and micronucleus assay. We assessed the correlation between both tests. Materials and Methods: Two groups of the human peripheral lymphocytes and mouse bone marrow cells were exposed to low dose (conditioning dose, 0,18 Gy) or high dose (challenging dose, 2 Gy) ${\gamma}$-rays. The other 4 groups were exposed to low dose followed by high dose after several time intervals (4, 7, 12, and 24 hours, respectively). The frequencies of chromosomal aberrations in metaphase analysis and micronuclei in micronucleus assay were counted. Results: Chromosomal aberrations and micronuclei of preexposed group were lower than those of the group only exposed to high dose radiation. Maximal reduction in frequencies of chromosomal aberrations were observed in the group to which challenging dose was given at 7 hour after a conditioning dose (p<0.001). Metaphase analysis and micronucleus assay revealed very good correlation in both human lymphocytes and mouse bone marrow cells (r=0.98, p<0.001 ; r=0.99, p=0.001, respectively). Conclusion: Radiation adaptive response could be induced by low dose irradiation in both human lymphocytes and mouse bone marrow cells. There was a significant correlation between metaphase analysis and micronucleus assay.
