Considerable effort has been directed toward the use of silk fibroin as a biotechnological material in biomedical applications on account of its excellent biodegradability, biocompatibility, and unique mechanical properties. For use in tissue engineering, it is very important to design and control the pore architecture of polymeric scaffolds, which provide the vital framework for seeded cells to organize into functioning tissue. In the present study, a silk fibroin scaffold with controlled interconnectivity and pore size was prepared using an electrospinning method with poly(ethylene oxide).
Poly(lactide-co-glycolide)(PLGA) and hyaluronic acid (HA) has been widely used as biocompatible scaffold materials to regenerate tissue. In this present study, we fabricated microporous PLGA and HA loaded PLGA scaffolds by a emusion freeze-drying method. In order to confirm that the release profile of cytokine or water-soluble drugs, we manufactured the granulocyte macrophage colony stimulating factor(GM-CSF) loaded PLGA and HA-PLGA scaffold. All scaffolds were characterized using scanning electron microscope(SEM), mercury porosimeter and wettability measurement. Cell proliferation and viability were assessed by a 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide (MTT) test. The porosity of HA-PLGA scaffold was greater than 95% with the total pore area of $261\;m^2/g$. The HA-FLGA scaffold exhibited well interconnected pores to allow greater cell adhesion and prolixferation. It was proven by higher cell viability in the HA-PLGA scaffold than PLGA alone. This may be due to the enhanced natural properties and higher water retention capacity of HA.
PLLA scaffold loaded with gentamicin sulfate (GS) was prepared by emulsion freeze-drying method for the prevention of infection and the improvement of wettability. i.e., the cell- and tissue-compatibility. GS-loaded PLLA scaffolds were characterized by scanning electron microscopy (SEM), mercury porosimetry and blue dye intrusion, and the GS release pattern was analyzed by high performance liquid chromatography (HPLC). GS-loaded PLLA scaffolds with porosity above 50%, medium pore size ranging from 30 to 57 ${\mu}{\textrm}{m}$ (with larger pore diameters greater than 150 ${\mu}{\textrm}{m}$), and specific pore area in the range of 35 to 75($m^2$ /g )were manufactured by varying processing parameter as GS concentration. It was observed that GS-loaded PLLA scaffolds were highly porous with good interconnections between pores for allowing cell adhesion and growth. These scaffolds may be applicable for scaffold as structures that facilitate either tissue regeneration or repair during reconstructive operations.

Tissue engineering techniques require the use of a porous biodegradable/bioresorbable scaffold, which server as a three-dimensional template for initial cell attachment and subsequent tissue formation in both in vitro and in vivo. Small intestinal submucosa (SIS) has been investigated as a source of collagenous tissue with the potential to be used as biomaterials because of its inherent strength and biocompatibility. SIS-loaded poly(L-lactide-co-glicolide)(PLGA) scaffolds were prepared by solvent casting/particle leaching. Characterizations of SIS/PLGA scaffold were carried out by SEM, mercury porosimeter, and so on. Muscle-derived stem cells can be differentiated in culture into osteoblasts, chondrocytes, and even myoblasts by the controlling the culture environment. Cellular viability and proliferation were assayed by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyltetrazolium-bromide(MTT) test. Osteogenic differential cells were analyzed by alkaline phosphatase(ALP) activity. SIS/PLGA scaffolds were implanted into the back of athymic nude mouse to observe the effect of SIS on the osteoinduction compared with controlled PLGA scaffolds. Thin sections were cut from paraffin embedded tissues and histological sections were conducted hematoxylin and eosin (H&E), Trichrome, and von Kossa. We observed that bone formatioin of SIS/PLGA hybrid scaffold as natural/synthetic scaffold was better thean that of only PLGA scaffold. It canb be explained that SIS contains various kinds of bioactive molecules for osteoinduction.

최근 골손상이 있을 경우 골 형성을 유도하고 기능을 부여하여 단순한 골조직의 대체를 위한 지지체가 아닌 한층 더 나아간 지지체의 연구가 활발히 진행되고 있다. 뼈 형성 억제 인자를 억제하거나 촉진인자를 첨가하여 뼈의 형성이 증가시키고, 뼈 형성과정에 관여하는 신호체계를 유도하는 어떤 물질을 첨가하여 뼈의 형성을 증가시킬 수 있다. 줄기세포는 다양한 세포로 분화할 수 있는 능력이 있는데 그 과정에서 여러 가지 signal이 관여한다. 그 중 wnt signaling은 줄기 세포가 분화하는 과정뿐만 아니라 세포의 사멸, 이동에 있어서도 매우 중요한 역할을 하며, 줄기세포의 운명 결정에 영향을 미친다고 알려져 있다. Silicon은 조골세포의 부착과 증식, 세포의 활성을 증가시키며 뼈의 형성과정과 석회화 과정에서 중요한 역할을 한다. 또한 BMP-2, collagen 등과 같은 유전자의 발현을 증가시킨다. 따라서 본 연구에서는 Silicon이 조골세포로의 분화과정에 관여하는 신호전달 중 wnt 신호에 미치는 영향에 대해 유전자의 발현 양상과 단백질의 발현 양상을 살펴보기 위해 각각 RT-PCR과 western-blotting을 수행하였다.
The retinae of Todarodes pacificus and Octopus minor are divided into four layers that are an outer segment, a rod base region, an inner segment, and a plexiform layer, respectively. The retina of Octopus minor is about $20{\mu}m$ thicker ($400{\sim}420{\mu}m$) than that of Todarodes pacificus ($385{\sim}400{\mu}m$). A retina is composed of visual cells and supporting cells. The microvilli of length $0.6{\sim}0.7{\mu}m$ are packed densely on top of the supporting cells of Octopus minor while they are not found in Todarodes pacificus. The visual cells and supporting cells have pigment granules that exclude light. In case of Todarodes pacificus, the pigment granules of the visual cell are larger ($2.0{\times}0.5{\mu}m$) than those of the supporting cell ($1.0{\times}0.3{\mu}m$). But, the sizes of both cells are similar in Octopus minor. In the upper portion of a visual cell, microvilli shaped like a comb are forming a rhabdome (diameter, 60 nm) of a hexagonal structure. The rhabdome consists of 4 rhabdomere and the total area of a rhabdom of Octopus minor is larger than that of Todarodes pacificus. The synaptosome constructing a plexiform layer in Todarodes pacificus are divided into two types, each of which possess electron dense-core vesicles and electron lucent vesicles, respectively. Octopus minor also has two types of synaptosomes but each type comprises a mixture of electron dense vesicles and electron lucent vesicles, and electron lucent vesicles only, respectively, which is different from the case of Todarodes pacificus.

To utilize as highly functional scaffolds for tissue engineering by improving hydrophobicity and cell compatibility of the exist polymer scaffolds, the biodegradable poly(L-lactic acid) (PLLA) films and scaffolds having the optimal hydrophilicity were prepared by in situ plasma treatment and grafting of a carboxyl acid-containing monomer, acrylic acid (AA) in the chamber. From the results of surface analyses, surface-modified nonporous PLLA film and dual pore scaffold surfaces showed high hydrophilicity due to the decrease in contact angle and the increase in carboxylic groups as compared with untreated PLLA control. In particular, among various surface modification methods, Ar(argon)+AA+AA sample prepared by Ar plasma and then acrylic acid treatments displayed lower contact angle and more carboxylic groups thar Ar/AA and Ar+TP(thermal polymerization) samples, indicating that Ar+AA+AA sample was optimally treated for improving its hydrophilicity. In the cases of surface modified nonporous PLLA films and dual pore scaffolds, the adhesion and proliferation of chondrocytes increased with increasing their hydrophilicity.

Calcium phosphates are very interesting materials for use as scaffolds for bone tissue engineering. These materials include hydroxyapatite (HA) and tricalcium phosphate (TCP), which are inorganic components of human bone tissue and are both biocompatible and osteoconductive. Although these materials have excellent properties for use as bone scaffolds, many researchers have used these materials as additives to synthetic polymer scaffolds for bone tissue regeneration, because they are difficult to manufacture three-dimensional (3D) scaffolds. In this study, we fabricated 3D calcium phosphate scaffolds with the desired inner and outer architectures using solid freeform fabrication technology. To fabricate the scaffold, the sintering behavior was evaluated for various sintering temperatures and slurry concentrations. After the fabrication of the calcium phosphate scaffolds, in-vitro cell proliferation and osteogenic differentiation tests were carried out.

Scaffold fabrication technology using a 3D printer was developed for damaged bone tissue regeneration. A scaffold for bone tissue regeneration application should be biocompatible, biodegradable, and have an adequate mechanical strength. Moreover, the scaffold should have pores of satisfactory quantity and interconnection. In this study, we used the polymer deposition system (PDS) based on fused deposition modeling (FDM) to fabricate a 3D scaffold. The materials used were polycaprolactone (PCL) and alginic acid sodium salt (sodium alginate, SA). The salt-leaching method was used to fabricate dual pores on the 3D scaffold. The 3D scaffold with dual pores was observed using SEM-EDS (scanning electron microscope-energy dispersive spectroscopy) and evaluated through in-vitro tests using MG63 cells.

This study was a preparatory experiment aimed the development of membrane scaffolds for tissue engineering. A PCL composite solution contained sodium chloride(NaCl). PCL porous membrane scaffolds were formed on a glass casting plate using a film applicator and immersed in distilled water to remove the NaCl reaching after drying. NaCl was used as a pore former for a 3 dimensional pore net-work. The dry condition parameters were $4^{\circ}C$, room temperature (RT) and $40^{\circ}C$ for each different temperatures in the drying experiment. SEM revealed the morphology of the pores in the membrane after drying and evaluated the in vitro cytotoxicity for basic bio-compatibility. The macro and micro pores existed together in the scaffold and showed a 3-dimensional pore net-working morphology at RT. The in vitro cytotoxicity test result was "grade 2" in accordance with the criterion for cytotoxicity by ISO 10993-5. The dry condition affected the formation of a 3 dimensional pore network and micro and macro pores. Therefore, these results are expected provide the basic process for the development of porous membrane scaffolds to control degradation and allow drug delivery.
