Immunogold Labellings and Expression of Metallothionein in Regenerating Rat Liver (재생중인 흰쥐 간의 메탈로사이오닌에 대한 면역-금 표지 및 발현에 관한 연구)

  • Metallothionein (MT) is a low molecular weight, cysteine-rich, metal-binding, and non-enzymatic protein. The present study was carried out to investigate the expression of MT gene as well as the localization of MT in regenerating rat liver. In partial hepatectomized rats, MT mRNA was detected as early as 1 hr and reached a maximal level by 8 hr after the operation. Thereafter, this level decreased gradually until 24 hr, and it became similar to that of sham control. Meanwhile, time course of MT immunoreactivity using immunogold-labelling revealed that the number of gold particles in hepatocytes increased significantly by 12 hr, but decreased at 48 hr after partial hepatectomy. Ultrastructurally, immunogold particles indicating the presence of MT were distributed in both the cytoplasm and the nucleus of the rat hepatocytes, particularly the particles were distributed at rough endoplasmic reticulum and nucleolus and did not seem to adhere to mitochondria or lysosomes in proliferating hepatocytes. Briefly, high level of MT mRNA expression and the intense immunoreactivity and/or the specific localization of MT was observed during liver regeneration. These results suggest that MT possibly involves hepatocyte proliferation via the storage or the supply of various metal ions in the regenerating rat liver.

Anti-stress Effect by the Treatment of Vitamin C (Vitamin C 투여에 의한 항스트레스 효과)

  • The anti-stress effect by the treatment of vitamin C was investigated in this study. The treatment of ascorbic acid in the presence of $Cu^{2+}$ion induced strong time- and dose-dependent degradation of hitamine, and also the addition of histamine accelerated time-dependent decomposition of ascorbic acid in vitro. The treatment of ascorbic acid in $ODS^{od}/_{od}$rats, which cannot synthesize ascorbic acid, significantly decreased the urinary histamine. The protreatment of ascorbic acid, dexamethasone and promethazine inhibited the lethal effect induced by immobilization stress, but that of dimethylsulfoxide did not. The addition of ascorbic and to a culture of spleen cells of $ODS^{od}/_{od}$rats significantly increased the Con A-dependent T lymphocyte proliferation.

Optimization of Retrovirus Mediated-Gene Transfer into Hematopoietic Stem Cells (Retrovirus를 이용하여 조혈모세포에 유전자를 전달하기 위한 최적화)

  • In this study, optimal conditions to infect CD34 positive cells containing hematopoietic stem cells obtained from cord blood and bone marrow were found using two different retroviral vectors expressing human growth hormone (hGH) and $\beta$-galactosidase. CD34 positive cells were successfully infected with recombinant retroviruses only when the CD34 positive cells were co-cultured with packaging cells secreting recombinant retroviruses. To find the highest infection efficiency for the gene transfer, CD34 positive cells from cord blood were co-cultured with packaging cells secreting recombinant retroviruses encoding E. coli lacZ gene. The highest infection efficiency was obtained when CD34 positive cells were cultured for 3 days, and then co-culturing was done for another 2 days. When CD34 positive cells from bone marrow were co-cultured with packaging cells secreting recombinant retroviruses encoding hGH gene, the maximum amount of hGH was also secreted at the same conditions found above, i.e. 3 days of culture and 2 days of co-culture. These results show that there are optimal conditions for the gene transfer into hematopoietic stem cells regardless of sources of target cells or retroviral vectors used to infect.

Antimicrobial and Biogenic Amine-Degrading Activity of Bacillus licheniformis SCK B11 Isolated from Traditionally Fermented Red Pepper Paste (전통고추장에서 유해균 억제 및 Biogenic Amines 분해 능력을 가지는 Bacillus licheniformis SCK B11의 분리)

  • In order to inhibit the growth of pathogens and degrade biogenic amines during the fermentation of soybean products, an isolate with antimicrobial activity against pathogens and biogenic amine-degrading property was obtained from 83 traditionally fermented soybean products. The morphological and biochemical tests and the phylogenetic relationship among 16S rRNA gene sequences indicated that the isolate named as the strain SCK B11 was most closely related to Bacillus licheniformis. The cell-free supernatant of two day cultures was active against several pathogens including Enterococcus faecalis, Listeria monocytosis, Micrococcus luteus, Pseudomonas aeruginosa, Bacillus cereus, and Staphylococcus aureus. PCR analysis was conducted to determine relatedness to antimicrobial lantibiotics and biosurfactants produced by Bacillus spp., but showed negative for the genes encoding surfactin, lichenysin, and lichenicidine. Electron microscopic observation indicated that the antimicrobial agent seemed to attack the membrane of the pathogens, leaving the ghost or shrunken cells. The strain was found to degrade histamine by 72% and tyramine by 66% in the cooked soybean containing 5.3% of biogenic amine over 10 days of fermentation time. The use of selected strain would be a potential control measure in manufacturing traditionally fermented soybean products that are difficult to control pathogens and biogenic amine levels.

Physiological Activity of Methanol Extracts from Ligularia fischeri and Their Hyperplasia Inhibition Activity of Cancer Cell (곰취 메탄올 추출물의 생리활성 및 암세포 증식억제 효과)

  • This study was conducted to gather the basic data on making good use of a kind of groundsel (Ligularia fischeri). We have made methanol extracts from Ligularia fischeri and have also determined the effects of extracting temperature and time on the physiological activities of methanol extracts from Ligularia fischeri. Total phenolic compound and flavonoid contents in the methanol extracts from Ligularia fischeri at the extracting concentration of $1,000mg{\cdot}L^{-1}$ were $75.8-297.7mg{\cdot}L^{-1}$ and $45.6-173.6mg{\cdot}L^{-1}$. Total phenolic compound and flavonoid contents, and DPPH radical scavenging activities were most increased when Ligularia fischeri was extracted with methanol at $95^{\circ}C$ for 6 hours, however, nitrite radical scavenging activities were extremely increased at $75^{\circ}C$ for 12 hours by 97.4%. At $200mg{\cdot}L^{-1}$ and $400mg{\cdot}L^{-1}$ methanol extracting concentration, the hyperplasia of lung cancer cells (Calu-6) and stomach cancer cells (SNU601) were effectively inhibited over 90%. Consequently, it was assumed that Ligularia fischeri was a functional vegetable with a higher physiological activities. Making the processed foods, it had better make the extracts from Ligularia fischeri with methanol at $95^{\circ}C$ for 6 hours.

The Environment-Friendly High Quality Pork Production by feeding the Fermentation Products of Natural Clay Mineral (천연의 점토광물질 발효산물 급여에 의한 친환경 고품질 원료돈육 생산)

  • We investigated the effect of the clay mineral fortified with complex trace elements by specific fermentation microbes on growth performance and a quality of pork in this study. For the declared experimental animal, 80 heads of crossbred with Large White and Yorkshire were randomly splitted into a control group and a test group. Each group were assigned with 4 replicates and 10 heads of each replicate. When 0.3% of the fermentation product of the clay mineral (FCM) was added into feed stuff and fed for 9 weeks of experimental feeding period, the results of the feed intake, weight gains, pH of carcass, lightness of meat color, and brightness of the muscle semimembranosus were similar to those of the control group. However, dressing percentage, reddishness of carcass, and water holding capacity were improved, respectively, as much as 2.7%, 12%, and 10%, and reduced by 6% in cooking loss. In order to investigate the immune modulatory effect of fermented clay mineral, pheripheral blood mononuclear cells (PBMC) were isolated and Immune modulatory parameters were measured. The proliferation activity of PBMC from pigs fed the fermented clay mineral were significantly increased compared to control group pigs, and also those results were more clearly observed as activated with lipopolysaccharide and concanavaline A. The secretion of TNF-${\alpha}$ of the FCM group pigs showed an increasing tendency. Therefore it was suggested that the feeding of FCM which was high in cation metathesis and the value of infrared ray, activated the immune responses, and thus the production of the environment-friendly high quality pork without the use of antibiotics would be possible.

Effect of Some Herbal Plant Extracts on the Activation of Dendritic Cells (일부 한약재의 수지상세포 활성화 효과)

  • Dendritic cells (DCs) play a pivotal roles in the initiation of T cell-mediated immune responses, making them an attractive in immuno vaccines. Angelica gigas and Cnidium officinale were a medicinal herb widely used in Asian countries. In this study, we examined the effects of A. gigas and C. officinale extracts on the DCs functional maturation and phono-type. Immature DCs were cultured in the presence of GM-CSF and IL-4, and the generated immature DCs were stimulated with OVA in the presence or absence A. gigas and C. officinale extracts, respectively, for 24 hours. The antigen-presenting capacity of A. gigas and C. officinale extracts-treated DCs as analyzed by $CD4^+$ helper T cell clone (OVA-specific) proliferation and cytokines (IL-2 and $IFN-{\gamma}$) production were significantly increased. But A. gigas and C. officinale extracts were not directly effected $CD4^+$ helper T cell clone function. Also, the expression of surface co-stimulatory molecules, including major histocompatibility complex (MHC) class II, CD86 and CD11c, is increased on DCs that were stimulated with A. gigas and C. officinale extracts. These results indicate the immunomodulatory properties of A. gigas and C. officinale extracts, which might be medical supplies or health foods.

Yangkyuksanhwa-Tang Attenuates Ischemic Brain Injury in a Focal Photothrombosis Stroke Model (뇌허혈 마우스모델에서 양격산화탕이 뇌 손상 완화에 미치는 효과)

  • Yangkyuksanhwa-Tang (YKSH), consisting of nine different herbs, is commonly used in Soyangin-type individuals with stroke, based on the Sasang Constitution Theory in Korea. However, no evidence has yet confirmed a beneficial effect of YKSH in ischemic stroke treatment. In this study, we investigated the effects of YKSH on ischemic brain injury in a mouse model of cerebral ischemia. Focal cerebral ischemia in mice was induced by photothrombosis, and behavioral recovery was evaluated. Infarct volume, inflammation, and newly generated cells were evaluated by histology and immunochemistry. YKSH treatment resulted in a significant recovery from the motor impairments induced by focal cerebral ischemia, as determined with wire grip and rotarod tests. YKSH treatment also decreased the infarct volume and the number of cells positive for tumor necrosis factor-${\alpha}$ and myeloperoxidase when compared with a vehicle-treated control group. By contrast, YKSH treatment considerably increased the number of cells positive for glial fibrillary acidic protein and ionized calcium-binding adapter molecule 1, as well as the number of cells doubly positive for Ki67/doublecortin when compared with the vehicle-treated group. These results suggest that YKSH treatment attenuated the infarct size by anti-inflammatory action, astrocyte and microglia activation, and neuronal proliferation, thereby facilitating neurofunctional recovery from a cerebral ischemic assault. YKSH could therefore be a potential treatment for neurofunctional restoration of the injured brains of patients with stroke.

Antiangiogenic activity of non-aqueous fraction from Sparassis crispa extract in human umbilical vein endothelial cells (혈관내피세포에서 꽃송이버섯(Sparassis crispa) 소수성 추출물의 항혈관신생 활성)

  • Sparassis crispa is an edible mushroom that is distributed in Korea, Japan, Europe, and North America. It exerts various biological activities such as immunopotentiation, anti-diabetic, anti-cancer, and anti-inflammatory effects. Recently, we separated the health functional non-aqueous fraction from the chloroform extract of S. crispa (SCF4). In this study, we evaluated the antiangiogenic activity of SCF4 in human umbilical vein endothelial cells (HUVECs). SCF4 effectively inhibited vascular endothelial growth factor (VEGF)-induced cell growth at concentrations ($5-25{\mu}g/mL$) showing no cytotoxic effects. SCF4 inhibited VEGF-induced invasiveness and tube formation ability, which are in vitro angiogenic features of HUVECs, in a dose-dependent manner. In addition, SCF4 markedly suppressed in vivo angiogenesis of chorioallantoic membrane from growing chick embryos without cytotoxicity. Furthermore, SCF4 downregulated the phosphorylation of VEGFR2, AKT, and ERK1/2, which are major angiogenic signal mediators. These results suggest that SCF4 inhibited angiogenesis by suppressing the VEGFR2 signaling pathways without cytotoxicity.

Anti-aging and Anti-inflammatory Activities of the Extracts of Calamagrostis arundinacea (Calamagrostis arundinacea (실새풀) 추출물의 항노화 및 항염증 활성)

  • The anti-aging and anti-inflammatory activities of hot-water (Ca-HW) and 70% ethanol (Ca-E70) whole-plant Calamagrostis arundinacea extracts, as well as their bioactive potentials, were investigated using cell-free and cell-mediated experimental systems. Use of the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical decolorization assay to evaluate the antioxidant activity of the Ca-HW and Ca-E70 extracts revealed DPPH radical scavenging activities of 27% and 48%, respectively. Neither extract caused significant cytotoxicity, and both showed cell proliferation and promotion effects using RAW 264.7, B16F10, and CCD986sk cells. B16F10 melanoma cells showed higher melanin synthesis when treated with 100 mg/ml Ca-HW or Ca-E70 than with arbutin, indicating a stronger inhibitory effect of arbutin on melanin synthesis. Ca-HW and Ca-E70 increased pro-collagen biosynthesis in the human fibroblast CCD986-SK cell line by 24.69% and 12.55%, respectively. Analysis of the anti-inflammatory effects of different concentrations of Ca-HW and Ca-E70 in RAW264.7 cells revealed that Ca-E70 appeared to inhibit the lipopolysaccharide-induced production of nitric oxide and IL-6, a proinflammatory cytokine; therefore, Ca-E70 showed an anti-inflammatory effect. These results suggested that C. arundinacea extracts could have skin anti-aging and anti-inflammatory properties.
