Effete of Ozone Uptake Rate on Photosynthesis and Antioxidant Activity in the Leaves of Betula Species (자작나무류 잎의 오존흡입량이 광합성 및 항산화효소 활성에 미치는 영향)

  • This study was conducted to compare the physiological and biochemical responses of four Betula species in response to ozone, and to find out the relationship between ozone uptake rate and photosynthesis or antioxidant activity. One-year-old seedlings of four Betula sp, B. costata, B. davurica, B. platyphylla var, japonica, and B. ermani, exposed to 100 pub ozone concentration for 8h day$^{-1}$ for 5 weeks in fumigation chamber. Ozone uptake rate, photosynthesis, SOD and GR activity were measured in the leaves of four species once a week. Cumulative ozone uptake rate was largest in the loaves of B. costata(53.8 mmol m$^{-2}$ ), smallest in the leaves of B. davurica(35.5 mmol m$^{-2}$ ). Photosynthesis of four Betula sp. exposed to ozone reduced relative to control, but the photosynthetic responses with changing stomatal conductance were different among species. Ozone exposure increased SOD activities of four species at the early exposing period, but after a critical point SOD activity decreased gradually. GR activity of B. costata was similar to the change of SOD activity, but the others showed the different patterns from B. costata. In conclusion, decreasing both SOD and GR activity at the critical point, B. costata may be sensitive species in response to ozone. In contrast, the others may be resistant species, which gradually increase GR activity following ozone exposure. GR activity was not always in accord with the change of SOD activity against ozone uptake, and the different responses between species were supposed to be affected by the cumulative ozone uptake.

Effects of Composted Pig Manure Application on Enzyme Activities and Microbial Biomass of Soil under Chinese Cabbage Cultivation (돈분퇴비의 시용이 토양의 미생물체량 및 효소활성에 미치는 영향)

  • To elucidate the effects of composted pig manure on soil biochemical properties, composted pig manure was amended in a sandy loam soil and Chinese cabbage was grown. Composted pig manure treatments included 8, 29 and $57Mg\;ha^{-1}$ for CM-08, CM-29, and CM-57 plots, respectively. Biomass contents and enzymes activities in the non-rhizophere soil were measured. Activities of protease, phosphomonoesterase and dehydrogenase in the plot CM-57 increased to 2.3, 1.6, and 2.4 fold as compared with those of the control plot. Soil microbial biomass contents increased in proportion to the application rates of compost and biomass C, N, and P in the plot CM-59 were 4.3, 3.4, 2.8-fold higher than those of control p1ot(no fertilizer), respectively. During cultivation of Chinese cabbage, biomass C and N were higher in the middle growth stage, although biomass P was the highest in the early growth stage. The average ratio of biomass C:N:P was 11:2:1, and proportion of biomass C and N in the soil organic C and N was 1.1 and 3.6%, respectively. Activities of protease and dehydrogenase had significant correlations with biomass C and P.

Bacterial Stripe of Proso Millet Caused by Acidovorax avenae subsp. avenae in Korea (Acidovorax avenae subsp. avenae에 의한 기장 세균성줄무늬병)

  • In July, 2009, proso millet (Panicum miliaceum), which showing the bacterial brown stripes on leaf sheaths, was collected in Miryang in Korea. Symptoms were systemic brown necrotic stripe lesions on the leaf sheaths and stems, and these symptoms were found in the entire field. The causal agent isolated from symptomatic plants was identified as an Acidovorax avenae subsp. avenae, based on its biochemical and physiological characteristics and also confirmed by the Biolog data and 16S rRNA gene sequence analysis. Also it caused hypersensitive response (HR) when it was inoculated onto the tobacco and tomato. It caused similar symptoms when inoculated onto proso millet. This is the first report of A. avenae subsp. avenae, the causal agent of bacterial brown stripe of the proso millet in Korea.

Mannanase Production by a Soybean Isolate, Bacillus subtilis WL-7. (된장 분리균 Bacillus subtilis WL-7에 의한 Mannanase 생산)

  • A bacterium producing the extracellular mannanase was isolated from Korean soybean paste. The isolate WL-7 has been identified as Bacillus subtiis on the basis on its 16S rRNA sequence, fatty acid composition, morphology and biochemical properties. The mannanase of culture supernatant was the most active around $55^{\circ}C$ and pH $6.0^{\circ}C$, and retained 90% of its maximum activity at range of pH 5.0∼7.5 and $50∼60^{\circ}C$. The additional carbohydrates including lactose, $\alpha$-cellulose, avicel, locust bean gum (LBG), wheat bran and konjak increased dramatically the mannanase productivity of strain WL-7. Especially, the maximum mannanase productivity was reached to 224 U/ml in LB medium supplemented with both 0.5% LBG and 0.5% konjak, which was approximately 200-folds more than that in LB medium. It was suggested that the increase of mannanase production was owing to induction of mannanase biosynthesis by both LBG and konjak hydrolysates transported following initial hydrolysis by extracellular mannanase during the cell growth.

Identification and Biochemical Characterization of a New Xylan-degrading Streptomyces atrovirens Subspecies WJ-2 Isolated from Soil of Jeju Island in Korea (제주도 토양으로부터 자일란 분해 Streptomyces atrovirens subspecies WJ-2 동정 및 효소의 생화학적 특성 규명)

  • A bacterial strain was isolated from a soil sample collected on Jeju Island, Korea. The strain, designated WJ-2, exhibited a high xylanase activity, whereas cellulase activity was not detected. The 16S rRNA gene sequence of WJ-2 was highly similar to type strains of the genus Streptomyces. A neighbor-joining phylogenetic tree based on 16S rRNA gene sequences showed that strain WJ-2 is phylogenetically related to Streptomyces atrovirens. Furthermore, DNA-DNA hybridization analysis confirmed that strain WJ-2 is a novel subspecies of Streptomyces atrovirens. The genomic DNA G+C content was 73.98 mol% and the major fatty acid present was anteiso-C15:0 (36.19%). The growth and xylanase production of strain WJ-2 were significantly enhanced by using soytone and xylan as nitrogen and carbon sources, respectively. Crude enzyme preparations from the culture broth of strain WJ-2 exhibited maximal total xylanase activities at pH 7.0 and $55^{\circ}C$. Thin-layer chromatography analysis revealed that the crude enzyme degrades beechwood xylan to yield xylobiose and xylotriose as the principal hydrolyzed end products.

  • Host rock or the standing sculptured Buddha in the Yongamsa temple is macular porphyritic biotite granite, which has gone through mechanical and chemical weathering. The rock around the Buddha statue is busily scattered with steep inclinations that are almost vertically discontinuous planes with the strikes of $N8^{\circ}E$. Especially the development of the joints that cross the major joints causes the structural instability of the rock. The rock of the Buddha statue is separated into several rock blocks because of many different discontinuity. Thus it is estimated that the bed rock has not only plane and toppling failure but also wedge failure in all the sides. Since the differential pressure is imposed on the body of the Buddha in the host rock, it is urgent to give a reinforce treatment of geotechnical engineering for the safe of its structural stability. Very contact area of joints have turned into soil, which promotes the growth of weeds and plant roots, then aggravates the mechanical weathering of the rock. Thus conservational treatments should also be considered to get rid of secondary contaminants and vegetation along the discontinuities and to prevent further damages.

Biochemical Characterization of Cysteine(-) Mutant Alanine Racemase from Bacillus pseudomycoides (Bacillus pseudomycoides로 부터 분리된 alanine racemase 유전자의 cysteine 치환 및 생화학적 특성)

  • A gene encoding an alanine racemase in B. pseudomycoides was cloned and one (Cys316) or both of two cysteines (Cys316 and Cys365) was (were) substituted with alanine. The cysteine (-) alanine racemases were expressed in E. coli BL21 (DE3) using a pET-21 vector. The expressed enzymes were purified through affinity chromatography using 6xHis ligand. The purified enzymes all showed major one bands by SDS-PAGE analysis, corresponding to 46 kDa. The cysteine (-) alanine racemases as well as the wild type enzyme showed alanine racemase activities, indicating that the enzyme is an alanine racemase and the cysteines in the enzyme may not be involved in the catalysis and/or substrate binding. Thermal stabilities of Cys (-) alanine racemases decreased considerably and half-lives were 26 (wild type), 21 (C316A) and 18 min (C316-365A), respectively at $60^{\circ}C$ pH 8.0, suggesting that cysteine is considerably contributive to the thermal stability of the alanine racemase.

Isolation of salt-tolerant bacteria from rhizosphere and rhizoplane of halophyte plant Suaeda japonica in Gochang·Buan tidal flat (고창·부안 갯벌에 자생하는 염생식물 칠면초 근권 및 근면으로부터 내 염성 세균 분리와 그 특성화)

  • Nine strains of high concentrations of salt-tolerant bacteria were isolated from the rhizosphere and rhizoplane of the halophyte plant Suaeda japonica grown in Gochang Buan tidal flat. The isolated bacteria were classified as genera Vibrio (strains JRS-1, -2, -3, -4, and -5, and JRL-1 and -4) and Bacillus (strains JRL-2 and -3) based on the 16S rRNA gene sequence similarity. The optical growth condition for salt concentration was examined on the selected, representative strains. Strain JRS-1 with the closest relative of Vibrio neocaledonicus showed the highest growth rate at the total salt concentration of 6% among the incubation conditions of 3-8% salt concentrations. Strain JRL-2 with the closest relative of Bacillus thuringiensis showed the tendency that growth rate increased with increasing salt concentrations and the maximum growth rate at 7% of the total salt concentration. The isolated bacteria showed salt-resistances to higher salt concentrations than their habitat soils with 3%. In addition, we identified evidences of potentially plant interaction-relevant enzymatic activities, from utilization of some substrates rich in plants, such as triglyceride, ${\rho}$-nitrophenyl-${\alpha}$,$\text\tiny{D}$-glucoside, and ${\rho}$-nitrophenyl-${\beta}$,$\text\tiny{D}$-glucoside.

A Thermostable Protease Produced from Bacillus sp. DF 218 (Bacillus sp. DF218이 생산하는 내열성 단백질 분해효소)

  • Microorganism (strain DF 218) producing thermostable pretense was isolated from Korean soil and compost. It was Gram-positive, rod-shaped, aerobic, and spore-forming with yellowish white colony color, Temperature range for growth at pH 6.5 was $30-65^{\circ}C$, with optimum growth at $60^{\circ}C$. pH range for growth at $60^{\circ}C$ was 5-7 with optimum of 6.5, which indicates strain DF 218 to be thermophilic. The 16S rDNA sequence of strain DF 218 had 95% sequence similarity with that of Bacillus flexus. Based on physiological properties and phylogenetic analysis, we proposed the isolated strain as Bacillus sp. DF 218. Pretense was produced aerobically at $60^{\circ}C$ for 32 hr in a medium (pH 6.5) containing 1% each trypton, glucose, and NaCl. Its molecular weight was estimated as 61 kDa, with optimum temperature and pH of $60^{\circ}C$ and 7.5, respectively.

  • The ratio of the eatable flesh taken from the filefish and its chemical components were investigated from March 1980 to February 1981 and the results are as follows :1. The ratio of the eatable flesh taken from the fish was low during March to June and high during July to December. 2. The moisture content was $81.4-82.0\%$ during March to June, $77.5-80.1\%$ during July to December and $79.4-81.4\%$ during January to February. 3. The protein content was $15.4-15.7\%$ during March to June, $17.0-19.9\%$ during July to December and $16.1-17.0\%$ during January to February. 4. The Lipid content was $0.31-0.41\%$ during March to January, $0.46-0.84\%$ during July to January and $0.43\%$ during February. 5. The carbohydrate content was $0.3-0.4\%$ during March to July, $0.5-0.7\%$ during August to January and $0.3\%$ during February. In conclusion, the ratios of the eatable flesh taken from the filefish and its protein content were highest during July to December in one year.
