Adipocyte Culture Medium Stimulates GITRL Expression in MDA-MB-231 Cells (지방세포배양액이 유방암세포주에서 GITRL의 발현을 유도)

  • Little is known about roles of adipose tissue, although obesity is one of the potential risk factors in causing breast cancer and adipose tissue surrounding breast ductal cells is the largest organ. To identify the genes that are regulated by factors secreted from adipocytes in breast cancer cells, MDA-MB-231 cells were treated with the culture medium of adipocytes. In present study glucocorticoid-induced TNF receptor-related protein ligand (GITRL) gene was studied among the induced genes. It was found that GITRL was significantly increased by the culture medium of adipocytes. Proteinase K-treated adipocyte culture supernatants failed to induce GITRL expression. These findings indicate that unknown protein factors are responsible for the induction of GITRL expression. The expression of GITRL did not affect the invasive ability of MDA-MB-231 cells, but coculture with NK92 expressing GITR suppressed the invasiveness of MDA-MB-231 cells.

Effect of Gastrodiae rhizoma Fractions on Serum Lipid Concentrations in Rats Fed with High Fat Diet (천마 분획물이 고지방식이를 급여한 흰쥐의 혈청 지질농도에 미치는 영향)

  • This study was carried out to investigate the effect of Gastrodiae rhizoma fractions on serum lipid profiles and atherogenic index (AI) in male S.D. rats fed a high fat diet supplemented with 10 : 2 : 1% (w/w) of lard, corn oil, and cholesterol during the entire 12-week experimental period. Forty-eight rats were randomly divided into four groups; A (low molecule), B (polysaccharide), C (protein) fractions of Gastrodiae rhizoma, respectively, and D (high fat diet as control). Body weight gain, diet intake and feed efficiency ratio did not differ significantly among the groups during the experimental period, but final body weight was on the average 44 g higher in control compared with the three groups of Gastrodiae rhizoma $(A{\sim}C)$. TC and TG levels of group B when compared with control were each decreased by an average of 21.5% and 39.6%, respectively. HDL-cholesterol level was markedly higher in group C than group A and B of Gastrodiae rhizoma. LDL-cholesterol levels of Gastrodiae rhizoma groups $(A{\sim}C)$ were significantly lower than control. AI was significantly lower in group C at 1.45 than the other two Gastrodiae rhizoma at $1.94{\sim}2.05$ and control of 2.12. From the findings, it is feasible for water soluble and high molecular weight components of Gastrodiae rhizoma like polysacchride and protein fractions to be considered as functional components for improving hyperlipidemia.

Functional Characteristics from the Barley Leaves and its antioxidant mixture - Study on the Nitrite Scavenging Effect - (맥엽 및 항산화 조성물의 생리 기능적 특성에 관한 연구 - 아질산염 소거능을 중심으로 -)

  • This study was conducted to estimate the biofunctional characteristics, namely nitrite scavenging effect of barley leaves (BL) and their mixture (AM) with other antioxidants. Aqueous or methanol extracts were obtained from BL and AM. Aqueous and methanol extracts from barley leaves had relatively higher nitrite scavenging effects and its activities and contents increased in a concentration-dependent manner. The activities and contents of methanol extracts obtained from BL and AM were higher than those of aqueous extracts. Especially, AM containing BL and other antioxidant mixture had the highest activities and contents increased in a concentration-dependent manner. BL or AM were added to macrophage cells of RAW 264.7. Survival rates of the cells treated with BL and AM were measured to be different. $IC_{50}$ value decreased in a concentration-dependent manner by the addition of aqueous or methanol extracts from BL and AM. Especially, methanol extracts of AM had the highest nitrite scavenging effects. Thus, BL and AM may have protective effect against carcinogen and immune ability against reactive molecules through NO (nitric oxide) signal pathway. From the above results, barley leaves appear to contain natural anticancer and immune-related agents and may have potentiality to be used as functional food ingredients.

Lactic Acid Fermentation and Biological Activities of Rubus coreanus (복분자의 유산발효와 생리활성 평가)

  • The puree of Rubus coreanus was fermented using lactic acid bacteria and its biological activities were examined. Lactobacillus acidophilus KCCM 32820, L. casei KCCM 12452, Lactococcus lactis subsp. lactis KCCM 40104, and Streptococcus thermophilus KCCM 40430 were used as a single or mixed starter for the lactic acid fermentation, and their cultures at the late logarithmic growth phase were inoculated to final concentration of 2% (v/v). L. casei fermented the puree of Rubus coreanus best when used as a single starter, and the culture of L. casei and L. lactis with the inoculation ratio of one to one showed the highest fermentation activity when used as a mixed starter. However, the fermented broth of the puree of Rubus coreanus using L. acidophilus and S. thermophilus showed the best results in the sensory evaluation. The optimal lactic acid fermentation conditions were as follows; the concentration of oligosaccharide added was 1% (w/v), pH of puree and fermentation temperature were 4.0 and $37^{\circ}C$, respectively, and fermentation time was $72{\sim}96$ hours. Glucose and fructose were major free sugars, and the content of lactic acid was 698.2 mg/100 g in the fermented broth. The fermented broth of the puree of Rubus coreanus showed the electron donating ability and nitrite scavenging ability with the value of 69% and 38.3% at pH 1.2, respectively. SOD-like activity and inhibitory activity on xanthine oxidase were also found in the fermented broth with the value of 60.3% and 41.8%, respectively. When the antimicrobial activities of the fermented broth were examined, it showed the highest growth inhibitory activity against Escherichia coli O-157:H7, and also contained antimicrobial activities against Salmonella typhimurium, Bacillus cereus, and Staphylococcus aureus.

  • Physicochemical and gelatinization properties of Suwon-415 pigmented rice starch were determined. The median and mean particle sizes of Suwon-415 pigmented rice starch were 5.23 and $6.27\;{\mu}m$, whereas those of non-glutinous rice starch were 4.71 and $5.43\;{\mu}m$ respectively. Water-binding capacity, iodine reaction and intrinsic viscosity of Suwon-415 pigmented rice starch were lower than those of non-glutinous rice starch. X-ray diffraction patterns showed traditional A type of cereals and relative crystalline of both samples showed no difference. Peak viscosity, breakdown and setback were increased with increasing weight of starches.

Preparation and Characteristics of Soybean Curd Using Cuttle Bone Powder Treated with Acetic Acid (유기산처리 갑오징어갑 분말을 이용한 두부의 제조 및 특성)

  • Soybean curd was prepared using cuttlebone powder treated with acetic acid (ATC), and its characteristics were determined. Soybean protein was coagulated by adding calcium chloride, magnesium chloride, and ATC at 0.7, 1.2, and 0.9%, respectively, or higher. Yield, white index, texture, and sensory evaluations revealed the optimal concentration of ATC for preparation of soybean curd was 2.0% of total soybean weight. The proximate compositions of soybean curd prepared by adding 2.O% ATC were 75.9%, 12.6%, 1.9%, 5.3%, 6.60, and $1.0×10^4$ CFU/g in for moisture, crude protein, crude ash, crude lipid, pH, and viable cell count, respectively. Although white index and mineral content of both soybean curds were similar, texture of ATC-prepared soybean curd was superior to that of soybean curd prepared with 1.5% calcium chloride.

Screening test for Dendropanax morbifera Leveille extracts: in vitro comparison to ox-LDL-induced lipid accumulation, ethanol-induced fatty liver and HMG-CoA reductase inhibition (황칠나무 추출물의 고지혈증 완화 효과 스크리닝)

  • The objective of this study was to compare the antihyperlipidemic effects of different Dendropanax morbifera leaf extracts in vitro. The extracts differed in terms of specimen age, harvesting season, and extraction method. RAW 264.7 cells were pretreated with these extracts and stimulated by oxidized low-density lipoprotein. Ethanol was used to induce toxicity in HepG2 cells. Cellular lipid accumulation was quantified using oil red O staining in both these cells. The extracts were evaluated for their inhibitory effects on 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase. RAW 264.7 cells treated with the 60% ethanol extract of an 8-year-old specimen harvested in November exhibited the lowest lipid accumulation. The 30% ethanol extract of a 5-year-old specimen harvested in May exhibited the greatest protection from ethanol-induced cytotoxicity in HepG2 cells. The hot water extract of an 8-year-old specimen harvested in May showed the greatest inhibition of HMG-CoA reductase. These results showed that D. morbifera extracts prepared from leaves that are harvested in May possess the highest antihyperlipidemic effects.

Studies for Component Analysis, Antioxidative Activity and ${\alpha}-glucosidase$ Inhibitory Activity from Equisetum arvense (쇠뜨기(Equisetum arvense) 추출물의 항산화 활성 분석 및 ${\alpha}-glucosidase$ 저해활성)

  • This study was carried out to investigate the chemical components, and antioxidative and $anti-{\alpha}-glucosidase$ activities of Equisetum arvense extracts. In Equisetum arvense extracts were composed of 53.20% of crude fiber, 20.42% of crude ash, 15.32% of crude protein and 2.21% of crude fat. Potassium was the most predominant mineral and followed by phosphorus, calcium, magnesium, and sodium. The contents of the unsaturated fatty acids, such as linolenic acid, linoleic acid, and palmitic acid, were higher than those of saturated fatty acids. Seventy percent ethanol extract exhibited antioxidative activity with $IC_{50}$ of $168.1\;{\mu}g/ml$. The Seventy percent methanol extract showed higher ${\alpha}-glucosidase$ inhibitory activity than other solvent extracts.

  • The physiological activity of St. John's wort extracts were examined. Total phenol contents in the ethanol extracts $(246.0{\pm}10.5\;{\mu}g/ml)$ with St. John's wort leaf was higher than that in water extract $(237.4{\pm}13.2\;{\mu}g/ml)$. The electron donating ability in the water extracts and in the ethanol extracts were 95.0% and 95.2% respectively. Antioxidant protection factor of the ethanol extract was higher than that of the water extract. The water extract from St. John's wort leaves did not show an antimicrobial activity against Helicobacter pylori, but the ethanol extract revealed high antimicrobial activities such as 11 mm of clear zone in $100\;{\mu}g/ml$ of phenol content and 13 mm of clear zone in $150\;{\mu}g/ml$ of phenol content. The hot water extract showed an angiotensin converting enzyme inhibitory activity of 19.2%. The xanthin oxidase inhibitory activity of hot water and ethanol extract were very high, amounting to 84.8% and 100% respectively. The results suggested a possibility for developing the phenol compounds in St. John's wort as anti Helicobacter pylori, anti-oxidant and anti-gout agents.

Moderating Effects of Skin Hyperpigmentation from Lycii fructus and Lycii folium Extracts (구기자 및 구기엽 추출물의 피부과색소 조절효과)

  • The objective of the present study was to evaluate the skin whitening effect of the extracts, Lycii fructus (LF), Dry-L. foilum (DLF) and Fresh-L. folium (FLF). Tyrosinase inhibition activities was 44% in DLF ethanol extracts at a $500{\mu}g/mL$. When the tyrosinase activities in B16F10 murine melanoma cell were tested, the activities in DLF ethanol extracts was 14% at a $50{\mu}g/mL$ concentration. The protein expression of microphthalmia-associated transcription factor, tyrosinase related protein 1 (TRP-1), TRP-2, and tyrosinase, which are all melanin related factors, showed that LF, DLF and FLF extracts inhibited the protein bio-synthesis in B16F10 melanoma cell. Especially the DLF extract showed greater decrease of protein expressions. Results indicate that the DLF extract tested in the present study had skin whitening activity and can be used as a function a ingredients for food and cosmetic compositions.
