Effects of Tarak, Korean Traditional Fermented Milk, on Proliferation of Immune Cells and Melanin Biosynthesis (전통발효유 타락의 면역세포 증식 및 멜라닌 생성에의 효과 - 연구노트 -)

  • Tarak is a Korean traditional fermented milk product that is fermented by adding rice wine to milk. Tarak was produced with Lactobacillus paracasei ssp. paracasei M13-65-3 isolated from rice wine, and its effects on immune cell proliferation and melanin biosynthesis were investigated. Tarak extract significantly increased proliferation of T lymphocyte Jurkat clone E6-1 cells at concentrations from 10 to $100{\mu}g/mL$. Tarak inhibited activities of tyrosinase and ${\alpha}$-melanocyte-stimulating hormone-induced melanin biosynthesis in mouse skin B16-F10 cells at a concentration of $100{\mu}g/mL$. These results suggest that tarak might have functionalities for enhancing the immune system by increasing immune cell proliferation and regulating melanin biosynthesis.

Elimination of R-Plasmid in Streptomyces bobili (YS-40) by Ethldium Bromide (Ethidium Bromide에 의한 Streptomyces bobili(YS-40)의 R-Plasmid 제거)

  • Streptomyces bobili (YS-40) isolated from soil was tested that it had drug resistance against penicillin, cephalosporin series antibiotics and other antibiotics in the previous paper. The treatment of Streptomyces bobili, (YS-40) with ethidium bromide (EtBr), acriflavine and sodium dodecyl sulfate. (SDS) resulted in the elimination of R-plasmid from the host strain. Minimum growth inhibitory concentrations (MIC) of Hg, Ag, penicillin-G, ampicillin, chloramphenicol, oxytetracycline, streptomycin and kanamycin were found to be 15, 10, > 3, 000, > 100, > 1, 000, > 100, < 5 and < 5$\mu\textrm{g}$/$m\ell$ respectively. Among the curing agents, EtBr was proved to be the most powerful compound for the elimination of R-plasmid in the strain and the elimination rate with EtBr(10$\mu\textrm{g}$/$m\ell$) was about 98%. Optimal pH to. the elimination of R-plasmid was pH 7.0 and the R-plasmid in the cells incubated for 24 hrs was proved to be eliminated most effectively. Aerial mass color, soluble pigment formation and reverse side color were reported to be often the plasmid associated characteristics of the R-plasmid bearing bacteria. But these characteristics of the uncured and cured Streptomyces bobili, (YS-40) showed no changes in the most of the pigment formation media tested in this work.

Antioxidative Properties of Phenolic Compounds Extracted from Black Rice (흑미 색소물질에 함유된 페놀화합물의 항산화 특성)

  • The composition and antioxidative effects of phenolic compounds in black rice were studied. The contents of free and bound phenolic compounds extracted from black rice were 845.4 and 401.6 mg respectively per 100g sample weight. Free phenolic compounds had higher antioxidation ability than those of bound phenolic compounds. Solvent fractionation of free phenolic compounds revealed that butanol fraction had the highest phenolic compounds contents and antioxidative activity among other solvent fractions. Although butanol fraction showed lower lipid peroxidation inhibition (LPI) ability than that of $\alpha$-tocopherol and BHT, free radical scavenging ability was much higher than that of $\alpha$-tocopherol and BHT, as evidenced by electron donating ability (EDA) and benzoic acid hydroxylation inhibition (BAHI) assays.

  • Cultivation conditions for the production of extracellular alkaline protease by a nonpiamentation Serratia sp. and purification of the enzyme were studied. The maximum enzyme level was obtained at the beginning of stationary phase when the organism was cultured on brain heart infusion medium at $25^{\circ}C$ under aeration (gyratory shaking, 180 cycles/min). The enzyme was purified about 100 fold with 16.5% yield by ammonium sulfate precipitation, ammonium sulfate fractionation followed by DEAE-cellulose chromatography (1st and 2nd). The purified enzyme moved as a single symmetrical peak in the analytical ultracentrifuge. The enzyme demonstrated its maximum activity at pH 8.5-9.0 and 4$0^{\circ}C$ when vitamin-free casein was used as a substrate.

Characteristic and Extraction of Fucoxanthin Pigment in Undaria pinnatifida (미역에 함유된 Fucoxanthin 색소의 추출 및 특성)

  • The contents of fucoxanthin in Undaria pinnatifida blade, stem and sporophyll were 87.6 mg/100 g, 62.4 mg/100 g and 127.7 mg/100 g, respectively. The fucoxanthin was analysed by using solvent fractionation, silica gel adsorption chromatography and HPLC techniques. Absorption spectrum of carotenoids extracted from Undaria pinnatifida was similar to the fucoxanthin carotenoids in sea mustard. The cleavage products formed by autoxidation of fucoxanthin were evaluated in order to elucidate possible oxidation products of fucoxanthin in liposomal suspension. Fucoxanthin solubilized at 50 $\mu$M in liposomal suspension formed five oxidized products. These results suggest that fucoxanthin might be cleaved to a series of cleavage products under the oxidative condition in liposomal suspension.

  • Recently, the developments in generating and amplifying ultrashort optical pulses $(ps=10^{-12}s or fs=10^{-15}s)$ have imposed on great advances in the time-resolved laser spectroscopy. Especially, the transient absorption spectroscopy has a wide application range and the main idea of this technique is pump & probe method. After the pump pulse makes the material an excited or a transient states, the probe pulse is sent through the material to measure the absorbance change due to the transient states. 흰목이버섯 유래 고순도 다당체의 광노화 개선 Global 화장품 신소재 개발

  • 최근 길어진 여름 및 이상고온 현상이 지속됨에 따라 심화되는 광노화 피부의 특징으로는 건조, 굵고 깊은 주름, 탄력저하 및 불균일한 색소침착 등이 나타나게 됨. 화장품 소재는 기존 광노화 관련 화학물질인 Retinol 등을 대체하기 위해 자연 유래 성분을 적용한 신소재 연구를 진행하고 있음. 흰목이버섯(Tremella fuciformis)은 흰목이목에 속하는 버섯류로 자실체는 한천질로서, 주름이 되어 갈라져 있거나 또는 귓불 모양을 이루고 있으며, 크기는 10 cm 정도이다. 중국에서는 보양식의 주재료로 쓰일 만큼 탁월한 항노화 효능이 알려져 있다. 본 연구에서는 흰목이버섯의 자실체에서 추출하여 정제한 β-Glucan의 성분 확인, in vitro 수준의 피부 항노화 효과, 동물대체 독성 시험을 통한 피부독성 확인 및 인체 피부유효성 평가를 통한 항노화 효과를 확인하였다. 흰목이에서 추출, 정제 후 Bio-LC를 통한 유리당 분석 결과 Mannose, Fucose, Glucose를 확인하였으며, Human Keratinocyte에 UVB를 조사하여 광노화를 유발한 피부세포에 피부 자극 및 탄력저하 인자인 IL-6, TNFa 및 MMP-1을 평가한 결과 농도 의존적으로 현저히 개선됨을 확인하였다. 또한 보습 및 피부장벽 개선 인자인 Filaggrin과 Involucrin 생성효능을 평가한 결과 매우 높이 생성됨을 확인하였다. 본 연구결과를 토대로 광독성, 피부감작성 및 안점막 동물대체 독성시험을 실시한 결과 무독성임을 확인하여 피부에 안전하면서 효능이 우수한 것을 in vitro 수준에서 확인하였고, 피부 홍반완화, 주름개선, 탄력개선 및 보습증가 등 광노화 예방효과를 인체를 대상으로 평가한 결과 유의적인 홍반완화, 주름개선, 탄력 및 보습증가효과를 확인하였다. 본 연구결과를 종합하여 볼 때 흰목이버섯에서 추출, 정제한 β-Glucan은 in vitro 수준에서 자외선으로 인한 피부 트러블 완화, 탄력 및 보습개선을 확인하였고 독성시험을 통해 무자극임을 판정하였으며, 인체유효성 평가를 통해 광노화 예방효과를 확인하였으며 본 결과를 통해 아시아 및 글로벌 시장으로 천연유래 항노화 소재로 확장하고자 한다.

Antioxidant and Nitric Oxide Inhibitory Activities of Pigments from Chionoecetes japonicas Rathbun (홍게 (Chionoecetes japonicas Rathbun) 껍질 색소의 항산화 활성 및 Nitric Oxide 생성억제 효과)

  • In the present study, antioxidant activities of two crude pigments (acetone and MeOH) and their solvent fractions (n-hexane, 85% aq.MeOH, n-BuOH, and water fractions) from red crab shell were evaluated by measuring 1,1-diphenyl-2-picryl hydrazyl (DPPH), peroxynitrites, and degree of production of reactive oxygen species (ROS) in HT 1080 cells as well as the extent of oxidative damage of genomic DNA purified from HT 1080 cells. From comparative analysis, 85% aq.MeOH fraction showed the strongest scavenging effect on both peroxynitrite in vitro and intracellular ROS in HT 1080 cells. Protective activities of these samples against hydroxyl radical-mediated genomic DNA damage were also investigated. 85% aq.MeOH and n-BuOH fractions significantly inhibited oxidative damage of purified genomic DNA. On the other hand, we investigated their inhibitory effects on nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated Raw 264.7 cells. All samples significantly reduced NO production. Among the samples, n-hexane and water solvent fractions most effectively inhibited NO.

농산물의 효소적 갈변의 저해제 개발 및 그 응용

  • 농산물의 수확 후 취급이나 가공중에 일어나는 효소적갈변 현상은 외관의 변색에 의한 소비자의 기호성을 저하시킬 뿐만 아니라 상품의 가치를 하락시키며, 경제적인 손실을 초래함으로서 현재 식품에 종사하는 기술자나 가공업자에게 가장 관심이 있는 분야이다. 효소적갈변의 주역을 담당하고 있는 polyphenol oxidase(PPO)(EC1.14, 18.1)은 대부분의 식품에 존재하고 있으며 monophenol의 hydroxylation에 의한 o-diphenol화합물의 형성반응(cresolase)과 o-diphenol화합물의 o-quinone으로의 산화반응(catecholase)을 촉배한다. 그리고 여기서 일단 생성된 o-quinone은 다시 다른 o-quinone이나 그밖의 단백질, 아미노산, 환원당 등의 여러 성분들과 상호 반응하여 갈변식품의 특유의 색소인 melanine을 형성한다. 효소적갈변에 의한 품질의 저하는 식품가공산업에서 해결되어야 할 하나의 큰 과제이다. 현재까지 식품산업에서 가장 널리 쓰이고 있는 효소적갈변 저해제는 sylfite류 이다. 그들은 새우의melanosis(blackspot)를 억제하며, 감자, 버섯, 사과와 다른 과채류의 갈변을 방지할 뿐 아니라 쥬우스와 포도주의 off flavors를 일으키는 폴리페놀 플리머의 형성을 감소 시킨다. 그러나 최근 sulfite가 사람의 건강에 좋지않는 부작용을 일으키며, 특히 천식을(asthma)을 지니고 있는 사람이 sulfite가 함유된 식품을 먹었을 경우 사망을 초래할 수 있어 식품첨가물로서 그들의 사용이 미국 FDA에 의해 재평가되고 있다. 최근에 FDA는 셀러드바에서의 sulfite사용을 금지 시켰으며 점차 감자가공품에의 사용을 금지하는 방향으로 나아가고 있다. 따라서 현재 실질적이고 기능적인 sulfite의 대체 화합물의 개발이 절실히 필요한 실정이다. 이러한 배경으로 본 강연에서는 효소적갈변 저해제의 개발과 그들의 식품가공에의 적용 현환 및 화장품, 의약품으로의 응용에 대해 설명하고자 한다.

  • This study was conducted to obtain a supply of conidia sufficient for screening mungbean mutant lines for a source of resistance to Cercospora leaf spot caused by Cercospora canescens Ellis and Martin. Abundant sporulation occurred in cultures on mungbean leaf decoction oatmeal agar(MOA) exposed to about 2,500 Lux of fluorescent light. but it did not occur in continuous darkness. The conditions that produced maximum number of conidia was not coincided with those for vegetative growth and pigmentation in culture medium. Removal of aerial mycelium in culture by brushing with sterile water so enhanced the conidial production that oatmeal agar medium(OA) could be useful for production of abundant conidia by the treatment.
