  • An optimal design to improve the ride quality was performed with the time and frequency domain analysis based on both of deterministic and random road profiles. The objective function is established to minimize the absorbed power while the constraints are taken so as to satisfy the condition for the stability of vehicle. The result of the optimal design shows that the rms for the acceleration of a driver and his seat is within the critical values for the ride quality from ISO. The optimal values obtained show that the maximum absolute acceleration of the driver and his seat has significantly been reduced and the reference limits on the relative displacement have satisfied their feasibility. As the optimal value according to a specific speed is the results from the optimization process, a global optimum value should be determined to be the one which gives th minimum values of total sum of absorbed power with respect to various speed.

Quantitative Analysis of Cobalt with Continuous Flow Preconcentration Using 1-Nitroso-2-naphthol as an Organic Precipitant (1-Nitroso-2-naphthol 침전제를 사용한 연속흐름 선농축법에 의한 코발트의 정량분석)

  • Trace amount of cobalt in water samples was preconcentrated continuously with an organic precipitant and determined by flame atomic absorption spectrometry. The flow injection technique was used to preconcentrate cobalt by on-line direct precipitation with 1-nitroso-2-naphthol. The precipitation was dissolved with methyl isobutyl ketone (MlBK) and was sent to the flame. The optimum conditions for cobalt determination were determined and used to analyze Co samples. For 1.0 mL of sample, the enrichment factor was 13 and the sample throughput was about lO per hour for 0.5 ppm Co solution. The enrichment factor was increased to 68 fold for 10.0 mL. A semi-reference biologicaI sample was prepared and analyzed. The result was in good agreement with the expected value with RSD of 4%.

Optimization of Growth Environment in the Enclosed Plant Production System Using Photosynthesis Efficiency Model (광합성효율 모델을 이용한 밀폐형 식물 생산시스템의 재배환경 최적화)

  • This study was aimed to assess the effects of microclimate factors on lettuce chlorophyll fluorescent responses and to develop an environment control system for plant growth by adopting a simple genetic algorithm. The photosynthetic responses measurements were repeated by changing one factor among six climatic factors at a time. The maximum Fv'/Fm' resulted when the ambient temperature was $21^{\circ}C,\;CO_2$ concentration range of 1,200 to 1,400 ppm, relative humidity of $68\%$, air current speed of $1.4m{\cdot}s^{-1}$, and the temperature of nutrient solution of $20^{\circ}C$. In PPF greater than $140{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, Fv'/Fm' values were decreased. To estimate the effects of combined microclimate factors on plant growth, a photosynthesis efficiency model was developed using principle component analysis for six microclimate factors. Predicted Fv'/Fm' values showed a good agreement to measured ones with an average error of $2.5\%$. In this study, a simple genetic algorithm was applied to the photosynthesis efficiency model for optimal environmental condition for lettuce growth. Air emperature of $22^{\circ}C$, root zone temperature of $19^{\circ}C,\;CO_2$ concentration of 1,400 ppm, air current speed of $1.0m{\cdot}s^{-1}$, PPF of $430{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, and relative humidity of $65\%$ were obtained. It is feasible to control plant environment optimally in response to microclimate changes by using photosynthesis efficiency model combined with genetic algorithm.

Comparison of Oxidative Stability for the Thermally-oxidized Vegetable Oils using a DPPH Method (DPPH법에 의한 식용유지의 열산화 안정성 비교)

  • The 2,2-diphenyl picrylhydrazyl (DPPH) method, which can be used to predict the oxidative stability of edible oils, was previously reported by our research group. Not only free radical scavenging antioxidants but also radicals from oxidized oils are capable of reacting with DPPH radicals, thereby reducing the absorbance of DPPH. In this study, the optimum sample size of edible oils for the DPPH method was determined, and the oxidation of the edible oils was monitored via DPPH, coupled with other conventional methods. The optimum sample size was determined as 1.5 g using soybean oil. Soybean, corn, virgin olive, and refined olive oils were thermally oxidized for 3 hr at $180^{\circ}C$ and analyzed via DPPH, conjugated dienoic acid (CDA) value, and p-anisidine value (p-AV) protocols. Soybean and corn oils were found to be more sensitive to thermal oxidation than virgin and refined olive oils, on the basis of the CDA value and p-AV measurements. The DPPH method can indicate the inherent radical scavenging activity of unoxidized samples, the time required for the depletion of antioxidants, and the rate of degradation of the antioxidants. The soybean and corn oils evidenced higher levels of free radical scavenging compounds, required more time for the consumption of inherent antioxidants, and also manifested steeper antioxidant degradation rates than olive oils, based on the results of DPPH analysis. The DPPH method, accompanied by other conventional methods, may prove useful in predicting the degree of oxidation of vegetable oils.

  • A sensitive stripping voltammetric study of the complex of indium with 8-hydroxyquinoline at a hanging mercury drop electrode was investigated in 0.1M acetate buffer solution. The effects of various analytical conditions on the reduction peak current of the adsorbed complex were discussed. Optimal analytical conditions were found to be the ligand concentration of $2 {\times}10^{-5}$M, solution pH 4.75, scan rate of 10 mV/s, deposition potential of -0.450V, a deposition time of 90 second. Interferences by other trace metals and Triton X-100 were also discussed. Detection limit was 0.2 ppb of indium after 90 sec. Deposition time, and the relative standard deviation(n = 10) at 4 ppb was 3.2%.

  • Spectrophotometric determination of cobalt by flow injection method is described. 2-(5-Bromo-2-pyridylazo)-5-(N-propyl-N-sulfopropylamino) aniline rapidly forms a water-soluble complex with cobalt in $NH_3-NH_4Cl$ buffer solution at pH 10.5. The absorption maxima of this complex is at 545 nm with molar absorptivity of $58000L\;mol^{-1}\;cm^{-1}$. The calibration curve of cobalt is linear over the range of 0.1 to 0.6ppm and the detection limit is 25ppb. The relative standard deviation is ${\pm}0.72%$ for 0.5ppm and the sampling rate is $60samples\;hr^{-1}$. The interfering effect of some cations and anions was investigated. Ni(II), Cu(II), Fe(III) and $CN^-$ interfered severely. The interfering effect of these matallic ions could be decreased by adding $1.0{\times}10^{-3}M$ EDTA solution to the carrier stream.

Water wave reflection over shear currents and dredged multi-arrayed trenches (외부 흐름과 준설된 다열 함몰지형에 의한 파랑의 반사)

  • Understanding the wave characteristics near the outlet of coastal power plants for cooling water in the vicinity of the dredged areas is critically important for the construction and operation of the plants. By Employing the eigenfunction expansion method, in this study, we analyzed the reflection of monochromatic water waves over (1) shear currents near the outlet and (2) multi-arrayed trenches representing dredged areas. We firstly optimized the number of grids expressing shear currents and the number of evanescent modes based on a convergence test. We then analyzed the sensitivity of the reflection coefficients depending on (1) magnitude of shear currents, (2) width of shear currents, (3) a distance between adjacent trenches, and (4) a number of trenches. The results showed that the reflection coefficient was more sensitive to the number of trenches and the distance between trenches than the velocity of shear currents and the width of shear currents. We also found that even the effect of shear currents is relatively small, the effect is not negligible in a relative water depth from shallow to near shallow water waves (0.01 < kh ≦ 0.70).

수소처리와 후성장층의 특성이 다이아몬드 박막의 전계방출 특성에 미치는 영향

  • 화학증착법으로 증착된 다이아몬드 박막은 우수한 전기적 특성과 뛰어난 화학적, 열적 안정성 때문에 전계방출소재로 많은 관심을 불러 일으키고 있다. 다이아몬드 박막의 전계방출은 저전계에서 일어나는 것으로 알려져 있으며, 저전계방출의 원인을 규명하려는 많은 연구가 진행되어 왔다. 한편, 다이아몬드 박막의 전계방출전류는 금속기판의 사용에 의한 기판/다이아몬드 접촉의 개선, 다이아몬드 박막내의 흑연성분의 조절에 의한 구조변화, 보론이나 인 (P), 질소의 도핑, 수소 플라즈마나 cesium 등의 금속을 이용한 표면처리 등의 여러 방법에 의하여 향상된다는 것이 입증되었다. 그 외에 메탄과 대기 분위기 처리, 암모니아 분위기에서의 레이저 조사도 전계방출특성을 향상시키는 것으로 보고되었다. 그러나, 다이아몬드 박막의 성장후 구조적 특성이 다른 박막의 후성장이나 열분해된 운자수소 처리가 다이아몬드 박막의 전계방출특성에 미치는 영향에 관한 연구는 지금까지 이루어지지 않았다. 본 연구에서는 수소처리와 후성장이 다이아몬드 박막의 전계방출특성에 미치는 영향을 고찰하고 이로부터 그 원인을 규명하고자 하였다. 다이아몬드 박막은 hot-filament 화학증착법을 이용하여 증착하였다. 후성장한 다잉아몬드 박막내의 흑연성분과 박막의 두께를 체계적으로 조절하여 후성장 박막의 구조적 특성과 그 두께의 영향을 확인할 수 있었다. 후성장층내의 흑연성분과 두께가 증가할수록 전계방출특성은 향상되다가 저하되었다. 한편, 다이아몬드 박막을 성장시킨 후 수소분위기 처리를 함에 따라 전계방출특성은 향상되었지만 수소처리시간이 5분 이상으로 증가함에 따라 그 특성은 저하되었다. 본 연구에서는 수소처리와 후성장시 나타나는 전계방출특성의 변화 원인을 규명하고자 한다.기판위에서 polymer-like Carbon 구조는 향상되는 경향을 보였다.0 mm인 백금 망을 마스크로 사용하여 실제 3차원 미세구조를 제작하여 보았다. 그림 1에서 제작된 구조물의 SEM 사진을 보여주었으며, 식각된 면의 조도가 매우 뛰어나며 모서리의 직각성도 우수함을 확인할 수 있다. 이와 같이 도출된 시험 조건을 기초로 하여 리소그래피 후에 전기 도금을 이용한 금속 몰드 제작 및 이온빔 리소그래피 장점을 최대한 살릴수 있는 미세구조 제작에 대한 연구를 계속 추진할 계획이다. 비정질 Si1-xCx 박막을 증착하여 특성을 분석한 결과 성장된 박막의 성장률은 Carbonfid의 증가에 따라 다른 성장특성을 보였고, Silcne(SiH4) 가스량의 감소와 함께 박막의 성장률이 둔화됨을 볼 수 있다. 또한 Silane 가스량이 적어지는 영역에서는 가스량의 감소에 의해 성장속도가 둔화됨을 볼 수 있다. 또한 Silane 가스량이 적어지는 영역에서는 가스량의 감소에 의해 성장속도가 줄어들어 성장률이 Silane가스량에 의해 지배됨을 볼 수 있다. UV-VIS spectrophotometer에 의한 비정질 SiC 박막의 투과도와 파장과의 관계에 있어 유리를 기판으로 사용했으므로 유리의투과도를 감안했으며, 유리에 대한 상대적인 비율 관계로 투과도를 나타냈었다. 또한 비저질 SiC 박막의 흡수계수는 Ellipsometry에 의해 측정된 Δ과 Ψ값을 이용하여 시뮬레이션한 결과로 비정질 SiC 박막의 두께를 이용하여 구하였다. 또한 Tauc Plot을 통해 박막의 optical band gap을 2.6~3.7eV로 조절할 수 있었다. 20$0^{\circ}C$이상으로 증가시켜도 광투과율은 큰 변화를 나타내지 않았다.부터

Increased Production of an Alkaline Protease from Bacillus clausii I-52 by Chromosomal Integration (Bacillus clausii I-52의 Chromosomal Integration에 의한 Alkaline Protease의 생산성 향상)

  • TTo increase productivity of a strong extracellular alkaline protease (BCAP), stable strains of Bacillus clausii I-52 carrying another copy of BCAP gene in the chromosome were developed. Integrative vector, pHPS9-fuBCAP carrying BCAP promoter, ribosome binding site, signal sequence and active protease gene was constructed and transferred into B. clausii I-52, and integration of the constructed plasmid into chromosome was identified by PCR. An investigation was carried out on BCAP production by B. clausii I-52 and transformant C5 showing the highest relative activity of alkaline protease using submerged fermentation. Maximum enzyme activity was produced when cells were grown under the submerged fermentation conditions at $37^{\circ}C$ for 48 h with an aeration rate of 1 vvm and agitation rate of 650 rpm in a optimized medium (soybean meal 2%, wheat flour 1%, sodium citrate 0.5%, $K_2HPO_4$ 0.4%, $Na_2HPO_4$ 0.1%, NaCl 0.4%, $MgSO_47H_2O$ 0.01%, $FeSO_47H_2O$ 0.05%, liquid maltose 2.5%, $Na_2CO_3$ 0.6%). A protease yield of approximately 134,670U/ml was achieved using an optimized media, which show an increase of approximately 1.6-fold compared to that of non-transformant (83,960 U/ml). When the stability of transformant C5 was examined, the integrated plasmid pHPS9-fuBCAP was detected in the transformant after cultivation for 8 days, suggesting that it maintained stably in the chromosomal DNA of transformant C5.

Studies on the ${\beta}-Tyrosinase$ -Part 2. On the Synthesis of Halo-tyrosine by ${\beta}-Tyrosinase$- (${\beta}-Tyrosinase$에 관한 연구 -제2보 ${\beta}-Tyrosinase$에 의한 Halogen화(化) Tyrosine의 합성(合成)-)

  • L-Tyrosine, 2-chloro-L-tyrosine, 2-bromo-L-tyrosine, and 2-iodo-L-tyrosine were synthesized by ${\beta}-tyrosinase$ obtained from cells of Escherichia intermedia A-21, through the reversal of the ${\alpha},{\beta}-elimination$ reaction, and their molecular structures were analyzed by element analysis, NMR spectroscopy, mass spectrometry and IR spectroscopy. Rates of synthesis and hydrolysis of halogenated tyrosines by ${\beta}-tyrosinase$, inhibition of the enzyme activity by halogenated phenols, and effects of addition of m-bromophenol on the synthesis of 2-bromotyrosine were determined. The results obtained were as follows: 1) In the synthesis of halogenated tyrosines, the yield of 2-chlorotyrosine from m-chlorophenol were approximately 15 per cent, that of 2-bromotyrosine from m-bromophenol 13.8 per cent, and that of 2-iodotyrosine from m-iodophenol 9.8 per cent. 2) Rate of synthesis of halogenated tyrosines by ${\beta}-tyrosinase$ was slower than that of tyrosine and the rates were decreased in the order of chlorine, bromine and iodine, that is, by increasing the atomic radius. Relative rate of 2-chlorotyrosine synthesis was determined to be 28.2, that of 2-bromotyrosine to be 8.13, and that of 2-iodotyrosine to be 0.98, respectively, against 100 of tyrosine. However 3-iodotyrosine was not synthesized by the enzyme. 3) The relative rate of 2-chlorotyrosine hydrolysis by ${\beta}-tyrosinase$ was 70.7, that of 2-bromotyrosine was 39.0, and that of 2-iodotyrosine was 12.6 against 100 of tyrosine, respectively. The rate of hydrolysis appeared to be decreased in the order of chlorine, bromine and iodine, that is, by increasing the atomic radius or by decreasing the electronegativity. But 3-iodotyrosine was not hydrolyzed by the enzyme. 4) The activity of ${\beta}-tyrosinase$ was inhibited by phenol markedly. Of the halogenated phenols, o-, or m-chlorophenol and o-bromophenol gave marked inhibition on the enzyme action, however inhibition by iodophenol was not strong. Plotting by Lineweaver-Burk method, a mixed-type inhibition by m-chlorophenol was observed and its Ki value was found to be $5.46{\times}10^{-4}M$. 5) During the synthesizing reaction of 2-bromotyrosine by the enzyme, sequential addition of substrate which was m-bromophenol with time intervals and in a small amount resulted in better yield of the product. 6) The halogenated tyrosines which were produced by ${\beta}-tyrosinase$ from pyruvate, ammonia and m-halogenated phenols were analysed to determine their molecular structures by element analysis, NMR spectroscopy, mass spectrometry, and IR spectroscopy. The result indicated that they were 2-chloro-L-tyrosine, 2-bromo-L-tyrosine, and 2-iodo-L-tyrosine, respectively.
