In Vitro Properties and Biodistribution of Tc-99m and Re-188 Labeled Monoclonal Antibody CEA79.4 (Re-188과 Tc-99m 표지 단일클론항체 CEA79.4의 생체외 특성과 생체내 분포)

  • Purpose: Radiolabeled CEA79.4 antibody has a possibility to be used in radioimmunoscintigraphy or radioimmunotherapy of cancer. We investigated the in vitro properties and biodistribution of CEA79.4 antibody labeled with Re-188 or Tc-99m. Materials and Methods: CEA79.4 was reduced by 2-mercaptoethanol to produce-SH residue, and was labeled with Re-188 or Tc-99m. For direct labeling of Tc-99m, methylene-diphosphonate was used as transchelating agent. CEA79.4 in 50 mM Acetate Buffered Saline (ABS, pH 5.3) was labeled with Re-188, using stannous tartrate as reducing agent. In order to measure immunoreactivity and the affinity constant of radiolabeled antibody, cell binding assay and Scatchard analysis using human colon cancer cells SNU-C4, were performed. Biodistribution study of labeled CEA79.4 was carried out at 1, 14 and 24 hr in ICR mice. Results: Labeling efficiencies of Tc-99m and Re-188 labeled antibodies were $92.4{\pm}5.9%$ and $84.7{\pm}4.6%$, respectively, In vitro stability of Tc-99m-CEA79.4 in human serum was higher than Re-188-CEA79.4. Immunoreactivity and affinity constant of Tc-99m-CEA79.4 were 59.2% and $6.59{\times}10^9\;M^{-1}$, respectively, while those of Re-188-CEA79.4 were 41.6% and $4.2{\times}10^9\;M^{-1}$, respectively. After 24 hr of administrations of Re-188 and Tc-99m labeled antibody, the remaining antibodies in blood were 6.32 and 9.35% ID/g respectively. The biodistribution of each labeled antibody in other organs was similar because they did not accumulate in non-targeted organs. Conclusion: In vitro properties and biodistribution of Re-188-CEA79.4 were similar to those of Tc-99m-CEA79.4. It appears that Re-188-CEA79.4 can be used as a suitable agent for radioimmunotheraphy.

Effects of Dietary Supplementation with Rosemary and α-Tocopherol Acetate on Performance and Meat Quality of Chicken Meat during Refrigerated Storage (로즈마리와 α-Tocopherol Acetate의 급여가 육계의 생산성 및 냉장 저장 중 계육의 품질에 미치는 영향)

  • The effects of rosemary and $\alpha$-tocopherol, added individually or in combination, on broiler performance, thiobarbituric acid reactive substance (TBARS), total plate count (TPC) and meat color of chicken thigh meat were investigated. Three hundred broiler chicks divided into five groups were fed a basal diet (control) or basal diet supplemented with 5 g rosemary/kg (T1), 10 g rosemary/kg (T2), 200 mg $\alpha$-tocopherol/kg (T3), or 5 g rosemary/kg + 200 mg $\alpha$-tocopherol/kg (T4) for 5 weeks. Following slaughter, chicken meat was stored at $4^{\circ}C$ for 10 days. All treatments did not influence the performance. Rosemary supplementation delayed lipid oxidation in thigh meat during refrigerated storage. T2 was significantly (p<0.05) more effective in delayed lipid oxidation compared to T1, but was inferior to T3. Samples containing a combination of antioxidant had lower TBARS values than those containing the individual antioxidants, indicating a synergistic effect. TPC was significantly increased (p<0.05) in thigh meat of all groups throughout the refrigerated storage. The T3 and control groups showed TPC counts that did not differ from each other during the entire storage period. However, rosemary supplementation was associated with bacterial counts that were significantly lower (p<0.05) than the control and $\alpha$-tocopherol groups at day 3 of storage and thereafter. For this period, T1 presented TPC counts that were significantly higher than the T2 group (p<0.05). At all storage times, the thigh meat of rosemary-fed chickens was redder than control (higher $a^*$), while no differences in $L^*$ and $b^*$ values were found. A synergistic effect was obtained from the combination of rosemary with $\alpha$-tocopherol, whereas individual use of the antioxidants significantly improved color stability compared to the control.

Characterization of Laccase Purified from Korean Trametes hirsuta S1 (한국산 흰구름버섯(Trametes hirsuta S1)으로부터 정제된 Laccase의 특성)

  • Laccase produced by Trametes hirsuta S1 isolated from Korea was partially purified and characterized using ultrafiltration, anion exchange chromatography and affinity chromatography. The laccase was produced as the predominant extracellular enzyme during primary metabolism. Neither lignin peroxidase nor veratryl alcohol oxidase (VAO) were detected in the culture fluid. Addition of 2,5-xylidine enhanced 4-fold laccase production. Purified laccase was a single polypeptide having a molecular mass of approximately 66 kDa, as determined by SDS-polyacrylamide gel electrophoresis, and carbohydrate content of 12%. $K_{m}\;and\;V_{max}$ values for laccase with ABTS [2,2-azinobis (3-ethylbenzthiazoline 6-sulfonic acid)] as a substrate (Lineweaver-Burk plot) was determined to $51.2\;{\mu}M\;and\;56.8\;{\mu}mole$, respectively. The optimal pH for laccase activity was found to be 3.0. The enzyme was very stable for 1 hour at $50^{\circ}C$. Half-life ($t_{1/2}$) of the enzyme was about 20 min at $70^{\circ}C$. Spectroscopic analysis of purified enzyme indicated that the enzyme was typical of copper-containing protein. Substrate specificity and inhibitor studies for laccase also indicated to be a typical fungal laccase. The N-terminal amino acid sequence of the T. hirsuta S1 laccase showed 100% of homology to those of laccase from C. hirsutus.

Effect of Various Interlayer Deposition on Room Temperature and High Temperature Properties of CrAlN Coatings (다양한 중간층 증착이 CrAlN 코팅의 상온 및 고온 특성에 미치는 효과에 관한 연구)

  • CrAlN 코팅은 높은 경도, 낮은 표면 조도 등의 상온에서의 우수한 기계적 특성 이외에 고온에서 안정한 합금상의 형성으로 인하여 우수한 내산화성 및 내열성을 보유하여 공구 코팅으로의 적용 가능성이 크다. 그러나 최근 공구사용 환경의 가혹화로 인하여 코팅의 내마모성 및 내열성 등의 물성 향상을 통한 공구의 수명 향상이 필요시 되고 있으며, 다양한 코팅 물질을 활용하여 다층 코팅을 합성함으로써 난삭재용 공구 코팅의 물성을 높이는 연구들이 진행되고 있다. 본 연구에서는 CrAlN 코팅과 WC-Co 6wt.% 모재 사이에 CrZrN, CrN, CrN/CrZrSiN 등의 중간층을 합성하여 CrAlN 코팅의 상온 및 고온 특성을 향상시키는 연구가 진행되었다. 합성된 코팅의 구조 및 물성을 분석하기 위해 field emission scanning electron microscopy(FE-SEM), nano-indentation, atomic force microscopy(AFM) 및 ball-on-disk wear tester를 사용하였다. 코팅의 고온 특성을 확인하기 위해 코팅을 furnace에 넣어 공기중에서 30분 동안 annealing 한 후에 nano-indentation을 사용하여 경도를 측정하였고, $500^{\circ}C$ annealing 코팅의 표면 조도 분석 및 $500^{\circ}C$에서 마찰마모시험을 실시하였다. CrAlN 코팅의 상온 특성을 분석한 결과, 모든 코팅의 경도(35.5-36.2 GPa)와 탄성계수(424.3-429.2 GPa)는 중간층의 종류에 상관없이 비슷한 값을 보인 것으로 확인됐다. 그러나, CrN/CrZrSiN 중간층을 증착한 CrAlN 코팅의 마찰계수는 0.33로 CrZrN 중간층을 증착한 CrAlN 코팅의 마찰계수(0.41)에 비해 향상된 값 보였으며, 코팅의 마모율 및 마모폭도 비슷한 경향을 보인 것으로 보아 코팅의 내마모성이 향상된 것으로 판단된다. 이것은 중간층의 H/E ratio가 코팅의 내마모성에 미치는 영향에 의한 결과로 사료된다. H/E ratio는 파단시의 최대 탄성 변형율로써, 모재/중간층/코팅의 H/E ratio 구배에 따라 코팅 내의 응력의 완화 정도가 변하게 된다. WC 모재 (H/E=0.040)와 CrAlN 코팅(H/E=0.089) 사이에서 CrN, CrZrSiN 중간층의 H/E ratio는 각각 0.076, 0.083 으로 모재/중간층/코팅의 H/E ratio 구배가 점차 증가함을 확인 할 수 있었고, 일정 응력이 지속적으로 가해지면서 진행되는 마모시험중에 CrN과 CrZrSiN 중간층이 WC와 CrAlN 코팅 사이에서 코팅 내부의 응력구배를 완화시키는 역할을 함으로써 CrAlN 코팅의 내마모성이 향상된 것으로 판단된다. 내열성 시험 결과, CrN/CrZrSiN 중간층을 증착한 코팅은 $1,000^{\circ}C$까지 약 28GPa의 높은 경도를 유지한 것으로 확인 되었다. $500^{\circ}C$ annealing 후 진행된 표면 조도와 마모시험 결과, 모든 코팅의 조도 값 및 마찰계수는 상온 값에 비해 증가하였으며 CrN/CrZrSiN 중간층을 증착한 CrAlN 코팅의 변화량이 가장 낮은 값을 보였다. 이는 CrZrSiN 중간층 내에 존재하는 $SiN_x$ 비정질상이 고온 annealing시에 산소 차폐막 역할을 하여, 코팅내의 잔류 산소에 의한 산화작용을 효과적으로 방지함으로써 코팅의 고온 특성이 향상된 것으로 판단된다.

Production and Characterization of Cyclodextrin Glucanotransferase fronm Bacillus sp. JK-43 Isolated from Kimchi (김치 분리균인 Bacillus sp. JK-43이 생산하는 Cyclodextrin Glucanotransferase의 생산 및 특성)

  • A bacterial strain, designated as JK-43, producing extracellular cyclodextrin glucanotransferase (CGTase)[EC 2.4.1.19] was isolated from kimchi. The CGTase from isolated strain JK-43 showed the transglucosylation activity from soluble starch to L-ascorbic acid(AA) compared to those obtained from other strains. A main product formed by this reaction was identified as $2-O-{\alpha}-glucopyranosyl$ L-ascorbic acid(AA-2G) by testing its susceptibility to ${\alpha}-glucosidase$ hydrolysis, the HPLC profiles, and through the elementary analysis. the ${\beta}-CD,\;{\gamma}-CD$, potato starch and corn starch were identified to be suitable glucosyl donor for transglucosylation reaction on AA by CGTase. Acceptor specificity on AA-2G production was examined by use of AA, Iso-AA and AA-2P. Transglucosylation was observed toward AA-2P as well as AA and Iso-AA. The microorganism isolated from kimchi was identified as a strain of Bacillus sp. JK-43 based on the morphological, cultural, biochemical characteristics and partial 16SrDNA sequence analysis. The maximal CGTase production was observed in a medium containing 1.0% soluble starch, 1.0% yeast extract, 1.0% $Na_2CO_3\;0.1%\;K_2HPO_4,\;and\;0.02%\;MgSO_4{\cdot}7H_2O$ with initial pH 7.0. The strain was cultured at $37^{\circ}C$ for 26 hrs with reciprocal shaking.

  • A new peanut variety "Seonan" (Arachis hypogaea ssp. hypogaea L.) was developed at the Yeongnam Agricultural Research Institute, NICS, in Milyang in 2007. It was developed from the cross between the erect short stem cultivar "SP9128" and the high-yielding cultivar "Daekwang". "Seanan" which is Virginia plant type has 18 branch number per plant with early maturing and long-ellipse shaped large kernel. Each pod has two grains with brown testa and 100 seed weight was 87 g in the regional yield trials (RYT). Especially this variety has resistance to lodging owing to short stem. The free sugar and tannin content of fresh peanut are 3.7% and 0.48%, respectively, similar to check variety. In the regional yield trials "Seonan" was outyielded than check variety by 14% with 8.59 MT/ha for fresh pod and by 7% with 3.75 MT/ha for grain.

  • Thermal batteries are also called molten-salt batteries as the electrolyte is mainly composed of molten salt. The molten-salt electrolyte is a solid that does not conduct electricity at room temperature, but when it is melted by a pyrotechnic heat source, it becomes an excellent ionic conductor. Thermal batteries are a kind of pyrotechnic battery because they operate only when the solid electrolyte is melted by the heat energy provided by pyrotechnic materials. Pyrotechnic components used in a thermal battery include heat sources, fuse strips, and an igniter. The reliability of these pyrotechnic components critically affects the reliability and performance of the battery that must supply electricity stably to guided munitions even under extreme environmental conditions. Different igniter types offer different advantages: notch-type igniters offer improved ignition probability, whereas film-type igniters offer improved safety. The addition of metal oxides to the heat paper could improve the burn rate, and the ignition reliability could be greatly improved by using it with a flame igniter at the same time. Using a two-step reduction process, high-purity Fe particles in coral form can be safely obtained.

Nanofibers Comprising Mo2C/Mo2N Nanoparticles and Reduced Graphene Oxide as Functional Interlayers for Lithium-Sulfur Batteries (Mo2C/Mo2N 나노 입자와 환원된 그래핀 옥사이드가 복합된 나노 섬유 중간층이 적용된 리튬-황 전지)

  • Nanofibers comprising reduced graphene oxide (rGO) and Mo2C/Mo2N nanoparticles (Mo2C/Mo2N rGO NFs) were prepared for a functional interlayer of Li-S batteries (LSBs). The well-dispersed Mo2C and Mo2N nanoparticles in the nanofiber structure served as active polar sites for efficient immobilization of dissolved lithium polysulfide. The rGO nanosheets in the structure also provide conductive channels for fast ion/electron transport during charging-discharging and ensured reuse of lithium polysulfide during redox reactions through a fast charge transfer process. As a result, the cell assembled with Mo2C/Mo2N rGO NFs-coated separator and pure sulfur electrode (70 wt% of sulfur content and 2.1 mg cm-2 of sulfur loading) showed a stable discharge capacity of 476 mA h g-1 after 400 charge-discharge cycles at 0.1 C. Furthermore, it exhibited a discharge capacity of 574 mA h g-1 even at a high current density of 1.0 C. Therefore, we believe that the proposed unique nanostructure synthesis strategy could provide new insights into the development of sustainable and highly conductive polar materials as functional interlayers for high performance LSBs.

Effect of Dietary Supplementation of Quercetin on Oxidative Stability of Chicken Thigh (Quercetin의 급여가 육계 다리육의 산화 안정성에 미치는 영향)

  • This study was carried out to determine the effect of dietary supplementation of quercetin and methoxylated quercetin extracted from onions on oxidation of chicken thigh during cold storage. For 35 days, 1-day-old 320 broiler chicks (Ross) were divided into 8 groups and supplemented the diet; basal diet only (T1), T1 with antibiotics (T2), vitamin E 20 IU (T3), vitamin E 200 IU (T4), quercetin 20 ppm (T5), quercetin 200 ppm (T6), methoxylated quercetin 20 ppm (T7), methoxylated quercetin 200 ppm (T8). After slaughtering the broilers, thigh meats were collected and analyzed the oxidation stability during storage at $4^{\circ}C$ for 7 days. Volatile basic nitrogen (VBN) was ranged from 10.98~15.37 mg% until day 3 and quercetin 20 ppm (T5) significantly reduced (p<0.05) the VBN value of thigh as much as antibiotics treatment group (T2). The supplementation of quercetin at a 200 ppm level significantly reduced 2-thiobarbituric acid reactive substances value when compared with the control. Supplementation of 20 ppm quercetin also affected fatty acid composition of chicken thigh and resulted in increase of C18:1w9 and decrease of C18:2w6 on day 3. In microbes of chicken thigh during storage, supplementation of 20 ppm quercetin significantly reduced total microbes compare to control on day 7 (p<0.05). There was no effect on the number of coliforms in thigh with quercetin diet. From these results, the dietary supplementation of 20 ppm of quercetin may have effects on enhancing oxidation stability.

Effects of Dietary Vitamin C and E on Lipid Oxidation and Stability of Color in Hanwoo Steer Beef (비타민 C 및 E 급여가 한우 거세우 육의 지방산화 및 육색 안정성에 미치는 영향)

  • This experiment was earned out to investigate the effects of dietary vitamin C and E on lipid oxidation and stability of color in Hanwoo steer beef. Thirty seven Hanwoo steers were randomly assigned to one of four treatments. The four treatments were control(vitamin E 20IU/kg feed), vitamin C(vitamin C 0.l%/kg feed and vitamin E 20IU/kg feed), vitamin E(vitamin E 220IU/kg feed) and vitamin C plus E(vitamin C 0.l%/kg feed and vitamin E 20IU/kg feed). Hanwoo steers were fed the experimental diets for 3 months at the last finishing period and pH, color, TBARS, concentration of myoglobin, water holding capacity, reduction by heat, drip loss and purge loss of longissimus dorsi were analyzed at 1, 3, 5, 7 and 10th day of storage, respectively. pH value, Hue angle, metmyoglobin concentration, TBARS, drip loss and purge loss in longissimus dorsi were increased as the storage period elapsed. However, water holding capacity in longissimus dorsi was decreased as the storage period elapsed. The animals fed the vitamin C plus E diet were higher in pH value and oxymyoglobin concentration, and lower in metmyoglobin concentration than in those fed the other diets during the whole storage period and lowest in TBARS. TBARS and Hue angle in longissimus dorsi were 10wer(P<0.05) in the animals fed the vitamin-supplemented diets than in those fed the control diet. Reduction by heat in longissimus dorsi was higher(P<0.05) in the animals fed the diets supplemented with vitamin E or vitamin C plus E than in those fed the control diet or vitamin C-supplemented diet. According to these results, it may be concluded that feeding the diets containing 0.1% vitamin C or 220IU vitamin E per kg feed or the diet mixed 0.1% vitamin C with 220IU vitamin E to Hanwoo steers can retard lipid oxidation of longissimus dorsi and stabilize beef color.
