Identification of a Single Nucleotide Polymorphism (SNP) Marker for the Detection of Enhanced Honey Production in Hoenybee (수밀력 우수 꿀벌 계통 판별을 위한 계통 특이 분자마커 개발)

  • Honeybees (Apis mellifera) are common pollinators and important insects studied in agriculture, ecology and basic research. Recently, RDA (Rural Development Administration) and YIRI (Yecheon-gun Industrial Insect Research Institute) have been breeding a triple crossbred honey bee named Jangwon, which have the ability to produce superior quality honey. In this study, we identified a single nucleotide polymorphism (SNP) marker in the genome of Jangwon honeybee, particularly, in the paternal line (D line). Initially, we performed Sequence-Based Genotyping (SBG) using the Illumina Hiseq 2500 in 5 honeybee inbred lines; A, C, D, E, and F; and obtained 1,029 SNPs. Seventeen SNPs for each inbred line were generated and selected after further filtering of the SNP dataset. The 17 SNP markers validated by performing TaqMan probe-based real-time PCR and genotyping analysis was conducted. Genotyping analysis of the 5 honeybee inbred lines and one hybrid line, $D{\times}F$, revealed that one set of SNP marker, AmD9, precisely discriminated the inbred line D from the others. Our results suggest that the identified SNP marker, AmD9, is successful in distinguishing the inbred honeybee lines D, and can be directly used for genotyping and breeding applications.

Control Effect of Botanical Pyrethrum to Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae) in Green Perilla (잎들깨에서 차먼지응애에 대한 천연 피레스럼 방제효과)

  • We investigated an ecologically-sensitive method to control Polyphagotarsonemus latus in a green perilla polyvinyl house using concentrated pyrethrum. The survival rates of Polyphagotarsonemus latus nymphs and adults to 2%, 4%, and 6% pyrethrum 3 days after spraying were 58.1%, 27.5%, and 22.7% respectively, and 73.4%, 37.3%, and 30.6% at 5 days after spraying. These results show that the most effective control occurred using 6% pyrethrum. To investigating the densities of P. latus on a 1 cm diameter leaf-disk of green perilla, we sprayed the leaf every 10 days from May 10 with 6% pyrethrum 1,000 times and milbemectin EC 2% 1,000 times. The period chosen was one where the incidence of P. latus could be predicted. The density was kept low during the treatment period. However, if leaf damage had already occurred from P. latus, the density could not be decreased by spraying 1 or 2 times each week where milbemectin EC 2% was used, but the population could be reduced when sprayed 3 times. Therefore, in order to effectively control P. latus in green perilla greenhouses, it is important to begin treatment at an early stage when P. latus are first observed.

  • The populations of Polyphagostarsonemus latus (Acari: Tarsonemidae) have been causing serious economic damages to farmers by directly feeding on leaves of green perilla. However, sampling of P. latus is impossible because there is no suitable sample unit until now. Thus, this study was conducted to suggest suitable sample unit and required sample number for P. latus on green perilla. Individuals of P. latus started to occurr from the leafstalk within a leaf. They required approximated 25 days to spread to all areas within a leaf. New shoots would be a suitable sample unit for P. latus on green perilla because its density on shoots per ㎠ was always higher than the densities on fully grown and moderately grown leaves. Required sample number at D = 0.25 was estimated as a shoot when eggs, nymphs, and adults of P. latus were observed within a leaf. Therefore, Using this small sample unit for sampling of P. latus on green perilla could be economical and efficient.

  • Humans have been breeding the mulberry silkworm for the long period of time to obtain silk fabric and nutrient-rich pupae. Currently, silkworm larvae, pupae, and silk-Fibroin hydrolysates are registered as food raw materials, while silkworm feces and Bombyx batryticatus are registered as Korean traditional medicines. Among sericulture products, individually recognized health functional food ingredients include silk-protein acid-hydrolysates for immunity enhancement, Fibroin-hydrolysates for memory improvement, and freeze-dried 5th instar and 3rd-day-silkworm powder for lowering-blood sugar. Recently, HongJam produced by steaming and freeze-drying mature silkworms were reported to have various health-promoting effects such as preventing the onset of Alzheimer's disease and Parkinson's disease, enhancing gastro-intestinal functions, improving skin-whitening and hair growth, and extending healthspan. By consuming silkworm products with various health-promoting effects, it is possible to increase the healthspan of human beings, thereby reducing personal and national medical expenses, resulting in increasing the individual's happiness.

Establishment of Optimal Rearing Conditions for the Production of Tenebrio molitor Larvae (갈색거저리 유충 생산을 위한 효율적인 사육조건 확립)

  • Tenebrio molitor larvae contain large amounts of proteins, lipids and other functional materials, enabling this insect to be used as an edible food source in animal feeds and for industrialization. Although many efforts have been made to set up mass rearing systems, few studies have been conducted to establish optimal rearing conditions for the production of high quality T. molitor larvae. Herein we investigated 1) the effects of additional diets on the survival and fecundity of the insect, 2) the relationship between oviposition period and the uniformity of larval size, 3) the effects of rearing density and temperature on insect development, and 4) the storage stability of eggs and pupae at low temperatures given possible temporary production discontinuation. The addition of carrot and zucchini to the traditional wheat bran diet significantly increased the survival and fecundity rate of adult T. molitor. Of the three different oviposition sampling periods (3, 7, and 14 days) used to investigate the uniformity of the hatched larvae in each treatment, the period of 3 and 7 days provided higher uniformity than the 14 days oviposition period. Larval development was faster at $30^{\circ}C$ than at 20, 25, and $35^{\circ}C$. Interestingly, oviposition rates were highest at $20^{\circ}C$ but showed much slower larval development and lower uniformity at $30^{\circ}C$. Regarding the effect of larval rearing densities (1, 10, 20, 30, 40, and 50 larvae per 90 mm diam. dish), larval weight was significantly reduced at higher rearing densities, but larval longevity and length were not influenced by rearing density. The 30 larvae/dish is suggested to be a reasonable density to be applied to mass production systems. When kept at $4^{\circ}C$, T. molitor eggs showed a significant reduction in hatching rate; however, when stored under the same conditions, pupae emergence rates remained high until 10 weeks, suggesting that storage at low temperatures is more suitable for the pupal stage than the egg stage. Our findings suggest that an increase in T. molitor adult survival and fecundity rates and a uniformity of hatched larval development can be achieved with the following recommendations: a combination diet (including wheat bran), a 7-day oviposition period; a larvae-rearing temperature of $30^{\circ}C$, a rearing density of 30 larvae/dish, and the storage of pupal stages at low temperatures in the case of rearing discontinuation. This study serves as a strong foundation for the successful mass production of high quality T. molitor larvae.

  • The cowpea aphid Aphis craccivora Koch (Hemiptera: Aphididae) is a polyphagous species with a worldwide distribution. We investigated the temperature effects on development periods of nymphs, and the longevity and fecundity of apterous female of A. craccivora. The study was conducted at six constant temperatures of 10.0, 15.0, 20.0, 25, 30.0, and $32.5^{\circ}C$. A. craccivora developed successfully from nymph to adult stage at all temperatures subjected. The developmental rate of A. craccivora increased as temperature increased. The lower developmental threshold (LT) and thermal constant (K) of A. craccivora nymph stage were estimated by linear regression as $5.3^{\circ}C$ and 128.4 degree-days (DD), respectively. Lower and higher threshold temperatures (TL, TH and TH-TL, respectively) were calculated by the Sharpe_Schoolfield_Ikemoto (SSI) model as $17.0^{\circ}C$, $34.6^{\circ}C$ and $17.5^{\circ}C$. Developmental completion of nymph stages was described using a three-parameter Weibull function. Life table parameters were estimated. The intrinsic rate of increase was highest at $25^{\circ}C$, while the net reproductive rate was highest at $20^{\circ}C$. Biological characteristics of A. craccivora populations from different geographic areas were discussed.

A Loop-mediated Isothermal Amplification Method for White-backed Planthopper-specific Detection (고리매개등온증폭법(LAMP)을 이용한 흰등멸구 특이 판별법)

  • A loop-mediated isothermal amplification (LAMP) primer set (WBPH-65) was designed for the species-specific detection of white-backed planthopper (WBPH) Sogatella furcifera based on the full-length sequence of the internal transcribed spacer 2 (ITS2) (KC417469.1). The WBPH-65 primer set consists of six primers (total 165 bp), F3 (18 bp), B3 (18 bp), FIP (43 bp), BIP (40 bp), LF (21 bp), and LB (25 bp). After the LAMP reaction of three rice planthoppers, S. furcifera, Nilaparvata lugens, and Laodelphax striatellus, with the WBPH-65 primer set for 60 min at $65^{\circ}C$, the LAMP products were observed in the genomic DNA of S. furcifera only. According to the DNA amount of S. furcifera and incubation duration at $65^{\circ}C$, the difference of fluorescence relative to the negative control (0 ng) was clearly observed in a 40-min incubation with 10 and 100 ng or in case of 60-min incubation with 0.01, 0.1, 1, 10, and 100 ng. There was little difference in fluorescence between the negative control and all the other DNAs tested in 20- and 30-min incubations. On the other hand, the WBPH-65 primer set without LF and LB primers showed little amplification in the genomic DNAs of the three rice planthoppers, S. furcifera, N. lugens, and L. striatellus in a 60-min incubation. These results suggest that all six primers (F3, B3, FIP, BIP, LF, and BF) are necessary for the WBPH-65 primer set to detect S. furcifera within a 60-min incubation, and is able to discriminate S. furcifera from at least N. lugens and L. striatellus.

  • The sweet potatoes infected with witches'-broom disease were collected in the growing fields in Jeon-bug area, Korea. As a possible control plant, Ipomoea batatas L.var. Suwon 147 was selected. The pathogen was identified by various methods ; such as mechanical transmission, antibiotic reactions and electron microscopy. In the results attained the author believed the pathogen of the sweet potato infected with witches' broom to be a mycoplasma-like organism. the results are as follows : 1. Mycoplasma-like bodies were occurred in the phloem region of the sweet potatoes infected with witches'-broom and its particles were sized in the range of about 200-2,500m.mu.. The membrane of the pathogen was observed to be made of an unit. 2. Responsibilities to the antibiotices were sensitive in case of tetracycline and terramycin, and root dipping method showed remarkable than foliage spray. 3. The infection was developed by the grafting transmission but by the insects, Myzus persicae and Cicadella viridis. 4. rosette, witches'-broom, stunt, yellowish, mosaic and necrosis were observed as the symptomps of the disease.

뽕밭의 생산성의 저해 환경요인과 대책

  • 생산성 저해 요인의 제거는 바로 생산성 향상으로 연결된다. 뽕밭의 생산성을 저해하는 요인은 여러가지지만 그 중에서 토양의 이화학적 불량과 농약 공해가 비중이 가장 크다고 볼 수 있다. 우리나라 뽕밭은 입지가 경사지에 많으므로 토심이 얕은데 비해, 뽕나무는 영년성 심근성 작물이므로 식재후 근권의 물리성에 따라 생산성이 좌우되는 비중이 매우 크다. 그러나 식재후 토양 물리성 개량은 화학성 개량보다도 어려우므로 그 결과 생산성이 낮은 상태를 벗어날 수 없다. 따라서 정기적인 심경이나 심토 파쇄를 통한 물리성 개량이 요망된다. 계속적인 금비위주의 비배관리에 의해 강산성 반응을 보이는 토양이 대부분이므로 석회에 의한 산성개량이 필요하다. 질소질의 과다시비는 수량을 감소시킬 뿐만 아니라, 엽질을 악화시키고 질산태 질소가 지하로 흘러 들어가 지하수의 오염을 일으켜 청색증과 암을 일으키는 원인이 될 수 있다. 심각한 정도로 축적된 인산과 칼리도 수량을 떨어뜨리고 환경을 해치는 원인이 되므로, 축적 양분의 재활용에 대한 연구가 계속되어져야 한다. 특히 사용의 편리성 때문에 고농도의 복합비료 위주의 비배관리가 반복되고 있는 실정이므로 토양검정을 받아서 단비를 이용하여 적정한 시비를 해야 할 것이다. 이러한 방법은 생산성의 향상, 영농비의 절감, 공해의 유발을 억제하는 여러가지 효과가 있다. 농약과 산업공해로부터 양잠의 보호를 위해서는 안전지대에 잠업의 단지화, 인근 농과와의 대화를 통해서 양잠에 안전한 농약을 사용토록 하는 한편, 연구분야에서는 심도 있는 연구를 통해 양잠농가를 보호할 수 있는 대책의 수립이 요망되고 있다. 종합해서 말하자면 연구기관의 심도있는 연구가 더욱 활발히 이루워져야 할 것이며, 농민은 개발된 새기술에 보다 깊은 관심을 갖고 활용하는 한편, 필요로 하는 기술을 연구기관에 개발을 요청할 정도의 적극적인 자세가 요구된다. 또한 연구기관의 농민의 중간 교량역할을 하는 지도부문에서도 보다 적극적인 활동이 요청된다.

  • Because of human population growth, longer lifespans, and climate change, there is growing concern around world to produce enough food and feed. Insects are regarded as an alternative with high potential because the production of insects demands limited amounts of water and land, and they can add value to low-value by-products. Insects have high levels of crude protein, lipids and minerals. The relative amount of protein can vary substantially, with crude protein content ranging from 23% to 76%, depending on insect species. Their amino acid composition is good and protein digestibility is high. Insect to be a significant sustainable source as a replacement of ingredients such as soya or fishmeal in the feeds of terrestrial livestock or fish. This review provides an overview of nutritional value of insect in animal feed and challenges required to develop a sustainable, safe, and affordable insect farming industry.
