Nuclear DNA inheritance of intra-specific somatic hybrids by di-mono cross in Pleurotus ostreatus based on URP-PCR analysis (URP-PCR 분석에 의한 느타리 이핵-단핵 계통간 교잡주의 핵 DNA 유전)

  • The primary objective of the present study is the characterization of the somatic hybrids of dikaryon-monokaryon (di-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus ostreatus strains with 48 intraspecific hybrids of 12 combinations between six P. ostreatus strains and one P. florida strain. The results on the experiments of hybridization rate, nuclear DNA patterns, and colors and morphology of fruit-bodies, are presented. In di-mono crosses, somatic hybrids among Pleurotus strains showed 100% of crossability as seen in those between P. ostreatus and P. florida strains indicating that the nuclei of a dikaryon is inferred to be migrated to a recipient. 87.5% of the somatic hybrids among Pleurotus strains were similar to the donor dikaryons, and 12.5% of the somatic hybrids presented DNA patterns of both parents. In 16.6% of di-mono crosses between P. ostreatus and P. florida, the nuclear DNA patterns of all hybrids showed the same or similar patterns compared to the donor dikaryons. 70.9% of the hybrids between P. ostreatus and P. ostreatus were similar to the donor dikaryons and 12.5% of them presented the DNA patterns of both parents. 79.2% of fruiting body morphology of the hybrids among Pleurotus strains were similar to the dikaryons and 20.8% of them were similar to both parents. Interestingly, the morphology of all dikaryons were dissimilar each other. All hybrid strains between dikaryon P. florida and monokaryon P. ostreatus showed the fruiting body of which colors were similar to those of the dikaryon, while the hybrids between dikaryon P. ostreatus and monokaryon P. florida were showed the combined colors of both parents. Therefore, the fruiting body color of P. florida tends to be generally dominant. In conclusion, the present study provides a way to find out and suggest superior hybrid strains using the nuclear DNA patterns of hybrids between Pleurotus strains as well as the characteristics of their fruiting bodies. The advantages of the di-mono crossing are needs to be fully utilized in mushroom breeding because it is an ideal way to develop the superior strains of Pleurotus.

  • This study was performed to investigate the high yielding potential of Pleurotus ostreatus (Fr.) Quel derived from intermating and intramating of three strains: Local strain, Nonggi 201, and Pleurotus florida Eger. All the strains tested were tetrapola mating systems, and hybridable. Intermating system resulted in more fresh sporophores than those from intramating. The highest yield of fresh sporophores of P. ostreatus was obtained from Nonggi 201 crossed with P. florida, when cultivated on the rice straw substrates.

  • F. velutipes Leu 2 gene (${\beta}-isopropylmalate$ dehydrogenase gene) was used for transformation of P. florida leucine requiring auxotrophic mutant P101. Transformation frequency was very low but the transformed colony can grow on minimal medium very slowly. Transformation was identified by Southern hybridization and reverse transformation into E. coli using chromosome DNA isolated from transformed P. florida.

  • This study aimed to investigate possible cause of slow and abnormal growth of oyster mushrooms, Pleurotus florida and P. ostreatus collected from bad crop farms. Spherical virus particles of 30 nm in diameter from P. florida, 23 nm particles from P. ostreatus, and both 23 and 30 nm particles were also found from interspecies mated culture between P. florida and P. osreatus. The virus particles might be associated with the bad crop of Pleurotus species.

  • Mutagenesis of protoplast could serve a great potential tool for improvement of strains and genetics in higher fungi. For the isolation of auxotrophic mutants from protoplasts of Pleurotus ostreatus and Pleurotus florida, viability levels of ultraviolet lights were determined. Seven auxotrophs were obtained from protoplasts irradiating UV to give $0.83{\sim}15%$ survival. The mutants showed a single requirement for each of Arg, Ribo-l, Ribo-2 or Phen for growth. Some of them showed two or three kinds of requirements, Gly Ser, Ade Hypo or Ala Om Tryp for growth.

  • The experiment of protoplast regeneration and reversion were undertaken to provide a basic techniques for protoplast manipulation. Protoplasts of Pleurotus florida and Pleurotus ostreatus were reverted to normal hyphal growth and the reversion frequency of both fungal protoplasts were $0.24{\sim}3.19%$. Reversion medium stabilized with 0.6 M potassium chloride and sucrose was better than the other stabilized one. The protoplast reversion frequency was increased when various amino acid and vitamin compounds were added to the hypertonic mushroom complete agar medium.

  • We describe a new taxon of Goodyera (Orchidaceae), G. ${\times}$ tamnaensis N.S. Lee, K.S. Lee, S.H. Yeau & C.S. Lee, sp. nov., from Jeju Island. This taxon is presumed to be a hybrid between G. schlechtendaliana and G. velutina based on several morphological characteristics, i.e., leaf venation, a patterned leaf surface and raised leaf epidermal cells, the color of the flower and the bract, the lateral sepal shape, and the hair length. The morphological characteristics and illustrations of the species based on the holotype are provided together with photographs of the habitat.

A Comparative Study on the Surface Patterns Applied to the Traditional Refining and Forge Welding Process Using Iron (철을 이용한 전통 정련·단접 과정 적용 소재별 표면무늬 금속학적 비교 연구)

  • This research has analyzed SI, the traditional steel, and SIHS(SI + HS), SICS(SI + CS), and SINiS(SI + NiS), the materials that were produced through welding and reprocessing three modern steel- HS, CS, and NiS- that have different carbon content. The purpose of the analyzation was to improve the definition of the multi-layered pattern that appears in the forging process. In observing modified structures on the commissures of three modern steel that have different carbon component to the SI, SINiS produced the most significant multi-layered pattern as well as the excellent welding quality. The excellent welding quality was due to the content of nickel which helped the forge welding process with other materials. There was no significant difference in crystal grain per materials, and SICS showed the highest hardness. At the measurement of EPMA for commissures of the materials, SINiS showed the highest definition of the multi-layered pattern due to the nickel and carbon content. The results above showed that the carbon steel with nickel content is the best material for the most definite multi-layered pattern, expressed from the multi-layered structure which is a characteristic of traditional forge welding technology. It is expected that the result of this research can be utilized as the technical data in further researches regarding the relics excavated from ancient welding process and their multi-layered structure and patterns.

Effects of Extraction Methods on the Antioxidative Activity of Artemisia sp. (추출 방법에 따른 쑥 추출물의 항산화 활성)

  • The effect of extraction methods, such as reflux extraction (RE), autoclave extraction (AE), low temperature high pressure extraction (LTPE) and ultrasonification extraction (USE) on antioxidant activity of various species of Artemisia (Artemisia capillaris T., Artemisia princeps P., Artemisia annua L.) was investigated. The extraction yield of RE and AE was higher than other methods tested for all Artemisia. The total polyphenol and flavonoid content of Artemisia sp. extracts from RE was highest of the extraction methods tested. The total polyphenol and flavonoid content of A. capillaris T. extracted by RE was 260.82 mg GAE/g and 11.52 mg RHE/g, respectively. The A. capillaris T. extract showed higher DPPH and ABTS radical scavenging activity than that of the other tested Artemisia sp. Nitrite scavenging activity and superoxide dismutase (SOD)-like activity of various extracts from RE was 45.48% and 68.29% (A. capillaris T.), 45.73% and 61.43% (A. princeps P.), and 44.25% and 58.19% (A. annua L.), respectively. The RE method was the most effective method for extracting antioxidant substances from various A. capillaris T. compared with AE, LTPE and USE. These results suggest that extracts of Artemisia sp. from RE can be used as bioactive and functional materials in the food industry.

Intergeneric Transfer of Isolated Nuclei from Lentinus edodes into Protoplasts of Pleurotus florida (사철느타리버섯 원형질체내에 표고 핵의 전이)

  • Transfer of the isolated nuclei from Lentinus edodes into protoplasts of Pleurotus florida was induced with polyethlene glycol (PEG) and $CaCl_2$. The intergeneric transfer products were classified into nuclear hybrid, heterokaryon or synkaryon, and reconstituted cell. These progenies except nuclear hybrids formed mature fruiting bodies on sawdust rice bran medium. Formation of fruit bodies was influenced by several factors such as light, temperature, nutrition and physic state of the culture media. Most of fruiting body characters were similar to those of P. florida in synkaryon and L. edodes in reconstituted cell, respectively. All these basidiocarps had clamp connections though initial heterokaryon colonies were lacking. Isozyme patterns of intergeneric progenies were quite different from those of parents. DNA polymorphisms of transfer products were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerase chain reaction. The RAPD patterns were different from those of donor and recipient. DNA fingerprints ranged in size from 0.25 to 4.0 Kb. On the basis of RAPD, the transfer products were classified into five groups. Two synkaryon were analysed with distribution of progenies and segregation of genetic markers by random spore analyses. The genetic markers were segregated into wild type and riboflavine requiring auxotrophs.
