The Effect of Nicotine-Contaminated Mulberry Leaf in the Vicinity of Tabacco Drying Plant on Cocoon Crop (연초건조장 부근의 뽕잎이 잠작에 미치는 영향)

  • The objective of the present study was to investigate the effect of nicotine-contaminated mulberry-leaf, which was grown in the vicinity of tobacco drying plant (TDP), on cocoon crop. Mulberry-leaf harvested from the field at Sericultural Experiment Station (SES), Suweon, Korea, was used as control, supposedly nicotine-free leaf. Leaf harvested from the field in the distances of 30-50m, 300-400m and 700-800m from TDP was fed during the whole larval stage of the silkworm at. SES. The effect of leaf in each treatment level on the quantitative characters of the silkworm was summarized as follows; 1. Larval duration from 4th instar on was significantly longer in the TDP-leaf treatments than for the control. 2. Duration of matured silkworm appearance became longer as the distance of the mulberry-field from TDP got shorter, because the larval duration and growth of the silkworm were not uniform in the TDP-leaf treatments. 3. Mortality rates during the late larval, cocoon spinning, and pupal stages were highest for the 30-50m leaf, especially mortality rates during the late larval and pupal stage were serious. 4. Pupation rate was lowest for the 30-50m leaf and those for the 300-400m and the 700-800m leaf were not significantly different from that of the control. 5. Nicotine damage to cocoon weight and cocoon shell weight was significant in each TDP-leaf level. Cocoon shell ratio was reduced at the same extent in each level, compared with the control. 6. The ratio among cocoon-classes was significantly different between treatments, compared with best-cocoon ratio of 87.1% for the control. Cocoons were not uniform for the 30-50m leaf, and those for the 300-400m and the 700-800m leaf were as almost uniform as those for the control. 7. Loss of fresh cocoon yield became greater as the distance of the mulberry-field from the TDP-got shorter. In conclusion, the critical distance of mulberry-field, which influences larval health, cocoon quality and yield, appeared to be 800m from the TDP. Such other factors as wind direction and topographic location may be involved in the critical distance. 8. From the present experiment, we could obtain only the effect of nicotine on the silkworm through digestive system, since the silkworm was raised at SES in Suweon. If the silkworm.. were raised in the vicinity of the TDP, poison effect of nicotine on the silkworm could beo expected through exoskeleton and tracheal system as well as through digestive system.

Quality Characteristics of Cookies Fortified with Soymilk Cake Fermented by Bacillus subtilis GT-D (고초균에 의한 비지 발효물을 첨가한 쿠키의 품질특성 연구)

  • Soymilk cake (SMC) fortified with defatted soy flour was fermented by a novel Bacillus subtilis GT-D in the solid-state fermentation with chlorella, mulberry leaves or green tea as food ingredient. The fermentation of SMC resulted in higher content of tyrosine and higher activities of protease, amylase and fibrinolytic enzymes. Fermented SMC including mucilage indicated six-fold higher content of dietary fiber compared to that of non-fermented SMC. The SMC fermented without additional ingredient allowed the highest spread ratio and width of cookies. Cookies prepared without SMC showed the lowest value of spread ratio and width compared to those of fermented SMC. In addition, these cookies showed the highest hardness value of 8,975 g, and thickness value of 5.97 and slightly higher bright color. Cookies fortified with SMC fermented with chlorella or mulberry leaves showed higher sensory evaluation value. Fortification of SMC fermented without additional ingredient resulted in the highest preference in the sensory evaluation of cookies.

Effect of Fermentation Time on the Chemical Composition of Mulberry (Morus alba L.) Leaf Teas (발효시간이 뽕잎차 구성성분에 미치는 효과)

  • Morus alba (Mulberry) leaves were exposed to fermentation for varying amounts of time: no fermentation (0 min, MANF), medial fermentation (10 h, MAMF), and full fermentation (24 h, MAFF). The chemical compositions of the teas were determined and compared with those of commercial Camellia sinensis teas. The results showed that mulberry leaf teas contained significantly higher amounts of ash and fat than Camellia sinensis tea. Compared with Camellia sinensis teas, all mulberry leaf teas contained significantly more total free amino acids (24.26~54.25mg L-glutamic acid equivalent $g^{-1}$), but the concentration of caffeine was relatively low for mulberry leaf teas. High thiamine, riboflavin, and niacin contents were found in all mulberry leaf teas, but ascorbic acid and pyridoxine were found at higher levels in Camellia sinensis teas than in mulberry leaf teas. Color measurements demonstrated that mulberry leaf tea infusions generally had lower $a^*$ (greenness) and $b^*$ (yellowness) values than Camellia sinensis tea infusions. All infusions exhibited low turbidity levels (less than 10%). The contents of total phenols were measured as 71.8 and 74.9mg 100 $ml^{-1}$ infusion in MANF and MAFF, respectively, but the MAMF tea infusion showed significantly lower total phenols (64.6mg 100$ml^{-1}$ infusion). The total flavonoid contents of mulberry leaf tea infusions were lower (8.9~20.6mg 100 $ml^{-1}$ infusion) than those of Camellia sinensis teas and thus had lower antioxidant capacities (DPPH: 326.8~526.9 ${\mu}M$ trolox equivalent $g^{-1}$ and FRAP: 364.6~387.6 ${\mu}M$ trolox equivalent $g^{-1}$) than Camellia sinensis teas. The amounts of ${\gamma}$-aminobutyric acid (GABA) and rutin were higher in fermented mulberry leaf teas; the level of GABA increased with increasing fermentation time and the content was highest in MAFF, but rutin content was highest in MAMF.

  • Yongchonppong is liable to withering of leaf and it is lowering leaf value for the feed. In this respect, covering of the vinylon gauze over the feed has been carried out in the branch rearing system to sustain leaf value as itself. The covering of the vintlon gauze over Yongchonppong marks 7% and 9% increment of cocoon yield in dry season, spring rearing season, as compared to the non-covering of the vinylon gauze over Kaeryangppong and Yongchonppong, respectively. There was no significant differences in cocoon shell percentages of them. In more humid environment than average relative humidity in Korea the rearing results of "Yongchonppong" was improved by covering with vinylon gauze after feeding compared with the non-covered rearing. However, the vinylon guage covering was not effective for improving the rearing results of "Kaeryangppong". No effect of the vinylon gauze covering was seen in the area where it frequently rains and raining season lasts. In case of the non-covering of the vinylon gauze, Yongchonppong reduces repidly in the first three hours of exposure to the air, as compared to Kaeryangppong. The water content reductions rate of Yongchonppong in 6 hours of exposure to the air marks 7% to so when the dry vinylon gauze cover and 17 to 26% when the wet vinylon gauze covers, as compared to 58.8% of it for the noncovering of the vinylon gauze.f the vinylon gauze.

Study on Spring Cocoon Crops with the Leaf Produced in the Mulberry Field close to the Totacco Field (개량 Mulching 담배밭 부근뽕잎이 춘잠작에 미치는 영향에 관한 연구)

  • The studies are to know how much cocoon crops is damaged by the stained leaf with nicotine produced from the tobacco field cultivated in mulching system in spring season and by residual nicotine in autumn season. Furthermore, the new knowledges are to make both industries keep up with their development. In spring season mulberry Held is located higher on the West-North of tobacco held below 20 degrees of slope and with 36 per cent of East-South wind and 18 per cent of South wind blowing from tobacco fold to the mulberry fold. In addition, silkworm larvae are fed with the mulberry leaf produced in the different plots placing by the different distances, l0m, 25m, 50m, 80m, and loom far from the tobacco Held as a control and it is also considered that narcotic larvae including the dead larvae are not observed. On the other hand, it is noted that better leaf quality and abundant growth of mulberry tree is produced from the mulberry fold closer to the tobacco field and with a low slope. 1) Maximum weight of larval body at the 5th stage is damaged by the stained leaf with the nicotine up to 25m far from the tobacco held. 2) The larvae fed with the mulberry leaf in mulberry Held up to 25m far from the tobacco fold produce small number of the fresh cocoons per 1 liter. 3) Low single cocoon weight and low cocoon shell weight are produced by the poison damaged larvae fed with the mulberry. leaf up to 25m far from the tobacco field and weight of cocoon shell is damaged higher than the single cocoon weight. It is resulted in low percentage of cocoon shell. 4) Cocoon yield including the double cocoon from 10,000 larvae is decreased by the larvae fed with the stained leaf in the mulberry fold up to 25m far from the tobacco fold and 19 per cent of cocoon yield is decreased with 2.4kg of cocoon yield in l0m plot and with 2.5kg of cocoon yield in 25m plot at the first season and at the 2nd season with 1.8kg o( cocoon yield in l0m plot and with 11.5kg of cocoon yield in 25m plot, 11 per cent and 9 per cent of cocoon yield including double cocoon from 10,000 larvae is decreased, as compared with the control, respectively. With these results, it is observed that nicotine damage is occurred to the silkworm larvae if the larvae are fed with the leaf in the mulberry Held within 25m-50m far from the tobacco field.

  • The effects of oral application of fenoxycarb, the commercial formulation Insegar, to the selected developmental stages of the silkworm, Bombyx mori, was investigated. An oral application of the chemical to the silkworm from the 2nd- to the 5th-instar larvae delayed the larval development upto more than 40 days and increased the larval body weight in the range of 1.1 to 1.7 folds. When the chemical was orally applied to the final instar larvae, spinning and pupation were prevented, and consequently permanent larvae occurred. The weight of a cocoon and its shell of silkworm(bombyx, mori, L) increased following the application of fenoxycarb to the 2nd- and the 3rd-instar larvae.

  • Water culture studies were conducted in the greenhouse with mulberry plants to investigate the nutrient uptake, especially Zn and Cu, under high phosphrous concentration. Mulberry plants were grown with five phosphorus levels(0, 0.2, 0.5, 2.0, 5.0 mM). Leaves and roots were analyzed for water content, total nitrogen, P, K, Ca, Mg, Fe, Mn, total Zn, soluble Zn, Cu, Cl, $NO_3HPO_4$ and $SO_4$. Dry matter increased upto 2.0mM phosphorus level, and then decreased. Water content, total nitrogen, P, K, and Fe in leaves increased with increasing phosphorus level. Total Zn content in leaves showed little change, whereas soluble Zn increased and Cu decreased with increasing phosphorus level. With increasing phosphous level. $SO_4$ and Cl decreased and then sharply increased above 2.0mM phosphorus. Lower uptake of Cu and higher uptake of $SO_4$ and Cl suggest a cause of mulberry yield decline with high accumulation of soil phosphorus.

  • To investigate the effects of applying animal feces to the mulberry field on the chemical properties and mulberry leaf yield, poultry, pig and cattle feces were applied to the mulberry field making-up the sand loam from 1988 to 1990. The chemical properties of the mulberry field have been improved by the application of the poultry, the pig, and the cattle feces, increasing pH level, organic matter, P2O5 and exchangeable cation, especially Mg and K. Applying the pig feces and poultry feces to the mulberry field increased leaf yield during autumn rearing season, but not increased during spring rearing season. Applying animal feces to the mulberry field increased the content of total-cargbohydrate in the leaf in autumn.

Development of Optimization Mixture Tea prepared with Roasting Mulberry Leaf and Fruit (반응표면분석을 이용한 로스팅 뽕잎과 오디 혼합차 개발)

  • In this study, we aimed to develop a mixed tea prepared with roasted mulberry leaf and fruit using response surface methodology (RSM). Roasting of mulberry leaf was by 6 stages, as shown in Fig. 1; and mulberry fruit was roasted in 4 stages, as shown in Fig. 2. Subsequently, physicochemical measurements such as total polyphenol content, nitric oxide production content, and ${\alpha}$-glucosidase inhibitory effect were obtained for each sample. Central composite design was applied to prepare samples containing varying contents of roasted mulberry leaf (RoML) and roasted mulberry fruit (RoMF); subsequently, sensory evaluation was conducted. The total polyphenol content of roasted samples (RoML and RoMF) were significantly higher than that of raw samples (RaML and RaMF), respectively. The nitric oxide (NO) production of roasted samples were significantly lower than that of control (LPS induced RAW 264.7 cell). The ${\alpha}$-glucosidase inhibitory effect of roasted samples was significantly higher than that of raw samples, respectively. Based on the RSM estimation for determination of optimum ratio by sensory evaluation (taste, color, and flavor) among 13 mixed samples, the optimum mixing ratio of RoML and RoMF for taste, color, and flavor were 1.64 g (RoML) and 0.88 g (RoMF), 1.35 g (RoML) and 0.92 g (RoMF), 1.65 g (RoML) 1.03 g (RoMF), respectively. Based on results of three sensory evaluations, mixing ratio comprising 1.54 g of RoML and 0.92 g of RoMF is desirable for delicious tea with functionality.

Effects of Acid, Salt, Heat Treatment and Natural Antimicrobials on Survival of Pathogens Isolated from Surface of Carcass in Minced Meat (분쇄육에서 산, 염, 열처리 및 천연항균물질 처리가 도체표면으로부터 분리한 병원성미생물의 생존에 미치는 효과)

  • Effects of acid, salt, heat treatment and natural antimicrobials on survival of E. coli O157:H7 CDF1, A. sobria CDF3 and S. aureus CDF2 isolated from surface of carcass in minced meat was investigated. The growth of E. coli O157:H7 CDF1 and A. sobria CDF3 inhibited in minced meat containing above 4% NaCl but not in 1% lactic acid. The growth of S. aureus CDF2 was not inhibited significantly by addition of 4% NaCl but inhibited completely in minced meat containing 1% lactic acid. Survival of A. sorbia CDF3 did not show any differences during storage at 4 and 10$^{\circ}C$. E. coli O157:H7 CDF10 and A. sobria CDF3 did not detect after heat treatment at 60$^{\circ}C$ for 10 min but S. aureus CDF2 decreased only 1 log after the same treatment. Viable cell of E. coli O157:H7 CDF1 decreased 2 log in TSB containing 0.5% Oolong tea extract after incubation for 12 hr compared with control but A. sobria CDF3 and S. aureus CDF2 did not detect at the same condition. The growth of E. coli O157:H7 CDF1, A. sobria CDF3 and S. aureus CDF2 was not inhibited by addition of 0.3% Oolong tea extract but inhibited by addition of 0.5% Oolong tea extract in minced meat at 20$^{\circ}C$ for 24hr.
