2,4-Dinitrochlorobenzene으로 유도된 아토피 피부염 동물모델에서 비파엽 및 삼백초 추출발효물의 항아토피 활성 (Anti-Allergic Activities of Fermented Eriobotrya japonica and Saurus chinensis Extracts in 2,4-Dinitrochlorobezene-Induced BALB/c Mice)

  • 본 연구는 in vitro 실험결과 아토피 피부염의 억제 효능이 기대되는 비파엽 추출물(BI군), 삼백초 추출물(SA군), 비파엽 추출물과 삼백초 추출 혼합물(FB군) 그리고 삼백초와 비파엽 추출 혼합 발효물(FA군)을 경구 투여하여 DNCB 처리한 아토피 실험동물을 대상으로 아토피 질환 개선에 대한 유효성을 평가하였다. 혈청 중의 IgE 수치를 측정한 결과, DNCB군에서 $227.21{\pm}60.04$ ng/mL를 나타내었으며, FA군에서는 $154.78{\pm}78.99$ ng/mL로 IgE 생성량이 감소되었다. 혈청내의 히스타민 농도는 DNCB군에서 $1.90{\pm}0.04$ ng/mL 였으며, FA, FB, BI, SA군에서 각각 $1.47{\pm}0.20$ ng/mL, $1.41{\pm}0.07$ ng/mL, $1.61{\pm}0.05$ ng/mL, $1.08{\pm}0.07$ ng/mL로 DNCB군과 비교 시 유의적인 감소를 나타내었다. 세라마이드 총 함량을 측정한 결과, FA군에서 $1,350{\pm}357\;{\mu}g/g$으로 DNCB와 비교 시 세라마이드 함량이 유의적으로 증가되었다. IFN-$\gamma$의 경우, DNCB군에서 $2.5{\pm}0.5$ ng/mL로 분석되 었으며, FA, FB, BI 및 SA군에서 각각 $7.1{\pm}1.3$ ng/mL, $7.7{\pm}0.2$ ng/mL, $11.5{\pm}0.6$ ng/mL 및 $6.9{\pm}0.2$ ng/mL로 DNCB 군과 비교 시 모두 유의적인 증가를 나타내었다. 특히 비파엽 및 삼백초 추출물 혼합 발효물은 등 부위 피부의 육안 증상을 관찰한 결과, 가피형성과 발적 감소가 가장 우수하여 아토피 피부염 질환 개선에 도움을 줄 수 있을 것으로 기대된다.

Scopolamine으로 유도된 기억력 손상 동물모델에서 비파엽 추출물의 학습 및 기억력 개선 효과 (Anti-amnesic Effect of Eriobotrya japonica Leaf Extract on Scopolamine-induced Memory Impairment in Rats)

  • 본 연구는 scopolamine으로 유도한 기억력 및 인지기능 감퇴 동물모델에서 비파엽 추출물의 기억력 및 인지기능 개선효과를 평가하기 위해 수행되어졌다. 6주령의 SD-rat(male)을 총 5개 군으로 분리하였으며, 양성대조군으로 donepezil(1 mg/kg b.w.)을 처리하였다. 15일간 24시간 간격으로 EJE를 50, 200 mg/kg b.w. 농도로 전처리(경구투여) 하였으며, 16일째에 농도별로 EJE 및 양성대조군으로서 donepezil을 투여하고 scopolamine을 복강투여(1 mg/kg b.w.) 하였다. Water maze test와 passive avoidance test를 통해 scopolamine으로 유도한 동물모델에서 EJE의 기억력 및 인지능력 개선 효과를 확인한 결과, scopolamine에 의해 감소된 기억력 및 학습능력이 EJE를 투여함으로써 유의적으로 회복 및 증가한 것을 확인할 수 있었다. Scopolamine에 의해 인지능력과 함께 감소된 운동능력의 평가를 위해 수행한 rotarod test 및 vertical pole test에서는 scopolamine의 투여로 감소된 균형감각, 협응력 그리고 grip strength와 같은 운동능력들이 EJE의 투여로 유의적으로 회복된 것을 확인하였다. 또한 scopolamine에 의해 증가한 AchE의 활성과 감소한 Ach의 함량이 EJE의 투여로 각각 개선되었다. 본 연구에서는 이와 같은 결과를 바탕으로 EJE의 scopolamine으로 유도한 동물모델에서 인지능력 개선과 감퇴한 운동능력의 향상 및 cholinergic neurotransmission system의 강화 효과를 확인할 수 있었으며, 이러한 연구 성과들로 비파엽 추출물이 퇴행성뇌질환인 AD 치료와 학습 및 기억력 개선에 있어 효과적인 대안으로서 보다 더 많은 분야에서 연구되기를 바라는 바이다.

  • Objective : The present study designed this study to investigate anti-oxidative effects of EF on HaCaT keratinocyte. Method : The present study measured the amount of polyphenoics and flavonoids, and also measured the levels of catalase, Ascorbate peroxidase (APX), SOD like activities and DPPH free radical scavenging activity. Then the effects of SB on viability and prolferation rates, and protective effects against oxidative stress induced by chemicals such as hydrogen peroxide and rotenone were also investigated. Results and conclusion : EF showed protective effect against cell death of HaCaT keratinocyte induced by rotenone and SNP significantly. In conclusion, these results suggest that EF may have anti-oxidantic action in human skin and also suggest that EF can be used as anti-aging agent.

통증완화를 위해 비파엽 찜질을 병행한 암환자 6례에 대한 증례보고 (The Case Report about Six Cancer Patients Added Eriobotryae Folium Fomentation to Mitigate Pain)

  • Objective: The purpose of this study is to see whether pain in cancer patients is decreased by Eriobotryae Folium Fomentation. Methods: We examined the change in pain among six cancer patients, prescribing Eriobotryae Folium Fomentation. As for evaluating methods, we used the case history of pain, the direct evaluation of pain, the indirect evaluation of pain. The direct evaluation of pain is subdivided by the language expression, the non-language expression, and Visual analog scale(VAS). The indirect evaluation of pain is subdivided by the condition of walking, mood and the condition of sleep. Results: After two weeks, any remarkable pain decrease is not observed in both of the direct evaluation and indirect evaluation.

비파엽 품질 비교 분석을 위한 Triterpenic Acid의 추출 방법 (Development of an Effective Extraction Method for the Quality Control of Eriobotrae Folium : Determination of Triterpenic Acids)

  • In result of analysis for the content determination of ursolic acid(UA) and oleanolic acid(OA) in extract of Eriobotryae folium using HPLC with UV detector, UA in chloroform extract of Moo-mok variety was showed highest content(2.7843 mg/g). And OA in ethyl acetate extract of Dae-bang variety was showed highest content(0.5898 mg/g). These result suggest that direct extraction using organic solvent(chloroform or ethyl acetate) was useful method for rapid quantitative analysis of UA and OA without preprocessing such as drying or fractionation.

  • We investigated the in vivo effect of an aqueous extract (referred to as EF) from Eriobotryae folium on glucokinase and hexokinase activities of diabetes mellitus induced by $interleukin-1{\beta}\;(IL-1{\beta})$. After 1 week of $IL-1{\beta}$ injection, the levels of serum glucose concentration and insulin secretion were dramatically increased. However, the insulin secretion was decreased with administration of EF. The level of glucose concentration was decreased by EF administration. Furthermore, it was observed that EF was effective in recovering the levels of insulin secretion. Enzyme activities of the glucokinase and hexokinase, which are key enzymes of glucose phosphorylastion, were decreased by $IL-1{\beta}$. EF administration to the mice allowed proportional increasing by stimulation of induction of enzyme activities as high as normal group. These results suggested that EF is highly effective in treatment of diabetes mellitus induce by $IL-1{\beta}$.

피부 각질세포에 대한 비파엽 에탄올 추출물의 PGE2 조절 작용과 상처치료 효과 (Prostaglandin E2 Up-regulation and Wound Healing Effect of the Ethanol Extract of Eriobotryae Folium in Human Keratinocyte)

  • Prostaglandin (PG) $E_2$ is an important mediator of skin wound healing without excessive scarring and gastric ulcer healing. However, $PGE_2$ has a short lifetime in vivo because it is metabolized rapidly by 15-hydroxyprostaglandin dehydrogenase (15-PGDH). Ethanol extract of Eriobotryae folium (EFEE) elevated intracellular and extracellular $PGE_2$ levels in HaCaT cells and inhibited 15-PGDH ($ED_{50}$ : $168.4{\mu}g/mL$) with relatively low cytotoxicity ($IC_{50}$ : $250.0{\mu}g/mL$). Real-time PCR analysis showed that mRNA expression of cyclooxygenase (COX)-1 and COX-2 enzymes were increased and prostaglandin transporter (PGT) was decreased in HaCaT cells by EFEE. Moreover, wound healing effect of EFEE ($168.4{\mu}g/mL$) was comparable to that of TGF-${\beta}1$ (300 pg/mL) as a positive control. These results demonstrate that EFEE may be valuable therapeutic materials for the treatment of $PGE_2$ level dependent diseases.

비파엽의 핵수용체 관련 신규 약물 작용점 연구 : 만성 질환 치료 가능성 (Eriobotrya japonica inhibits the transactivation of nuclear orphan receptor : the possibility to treat the chronic diseases)

  • Objectives : Oxidative damage has a variety of mechanism in the human organs including brain and liver. The purpose of this study is to examine the prevention against cellular damage, and the inhibitory effect on lipogenesis of the water extract of Eriobotrya japonica (EJE). Methods : The effect of EJE on cell viability was assessed using the MTT assay. To investigate the mechanism of EJE's inhibition of lipogenesis, we analyzed the relevant proteins using immunoblot analysis. Results : Induction of T0901317, a LXR agonist, significantly increased SREBP-1c expression, which was blocked by the pretreatment of EJE in the dose-dependent manners. This beneficial herb also regulated the genes related with SREBP-1c such as the acetyl-CoA carboxylase. Conclusion : These results suggested that EJE might have a possibility of the treatment of chronic diseases as mediated with the inhibition of LXRα/SREBP-1c pathway.

비파엽 열수 추출물에서 분리한 Caffeoylquinic Acid 3 종의 Nitric Oxide 생성 억제 효과 (Nitric Oxide Production Inhibitory Effects of Three Caffeoylquinic Acids Isolated from Hot Water Extract of Eriobotrya japonica L. Leaves)

  • Background: Research on hot water extracts of medicinal plants that are easily applicable in the clinical setting is essential. To confirm the anti-inflammatory-related active compounds present in the hot water extract of Eriobotrya japonica leaves, ability to inhibit nitric oxide (NO) production was measured and active compounds isolated from the extract were analyzed. Methods and Results: Sovent fractionation by solvent was performed to identify the active compounds present in the hot water extract, and the ability of the extract and the fractions obtained to inhibit NO production was measured. Subsequently, based on the results of liquid chromatography (LC) profile analysis of the n-butanol fraction that had a relatively high inhibitory ability of NO production, six subfractions were separated around the main peak. Among the separated subfractions spectra from mass spectroscopy (MS) were analyzed and standard comparisons were performed on the compounds of the three main peaks on the chromatogram. NO production inhibitory activity of subfraction 2 identified as neochlorogenic acid was the highest with an IC50 of 18.49 ㎍/㎖ followed by that of subfraction 5 identified as cryptochlorogenic acid with IC50 of 25.82 ㎍/㎖. Conclusions: Our result, it was confirmed that several caffeoylquinic acids, including neochlorogenic acid and cryptochlorogenic acid present in the hot water extract of E. japonica leaves have an important role as compounds exhibiting anti-inflammatory activity.

AMPK 활성화를 통한 중금속 유발 산화적 스트레스에 대한 비파엽의 세포 보호 효과 (Cytoprotective effect of Eriobotrya japonica L. against the iron-induced oxidative stress through AMPK activation)

  • Objectives : In this study, we investigated the cytoprotective effect of Eriobotrya japonica L. (EJ) extract against Arachidonic acid (AA)+iron-induced oxidative stress. Methods : To confirm the cytoprotective effect of EJ against AA+iron-induced oxidative stress in HepG2 cells, it was evaluated by MTT assay, immunoblot anaylsis, and Calcein-AM/propidium iodide (PI) staining. Additionally, the mechanism of action of the cytoprotective effect was evaluated through molecular mechanisms. Results : EJ (100 ㎍/mL) inhibited Arachidonic acid (AA)+iron-induced cell death in a concentration-dependent manner. It also inhibited AA+iron-induced mitochondrial dysfunction and ROS production. EJ activated the LKB1-AMPK signaling pathway. Conclusions : In conclusion, EJ has the ability to protect liver cells from oxidative stress, indicating that it is related to AMPK-LKB1 signaling pathways.
