Biological Treatment of Starch Waste Part 1. Isolation of Wheat Starch Waste Decomposing Organisms and Their Efficiency on Waste Treatment (전분폐수의 생물학적 처리에 관한 연구 1. 소맥 전분포수 처리균의 분리와 처리효과)

  • In order to develop an activated sludge which can be used for both waste treatment and protein source of animal feed, microorganisms were isolated from sewages of various wheat or sweet potato starch processing plants and their activities were tested. Out of 32 isolates which composed of two protozoan genera and 13 bacterial strains, were screened and three bacterial stranis were found to be most effective in both floe-formation and wheat starch waste liquid stabilization.

동치미 유래 생균제로서 가능성이 있는 Lactobacillus sp. FF-3의 분리와 특성

  • 생균제로서 가능성 있는 유산균의 선별을 위해 발효식품인 동치미로부터 분리된 균주들의 내산성, 내담즙성, 내췌장액성 및 유해균 억제능을 조사하였다. 특히 분리된 균종 중에서 FF-3가 인공위액과 담즙산에 대한 높은 내성을 나타내었을 뿐만 아니라, Salmonella sp. 과 Escherichia coli에 대하여 높은 항균 활성을 나타내었다. FF-3유래 박테리오신은 그람양성 및 음성 세균에서 넓은 항균 범위를 나타내었다. 본 연구에서 최종선별된 FF-3 균주를 16s rDNA sequence 동정한 결과 Lactobacillus sp.으로 확인되어 FF-3으로 명명하였다.

Nitrogen Fixation Screening and Plant Growth Assessment for Urban Greening (도시 녹화를 위한 질소고정 균 선별 및 식물 생장 평가)

  • Currently, urban greening projects and research are attracting attention as a way to mitigate urban heat island phenomenon. In this study, nitrogen fixative bacteria were isolated and their effects on plant growth were confirmed. First, enrichment was performed in a nitrogen-free medium to isolate the nitrogen-fixing bacteria, and the colony showing high growth in a medium with limited nitrogen source was isolated and purified. Separated bacterial isolates were reduced by more than 90% acetylene by ARA and indirectly confirmed the activity of nitrogenase by ethylene production. Cedecea sp. MK7 and Enterobacter sp. Y8 with confirmed reproducibility were selected as nitrogen fixative bacteria. Nitrogen fixing bacteria were applied to the growth of perennial rye grass, and it was found that the dry weight increased to 34.80 mg (186.60%) compared with the control with 18.65 mg dry weight. After plant growth, microbial community analysis of soil applied by bacteria showed similarity to the control group. Therefore, in this study, it is expected that the efficiency will be increased if plant growth is promoted by using nitrogen fixing bacteria in urban greenery system.

Isolation and Characterization of Microorganisms for the Development of Fermentation Accelerator of Animal Manure (가축분뇨 발효제의 개발을 위한 미생물 분리 및 특성조사)

  • Several microorganisms were isolated and characterized for the development of fermentation accelerator of animal manure. Firstly, 61 species were isolated from rice bran extract. Secondly, five strains of microorganisms were screened by the analysis of hydrolysis activities for organic compounds including protease, cellulase, amylase, and lipase. From a deodorization test for ammonia gas using the isolated strains, finally three bacterial strains were selected (NA 2, 12, 15). The selected strains, NA 2 and 15 were identified as Bacillus acidocaldarius and Planococcus sp. respectively. The media composition of key nutrients and pH for the mixed culture of the three selected strains were optimized using an experimental design method (response surface method) as follows : beef extract (4.59g/L), peptone (8.72g/L) and pH 6.3. Consequently, the isolated microorganisms seem to have potential applicability in the animal manure treatment.

  • In this study, waste car tau lamps that usually gets incinerated or buried has been recovered during the dispose process in order to increase the recycling rate of waste cars and the material separation research was performed by applying triboelectrostatic separation method. After a series of charging characteristic test, PMMA materials was confirmed of being an effective charging material for waste car tail lamp material separation, a cyclone charger using PMMA material for continuous process was developed. In optimum test conditions using triboelectrostatic separator developed in this study, high quality PMMA material with recovery rate of 99.0% and 90.2% was achieved from the waste car tail lamp. Therefore, a material separation technology that can recycle tail lamps of waste cars has been established.

Testing the Potential of Sewage Sludge Gasification Solid Residues as a Circulating Resource by Physical Separation (하수슬러지의 가스화 고형 잔재물의 순환자원으로서 물리적 선별에 의한 잠재성 검토)

  • In this study, physical property evaluation and physical separation of the target product were performed to investigate the possibility of using sewage sludge gasification solid residue (GSRs) as a circulating resource. Firstly, the GSRs used in this study was supplied by Sudokwon Landfill Management Corporation, and generally the GSRs was in the form of porous pellets with a particle size of several millimetres. In addition, the partially black areas were confirmed to be unburned and ungasified carbon, and the average carbon content was 5%. In addition, the content of silica, alumina and phosphorus oxide was more than 70% of the total content. It was confirmed that the metallic components of the wet grinding product were separated into individual elements. As a physical separation of metallic and non-metallic components was required, it was finally found that flotation screening was suitable. Accordingly, cationic and anionic surfactants were selected to separate metallic components in which a relatively large amount of non-metallic components were concentrated, and the separation characteristics were confirmed. As a result, it is expected that the concentration of non-metallic components such as silica, alumina and phosphorus will be easier than the separation of metallic components. Therefore, since it is possible to physically treat the gasified sludge residue, it is judged to have potential as a circular resource according to the proposed recycling method for the separated product.

토양균에서 항생물질의 분리와 구조연구

  • 국내의 토양에서 Streptomyces spp.를 분리하였고 이 중 항균활성이 우수한 균주들을 선별하였다. 이들 균주들을 적절한 조건에서 배양하여 얻은 배양액을 이온 교한 수지와 흡착 크로마토그래피를 행하여 항생물질들을 분리하였다. 이들 항생물질들은 Bacillus subtilis, Pyricularia oryzae, Alternaria mail, Candida albicans, Mycobacterium smegmatis, Pseudomonas aeruginosa , 및 Escherichia coli에 강한 항균활성을 나타내었다. 각 항생물질들을 산으로 가수분해하고 흡착관 크로마토그래피로 가수분해 생성물을 분리하였다. 분리한 항생물질과 가수분해 생성물들의 $^1$IH-NMR, $13^{C}$ -NMR, FAB-Mass 스펙트럼을 분석하여 구조를 연구하였다.

  • 방선균은 자연계에 널리 분포하고 있고 학문적으로 흥미롭거나 상업적으로 유용한 경우도 많이 알려져 있으나 실제 자연계에 있어서의 자세한 분포 및 역할은 아직 잘 알려져 있지 않은 경우가 많다. 적절한 균 분리원의 선택 및 이로부터의 신규 또는 유용한 방선균만을 효율적으로 분리하는 방법은 아직까지 논의의 여지가 있지만 자연계에는 아직 미지으 유용한 방선균이 많이 존재함에는 이의가 없다. 따라서 목적을 가지고 방선균을 효율적으로 분리하고 이용하는 기술이 향상되면 스크리닝은 성공적이 될 것이다. 이를 위해 본 고에서는 이에 필요한 적절한 균 분리원의 선정, 시료의 전처리, 적절한 배지 등의 선정, 배양조건의 검토 및 효율적인 균 선별 등에 대하여 간략하게 검토하였으며 차후 보다 기본적인 생태학적인 조사 및 분자생물학적인 연구 지원을 통하여 이 분야의 연구는 더욱 발전하리라 보여진다.

  • 색채선별기는 양질의 곡물 유통을 위해 불량곡물, 이물질을 선별하는 시스템이다. 기존의 색채 선별기는 PC기반 제어시스템으로서 비교적 고가이고 부피가 크며 현장 환경에서 기계적 충격이나 먼지등에 취약하다는 문제점을 가지고 있다. 본 논문에서는 이러한 문제점을 개선하기 위해 운영체제가 탑재된 임베디드 제어기 기반의 색채선별기를 제안한다. 색채선별기는 기본적으로 곡물의 유입량 및 속도를 조절해 주는 피터부, 곡물의 영상을 받아 이미지를 처리해 주는 카메라부, 카메라부에서 받은 정보를 이용해 양품과 불량품을 분리해 주는 이젝터부로 구성된다. 본 논문에서는 개발된 지능형 색채선별기용 ARM 프로세서 기반 임베디드 제어기의 하드웨어 구성, 피더부, 카메라부, 이젝터부 간의 통신프로토콜을 이용한 총괄관리 제어와 그래픽 사용자 인터페이스(GUI)를 소개한다.

Identification of Fluorescent Pseudomonads Producing Siderophore and Construction of Siderophore Biosynthesis Defective Mutant (Siderophore를 생성하는 Fluorescent Pseudomonads의 분리, 동정 및 돌연번이 유기)

  • The present study was performed to isolate the fluorescent pseudomonads from Kwang-Ju soil and to construct a mutant strain defective in siderophore biosynthesis. The siderophore-secreting pseudomonads were screened on Blue agar (Chrome Azuol S agar) plates and one strain of them was designated to Pseudominas fluorescens (P. fluorescens) PY002. To construct a mutant defective in siderophore biosynthesis, P. fluorescens PY002 was randomly mutagenized with a transposon Tn5. The location of Tn5 integrated into chromosomal of the mutants strain was determined by Southern blot analysis. The mutagenized strain showed non-fluorescent on a King's B agar plate and were defective in iron (III) acquisition ability.
