  • 2매의 금속을 맞대어 그 한 쪽에 접촉면과 평행하게 고주파진동을 가하면 단 시간에 접합된다. 이공정을 초음파 용접이라고 하며 그 물리적인 본질은 아직 불분명 하지만, 첫째로는 강한 마찰에 의해 금속 자유면의 산화물층이 제거되기 때문이라는 점과 둘째로는 마찰에 의해 금속 표면이 강하게 가열되어 이에 따른 연화에 의해 접합 이 된다고 하는 점이다. 그러나 이와 같이 가열된다고 하더라도 가열은 표면부에만 국한되고 다른 부분은 가열되지 않는다. 따라서 초음파용접은 냉간접합이라고도 한다. 또 가압력과 진동에 의한 힘이 동시에 작용하기 때문에 용접할 면을 미리 청정하게 할 필요는 없고 용접전의 단계에서 자연적으로 청정화가 이루어진다고 하는 사실이 간접 적으로 증명되고 있다. 초음파 용접(Ultrasonic welding)의 특징을 요약하면 고상용접 의 일종으로서 용접중에 국부적으로 고주파 진동에너지와 압력을 가하여 용접하는 방법이다. 이 때 모재를 용융시키지 않고 건전한 야금학적 결합부가 얻어진다는 데에 큰 특징이 있다. 또한 초음파용접은 다른 용접법에 비해 경제성이 매우 높고, 초음파 용접에 필요한 출력이 전기아크 용접에 필요한 출력의 5 - 10%로 충분한 경우가 많다. 초음파 용접은 통상의 방법으로는 용접하기 어려운 동종 금속이나 이종 금속의 용접에 널리 사용된다. 이 용접법은 반도체, 미세회로, 전기 접점의 형성에 대한 생산기술 로서 사용되고 있는데 소형 모터, 알루미늄 박의 가공, 알루미늄 합금의 조립 등에 이용되고 있다. 한편 최근에는 자동차, 우주항공산업 분야의 구조제 용접용으로도 채용되고 있다.출함이 바람직하다.분비되는 배설-분비 항원의 자극과 깊은 관계가 있음을 알 수 있었다.넌트 명세서를 대한 XML DTD(Document Type Definition)를 정의하고, HTML 기반 검색 방법과 XML 기반 검색 방법에 대한 컴포넌트 검색 성능을 비교한다.따라 NO 생산 및 세포독성이 증가하였고. Enhancing Effect of Acanthopanax senticosus Extracts on Mouse Spleen and Macrophage Cells Activation (가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과)

  • Acanthopanax senticosus is an herb that has been used as a traditional remedy and medicine source. Its anti-inflammatory and, anti-oxidative effects have been reported in previous studies. This study aimed to investigate the effect of Acanthopanax senticosus water extracts on mouse macrophage cell in vitro. Mouse splenocyte proliferation increased after application of Acanthopanax senticosus water extract supplement of 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after 48 h pre-treatment with a mitogen (ConA or LPS). The production of cytokines secreted by LPS and non LPS stimulated macrophages was detected by ELISA assay using a cytokine kit. Cytokine production (IL-2, IFN-${\gamma}$, and TNF-${\alpha}$) increased after water extract supplementation. The result of this in vitro study, showed that splenocyte proliferation and cytokine production by activated peritoneal macrophages were increased after Acanthopanax senticosus water extract in the range of $500{\sim}1,000{\mu}L/mL$. Thus, it is suggested that supplementation with Acanthopanax senticosus water extracts may enhance immune function by regulating splenocyte proliferation and enhancing cytokine production by activated macrophage.

Immunomodulating Effects of Red Ginseng on the Regulation of Cytokine Release in vivo (홍삼의 생체 내 사이토카인 분비에 대한 면역조절효과)

  • Cytokines released from innate immune cells play key roles in the regulation of the immune response. Red Ginseng (RG, steamed and dried root of Panax ginseng C.A. Meyer) is known to show different pharmacological effects by changed composition of saponins compared with Panax Ginseng. In this study, we examined the immunomodulatory effects of RG on the regulation of cytokine release in mice. RG was injected i.p at doses of 0.5, 5 and 50 mg/kg for 6 weeks. We assessed that the weight index of immune organs such as thymus, and spleen, and the mitogen blastogenesis of splenocytes. We also determined the levels of circulating cytokines in serum from RG-treated mice using ELISA assay. The weight index of thymus and spleen, and proliferation of mitogen response of splenocytes have increased in the RG-injected groups. In addition, the levels of IFN-${\gamma}$, TNF-${\alpha}$, IL-6, IL-12 and IL-2 concentrations have significantly increased in the serum of RG-treated mice, but that of IL-10 has not. These results suggest that RG has immune stimulating effects and could be useful as a immunoregulator of circulating cytokine release in vivo.

  • The authors inquired into what reactions comprise the response of mice(as a model) CD3, CD4 and CD8 monoclonal antibodies in spleen tissue when injected intraperitoneally by antigens of Clonorchis sinensis. The author is objective was focused on investigating the property of cellular immunity for liver fluke. In particular, the results of having examined the phenotype of the tissue of spleen were revealed as follows: a certain length of time after having been intraperitoneally injected with antigens of Clonorchis sinensis and Freund's adjuvant, the tissue of spleen was embedded and immunohistochemically stained by the avidin-biotin complex method. A strong reaction in response to CD3, while a feeble reaction resulted from CD4 and CD8. The tissue region showed a positive reaction to all antibodies, especially from capsules, vascular areas, white pulps and membrane of blood cells.

Anti-inflammatory Effect of MeOH Extract of Cibotium barometz in IFN-$\gamma$ and LPS-stimulated Mouse Peritoneal Macrophage (IFN-$\gamma$와 LPS로 자극된 쥐의 복강 대식세포에서 구척 메탄올 추출물의 항염증 효과)

  • The rhizome of Cibotium barometz has been used for variety of bone disease as a traditional medicine. In the present study, we examined the antioxidant and anti-inflammatory activities of 85% methanol extract of C. barometz. C. barometz exhibited potent scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, superoxide radical and nitric oxide radical. In IFN-$\gamma$/LPS-stimulated mouse peritoneal macrophage model, C. barometz suppressed nitric oxide production and IL-6 secretion dose-dependently. Moreover, C. barometz showed decreased iNOS and COX-2 expression without notable cytotoxicity. These results suggest that C. barometz may be an useful agent as an antioxidant and anti-inflammatory agent.

  • Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract from Pulsatilla koreana $N_{AKAI}$ (PK) on the peritoneal macrophage. Methods : To evaluate of anti-inflammatory of PK, we examined cytokines and NO production in lipopolysacchride (LPS)-induced macrophages. Furthermore, we examined molecular mechanism using western blot. Results : 1.Extract from PK reduced LPS-induced NO, tumor necrosis factor-a ($TNF-{\alpha}$), interleukin (IL)-6 and IL-12 production in peritoneal macrophages. 2.Extract from PK itself does not have any cytotoxic effect. PK inhibited the activation of extracelluar signal-regulated kinase(ERK 1/2) but not another mitogen-activated protein kinases (MAPKs) such as p38, c-Jun NH2-terminal kinase (JNK) and the degradation of inhibitory kappa B a ($I_{k}B_{a}$) does not any effect in the LPS-stimulated peritoneal macrophages. Conclusion : PK down-regulated LPS-induced NO and cytokines production, which may be provide a clinical basis for anti-inflammatory properties of PK.

Inhibitory Effects of Piperine on the Production of Nitric Oxide, Interleukin-10 and Interleukine-12 in Murine Peritoneal Macrophages (복강 대식세포에서 피페린의 일산화질소, 인테루킨-10과 인테루킨-12의 억제 효과)

  • The purpose of this study was to investigate the anti-inflammatory effects and cellular mechanism of piperine on murine peritoneal macrophages. To evaluate the effects of piperine, we examined the production of nitric oxide (NO), interleukin (IL)-10 and IL-12. To investigate inhibitory mechanism of piperine, we examined the MAPKs and Ik-Ba in murine peritoneal macrophages, Piperine itself does not have any cytotoxic effect and reduced lipopolysaccharid (LPS), Poly(I:C), CpG-ODN -induced production of NO, IL-10 and IL-12 in peritoneal macrophages. Piperine inhibited the activation of extracelluar signal-regulated kinase (ERK 1/2) and c-Jun NH2-terminal kinase (JNK 1/2) not the activation of p38 and the degradation of inhibitory kappa B a (Ik-Ba) in the LPS-stimulated murine peritoneal macrophages.ln conclusion, Piperine down-regulated LPS-induced production of NO, IL-10 and IL-12, which could provide a clinical basis for anti-inflammatory properties of piperine.

Anti-inflammatory Effect of Euphorbiae kansui Radix Extract in Lipopolysaccharide-stimulated Mouse Peritoneal Macrophages (LPS로 유도된 마우스 복강 대식세포에서 감수(甘遂)추출물의 염증억제 효과)

  • This study is aimed to investigate the anti-inflammatory effect of Euphorbiae kansui radix methanol extract (ERE) in lipopolysaccharide(LPS)-stimulated mouse peritoneal macrophages. Peritoneal macrophages were obtained from thioglycollate-injected Balb/c mice. Cells were stimulated with LPS or LPS plus interferon-gamma (IFN-${\gamma}$) in the presence of ERE and various inflammatory markers were assayed. Finally, LPS-induced signaling molecules were measured. ERE up to $400{\mu}g/m{\ell}$, was not cytotoxic to ERE inhibited LPS/IFN-${\gamma}$-induced nitric oxide (NO), inducible NO synthase. ERE also reduced the levels of cyclooxygenase-2 and the proinflammatory cytokines such as tumor necrosis factor-${\alpha}$, interleukin(IL)-6 and IL-12. The inhibitory effect of ERE on LPS-induced $I{\kappa}B{\alpha}$ degradation was weak but phosphorylation of JNK, p38 and ERK1/2 was strongly suppressed. Our data indicated that the anti-inflammatory effect of ERE in LPS-stimulated macrophages was partly mediated by its inhibition of JNK, p38 and ERK1/2.

Water Extract of Flowers of Magnolia Denudata Inhibits LPS-induced Nitric Oxide and Pro-inflammatory Cytokines Production in Murine Peritoneal Macrophage by Inhibiting $NF-{\kappa}B$ Activation (LPS로 활성화된 복강 대식세포에서 신이 추출물의 염증성 사이토카인 및 NO 억제 효과)

  • Flowers of Magnolia denudata has been reported to possess a variety of pharmacological activities. In this study, we investigated the anti-inflammatory effects and mechanism of the water extract of Flowers of Magnolia denudata(MD) in lipopolysacchride (LPS)-mediated inflammatory mediators in murine peritoneal macrophages. MD itself does not have any toxic effects in murine peritoneal macrophages. MD inhibits LPS-induced nitric oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, IL-6 and IL-12 production in murine peritoneal macrophages. Furthermore, we have found that MD inhibited LPS-induced $NF-{\kappa}B$ but not c-Jun N-terminal kinase (JNK), p38 and extracellular signal-ragulated kinase (ERK) activation. These results suggested that MD inhibit LPS-induced production of $TNF-{\alpha}$, IL-6 and IL-12 via suppression of the $NF-{\kappa}B$ activation.

Effects of Plant Water Extract Mixture Ixeris Sonchifolia Hance, Oenanthevjavanica, Fagopyrum Esculentum Moench, Hizikia Fusiforme, Zingiber Officinale Roscoe on Mouse Immune Cell Activation Ex vivo (5가지 (고들빼기, 돌미나리, 메밀, 톳, 생강)혼합식품 물 추출물의 마우스 면역세포 활성화 효과)

  • Ixeris sonchifolia Hance (Godulbaegi), Oenanthe javanica (Dolminari), Fagopyrum esculentum Moench (Buckwheat>, Hizikia fusiforme (Seaweed Fusiforme) and Zingiber ojficinale Roscoe (Ginger) have been used respectively as one of folk remedies as well as food materials. However, reportedly few studies on their immunomodulating effects have been made, although it has been known from other preceding studies that the ex vivo supplementation of each Ish, OJ, Fem, Hf, Zor water extracts tends to enhance the proliferation of splenocyte in comparison to the control group. This study on the combined immunomodulative effect of water extract mixture of these five food materials (Ish + Oj + Fem +Hf+Zor) lasted covering seven or eight weeks. The old mice (balb/c) was fed ad libitum on chow diet, and the water extract of plant mixture was orally administrated every other day for four weeks at two different concentrations (50 and 500 mg/kg B.W) . After preparing the single cell suspension, the proliferation of splenocyte was determined by MTT (3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide) assay. The production of cytokine ($IL-{\beta}$, IL-6, and $TNF-{\alpha}$) which was secreted by macrophages stimulated with LPS or not was detected by ELISA assay using the cytokine kit. After the 48 hours of incubation with the mitogen (ConA or LPS) stimulation, the proliferation of the mice splenocyt in the experimental group statisticaly increased at both of two different concentrations in comparison to the control group. The cytokines production was more significantly enhanced at the lower supplementation (50 mg/kg B.W.) group than at the higher concentration (500 mg/kg B.W.). The result of this study may suggest that the supplementation of water extract of plant mixture can regulate and enhance the immune function by increasing the splenocyte proliferation and regulating the cytokine production capacity by the activated macrophages in mice.
