Rapid Measure of Color and Catechins Contents in Processed Teas Using NIRS (근적외선 분광광도계를 이용한 차 제품의 색상 및 카테킨류의 신속 측정)

  • This study was done to measure the color and catechins contents in processed teas using the whole bands (400~2500 nm) with near-infrared spectroscopy(NIRS). The powder colors of 109 processed teas were measured with a colorimeter. The a/b ratios in Hunter color scale in processed teas accounted for about 98.9% of the variation in the fermentation degree(FD), indicating that the a/b ratio was a very useful trait for assessing fermentation degree. Also tea powders were scanned in the visible bands used with NIRSystem. The calibration equations for powder colors were developed using the regression method of modified partial least squares(MPLS) with the internal cross validation. The equations had low SECV (standard errors of cross-validation), and high $R^2$ (coefficient of determination in calibration) values with 0.996~1.00, indicating that the visible bands(400~700 nm) with NIRS could be used to rapidly measure the variables related to powder color and fermentation degree. Also another powders of 137 processed teas were scanned at 780~2500 nm bands in the reflectance mode. The calibration equations were developed using the regression method of MPLS with the internal cross validation. The equations had low SECV, and high $R^2$ (0.896~0.983) values, showing that NIRS could be used to rapidly discriminate the contents of EGC($R^2$=0.919), EC(0.896), EGCg(0.978), ECg(0.905) and total catechins(0.983) in processed teas with high precision and ease.

Taxonomy of tribe Neillieae (Rosaceae): Physocarpus (나도국수나무족의 분류: 산국수나무속)

  • The tribe Neillieae, a small group of about 18 species in the Rosaceae, comprises three taxonomically difficult genera, Neillia, Physocarpus, and Stephanandra. The tribe, characterized by lobed leaves with persistent or deciduous stipules and ovoid, shiny seeds with copious endosperm, is strongly supported as a monophyletic group by a variety of lines of molecular evidence. Due to the high amount of morphological variation across the three genera and the species in tribe Neillieae, conflicting classification schemes and numerous species have been proposed over the past three centuries. However, no comprehensive systematic study of the group, including all species across their geographic ranges, has ever been undertaken. As part of a taxonomic revision of tribe Neillieae, a revision of Physocarpus based on the morphological examination of herbarium specimens, including types, and field observation is presented. Artificial keys, comprehensive nomenclatural treatments, descriptions, distribution maps, and lists of specimens examined are provided. Six species in Physocarpus are recognized. A lectotype is here designated for the following species: Opulaster pubescens, Opulaster ramaleyi, Spiraea opulifolia var. parvifolia, Spiraea opulifolia var. tomentella, Physocarpus michiganensis, and Physocarpus missouriensis.

  • To obtain several foundamental information for the breeding of Korean Ginseng. 16 line selecteds in Korea were investigated and analyzed. The results obtained are as follows : 1. In all quantitative genetic characters of Korean Ginseng, the stem length showed the highest heritability(0.7050) and the leaf length the lowest(0.5674). 2. The genotypic correlation was showen the highest value (0.8649) at between the leaf length and the stem diameter and the lowest value(0.8649) at between the leaf length and the stem length, but the phnotypic correlation was shown the highest value (0.8788) at between the stem length and the stem dimeter and the lowest value (0.7950) at between the leaf length and stem length. 3. The co-heritability between the stem length and the stem diameter showed the highest value(0.6124) and that between the leaf length and the stem length showed the lowest value(0.5471). 4. The inter-class correlation among the planting location showed the highest value at the stem length and the lowest value at the stem diameter.

Biological Properties of Vero Cell-Adapted Newcastle Disease Virus (Vero 세포적응 뉴캣슬병 바이러스의 생물학적 특성)

  • Newcastle disease virus (NDV) Kr005/V strain was generated through 55 serial passages of NDV Kr005 strain in Vero cells. The Kr005/V virus yielded high infective titers of $10^{7.8}$ $TCID_{50}/mL$ in Vero cells and the infected cells showed cytopathic effects such as marked cell rounding, though less frequent syncytia. The Kr005/V virus was heat-stable and classified into the lentogenic type with a Mean Death Time (MDT) of 120h or greater while the Kr005 strain was heat-labile and velogenic (MDT of 49.6 h). Only the single amino acid substitution (T to S) was observed at position 433 of the HN protein of the Kr005/V strain, whereas no amino acid change was found in the F protein. The Kr005/V input virus correlated well (correlation coefficient $r^2$=0.97) with the Kr005 virus when ten field sera were tested by virus neutralization test. The biological properties and usefulness of Vero cell-adapted Kr005/V virus were discussed.

A Study on Analysis of Variant Factors of Recognition Performance for Lip-reading at Dynamic Environment (동적 환경에서의 립리딩 인식성능저하 요인분석에 대한 연구)

  • Recently, lip-reading has been studied actively as an auxiliary method of automatic speech recognition(ASR) in noisy environments. However, almost of research results were obtained based on the database constructed in indoor condition. So, we dont know how developed lip-reading algorithms are robust to dynamic variation of image. Currently we have developed a lip-reading system based on image-transform based algorithm. This system recognize 22 words and this word recognizer achieves word recognition of up to 53.54%. In this paper we present how stable the lip-reading system is in environmental variance and what the main variant factors are about dropping off in word-recognition performance. For studying lip-reading robustness we consider spatial valiance (translation, rotation, scaling) and illumination variance. Two kinds of test data are used. One Is the simulated lip image database and the other is real dynamic database captured in car environment. As a result of our experiment, we show that the spatial variance is one of degradations factors of lip reading performance. But the most important factor of degradation is not the spatial variance. The illumination variances make severe reduction of recognition rates as much as 70%. In conclusion, robust lip reading algorithms against illumination variances should be developed for using lip reading as a complementary method of ASR.

Development of a New Commercial Grain Cooler (곡물냉각기의 개발)

  • The objectives of this study were to develop a new commercial grain cooler suited to domestic weather and post-harvesting conditions for paddy, and to evaluate the performance. A prototype grain cooler capable of cooling paddy of 200 tons within 24 hours was developed. The grain cooler was designed to control the refrigeration capacity from 0 to 100% by controlling the capacity of compressor with unloading solenoid valve and by changing the flow rates of hot refrigerant gas flowing into reheater and evaporator from compressor. And a controller with one chip microprocessor was developed to control temperature and relative humidity of cooling air. The maximum cooling capacity of the grain cooler was 35,284㎉/hr at condensing/evaporating pressure of 16.5/3.6 kgf/$\textrm{cm}^2$. Maximum flow rate of cooling air was 120 ㎥/min at static pressure of 279 mmAq. The total maximum required power was 22.8㎾, and total required energy was saved from 26.7 to 33.3% of maximum power depending on operating conditions. The coefficient of performance of refrigeration devices and total coefficient of performance of the grain cooler were 4.71 and 1.8, respectively.

Studies on Genetic Variation of Soluble Solids, Acidity and Carotenoid Contents in Tomato Fruits from Germplasm (국내외에서 수집된 토마토에서 당도, 산도, 카로티노이드 색소의 유전변이에 관한 연구)

  • Tomato (Lycopersicon esculentum M.) is one of the most important crops to the fresh vegetable market and the food processing industry. To evaluate genetic variation in tomato fruits, major characteristics such as soluble solids, acidity and carotenoid contents were analyzed for 771 genetic resource lines. Lines in red color was about 85% which is the largest one followed by peach color, yellow, green, orange, and black. The sweetness of juice ranged from 2.2 to 11.5% (in brix), the average being 5.6%. The acidity ranged from 0.124% to 1.665%, and the average was 0.881%. The lycopine contents was up to 80.4 ${\mu}g/g$, and 43.4 ${\mu}g/g$ in average. ${\beta}$-carotine ranged 1.8 to 48.8 ${\mu}g/g$ and it average was 10.8 ${\mu}g/g$. Statistical analysis indicates that there is coefficient of correlation between acidity and sweetness, acidity and pH, pH and lycopine, lycopine and ${\beta}$-carotine. It is expected that the result of this study can be used for breeding more competitive species with respect to contents in sugar or functional chemicals from the selected characteristic species.

The Identity of Eggplant Powdery Mildews Collected in Korea (한국에서 채집된 가지 흰가루병균의 실체)

  • Three species of powdery mildew (Erysiphales) on eggplant (Solanum melongena L.) have been listed in Korea, namely Erysiphe cichoracearum (now genus Golovinomyces), Leveillula taurica, and Sphaerotheca fusca (now genus Podosphaera; syn. Podosphaera xanthii). Since E. cichoracearum was recorded on eggplant for the first time in Korea in 1969, it has been regarded as a major powdery mildew agent on that plant. In 1998, the causal agent of powdery mildew on eggplant was recorded as L. taurica, then as S. fusca in 2002. During our extensive field surveys in Korea, we collected 22 samples of eggplant powdery mildews. Our microscopic observations and molecular sequence analyses showed that all of our samples belonged to the genus Podosphaera, in the absence of either E. cichoracearum or L. taurica, suggesting that P. xanthii is the dominant agent of powdery mildew disease on eggplants in Korea. As there have been no additional findings on L. taurica after the first report on the species, it seems to be a minor species that is rarely found in greenhouses. The presence of E. cichoracearum (syn. Golovinomyces cichoracearum s. lat.) on eggplants is questionable, as the morphological characteristics of E. cichoracearum in the original description of the Korean collection deviate from the morphological variations of this species. In addition, no herbarium material of E. cichoracearum remains. Consequently, it seems that P. xanthii is the main species of powdery mildew on eggplants, whereas L. taurica occurs rarely on eggplants, in Korea. This review provides the historical and recent taxonomy of eggplant powdery mildews in detail.

An Overview for Molecular Markers in Plants (식물에서 분자 마커의 동향)

  • A molecular marker is a molecule contained within a sample taken from an organism or other matter. The development of molecular techniques for genetic analysis has led to a great contribution to our knowledge of plant genetics and our understanding of the structure and behavior of various genomes in plants. Recently, functional molecular markers have been developed to detect the presence of major genes from the analysis of pedigreed data in absence of molecular information. DNA markers have developed into many systems based on different polymorphism-detecting techniques or methods such as RFLP, AFLP, RAPD, SSR, SNP, etc. A new class of very useful DNA markers called genic molecular markers utilizing the ever-increasing archives of gene sequence information being accumulated under the EST sequencing projects on a large number of plant species. Functional markers are derived from polymorphic sequences, and are more likely to be involved in phenotypic trait variation. Based on this conceptual framework, the marker systems discussed below are all (gene)-targeted markers, which have the potential to become functional. These markers being part of the cDNA/EST-sequences, are expected to represent the functional component of the genome i.e., gene(s), in contrast to all other random DNA based markers that are developed/generated from the anonymous genomic DNA sequences/domains irrespective of their genic content/information. Especially I sited Poczai et al’ reviews, advances in plant gene-targeted and functional markers. Their reviews may be some useful information to study molecular markers in plants.

Thrombotic thrombocytopenic purpura with decreased level of ADAMTS-13 activity and increased level of ADAMTS-13 inhibitor in an adolescent (청소년기에 발생한 ADAMTS-13 활성도 저하와 항체 양성을 보인 혈전저혈소판혈증자색반병 1례)

  • Thrombotic thrombocytopenic purpura (TTP) is a thrombotic microangiopathy characterized by endothelial cell damage, resulting in microangiopathic hemolytic anemia, thrombocytopenia, and various degrees of neurological and renal impairment caused by microvascular thrombi. It is rare in children and frequently follows a fatal course. TTP is divided into 2 types: one is inherited and associated with ADAMTS-13 gene mutations and the other is acquired and associated with anti-ADAMTS-13 autoantibodies. The measurement of ADAMTS-13 activity in plasma, identification of ADAMTS-13 circulating inhibitor, anti-ADAMTS-13 IgG, and ADAMTS-13 gene sequencing are crucial to the diagnosis of TTP. Plasma exchanges are the first-line treatment for acquired TTP, combined with steroids and immunosuppressive drugs. Here, we describe the case of an adolescent patient with TTP, confirmed by decreased level of ADAMTS-13 activity and an increased level of ADAMTS-13 inhibitor, who was successfully treated by plasma exchanges.
