Antimicrobial Effect of Hedyotis diffusa Extracts on Food-Borne Pathogens (식중독유발 세균의 증식에 미치는 백화사설초 추출물의 영향)

  • This study was performed to investigate the antimicrobial effect of the Hedyotis diffusa extracts against food-borne pathogens. First, the Hedyotis diffusa was extracted with methanol at room temperature and the fractionation of the methanol extracts was carried out by using petroleum ether, chloroform, and ethyl acetate, and methanol respectively. The antimicrobial activity of the Hedyotis diffusa extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Hedyotis diffusa showed the highest antimicrobial activity against Staphylococcus aureus and Shigella flexneri. Synergistic effect in inhibition was observed when Hedyotis diffusa extract was mixed Sophora subprostrata extract as compared to each extracts alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Hedyotis diffusa against Staphylococcus aureus and Salmonella Typhimurium. The methanol extract of Hedyotis diffusa had strong antimicrobial activity against Staphylococcus aureus at the concentration of 4,000 ppm. At this concentration, the growth of Staphylococcus aureus was retarded more than 36 hours and up to 24 hours for Salmonella Typhimurium. In conclusion, the methanol extracts of Hedyotis diffusa inhibit efficiently Staphylococcus aureus and Salmonella Typhimurium.

백화사설초의 현탁세포배양에 의한 oleanolic acid 생산

  • Oleanolic acid is a triterpenoid compound that exists in Oldenlandia diffusa. Recently, oleanolic acid has been noted for antitumor effect. Application of both plant growth regulators, 2,4-D and kinetin, was found to be essential for the induction of callus and suspension cells. Optimum induction condition for callus and suspension cells of Oldenlandia diffusa was determined to be 0.5 mg/L 2,4-D and 0.1 mg/L kinetin. Chromatographic separation of oleanolic acid from its derivatives was achieved using Rexchrom S5-100-ODS column. The retention time of oleanolic acid was 12.6 min and the specific content of oleanolic acid was 0.41 mg/g dry weight.

  • From the whole plants of Oldenlandia diffusa ursolic acid together with sterols, ${\beta}-sitosterol$ and its $3-0-{\beta}-D-glucoside$ were isolated and characterized mainly by means of spectroscopic methods. Based on the present findings, it may be considered that this plant may contribute to be a rich source for ursolic acid.

Enhanced Production of Oleanolic Acid by the Elicitation in Oldenlandia diffusa Suspension Cell Cultures (백화사설초의 현탁세포배양에서 Elicitation에 의한 Oleanolic acid 생산성 증대)

  • Oldenlandia diffusa is a Chinese medicinal herb with antitumor activity capable of suppressing the growth of some cancer cell lines. Oleanolic acid and ursolic acid are triterpenoid compounds that exist in Oldenlandia diffusa. Recently, these have been noted for anti-inflammatory, anti-cancer, and hepato-protective effects. Application of both plant growth regulators, 2,4-D and kinetin, was found to be essential for the initiation of callus and suspension cells. Leaf blades of Oldenlandia diffusa was transformed into callus on Schenk and Hildebrandt medium supplemented with 0.5 mg/L 2,4-D and 0.1 mg/L kinetin, while optimum initiation condition for suspension cells of Oldenlandia diffusa was determined to be 0.75 mg/L 2,4-D and 0.1 mg/L kinetin. Chromatographic separation of oleanolic acid from its derivatives was achieved using Rexchrom S5-100-ODS column. Analytical conditions for oleanolic acid were determined as follows: flow rate at 1.0 mL/min, UV length at 200 nm and mobile phase of $80\%$ acetonitrile and $20\%$ water. Production of secondary metabolites was found to be increased by the treatment with elicitors or signal transducers. The maximum production of oleanolic acid was 99.6 mg/L in cultures with 0.5 mM salicylic acid. It is 1.74 times higher than that of control.

Immunomodulatory Effect of Methanol Extracts of Korean Oldenlandiae Herba and Radix (국내산 백화사설초 전초 및 뿌리 메타놀층의 면역조절 효과)

  • For the evaluation of immunomodulatory effect of Korean Oldenlandiae Herb (OH) and Radix (OR), our experiment was performed with methanol extracts of Korean Oldenlandiae Herba and Radix. After administration of methanol extracts of Korean OH and OR for 7 days, Balb/C mice were immunized with sheep red blood cells. Four days later, splenic leukocytes were isolated and immunological experiments were performed. Rosette forming cells and plaque forming cells were significantly increased in Korean OH and OR treated mice compared with PBS treated control. Korean OH and OR also enhanced T and B lymphocytes, macrophage and natural killer cells by flowcytometric analysis. LPS-induced TNF-α and IL-6 levels were increased by OH and OR compared with untreated control. These results suggest that Korean OH and OR have immunomodulatory activity through regulation of cell-mediated immune and humoral immune response.

Effect of Oldenlandiae Herba in Calves with Diarrhea (송아지 설사증에 대한 백화사설초의 투여 효과)

  • The therapeutic effects and immune responses with Oidenlandiae Herba administration(17.5%, 1g/30 kg, everyday, PO) were investigated in calves(1 to 3 months old) with diarrhea from 159 Korean native calves(76 heads from A farm and 83 heads from B farm) in Jeju. The causes of 47 calves with diarrhea were coronavirus(1 head) and rotavirus(1 head), coccidium(5 heads) and bacteria(40 herds). PCV values were significantly increased 2 weeks after administration of Oldenlandiae Herba in A form, compared by those of pre-treatment(p<0.05). However RBC, TP, WBC, fibrinogen levels were not significantly changed. The significant difference were not detected in changes of AST, BUN and creatinine. The CD14+ distribution rate were increased in 1 week after administration of Oldenlandiae Herba, compared by that of pre-treatment(p<0.05), however, certain tendencies of changes were not observed in distribution rates of CD4+ CD8+ and CD19+, respectively. The rate of death was 20.1%(36/179 calves) in 2004, however, it was decreased to 12.6%(20/159 calves) by Oldenlandiae Herba administration at 2 farms from Jeju in 2005. In conclusion, the administration of Oldenlandiae Herba was effective for treatment of calves with diarrhea.

Effects of Oldenlandidae Herba as an Alternative Medicine on the Swine Productivity (대체의학으로서 백화사설초 급여가 돼지 생산성에 미치는 효과)

  • As control of swine consumptive disease is the key of success in pig farming business, the effect of Oldenlandidae Herba on prevention of swine diseases as an alternative medicine was investigated in-vivo. As it contains substances such as ${\beta}$-sitisterol, ursolic acid and 3-O-${\beta}$-D-giucoside, it is effective on suppression of colonic tumor in rats and also, is widely used as an preventative medicine of cancer and inflammation in Chinese medicine. In this experiment, sows and piglets on two different farms were treated with Oldenlandidae Herba, and they recoded weaning rate greater than 80%, which were higher than national average 70% (PSY 68%). Meanwhile, neither organ accumulation nor toxicity has been detected on the basis of hematology and serum chemistry. In conclusion, the extracts of Oldenlandidae Herba is expected to be an good alternative medicines for swine productivity.

The Effects of Oldenlandiae Diffusae Herba Extract on Eosinophil, IgE and IL-4 in Experimental Asthma induced by Ovalbumin (백화사설초(白花蛇舌草)가 ovalbumin으로 유도된 천식동물모델에서 Eosinophil의 수, IgE 및 IL-4에 미치는 영향)

  • Objective : This study was performed to investigate the effect of oral administration of Oldenlandiae Diffusae Herba (ODH) on eosinophil, immunoglobulin-E and interleukin-4 in the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. And, it was observed the change of the concentrations of IL-4 in BALF and splenocyte, IgE in serum by ELISA. Results : 1. The number of eosinophil in BALF was significantly decreased in ODH group copared with control group. 2. Concentration of IL-4 in serum, BALF and splenocyte was significantly decreased in ODH group compared with control group, respectively. 3. Level of IgE in serum was significantly decreased in ODH group compared with control group. Conclusion : We found that the effect of ODH extract in asthma was implicated in reductions of IL-4 released from Th2 cell, and decreases of IgE from plasma cell. These findings suggest that ODH extract can produce anti-asthmatic effect, which may playa role in allergen-induced asthma therapy.

Apoptic and antimetastatic effects of ursolic acid isolated from Oldenlandia diffusae Herba (백화사설초(白花蛇舌草)로부터 분리(分離)한 ursolic acid의 자연살해효과(自然殺害效果)와 항전이작용(抗轉移作用))

  • Ursolic acid(UA) was isolated from Oldenlandiae diffusae Herba, one of the commonly used medicinal herbs for the treatment of cancer. IC50 of UA against cancer cell lines as SNU-1, B16-Fo. SK-OV3, HCT15, XF498, SK-MEL and A549 was $6{\mu}g/ml$, 4$4.4{\mu}g/ml$, $4.5{\mu}g/ml$, $4.6{\mu}g/ml$ and $4.2{\mu}g/ml$ respectively suggesting cytotoxicity against cancer cells. DNA fragmentation was expressed from the concnetration of $5.5{\mu}g/ml$ of UA by agarose electrphoresis. In the observation of morphological changes by phase contrast microscope, SEM and TEM, cell injury and condensation of cytoplasm from nucleus began 4 hr after UA treatment. fragmentaion of nucleus and injury of cell membrane was shown 24 hr after UA treatmeilt with SNU-1 cells. Aurin tricarboxic acid as endonuclease inhibitor. and nicotinamide as poly(ADP-ribose) polymerase inhibitor protected over 50% of cytotoxicity of UA against SNU-1 was at the concentrations of $3{\mu}M$ and $300{\mu}M$ respectively suggesting UA acts on nucleus. These results suggest that UA had antimetastatic effect and induced apoptosis.
