Correlation and Combining Ability of Days to Germination, Flowering and Ripening in F1 Hybrids of Petunia hybrida (페튜니아 일대 잡종에서 발아, 개화 및 종자 등숙소요일수 관련형질의 상호관계 및 조합능력)

  • The correlation among days to germination, days to flower, days to seed ripening and number of seeds per a pod and their combining were studied in 15 F1s from the partial six-parent diallel cross in Petunia hybrida. The days to flowering, days to germination and seed number per a pod showed high positive correlation together, however germination percentage and number of seed showed negative correlation with days to ability(SCA) were highly significant for all the characters. The estimates of variance components of SCA were greater than those of GCA for all the characters, showing preponderance of non-additive gene actions for these characters. The line of A and C showed relatively high GCA effect for days to germination and days to flower and the lines of C and G for germination, days to seed ripening and number of seeds per a pod. The crosses of D×G and E×F exhibited high SCA effect for decreasing days to flower and days to germination, and increasing germination percentage and number of seeds per a pod. The broad sense heritability was generally high compared to narrow sense one in the 5 characters. The higher heritability values of both broad and narrow sense were shown in the days to ripening and number of seeds per a pod.

Accelerated Germination of Orchardgrass(Dactylis glomerata) Seed Under Water Stress by Seed Pre-treatments (수분 장애시 종자 전처리가 오차드그라스의 발아에 미치는 영향)

  • It is important to improve the germination of grass seeds because they are poor germination under water stress. This experiment was conducted to investigate the response of seeds pre-treated individually with Polyethylene glycol 6000 (PEG) and growth regulators(GA$_3$, Kinetin, NAA) on the germination characteristics when two levels of osmotic potential (0, -5 bar) were put to seeds of Orchardgrass. In the untreated seeds, the total germination rate was decreased under water stress, and the mean germination time was delayed. Priming of Orchardgrass seeds using PEG or GA$_3$ resulted in improved germination under water stress(-5 bar), whereas the opposite was true of kinetin or NAA. The response of seeds primed in solutions of either -15 bar PEG or 100 ppm GA$_3$ was particulary marked compared with the untreated seeds. It is suggested that PEG and GA$_3$ may have positive regulatory effects in triggering the system for water stress alleviation. But the inhibitory effect of the water stress was not completely removed by allowing the seeds to pre-treat in solution of PEG or GA$_3$.

Response of Amylase and Peroxidase Activity of Emerging Rice Seeds to Different Temperatures (벼 종자 출아시 온도차이가 Amylase와 Peroxidase활성에 미치는 영향)

  • The periods of germination and seedling emergence, epidermal cell size and the activities of peroxidase and amylase of 6 rice cultivars were examined to clarify the response to 3 temperature conditions, constant temperatures of 27$^{\circ}C$ and 17$^{\circ}C$ and alternating temperature of 24/1$0^{\circ}C$, in the dark condition. The periods of germination and seedling emergence were increased and the germination was delayed greater than the seedling emergence under 17$^{\circ}C$ and 24/l$0^{\circ}C$, compared with 27$^{\circ}C$. Lengths of epidermal cell of coleoptile and first leaf were reduced, but the widths were increased in the 17$^{\circ}C$ and 24/1$0^{\circ}C$, compared with 27$^{\circ}C$. The activities of peroxidase in the emerging shoots and amylase in the germinating seeds were reduced in 17$^{\circ}C$ and 24/1$0^{\circ}C$. There were significant correlations between peroxidase activities and the widths of epidemal cell of first leaf and between amylase activities and periods of germination. Varietal differences of all observations were remarkable in 17$^{\circ}C$ and 24 /1$0^{\circ}C$.

Rooting and Budbreak of Single-Stemmed Roses (Rosa hybrida L.) as Affected by Axillary Bud Position and Leaf Area of Cuttings (장미 식물공장 생산에서 삽수의 채취 절위와 엽면적에 따른 단경삽목묘의 발근과 신초발아 특성)

  • This study was carried out to investigate the characteristics of rooting and bud break of single-stemmed roses (Rosa hybrida L. 'Rate Rose' and 'Teresa') as affected by axillary bud position and leaf area of cuttings derived from different growth stage of the mother shoots. In fact, both rooting and budbreak of single-stemmed roses were not influenced by growth stage of mother shoots, with showing more than 95% in all treatment related to mother shoot maturity. 'Rote Rose' required 34 days to average rooting and budbreak after cutting, whereas 'Teresa' did only 18 days to budbreak which was 9 days faster than rooting. Rooting and bud break needed more time and showed lower percent as the axillaty bud position for cuttings went down to the base of mother shoots. Especially 'Teresa' showed 12 days of delay to budbreak and 14.4% decrease in budbreak. Also, the increase in leaf area of cuttings accelerated rooting and budbreak, of which the time was shorter and the percent was higher.

Effect of Seedbed Environments, Hot Water Treatment, Cold Wet Storage, and Dry Storage on Seed Germination in Korean Native Lilies (자생나리의 종자발아에 미치는 파종상의 환경, 온탕처리, 습윤냉장과 건조저장의 영향)

  • Germination percentage of Korean native lily seeds was high at $20-25^{\circ}C$. It was almost 100% in L. cernuum, L. callosum, L. amabile, and L. concolor, 88.0% in L. lancifolium, and 73.0% in L. maximowitzii, respectively. Meanwhile, it was low rate of 34.0%-54.0% in L. distichum, L. hansonii, and L. tsingtauense. Germination was mostly delayed of $15^{\circ}C$ and days to germination were more shortened in species with higher germination percentage. Even though the effect of daylength was not considerable in germination rate, it was promoted in L. maximowitzii but it was delyed in L. hansonii under long day. The effect of soaking in hot PGRs solution in L. callosum, L. cernuum, L. amabile, L. lancifolium, and L. concolor did not show any difference in comparison with non-treatment. However, it was improved by BA in L. maximowitzii. Longer period of cold wet storage resulted in improved germination percentage in L. maximowitzii and L. lancifolium, while it affected decreased percentage in L. distichum and L. hansonii. Days to germination were shortened by longer period of cold wet storage regardless of species. Germination percentage in dry storage was higher under cold temperature than room temperature and under desiccator storage than outside desiccator, it was highest under desiccator storage at $4^{\circ}C$. It was drastically reduced by the non-use desiccator storage at room temperature L. concolor, however it was improved only by the use of desiccator L. maximowitzii for a long time.

Effect of Priming Conditions on Enhancing Germination of Onion (Allium cepa L.) Seeds (Priming 조건이 양파 종자의 발아율 향상에 미치는 영향)

  • Priming of seeds in 150 mM KCI at $10^{\circ}C$ for 6 days was most effective in increasing germination rate and shortening germination time. Germination rate of primed seeds was increased to 96% which is about 14% higher than that of non-primed seeds. Germination temperature for better priming effects from primed onion seeds was 10 to $15^{\circ}C$ which is relatively lower than that of normal germination temperature of $20^{\circ}C$. Drying primed seeds at $20^{\circ}C$ for 5 hours resulted in best priming effects.

Changes in the Seed Characters and Germination Properties of Three Tree Species at Different Storage Time (저장기간에 따른 3개 수종의 종자 및 발아 특성 변화)

  • This study was conducted to examine the changes in the seed physical characters and germinative properties of three tree species at three different storage times in order to determine the most suitable storage time and condition. Seed physical characters and germinative properties were examined from seeds of Fraxinus rhynchophylla, Zanthoxylum schinifolium and Staphylea bumalda that were stored at -18C for 1, 3 and 10 years, and the growth performance and physiological characters of their seedlings were analyzed. Seed physical characters and germinative properties showed significant differences between three storage times as well as three tree species. Seed moisture content of F rhynchophylla and S. bumalda and seed fresh weight of Z. schinifolium decreased with increase of storage time. Storage time represented negative correlation with moisture content and fresh weight (r = -0.822, P<0.01). Seed percent germination of Z. schinifolium and S. bumalda stored for 3 and 10 years decreased less than 15% and 10.7% respectively. Mean germination times for seed of F. rhynchophylla, Z. schinifolium and S. bumalda were delayed 11.9, 5.7 and 9.7 days after a storage time of 10 years, respectively. However the growth and viability of seedlings didn't show a special pattern for storage time and tree species.

Influence of Accumulated Hours of Low Temperature in Dormant and Changing Temperature after Bud Breaking on Flowering of Main Apple Cultivars in Korea (휴면기 저온 누적 시간 및 발아 후 변온이 국내 주요 사과품종의 개화에 미치는 영향)

  • This study was carried out to examine the base temperature to flowering and the average days to flowering by accumulated hours of low temperature ($5.0^{\circ}C$) or changing temperature after bud breaking. Over-all, the prediction of flowering time in the commercial apple cultivars ('Fuji' and 'Tsugaru') and apple cultivars ('Chukwang', 'Gamhong', 'Hongan', 'Honggeum', 'Hongro', 'Hongso', 'Hwahong', 'Summer dream', 'Sunhong') bred in Korea at the Gunwi region for 4 years (from 2009 to 2012) was investigated. Also, this study estimated the flowering time when the air temperature of Gunwi region rises at $5.0^{\circ}C$ was investigated using the same data. The range of accumulated hours of low temperature (chilling requirement) was from 0 hour to 1,671 hours, and the range of high temperature (heat requirements) to flowering after low temperature treatment was from $5.0^{\circ}C$ to $29.0^{\circ}C$. The treatments of changing temperature after bud breaking were classified as constant temperature treatment (control) and $5.0{\sim}10.0^{\circ}C$ elevation or descent treatments. The results show that the average days to flowering was longer with shorter accumulated hours of low temperature, and the average days from bud breaking to flowering of 0 hour treatment was longer about 2~4 weeks than that of 1,335~1,503 hours treatments. In comparing to apple cultivars, the all cultivars were not flowered under $10.0^{\circ}C$ after bud breaking, and the cultivars with low chilling requirements needed low heat requirements for flowering. The average days to flowering of treatments that the air temperature after bud breaking was controlled about $15.0^{\circ}C$ was shorter about 1~3 weeks than that of treatments was controlled about $10.0^{\circ}C$. In the treatment of changing temperature after bud breaking, the average days from bud breaking to flowering of temperature elevation treatment was shorter than that of constant temperature treatment. By use of these results, the base temperature to flowering of main apple cultivars in Korea was seemed to $10.0^{\circ}C$, and if the air temperature of Gunwi region rises about $5.0^{\circ}C$ than that of current, the flowering time was estimated to be delayed by 1 week.

Effects of Days after Pollination and Media on Asymbiotic Germination in Cross Combination of Cymbidium spp. (수분 후 경과일수 및 배지가 심비디움 교배조합의 무균발아에 미치는 영향)

  • This study was conducted to investigate the effective media and optimal days after pollination for germination of seeds which were crossed by selfing or reciprocal pollination in Cymbidium spp. The elongation of ovary stopped about 110 days after pollination regardless of cross combinations. The increase of ovary width stopped about 140 days after pollination showing some differences among cross combinations. The cross combinations expressing high capsule number and viability showed good germination when they were sown in vitro. The capsules collected 150 to 180 days after pollination showed good germination performance and short days of germination. Hyponex media was the most effective for protocorm development and germination.

Influence of low temperature and sowing depth on emergence in dry direct seeded rice (벼 건답직파재배(乾畓直播栽培)시 저온발아성(低溫發芽性)과 파종심(播種深)에 따른 출아특성(出芽特性))

  • This experiment was conducted to obtain the basic information on ability of germination at low temperature with 26 cultivars of Korea including USA cultivars and to identify the optimal sowing depth in dry-direct seeded rice. Varietal difference in germination rate and days to emergence at low temperature($13^{\circ}C$) was observed. Average germination was 81.2% and range of germination rate showed 64.3 to 94.8%. In days to germination there was varietal difference. Average days to germination was 17.4 days and those range showed 14 to 24days. Negative high correlation was observed between the ability of germination and days to germination. Considering germination and days to germination. Jakwangdo, Nagdongbyeo, Cheongmeongbyeo, Calom, Anjungbyeo, Gyehwabyeo, Daegwanbyeo, Dongjinbyeo and Seohaebyeo were selected as a adaptable cultivar. High correlation was observed among the lodging index and sowing depth, stem diameter, height of central gravity, breaking strength. Seedling emergence was increased and days to emergence was shortened at the seeding depth of 1 and 3cm. Lodging index was decreased as seeding depth was deep and so the reasonable seeding depth would be around 3cm in direct seeded rice.
