Recent Progress in Multiplexed Detection of Biomarkers Based on Quantum Dots (양자점 기반 다중 바이오마커 검출법의 연구동향)

  • Semiconductor quantum dots (QDs) are optical probes with excellent fluorescence properties. Therefore, they have been applied to various bio-medical imaging techniques and biosensors. Due to the unique optical characteristics of wide absorption and narrow fluorescence energy bands, multiple types of signals can be generated by the combination of fluorescence wavelengths from different QDs, which enables the simultaneous detection of more than two biomarkers. In this review, the advantages and applications of QDs and QD nanobeads (QBs) in multiple biomarker assays were described, and new developments or improvements in multiplexed biomarker detection techniques were summarized. In particular, recent reports were summarized, focusing on the design strategies in immunoassay construction and signal transducing materials for fluorescence-linked immunosorbent assays using QDs and immunochromatographic assays using QBs. New detection platforms will be developed for early diagnosis of diseases and other fields if multiplexed detection technologies of excellent accuracy and sensitivity are combined with artificial intelligence algorithms.

A Study on Biomaterial Detection Using Single-Walled Carbon Nanotube Based on Interdigital Capacitors (인터디지털 커패시트 기반의 단일벽 탄소 나노 튜브를 이용한 바이오 물질 검출에 관한 연구)

  • In this paper, we have studied on the possibilities of the biomaterial detection using single-walled carbon nanotube (SWNT) based on interdigital capacitors. For the four different configurations, such as interdigital capacitor, SWNT in the $5\;{\mu}m$ gap interdigital capacitor, biotinlated SWNT, and biotin and sreptavidin immobilization cases, the resonant frequency has been measured as 10.02 GHz, 11.02 GHz, 10.82 GHz, and 10.22 GHz, respectively. Assuming that the resonant frequency reflects the capacitance changes due to binding of two-different permittivity biomaterials, we have suggested an equivalent circuit model based on measured results, confirming the capacitance changes. For biotinlated SWNT and biotin-streptavidin immobilization cases, the capacitances are $C_b=0.55\;pF$ and $C_s=0.95\;pF$. In this work, we experimentally demonstrated that the specific biomaterial binding causes the capacitance change and therefore this gives rise to resonant frequency. In conclusion, we confirmed the sufficient possibility as CNT biosensor because an analyte biomaterial(streptavidin) binding arouses a considerable resonant frequency change.

Cellular Force Sensing for Force Feedback-Based Biological Cell Injection (힘 피드백 기반의 세포조작을 위한 세포막 침습력 측정)

  • In biological cell manipulation, manual thrust or penetration of an injection pipette into an embryo cell is currently performed by a skilled operator, relying on visual feedback information only. Accurately measuring cellular forces is a requirement for minimally invasive cell injections. Moreover, the cellular force sensing is essential in investigating the biophysical properties for cell injury and membrane modeling studies. This paper presents cellular force measurements for the force feedback-based biomanipulation. Cellular force measurement system using piezoelectric polymer sensor is implemented to measure the penetration force of a zebrafish egg cell. First, measurement system setup and calibration are described. Second, the force feedback-based biomanipulation is experimentally carried out. Experimental results show that it successfully supplies real-time cellular force feedback to the operator at tens of uN and thus plays a main role in improving the reliability of biological cell injection tasks.

  • Surface enhanced Raman scattering (SERS) is considered as one of promising medical and diagnostic technologies. The SERS effect is caused by the localized surface plasmon resonance (LSPR) from metal nanoparticles with narrow hot spots. The mechanism of LSPR, development of nanostructure fabrication, and corresponding researches are discussed. The flexible, label-free, low-cost, and highly-sensitive Au/ZnONRs/G is introduced. The Au/ZnONRs/G detects and distinguishes cataract, age-related macular degeneration, and diabetic macular edema from aqueous humor. Comprehension of SERS provides further improvement in bio sensing technology including early diagnosis and prolonged life expectancy.realize highly stretchable electrodes.

  • The mass change during the molecular interaction between explosive specific bio-receptors and target molecules has been measured using quartz crystal microbalance(QCM), which has a mass change detection limit up to ~ng/$cm^2$. The environmental effect on the molecular interaction has been evaluated. In the liquid phase molecular interaction experiments, the high selectivity of the bio-receptor to DNT compared with toluene has been shown and the sensitivity for various concentrations of DNT has been demonstrated.

  • 도서관에서 사용자들이 책을 빌리거나 반납하기 위해서는 'ㄱ'부터 시작하여 'ㅎ'까지 순서대로 진열이 되어 있는 책의 위치를 찾아가야 한다. 책을 반납할 때, 책을 다시 제자리에 놓는다면 문제가 없지만 사람들이 책을 읽고 진열대에 다시 넣는 과정에서 착오가 생겨, 다른 진열대 넣는 경우, 도선관 사서가 이를 찾아내, 올바른 위치에 놓아야 한다. 또한, 사용자들이 반납한 책들을 도서관 사서가 실수로 다른 곳에 진열할 수도 있다. 본 논문에서는 이러한 문제점을 줄이기 위해, 지능형 모바일 증강현실에 기반한 도서 위치 관리 방법을 제시한다. 제시된 방법에서는 책의 위치 정보, 각도 등 다양한 센싱 정보를 활용하여 진열된 책들이 제자리에 있는지를 실시간으로 점검하고 책의 위치가 잘못 되었을 경우, 도서관 사서에게 이를 알려 책의 위치를 바로 잡도록 하였다.

Rapid Detection Methods for Biogenic Amines in Foods (식품 내 바이오제닉아민 신속검출기술 개발 동향)

  • Biogenic amines have been used as chemical indicators to estimate bacterial spoilage of foods, particularly fish and fish products, cheese, and fermented foods. So far many chromatography methods have been developed to detect biogenic amines in foods. Although these instrumental analyses exhibit good sensitivity, they cannot be used as rapid detection methods due to the chemical treatment of the samples and the time-consuming process involved. For the rapid and simple detection of biogenic amines, enzyme linked immunosorbent assay kits are commercially available. In addition, analytical systems with enzyme-based amperometric biosensor detection have been increasingly developed. The biosensors used to detect the biogenic amines are based on the action of either amine oxidases or amine dehydrogenases that catalyzes the oxidative deamination of biogenic amines to the corresponding aldehydes and ammonia. This review mainly focused on the principle, development, and applications of the detection methods for rapid detection of biogenic amines in foods.

Integration of immunohistochemical reactions into Electrochemical and Optical Analyses of Biochips (면역 조직화학 반응이 통합된 바이오칩의 전기화학 및 광학적 분석)

  • We have addressed two important issues of immunosensing biochips, including the construction of antibody functionalized suface for efficient affinity reactions and the development of a signal registration strategy that converts biospecific reactions into highly quantifiable electrochemical and/or optical signals. The developed immunoassay reaction is an integrated version of enzyme-mediated immunoprecipitaion reaction, which is widely used in immunohistochemistry, and electrochemical signaling reaction. For the evaluation of analytical performance of fabricated immunosensing biochips, signaling for mouse IgG in antiserum was conducted. Applications of the developed strategy have been found for the evaluation of histology chemicals and for the signal amplification for array-type biochip analysis.

Nonenzymatic Sensor Based on a Carbon Fiber Electrode Modified with Boron-Doped Diamond for Detection of Glucose (보론 도핑 다이아몬드로 표면처리된 탄소섬유 기반의 글루코스 검출용 비효소적 바이오센서)

  • In this study, we demonstrated that the nonenzymatic glucose sensor based on the flexible carbon fiber bundle electrode with BDD nanocomposites (CF-BDD electrode). As a nano seeding method for the deposition of BDD on flexible carbon fiber, electrostatic self-assembly technique was employed. Surface morphology of BDD coated carbon fiber electrode was observed by scanning electron microscopy. And the electrochemical characteristics were investigated by cyclic voltammetry, electrochemical impedance spectroscopy and chronoamperometry. This CF-BDD electrode exhibited a large surface area, a direct electron transfer between the redox species and the electrode surface and a high catalytic activity, resulting in a wider linear range (3.75~50 mM), a faster response time (within 3 s) and a higher sensitivity (388.8 nA/mM) in comparison to a bare CF electrode. As a durable and flexible electrochemical sensing electrode, this brand new CF-BDD scheme has promising advantages on various electrochemical and wearable sensor applications.

A Study on the Blue Fluorescence Characteristics of Silica Nanoparticles with Different Particle Size (실리카 나노 입자의 크기에 따른 청색 형광 특성 연구)

  • Organic dye-doped silica nanoparticles are used as a promising nanomaterials for bio-labeling, bio-imaging and bio-sensing. Fluorescent silica nanoparticles(NPs) have been synthesized by the modified $St{\ddot{o}}ber$ method. In this study, dye-free fluorescent silica NPs of various sized were synthesized by Sol-Gel process as the modified $St{\ddot{o}}ber$ method. The functional material of APTES((3-aminopropyl)triethoxysilane) was added as an additive during the Sol-Gel process. The as-synthesized silica NPs were calcined at $400^{\circ}C$ for 2 hours. The surface morphology and particle size of the as-synthesized silica NPs were characterized by field-emission scanning electron microscopy. The fluorescent characteristics of the as-synthesized silica NPs was confirmed by UV lamp irradiation of 365 nm wavelength. The photoluminescence (PL) of the as-synthesized silica NPs with different size was analyzed by fluorometry. As the results, the as-synthesized silica NPs exhibits same blue fluorescent characteristics for different NPs size. Especially, as increased of the silica NPs size, the intensity of PL was decreased. The blue fluorescence of dye-free silica NPs was attributed to linkage of $NH_2$ groups of the APTES layer and oxygen-related defects in the silica matrix skeleton.
