Development of Probiotic Microcapsules for the Preservation of Cell Viability (생균활성 보존을 위한 유산균 미세캡슐 개발)

  • Lactobacillus fermentum YL-3 was encapsulated to increase acid tolerance and its total viability. After micro-encapsulation of L. fermentum YL-3 cells with sodium alginate and soybean oil, the morphology of the microcapsule was observed using confocal laser scanning microscopy (CLSM) after staining with pyronin Y and fluorescein isothiocyanate. The sizes of the microcapsules were 120-126 ${\mu}m$, 444-486 ${\mu}m$ and 401-463 ${\mu}m$ when manufactured at pH 2, 3 and 4, respectively. The microcapsule could hold live cells of L. fermentum YL-3 up to $1.2{\times}10^{7}$, $8.1{\times}10^{7}$ and $1.1{\times}10^{8}$ CFU/mL at pH 2, 3 and 4, respectively. The acid tolerance and preservative ability of L. fermentum YL-3 in microcapsule and macrocapsule at $4^{\circ}C$ and $25^{\circ}C$ were tested. L. fermentum YL-3 cells were evenly located in the alginate capsule matrix structure and the firmness of microcapsule was highest at pH 2. Micro-encapsulation showed the most effective acid tolerance at pH 2.0 and preservation of viability at $4^{\circ}C$. However, at $25^{\circ}C$, the macrocapsules showed more effective cell protection than the microcapsules. The application range for microcapsules could be wider than for macrocapsules in the food industry.

Nano-capsulation of L-Ascorbic Acid in Nonaqueous System (L-Ascorbic Acid의 비수계 나노 캡슐화)

  • In this work, the basic research for nano-capsulation of L-ascorbic acid (Vitamin C) in nonaqueous system was carried out. 500 nm-sized nano-capsules were prepared in nonaqueous system, and the emulsified capsule had mean size of 410 nm. The stability test on the temperature and the storage periods was performed at 4, 20, and $30^{\circ}C$ for 30 days. After 5 days, L-ascorbic acid was extricated 5.1, 9.3, and 12.5% at each temperature, but only 1~2 % was extricated after the time span. Likewise, the results of the skin susceptibility on women and men, each 10 persons, revealed that the very thin allergy was shown from only a woman after 2 days, but it was not shown from the others.

Stability and Processing Characteristics of Microencapsulated Squid Liver Oil by Fluidized Bed Coating (오징어 간유 미세캡슐의 유동층 코팅에 따른 품질 특성)

  • Squid oil is an abundant source of polyunsaturated fatty acids. This is particularly true for eicosapentaenoic acid and docosahexaenoic acid. The principal objective of this study was to extend the stability and improve the process aptitude of squid liver oil. Fluidized bed coatings were employed for coating with microencapsulated oil. The efficiency of the fluidized bed coating of the microencapsulated powder was over 90%. The apparent density with zein-DP was 0.6 g/mL, thereby indicating that flow ability had been improved as the result of an increase in specific gravity. The solubility of artificial gastric and enteric fluids with HPMC-FCC was 59.9 and 0%, respectively, whereas with zein-DP solubility was 0 and 31.0%, respectively. Polyunsaturated fatty acid retention results demonstrated that zein-DP coating was higher than HPMC-FCC, followed by the microencapsulated squid liver oil method. These results demonstrated that the application of microencapsulation and fluidized bed micro-coating techniques improved the stability and processing compatibility of squid liver oil.

Effect of Ratio of Demineralized Bone Powder with Alginate Microcapsules on Articular Cartilage Regeneration (탈미네랄 골분이 비율별로 포접된 알지네이트 미세캡슐을 이용한 조직공학적 연골재생)

  • Alginate, obtained from the seaweeds, is a widely used biomaterial for cell transplantation, since its positive effect on viability of capsulized cells and its easier encapsulation capability of living cells. Demineralized bone powder (DBP), derived from the natural bone tissue, is widely applied for clinical trials for its low rate of reaction and antigenicity. A chondrocyte was seeded into an alginate with DBP of different contents, and a microcapsule was produced. The adhesion and proliferation of cells was observed through the MTT analysis, and the PCR was applied to estimate the content of the glycosaminoglycan (sGAG) and collagen, and confirm the specific genetic pattern of the chondrocytes. Also, the alginate microcapsule where the chondrocyte is seeded was extracted after transplantation under the skin of a nude mouse, and was immunochemically stained. The experimental result confirmed that the alginate microcapsule containing 1% of DBP not only showed the highest proliferation of cell but had a positive effect of chondrocytes by the interaction between the alginates and the growth factor in DBP. It can be expected that the microcapsule with application of the alginates and DBP might be an appropriate scaffold for tissue engineering.

Preparation and Physicochemical Characteristics of Octenyl Succinated Rice Starches Based on Amylose Content (아밀로오스 함량에 따른 옥테닐호박산 쌀전분의 제조 및 이화학적 특성)

  • The purpose of this study was to investigate the preparation and physicochemical characteristics of n-octenylsuccinylated (OSAn) rice starches with different rice amylose contents. Amylose contents of Jinsumi flour and Milyang 261 flour were 15.42 and 20.31%, respectively. After processing by alkali treatment, amylose contents of Jinsumi rice starch and Milyang 261 rice starch were 34.21% and 39.32%, respectively. After OSAn treatment, the degree of substitution and reaction efficiency of Jinsumi starch were higher than those of Milyang 261. The stability of the modified Jinsumi emulsion was higher than that of the Milyang 261 emulsion. Viscosity of the Jinsumi emulsion was higher than that of the Milyang 261 emulsion. When the emulsions were spray dried, modified Jinsumi rice starch showed excellent coating efficiency compared to that of modified Milyang 261 rice starch. Therefore, Jinsumi was more suitable than Milyang 261 to apply for encapsulation as wall materials.

The Effects of Coating Treatments on Enteric Coating of the Soft Capsules Containing Omega-3 Fatty Acids (오메가-3 연질캡슐의 코팅 조건에 따른 장용성 코팅품질에 미치는 영향)

  • This article presents an evaluation of the effects of coating conditions on the enteric coating quality of soft gelatin capsules containing Omega-3 fatty acids. Three conditions were controlled: concentration of hydroxypropyl methylcellulose phthalate (6, 8, and 10 wt% in solution), temperature of the inlet air (32, 35, and $38^{\circ}C$), and the coating solution feed rate (7.5, 11.25, and 15.0 g/min). The transparency of the enteric coated soft gelatin capsules was evaluated by measuring the degree of whiteness of the surface using a spectrophotometer. Results showed that the most important parameter in the enteric coating process was the coating solution feed rate. As the coating solution feed rate decreased and inlet air temperature increased, the degree of whiteness of coating surfaces decreased. We also evaluated the disintegration properties of the enteric coated capsules in accordance with the Korea Health Functional Food Code.

  • Lauric acid (LA) which is also known as dodecanoic acid has been selected as the phase change material (PCM) owing to eco-friendly in nature. A systematic study has been conducted for encapsulation of LA (core) with silicon dioxide (SiO2) as shell material. Different core-shell ratio was chosen to microencapsulate the LA with 10 ml of tetraethyl orthosilicate (TEOS) as the precursor solution for the formation of SiO2. The synthesis of microencapsulated LA was carried out at 2.5 pH of precursor solution. The synthesized microencapsulated LA are characterized by Fourier transform infrared spectroscope (FT-IR) and X-Ray Diffraction (XRD) which confirmed the presence of SiO2 shell on the surface of LA.

  • The synthesis of stearic acid composite phase change material (PCM) was investigated and the samples produced were characterized for use in latent heat storage, using a simple chemical sol-gel process. The PCM was encapsulated to tetraethyl orthosilicate by various preparation ratios of stearic acid (5, 10, 15, 20, 30 and 50%). Fourier transformation infrared spectroscope (FT-IR) and X-Ray diffraction (XRD) were performed to determine the chemical structure and crystalloid phase of the microencapsulated PCM. SATEOS1 (5%) shows the best proportion for the PCM. With the presence of stearic acid as core materials and SiO2 as the supporting materials, it does not show any chemical reaction between both of them. SATEOS1 shows promising potential for thermal energy storage as it shows a better encapsulation efficiency and good thermal stability.

  • The aim of this study was to evaluate the effects of microencapsulation on the water absorption, DPPH radical scavenging activity (DRSA), hydroxyl radical scavenging activity (HRSA) and tyrosinase inhibition activity (TIA) in corn silk extracts. The lowest value (0.20) of water absorption index (WAI) and the highest value (95.23%) of water solubility index (WSI) were maltodextrin+cyclodextrin microencapulated corn silk extracts (MD+CD) and cyclodextrin encapulated corn silk extracts (CD), respectively. The 18.60% for DRSA value of control was increased to 89.25% for that of CD. The 16.89% for HRSA value of control was increased to 47.46%, and 7.16% for TIA value of control was increased to 39.35% for that of MD+CD, respectively. The MD+CD would be used for functional food and cosmetics materials as antioxidant and skin whitening agents. All investigated responses between control and treatment were statistically significant (p<0.05).

An Implementation of HBC System for Capsule Endoscope (캡슐내시경을 위한 HBC시스템 구현)

  • In this paper, a comprehensive design of HBC(Human Body Communication) system for capsule endoscope is presented. First, we propose a method of combining the signals received from multiple patches attached to the body of patient through differential operation and derive the signal SNR mathematically. To synchronize HBC transmission signal sent from capsule, we analyzed coarse timing synchronization method using PN code and fine timing synchronization performance among Manchester, NRZ and RZ modulation method using ZCD(Zero Crossing Detector). In addition, we evaluated the equalization performance of HBC signal frame in Rician and Rayleigh channel environments by applying LMS and RLS algorithm.
