Inactivation of Microorganisms and Enzymes in Foxtail Millet Yakju by High Hydrostatic Pressure Treatment (초고압 처리에 의한 좁쌀약주의 미생물 살균 및 효소 불활성화)

  • High hydrostatic Pressure was applied to Foxtail Millet Yakju to investigate the effects of high pressure on inactivation of microorganisms and enzymes. Total bacteria, lactic acid bacteria and yeast in untreated Yakju were $1.5{\times}$10$^4$,1.9${\times}$10$^4$ and 1.4${\times}$10$^4$ CFU/mL, respectively. Total bacterial count was reduced to 4.1${\times}$l0$^2$ CFU/mL, while lactic acid bacteria and yeast were sterilized completely in Yakju heated at $65^{\circ}C$ for 15 min. Lactic acid bacteria and yeast decreased greatly with the increase of treatment pressure, and were sterilized completely in Yakju treated at more than 300 ㎫ for 10 min/$25^{\circ}C$. Total bacteria were not completely sterilized with pressurization of even 600 ㎫ at room temperature and reduced to 2 log cycle even at $65^{\circ}C$. Total bacteria decreased by 2∼3 log cycle with the increase of treatment time from 10 to 60 min at $25^{\circ}C$/300 ㎫. Pressurization of Yakju caused a partial inactivation of $\alpha$ -amylase and glucosamylase, and the activities of $\alpha$ -amylase and glucoamylase decreased by 18.1% and 21.1%, respectively at $25^{\circ}C$/600 ㎫/10 min. Activities of $\alpha$ -amylase and glucoamylase decreased with the increase of temperature, and 22.2% and 32.1% of the original activity were remained with the treatment at $65^{\circ}C$/300 ㎫/10 min, respectively. Enzyme activities decreased slightly with the increase of treatment time at $65^{\circ}C$/300 ㎫.

Effect of Gamma Irradiation on the Sterilization of Red Pepper Powder (고추가루의 살균(殺菌)을 위한 ${\gamma}$-선(線) 조사효과)

  • In order to investigate the hygienic status of commercial red pepper powder, one sample was directly prepared from raw red pepper which was purchased at market and the other one was a commercial red pepper powder. They were used for the investigation upon the status of microbial contamination, effect of irradiation for sterilization and physicochemical changes during 3 months of storage. Total bacterial counts of commercial red pepper powders were 3.83 to $6.68{\times}10^6$ per gram and coliform group was shown to be positive in some products. Total bacteria and coliform group were sterilized by the irradiation of 9 kGy and 3 kGy, respectively and no microorganisms were grown up until 3 months of storage at room temperature. The $D_{10}$ values of total viable bacteria in red pepper powders were 1.52 to 1.58 kGy. Chemical components such as moisture, total and reducing sugars were slightly decreased during the storage period. The contents of capsanthin and capsaicin were partly affected by the high dose irradiation but the difference between unirradiated and irradiated groups was diminished with the elapse of storage period.

Analysis of Tubulysin Biosynthetic Genes in Archangium gephyra (Archangium gephyra의 tubulysin 생합성 유전자 분석)

  • Tubulysins are a group of bioactive secondary metabolites from myxobacteria exhibiting strong anticancer activity against various cancer cell lines. In this study, we describe the identification of putative tubulysin biosynthetic gene clusters (tubA~tubF) in the genome sequences of two tubulysin-producing myxobacterial strains, Archangium gephyra MEHO_002 and MEHO_004. The inactivation of the putative tubulysin biosynthetic genes resulted in a tubulysin-production defect. The DNA sequences of the A. gephyra MEHO_002 and MEHO_004 tubulysin biosynthetic genes were 97% identical, and the amino acid sequences of the encoded proteins shared a similarity of 97-100%. The nucleotide sequences of the tubulysin biosynthetic gene clusters in MEHO_002 and MEHO_004 were 86% identical to that in Cystobacter sp. SBCb004 known as a tubulysin-producing myxobacterium, and the organization of the clusters was identical except for the lack of a tubZ gene in the clusters in MEHO_002 and MEHO_004. The amino acid sequences of the proteins encoded by each gene were 88-97% similar to those encoded by SBCb004, and the domain compositions of the proteins were also identical.

Nutritional quality of Peucedanum japonicum Thunb. leaves in relation to ripening time, growing condition and blanching (재배조건, 수확시기 및 열처리에 따른 갯기름나물의 영양성분 변화)

  • This work aimed to investigate the changes in the nutrient contents and antioxidant activity of Peucedanum japonicum Thunb. by ripening stage, growing condition, and blanching. The crude protein content of the young leaves (3.6~4.3%) was higher than that of the mature leaves (3.1~3.9%). Higher calcium contents were observed in the greenhouse-cultivated samples (225.9~259.2 mg/100 g) compared to the field-cultivated samples (178.5~199.5 mg/100 g). The vitamin C and folate contents (18.1~83.8 and 175.8~220.2 mg/100 g, respectively) of the field-cultivated samples were significantly (p<0.05) higher than those of the greenhouse-cultivated samples (13.1~57.7 and 133.0~148.8 mg/100 g, respectively). The growing condition and blanching were significant factors affecting the changes in the vitamin and polyphenol contents. The ${\beta}$ carotene contents of the blanched samples increased 2.6-fold compared to those of the raw samples. The total polyphenol contents (10.2~17.1 mg/g extract) and DPPH radical scavenging activity ($IC_{50}=2.0{\sim}3.0mg/ml$) of the blanched samples were significantly (p<0.05) higher than those of the raw samples (1.8~4.3 and $IC_{50}=16.2{\sim}21.1mg/ml$, respectively).

  • 최근 근관 치료 영역에서는 적절한 기계적 근관 성형과 근관 세척으로만 효과적으로 근관 내 미생될의 숫자를 감소시킬 수 있어 다른 약제의 사용은 권장되고 있지 않다. 그럼 에도 불구하고 수종의 근관내 약제는 감염된 근관에서 미생물의 숫자를 줄이고 근관 내용물의 불활성화와 삼출액을 줄이기 위해 사용되고 있다. 그 중 포름 알데하이드를 함유하고 있는 Depulpin$^{(R)}$과 근관 치료학에서 오랫동안 널리 사용되어온 수산화 칼슘을 포함하고 있는 Tempcanal$^{(R)}$과 Vitapex$^{(R)}$ 등에 대한 세포 독성은 충분한 연구가 이루어지지 못한 상태이다. 이에 본 연구에서는 이들 약제가 백서에서의 세포 독성에 미치는 영향에 관하여 고찰하고자 하였다. Sprague-Dawley계 백서 숫놈 20마리를 사용하여, 각각의 쥐는 케타민과 럼푼을 근육내 주사하여 마취하였고, 복부의 피하 부위를 절개한 뒤 3개씩의 Teflon-coating된 매식체를 삽입하였다 매식체 안에는 각각 Tempcanal$^{(R)}$, Vitapex$^{(R)}$, Depulpin$^{(R)}$을 넣고, 백서 20마리를 6개군으로 나누어 매식체 삽입 후 1주 뒤, 4주 뒤에 희생시켜 매식체 주위 조직을 잘라내고 10%포르말린에 고정 후 파라핀에 포매 하였다 미세 절단기로 4$\mu$m로 연속 절단 후, hematoxy-line-eosin염색 후 3명의 관찰자가 광학 현미경으로 관찰하여 염증의 정도를 4단계로 평가한 뒤 Kruskall-Wallis test(P<0.05)로 통계 처리하였다. 그 결과 제 1군Tempcanal$^{(R)}$ 7일후 군), 제 2군(Vitapex$^{(R)}$ 7일후 군), 제 3군(Depulpin$^{(R)}$ 7일후 군) 모두 중등도의 염증도를 보였으나, 세 군간의 통계학적 유의성은 없었다. 그러나, 제 4군(Tempcanal$^{(R)}$ 30일후 군)과, 제 5군(Vitapex$^{(R)}$ 30일후 군)의 경우에서는 약한 염증도를 보여주었으나, 제 6군(Depulpin$^{(R)}$ 30일후 군)은 가장 심한 염증 반응과 함께 조직 괴사의 양상을 보여주었으며, 4, 5군과 6군간에 통계학적 유의성을 보였다. 본 실험 결과, Depulpin$^{(R)}$은 Tempcanal$^{(R)}$와 Vitapex$^{(R)}$에 비해 높은 세포 독성을 보여주공 있으나, 좀 더 많은 임상적 검증이 필요할 것으로 사료된다.

Water Quality Variation and Removal Characteristics of Poliovirus by Biological Activated Carbon (BAC) and Ozone Treatment Process in Nakdong River. (낙동강 원수의 생물활성탄 및 오존처리공정에 따른 수질 변화 및 폴리오바이러스의 제거특성)

  • Ozonation is a disinfection technique of harmful mi-crobes commonly used in the treatment of drinking water. And Biological Activated Carbon (BAC) treatment also provides numerous benefits for drinking water utilities, including removal of micro- pollutants, improved treatment processes. The multiful-stage ozonation and BAC play roles as effective methods for removing several materials in raw water. Water quality variation in Nak dong river and the removal efficiency of viruses by ozonation-BAC process were investigated on pilot scale. During the period of survey, most of water quality parameters including $NH_{4}^{+}-N$ were highly improved after passing through the BAC. The removal efficiency of poliovirus type III in water treatment process using pilot-plant,$ 99.6\% $ of viruses were removed by pre-ozonation, sedimentation and sand filteration process, $ 100\% $ were removed after in BAC filteration step. In the removal survey of viruses by ozonation, ap-proximately $ 61.1\% $ or polioviruses were inactivated by ozone of 0.4 mg/l within 5 min. and $ 100\% $ were inactivated by ozone of 0.8 mg/l over 10 min.

Inactivation of Xanthomonas citri subsp. citri and Effect on Infection of Citrus Canker by Gamma Irradiation (감마선에 의한 감귤 궤양병균의 불활성화와 감귤 궤양병 감염에 미치는 영향)

  • Citrus canker caused by Xanthomonas citri subsp. citri (Xcc) has been quarantined by many countries in the world. Recently, the usage of methyl bromide should be limited, application by gamma irradiation on the agricultural production is raised as an alternative method. In this study, the level of gamma irradiation which could decrease of population of Xcc in the suspension or on the surface of citrus fruit was investigated. The $D_{10}$ value of Xcc, which is radiation dose required to reduce the number of the microorganism, was 55 and 28 Gy in the suspension and on the surface of citrus fruit, respectively. Furthermore, disease severity was suppressed on the citrus leaves inoculated with Xcc suspension pre-treated with gamma irradiation. Based on this study, it is suggested that Xcc on the citrus fruit could be eradicated by gamma irradiation and the results of this study may be valuable for application of gamma ray in quarantine activity.

Biocontrol Efficacy of Endophytic Bacteria Flavobacterium hercynim EPB-C313 for Control of Chinese Cabbage Clubroot (Flavobacterium hercynium EPB-C313 균주를 이용한 배추 뿌리혹병 생물적 방제)

  • Clubroot of Chinese cabbage by Plasmodiophora brassicae, was found to be high virulent to the Chinese cabbage, turnips and cabbage. It this study, the endophytic bacteria Flavobacterium hercynium EPB-C313, which was isolated from tissues of Chinese cabbage, was investigated the antimicrobial activity on the inactivation of resting spores and its control effect on clubroot disease by bioassay. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivate the resting spores of P. brassicae with 90.4, 36.8, and 26.0%, respectively. The clubroot was inhibited with 100% by dipping the seedlings of Chinese cabbage in culture solutions of F. hercynium EPB-C313 before planting, however the chemmical 'fluazinam' was 91.7% in pot tests. Complex treatment were highly enhanced control efficacy with 63.7% at field of 50% diseased plants by soil incorporation with the pellet contains organic matter and F. hercynium EPB-C313, seedling drench of culture solution of F. hercynium EPB-C313 and soil drench with 10 days after planting. These results imply that the F. hercynium EPB-C313 is a very useful biological control agent of clubroot disease of Chinese cabbage.

Method for Increasing the Stability and Activity of Polygalacturonase and Its Application to the Production of Vegetable Single Cell (Polygalacturonase의 활성 증진 및 이를 이용한 식물 단세포 제조 방법)

  • This study was carried out to enhance the stability and activity of polygalacturonase (PGase) purified from Kluyveromyces marxianus IFO 0288. Gums such as xanthan gum, guar gum, and locust bean gum were capable of increasing the catalytic stability and activity of the PGase. At $30^{\circ}C$, the rate constants for the inactivation of the PGase with xanthan gum, guar gum, and locust bean gum were estimated to be $0.0003min^{-1}$, below $0.0001min^{-1},\;and\;0.0001min^{-1}$ respectively, whereas control was estimated to be $0.0082min^{-1}$. The yield of the maceration reaction catalyzed by the PGase for the production of carrot single cells increased by 13% in the presence of guar gum, where the relative enzyme activity supplemented with guar gum was two-fold greater than that of the PGase alone.

Pasteurization Efficiency and Physico-chemical Changes of Soymilk HTST Pasteurized Using Microwaves (두유의 마이크로파 고온단시간 살균시 살균효과 및 이화학적 성분 변화)

  • This work was to determine the microbial and physico-chemical changes of HTST-pasteurized soymilk using microwave energy. Soymilk was HTST pasteurized$(at\;90^{\circ}C\;for\;20\;sec)$ by three methods: by heating in a stainless steel tube immersed in a hot water bath(MP0), by heating in a microwave cavity to a defiled temperature and then holding in a hot water bath(MP1), and by both heating and holding in a microwave cavity(MP2). The microbial quality based on the total plate count was in the order of MP0, MP2 and MP1. The three samples pasteurized by different methods showed the similar microbial quality with respect to the coliform count, psychrotrophic bacterial count and phosphatase activity. The destruction of trypsin inhibitor was in the order of MP0, MP1 and MP2. There were no significant differences in pH, titratable acidity, viscosity and vitamin $B_2$ content before and after pasteurization and among the different pasteurization methods. The similar or higher quality retention of the MP1 or MP2 supports the possibility of using microwave energy for the HTST pasteurization of soymilk and other fluid food products.
