Investigation of Furfural Yields of Liquid Hydrolyzate during Dilute Acid Pretreatment Process on Quercus Mongolica using Response Surface Methodology (신갈나무 약산 전처리 공정 중 반응표면분석법을 이용한 액상 가수분해물의 furfural 수율 탐색)

  • In this study, furfural, which is one of the value-added chemicals, was produced from the hydrolyzate of Quercus mongolica using dilute acid pretreatment, and the optimal pretreatment condition was determined by Response Surface Methodology (RSM) to obtain high yield of furfural. Based on Central Composite Design, the pretreatment experiment was designed with parameters such as reaction temperature ($X_1$), acid concentration ($X_2$), and reaction time ($X_3$) as independent variables, while dependent variable was furfural concentration (Y), and furfural yield (Z) was shown as percentage of Y per a dry weight basis. According to results of RSM, it was confirmed that reaction temperature ($X_1$) was the most influence factor and reaction temperature ($X_1$)-acid concentration ($X_2$) was the most significant interaction factor on furfural yield. Also, the optimal condition for the highest furfural yield was predicted at reaction temperature of $184^{\circ}C$, acid concentration of 1.17%, and reaction time of 5 min by RSM, and expected maximum yield of furfural was 6.37%. Experimentally, the maximum yield of furfural produced at above optimal condition was 6.21%, and it was considerably similar with the predicted value, and therefore the model for furfural production from the hydrolyzate of Quercus mongolica during dilute acid pretreatment could be built using RSM.

Analysis of Characteristics and Optimization of Photo-degradation condition of Reactive Orange 16 Using a Box-Behnken Method (실험계획법 중 Box-Behnken(박스-벤켄)법을 이용한 반응성 염료의 광촉매 산화조건 특성 해석 및 최적화)

  • The aim of our research was to apply experimental design methodology in the optimization of photocatalytic degradation of azo dye(Reactive orange 16). The reactions were mathematically described as a function of parameters amount of $TiO_2(x_1)$, and dye concentration($x_2$) being modeled by the use of the Box-Behnken method. The results show that the responses of color removal(%)($Y_1$) in photocatalysis of dyes were significantly affected by the synergistic effect of linear term of $TiO_2(x_1)$ and dye concentration($x_2$). Significant factors and synergistic effects for the $COD_{Cr}$, removal(%)($Y_2$) were the linear term of $TiO_2(x_1)$ and dye concentration($x_2$). However, the quadratic term of $TiO_2(x_1^2)$ and dye concentration($x_2^2$) had an antagonistic effect on $Y_1$ and $Y_2$ responses. Canonical analysis indicates that the stationary point was a saddle point for $Y_1$ and $Y_2$, respectively. The estimated ridge of maximum responses and optimal conditions for $Y_1:(X_1,\;X_2)$=(1.11 g/L, 51.2 mg/L) and $Y_2:(X_1,\;X_2)$=(1.42 g/L, 72.83 mg/L) using canonical analysis was 93% and 73%, respectively.

Effect of Korean Mistletoe Extract and Lectin on the Preneoplastic Hepatic Lesion and Apoptosis in Experimental Hepatocarcinogenesis (실험적 간암모델에서 한국산 겨우살이 추출물 및 렉틴 투여가 전암성 병변의 생성 및 Apoptosis에 미치는 영향)

  • This study was done to investigate the effects of Korean mistletoe water extract and lectin on the apoptosis and preneoplastic lesion in chemically induced rat hepatocarcinogenesis. To attain the above objectives, weanling Sprague-Dawley male rats were fed modified AIN-76 diets containing 10% corn oil for 9 weeks. One week after feeding starts, rats were intraperitoneally injected twice with a dose of diethylnitrosamine (DEN, 50 mg/kg body weight (BW). Rats were provided with 0.05% phenobarbital (PB) in drinking water from one week after DEN treatment until the end of experiment. During the period of PB treatment, rats were injected with mistletoe extract (100 $\mu\textrm{g}$/kg BW) and lectin (10 $\mu\textrm{g}$/kg BW) twice a week. At the end of 9th week, rats were sacrificed and the formation of hepatic glutathione S-transferase placental form positive (GST-P$^{+}$) foci, apoptosis, DNA fragmentation and apoptosis related proteins were determined respectively. The formation of GST-P$^{+}$foci was significantly decreased by mistletoe extract or lectin treatment. Although there was no effect on apoptosis and DNA fragmentation in hepatic tissue by mistletoe extract or lectin treatment, caspase-9 and fas-L were increased. These results suggest that Korean mistletoe extract and lectin have a potential to inhibit hepatocarcinogenesis by increasing apoptosis.sis.

  • Background: Ethyl pyruvate (EP) is a derivative of pyruvate that has recently been identified by both various in vitro and in vivo studies to have antioxidant and anti-inflammatory effects. The aim of this study was to determine the effect of EP on lipopolysaccharide (LPS)-induced acute lung injury (ALI). Methods: 5 weeks old, male BALB/c mice were used. ALI was induced by an intratracheal instillation of LPS 0.5mg/Kg/$50{\mu}L$ of saline. The mice were divided into the control, LPS, EP+LPS, and LPS+EP groups. In the control group, balanced salt solution was injected intraperitoneally 30 minutes before or 9 hours after the intratracheal instillation of saline. In the LPS group, a balanced salt solution was also injected intraperitoneally 30 minutes before or 9 hours after instillation the LPS. In the EP+LPS group, 40mg/Kg of EP was injected 30 minutes before LPS instillation. In the LPS+EP group, 40mg/Kg of EP was injected 9 hours after LPS instillation. The TNF-$\alpha$ and IL-6 concentrations in the bronchoalveolar lavage fluid (BALF), and that of NF-$\kappa$B in the lung tissue were measured in the control, LPS and EP+LPS groups at 6 hours after instillation of saline or LPS, and the ALI score and myeloperoxidase (MPO) activity were measured in all four groups 24 and 48 hours after LPS instillation, respectively. Results: The TNF-$\alpha$ and IL-6 concentrations were significantly lower in the EP+LPS group than in the LPS group (p<0.05). The changes in the concentration of these inflammatory cytokines were strongly correlated with that of NF-$\kappa$B (p<0.01). The ALI scores were significantly lower in the EP+LPS and LPS+EP groups compared with the LPS group (p<0.05). In the EP+LPS group, the MPO activity was significantly lower than the LPS group (p=0.019). Conclusion: EP, either administered before or after LPS instillation, has protective effects against the pathogenesis of LPS-induced ALI. EP has potential theurapeutic effects on LPS-induced ALI.

About Short-stacking Effect of Illite-smectite Mixed Layers (일라이트-스멕타이트 혼합층광물의 단범위적층효과에 대한 고찰)

  • Illite-smectite mixed layers (I-S) occurring authigenically in diagenetic and hydrothermal environments reacts toward more illite-rich phases as temperature and potassium ion concentration increase. For that reason, I-S is often used as geothermometry and/or geochronometry at the field of hydrocarbons or ore minerals exploration. Generally, I-S shows X-ray powder diffraction (XRD) patterns of ultra-thin lamellar structures, which consist of restricted numbers of sillicate layers (normally, 5 ~ 15 layers) stacked in parallel to a-b planes. This ultra-thinness is known to decrease I-S expandability (%S) rather than theoretically expected one (short-stacking effect). We attempt here to quantify the short stacking effect of I-S using the difference of two types of expandability: one type is a maximum expandability ($%S_{Max}$) of infinite stacks of fundamental particles (physically inseparable smallest units), and the other type is an expandability of finite particle stacks normally measured using X-ray powder diffraction (XRD) ($%S_{XRD}$). Eleven I-S samples from the Geumseongsan volcanic complex, Uiseong, Gyeongbuk, have been analyzed for measuring $%S_{XRD}$ and average coherent scattering thickness (CST) after size separation under 1 ${\mu}m$. Average fundamental particle thickness ($N_f$) and $%S_{Max}$ have been determined from $%S_{XRD}$ and CST using inter-parameter relationships of I-S layer structures. The discrepancy between $%S_{Max}$ and $%S_{XRD}$ (${\Delta}%S$) suggests that the maximum short-stacking effect happens approximately at 20 $%S_{XRD}$, of which point represents I-S layer structures consisting of ca. average 3-layered fundamental particles ($N_f{\approx}3$). As a result of inferring the $%S_{XRD}$ range of each Reichweite using the $%S_{XRD}$ vs. $N_f$ diagram of Kang et al. (2002), we can confirms that the fundamental particle thickness is a determinant factor for I-S Reichweite, and also that the short-stacking effect shifts the $%S_{XRD}$ range of each Reichweite toward smaller $%S_{XRD}$ values than those that can be theoretically prospected using junction probability.

Effect of the Solvent Fractions of Zingiber officinale Roscoe against Thrombintreated Tumor Invasion in Human Oral Squamous Carcinoma YD-10B Cells (YD-10B 인간구강암세포주에서 생강 유기용매 분획물의 항산화, 트롬빈억제 및 thrombin에 의해 처리된 암 침윤 및 전이 억제 효과)

  • Oral squamous cell carcinoma (OSCC) is a common malignant tumor in the oral cavity, comprising up to 90% of oral cancer. Oral cancer is characterized by a marked tendency of local invasiveness and is good for early detection and treatment; therefore, it is recognized as a good model for cancer prevention. The present study investigated the antioxidant, thrombin inhibitory, and anti-invasive activities of the solvent fractions of Zingiber officinale Roscoe. Samples were fractionated into hexane, chloroform, ethyl acetate, butanol, and water fractions, and each of these was assayed individually. The water fraction showed the highest extraction yield at 9.79%(w/w). Anti-oxidative activity was analyzed by DPPH assay. Thrombin inhibitory activity was used to analyze thrombin inhibitor assay. Cell viability was detected by the MTS assay. The activity and mRNA expression of MMP-2 and MMP-9 in human oral squamous carcinoma YD-10B cells were examined by zymography and RT-PCR. The antioxidative activities of hexane and water fractions were 92.38% and 92.96%, respectively. In the thrombin inhibitory activity test, water fraction was the highest, with a value of 65.86%. MMP-2/-9 activation was increased in phorbol 12-myristate 13-acetate (PMA)-induced YD-10B cells. MMP-9 activation was increased in thrombin-treated YD-10B cells. In PMA- or thrombin-treated YD-10B cells, the increased mRNA expression and protein activation of MMP-2/-9 were significantly inhibited in the hexane fraction. Therefore, the hexane fraction obtained from a Zingiber officinale Roscoe water extract is a promising therapeutic anti-invasive agent in oral cancer.

Effect of Thyroid Hormone on the Ischemia-Reperfusion Injury in the Canine Lung (갑상선 호르몬이 잡견 폐장의 허혈-재관류 손상에 미치는 영향)

  • Background: Ischemia-reperfusion injury is one of the major contributing causes of early graft failure in lung transplantation. It has been suggested that triiodothyronine (T3) may ameliorate ischemia-reperfusion injury to various organs in vivo and in vitro. Predicting its beneficial effect for ischemic lung injury, we set out to demonstrate it by administering T3 into the in situ canine ischemia-reperfusion model. Material and Method: Sixteen adult mongrel dogs were randomly allocated into group A and B. T3 $(3.6\mug/kg)$ was administered before the initiation of single lung ischemia in group B, whereas the same amount of saline was administered in group A. Ischemia was induced in the left lung by clamping the left hilum for 100 minutes. After reperfusion, various hemodynamic parameters and blood gases were analyzed for 4 hours while intermittently clamping the right hilum in order to allow observation of the injured left lung function. Result: Arterial oxygen partial pressure $(PaO_2)$ decreased 30 minutes after reperfusion and recovered gradually thereafter in both groups. In group B the decrease of $PaO_2$ was less marked than in group A. The recovery of $PaO_2$ was faster in group B than in group A. The differences between the two groups were statistically significant from 30 minutes after reperfusion $(125\pm34$ mmHg and $252\pm44$ mmHg, p<0.05) until the end of the experiment $(178\pm42$mmHg and $330\pm37$ mmHg, p<0.05). The differences in the arterial carbon dioxide pressure, airway pressure and lung compliance showed no statistical significance. The malondialdehyde (MDA) level, measured from the tissue obtained 240 minutes after reperfusion, was lower in group B $(0.40\pm0.04\mu$M) than in group A $(0.53\pm0.05\mu$M, p<0.05). The ATP level of group B $(0.69\pm0.07\mu$M/g) was significantly higher than that of group A $(0.48\pm0.07\mu$M/g, p<0.05). The microscopic exami nation revealed varying degrees of injury such as perivascular neutrophil infiltration, capillary hemorrhage and interstitial congestion. There were no differences in the microscopic findings between the two groups. CONCLUSION T3 has beneficial effects on the ischemic canine lung injury including preservation of oxygenation capacity, less production of lipid peroxidation products and a higher level of tissue ATP. These results suggest that T3 is effective in pulmonary allograft preservation.

Anti-Allergic Activities of Fermented Eriobotrya japonica and Saurus chinensis Extracts in 2,4-Dinitrochlorobezene-Induced BALB/c Mice (2,4-Dinitrochlorobenzene으로 유도된 아토피 피부염 동물모델에서 비파엽 및 삼백초 추출발효물의 항아토피 활성)

  • This study was conducted to evaluate the effects of Eriobotrya japonica and Saurus chinensis extracts and their fermented extracts on immune parameters in BALB/c mice treated with 1% 2,4-dinitrochlorobenzene (DNCB). The groups were Eriobotrya japonica extract (BI), Saurus chinensis extract (SA), mixture with E. japonica extract and S. chinensis extract (FB) and fermented mixture with E. japonica extract and S. chinensis extract (FA) and distilled water treated control. The level of serum immunoglobulin (Ig) E was decreased in FA compared to control group, but significant difference was not observed (p<0.05). The histamine and contents in FA and control group were $1.47{\pm}0.20$ ng/mL and $1.90{\pm}0.04$ ng/mL, respectively (p<0.05). Ceramide contents were significantly increased in FA compared to BI, SA, FB and control group (p<0.05). These results suggest that FA supplementation in the DNCB treated BALB/c mice affect anti-allergic activities positively, and may be used as functional material for suppression of atopy dermatitis in food industry.

The effect of semantic categorization of episodic memory on encoding of subordinate details: An fMRI study (일화 기억의 의미적 범주화가 세부 기억의 부호화에 미치는 영향에 대한 자기공명영상 분석 연구)

  • Grouping episodes into semantically related categories is necessary for better mnemonic structure. However, the effect of grouping on memory of subordinate details was not clearly understood. In an fMRI study, we tested whether attending superordinate during semantic association disrupts or enhances subordinate episodic details. In each cycle of the experiment, five cue words were presented sequentially with two related detail words placed underneath for each cue. Participants were asked whether they could imagine a category that includes the previously shown cue words in each cycle, and their confidence on retrieval was rated. Participants were asked to perform cued recall tests on presented detail words after the session. Behavioral data showed that reaction times for categorization tasks decreased and confidence levels increased in the third trial of each cycle, thus this trial was considered to be an important insight where a semantic category was believed to be successfully established. Critically, the accuracy of recalling detail words presented immediately prior to third trials was lower than those of followed trials, indicating that subordinate details were disrupted during categorization. General linear model analysis of the trial immediately prior to the completion of categorization, specifically the second trial, revealed significant activation in the temporal gyrus and inferior frontal gyrus, areas of semantic memory networks. Representative Similarity Analysis revealed that the activation patterns of the third trials were more consistent than those of the second trials in the temporal gyrus, inferior frontal gyrus, and hippocampus. Our research demonstrates that semantic grouping can cause memories of subordinate details to fade, suggesting that semantic retrieval during categorization affects the quality of related episodic memory.

Alteration of Growth Factor Expression after Acute Ischemic Renal Injury (급성 허혈성 신손상 후 여러 성장인자 발현의 변화)

  • Purpose : Regeneration and repair after ischemic renal injury appears to be modulated by circulating or locally produced growth factors. This study examined the changes of serum insulin like growth factor(IGF-I) and renal expression of IGF-I and II, vascular endothelial growth factor(VEGF), transforming growth $factor-{\beta}$($TGF-{\beta}$), and connective tissue growth factor(CTGF) during the active regeneration period after acute ischemic injury. Methods : Sera and kidney tissue samples(whole kidney, cortex, outer medullae and inner medullae) were obtained before and after one, three, five and seven days of 40 minutes bilateral renal pedicle clamping. Acute renal failure was assessed by measuring the concentration of serum creatinine. Serum IGF-I level was measured by radioimmunoassay. The mRNA expression in kidney was measured by RT-PCR. The distribution of IGF-I and CTGF was detected by immunohistochemistry. Resuts : Serum IGF-I concentration after one day following acute ischemic renal injury was significantly decreased compared to preischemic value. The mRNA levels of IGF-I, IGF-II, $TGF-{\beta}1$ and VEGF in whole kidney were temporally decreased on day one of ischemic injury. IGF-I and IGF-II expressions in outer medullae were significantly decreased on day one after ischemic injury. $TGF-{\beta}1$, CTGF and VEGF expressions were markedly decreased in medullae after one day of ischemic injury compared to other kidney sections. IGF-I was markedly decreased in cortical tubules on day one of uremic rat. CTGF was markedly increased on tubule within three days of ischemic injury. Conclusion : These findings suggest that IGFs, $TGF-{\beta}1$ and CTGF may involve in the pathogenesis or the recovery from acute ischemic renal injury.
