Locusta migratoria is a widespread locust species in many parts of the world and is considered an alternative source for the production of protein for value-added ingredients. We previously identified putative antimicrobial peptides derived from L. migratoria through an in silico analysis of its transcriptome. However, its anti-inflammatory effect has not been studied. In this study, we investigated the anti-inflammatory activities of the antimicrobial peptide locustacin (KTHILSFFPSFLPLFLKK-NH2) derived from L. migratoria on lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. Locustacin (50, 100, and 200 ㎍/ml) significantly reduced the production of nitric oxide (NO) in LPS-stimulated macrophages without any cytotoxicity. Locustacin also inhibited the mRNA and protein expression of pro-inflammatory mediators, such as inducible NO synthase and cyclooxygenase-2, in contrast to the presence of LPS alone. Locustacin decreased the release of LPS-induced pro-inflammatory cytokines, including interleukin (IL)-6 and IL-1β, and their gene expression in a dose-dependent manner. Furthermore, locustacin (100 and/or 200 ㎍/ml) inhibited phosphorylation levels of extracellular signal regulated kinase, p38, and c-Jun N-terminal kinase. Locustacin also suppressed the degradation of inhibitory kappa B alpha, which was considered to be an inhibitor of nuclear factor kappa B (NF-κB). Collectively, these results demonstrate that locustacin can exert anti-inflammatory effects through the inhibition of mitogen-activated protein kinase (MAPK) phosphorylation, activation of NF-κB, and downstream inflammatory mediators in LPS-stimulated macrophage cells.

Particulate matters with a diameter of < $10{\mu}m$ (PM10) exert oxidative stress and inflammatory events in various organs. The purpose of this study was to examine the molecular mechanism of reactive oxygen species (ROS) production induced by PM10 in the human epidermal keratinocytes (HEKs). When cultured HEKs were exposed to PM10, ROS production was induced and it was inhibited by apocynin, an antioxidant. The mRNA expression of NADPH oxidase (NOX) family was analyzed in order to examine their role in PM10-induced ROS production. PM10 increased the mRNA expression of NOX1, NOX2, dual oxidase (DUOX) 1 and DUOX2. HEKs expressed DUOX1 and DUOX2 at higher levels compared to other NOXs. The mRNA expression of dual oxidase maturation factors, DUOXA1 and DUOXA2, was also increased by PM10. We examined whether these calcium-dependent enzymes, DUOX1 and DUOX2, mediate the PM10-induced ROS production. A selective intracellular calcium chelator, BAPTA-AM, attenuated ROS production induced by PM10 or calcium ionophore A23187. The small intereference RNA (siRNA)-mediated down-regulation of DUOX2, but not DUOX1, attenuated the ROS production induced by PM10. PM10 increased the expression of inflammatory cytokines such as interleukin $(IL)-1{\beta}$, IL-6, IL-8 and interferon $(IFN)-{\gamma}$. SiRNA-mediated down-regulation of DUOX2 suppressed the PM10-induced expression of $IFN-{\gamma}$ but not other cytokines. This study suggests that DUOX2 plays a crucial role in ROS production and inflammatory response in PM10-exposed keratinocytes.
Clay minerals, a major component of mud volcano (MV) sediments, are expected to provide important information for characterizing mud volcano formation mechanisms, but clay minerals have rarely been studied. The purpose of this study is to investigate the characteristics of 420 MV and surrounding marine sediments. Clay minerals and grain size were analyzed for 8 box cores from 420 MV and Mackenzie Trough. The relative proportions of the four major clay minerals in the Mackenzie Trough are almost constant in the order of illite, chlorite, kaolinite, and smectite, regardless of the distance from the Mackenzie River. However, the grain size tends to become fining as they move away from the Mackenzie River. Comparing the clay mineral characteristics of river (Colville River, Kuparuk River, Sagavanirktok River, Canning River, Mackenzie River) sediments entering the Beaufort Sea in order to determine the origin of the Mackenzie Trough and 420 MV sediments, the sediments of the Mackenzie Trough are characterized mainly by the Mackenzie River with a low ratio of smectite/illite and a high ratio of kaolinite/chlorite. In 420 MV sediments, the contents of clay minerals decrease in the order of illite, kaolinite, chlorite, and smectite, and the grain size with depth is almost constant. The content of smectite and coarse sediments is about two times higher than the reference core. No river with higher kaolinite content than chlorite exists in the Beaufort Sea, and the ratio of smectite/illite to kaolinite/chlorite is different from the reference core such as the ratio of the Mackenzie River. Compared to the reference core, the high contents of coarse sediments and the constant grain size with depth might be attributed to the ejection by MV. The reference core is interpreted as originating from Mackenzie River, and sediment of 420 MV is interpreted as originating from eruption of MV.
Muscle dysfunction may arise from skeletal muscle atrophy caused by aging, injury, oxidative stress, and hereditary disease. Powdered heat-killed Enterococcus faecalis (EF-2001) has anti-allergy, anti-inflammatory, and anti-tumor effects. However, its antioxidant and anti-atrophy effects are poorly characterized. In this study, we examined the effects of EF-2001 on muscle atrophy. To determine the protective effect of EF-2001 on oxidative stress, C2C12 myoblasts were treated with $H_2O_2$ to induce oxidative stress. This induced cell damage, which was reduced by treatment with EF-2001. The mechanism of EF-2001's effect was examined in response to oxidative stress. Treatment with EF-2001 reversed the expression of HSP70 and SOD1 proteins. Also, mRNA levels of Atrogin-1/MAFbx and MuRF1 increased under oxidative stress conditions but decreased following EF-2001 treatment. To evaluate muscle volume, two and three dimensional models of the muscles were analyzed using micro-CT. As expected, muscle volume decreased after sciatic denervation and recovered after oral administration of EF-2001. Therefore, EF-2001 is a candidate for the treatment of muscular atrophy, and future discovery of the additional effects of EF-2001 may yield further applications as a functional food with useful activities in various fields.
This study reviewed the change of theory of ecological niche(concepts and definitions) over time to provide a theoretical basis for habitat-related studies of animals and plants. Accordingly, it analyzed and summarized the major discussion trends of ecological niche worldwide in each period from the 1900s to the present. Countries advanced in ecological studies, such as the EU and the USA, have conducted theoretical and empirical studies on the ecological niche since the early 1990s. The concept of the ecological niche was introduced in the early 1900s, developed in the mid-1900s, and advanced from the mid-1900s to the late 1900s. Since the 2000s, the advanced concept has diversified with new developments in technologies and research methods. The factors suggested by theoretical and empirical studies in defining the ecological niche of a species include 1) population dynamics of the target species, 2) all biotic conditions to sustain a population (food relationship and material flow in the food chain), 3) all non-biotic conditions to sustain a population (physical environmental conditions), 4) all direct and indirect interactions between these environmental factors, and 5) response and adaptation mechanisms that include the migratory ability of the target species or genetic diversity and adaptability to change. Unlike such international advancement, there have not been sufficient theoretical, philosophical, and empirical studies of ecological niche in Korea. The concepts and definitions by Greennell, Elton, and Hutchinson were selectively and partially borrowed for empirical studies without full description. Considering that the theory of ecological niche becomes the foundation for habitat-based species conservation and restoration, it is necessary to seek diversification and advancement of theoretical and empirical research and research methods and technological development. It will provide an important foundation for the academic advancement of ecology and for establishing and implementing policies to preserve and restore ecology and biodiversity effectively and successfully in Korea.

The presence of the hydrate-bearing sediments in Ulleung Basin of South Korea has been confirmed from previous studies. Researches on gas production methods from the hydrate-bearing sediments have been conducted worldwide. As production mechanism is a complex phenomenon in which thermal, hydraulic, and mechanical phenomena occur simultaneously, it is difficult to accurately conduct the productivity and stability analysis of hydrate bearing sediments through lab-scale experiments. Thus, the importance of numerical analysis in evaluating gas productivity and stability of hydrate-bearing sediments has been emphasized. In this study, the numerical parametric analysis was conducted to investigate the effects of the bottom hole pressure and the depressurization rate on the gas productivity and stability of hydrate-bearing sediments during the depressurization method. The numerical analysis results confirmed that as the bottom hole pressure decreases, the productivity increases and the stability of sediments deteriorates. Meanwhile, it was shown that the depressurization rate did not largely affect the productivity and stability of the hydrate-bearing sediments. In addition, sensitivity analysis for gas productivity and stability of the sediments were conducted according to the depressurization rate in order to establish a production strategy that prevents sand production during gas production. As a result of the analysis, it was confirmed that controlling the depressurization rate from a low value to a high value is effective in securing the stability. Moreover, during gas production, the subsidence of sediments occurred near the production well, and ground heave occurred at the bottom of the production well due to the pressure gradient. From these results, it was concluded that both the productivity and stability analyses should be conducted in order to determine the bottom hole pressure when producing gas using the depressurization method. Additionally, the stress analysis of the production well, which is induced by the vertical displacements of sediments, should be evaluated.

An Alum-sludge based adsorbent (ASBA) was synthesized by the hydrothermal treatment of alum sludge obtained from settling basin in water treatment plant. ASBA was applied to remove fluoride and arsenic in artificially-contaminated aqueous solutions and mine drainage. The mineralogical crystal structure, composition, and specific surface area of ASBA were identified. The result revealed that ASBA has irregular pores and a specific surface area of 87.25 ㎡ g-1 on its surface, which is advantageous for quick and facile adsorption. The main mineral components of the adsorbent were found to be quartz(SiO2), montmorillonite((Al,Mg)2Si4O10(OH)2·4H2O) and albite(NaAlSi3O8). The effects of pH, reaction time, initial concentration, and temperature on removal of fluoride and arsenic were examined. The results of the experiments showed that, the adsorbed amount of fluoride and arsenic gradually decreased with increasing pH. Based on the results of kinetic and isotherm experiments, the maximum adsorption capacity of fluoride and arsenic were 7.6 and 5.6 mg g-1, respectively. Developed models of fluoride and arsenic were suitable for the Langmuir and Freundlich models. Moreover, As for fluoride and arsenic, the increase rate of adsorption concentration decreased after 8 and 12 hr, respectively, after the start of the reaction. Also, the thermodynamic data showed that the amount of fluoride and arsenic adsorbed onto ASBA increased with increasing temperature from 25℃ to 35℃, indicating that the adsorption was endothermic and non-spontaneous reaction. As a result of regeneration experiments, ASBA can be regenerated by 1N of NaOH. In the actual mine drainage experiment, it was found that it has relatively high removal rates of 77% and 69%. The experimental results show ASBA is effective as an adsorbent for removal fluoride and arsenic from mine drainage, which has a small flow rate and acid/neutral pH environment.
Natural soil was artificially contaminated with heavy metals (Cd, Pb, and Zn), and the movement of earthworm was characterized in real time using the ViSSET system composed of vibration sensor and the other components. The manifestation mechanism of ecological toxicity of heavy metals was interpreted based on the accumulative frequency of earthworm movement obtained from the real-time monitoring as well as the conventional indices of earthworm behavior, such as the change in body weight before and after tests and biocumulative concentrations of each contaminant. The results showed the difference in the earthworm movement according to the species of heavy metal contaminants. In the case of Cd, the earthworm movement was decreased with increasing its concentration and then tended to be increased. The activity of earthworm was severely increased with increasing Pb concentration, but the movement of earthworm was gradually decreased with increasing Zn concentration. The body weight of earthworm was proved to be greatly decreased in the Zn-contaminated soil, but it was similarly decreased in Cd- and Pb-contaminated soils. The bioaccumulation factor (BAF) was higher in the sequence of Cd > Zn > Pb, and particularly the biocumulative concentration of Pb did not show a clear tendency according to the Pb concentrations in soil. It was speculated that Cd is accumulated as a metallothionein-bound form in the interior of earthworm for a long time. In particular, Cd has a bad influence on the earthworm through the critical effect at its higher concentrations. Pb was likely to reveal its ecotoxicity via skin irritation or injury of sensory organs rather than ingestion pathway. The ecotoxicity of Zn seemed to be manifested by damaging the cell membranes of digestive organs or inordinately activating metabolism. Based on the results of real-time monitoring of earthworm movement, the half maximal effective concentration (EC50) of Pb was estimated to be 751.2 mg/kg, and it was similar to previously-reported ones. The study confirmed that if the conventional indices of earthworm behavior are combined with the results of newly-proposed method, the mechanism of toxicity manifestation of heavy metal contaminants in soils is more clearly interpreted.
As a cultivar of Korean wheat, 'Keumgang' wheat variety has a fast growth period and can be grown stably. Hexaploid wheat (Triticum aestivum) has moderately high salt tolerance compared to tetraploid wheat (Triticum turgidum L.). However, the molecular mechanisms related to salt tolerance of hexaploid wheat have not been elucidated yet. In this study, the candidate genes related to salt tolerance were identified by investigating the genes that are differently expressed in Keumgang variety and examining salt tolerant mutation '2020-s1340.'. A total of 85,771,537 reads were obtained after quality filtering using NextSeq 500 Illumina sequencing technology. A total of 23,634,438 reads were aligned with the NCBI Campala Lr22a pseudomolecule v5 reference genome (Triticum aestivum). A total of 282 differentially expressed genes (DEGs) were identified in the two Triticum aestivum materials. These DEGs have functions, including salt tolerance related traits such as 'wall-associated receptor kinase-like 8', 'cytochrome P450', '6-phosphofructokinase 2'. In addition, the identified DEGs were classified into three categories, including biological process, molecular function, cellular component using gene ontology analysis. These DEGs were enriched significantly for terms such as the 'copper ion transport', 'oxidation-reduction process', 'alternative oxidase activity'. These results, which were obtained using RNA-seq analysis, will improve our understanding of salt tolerance of wheat. Moreover, this study will be a useful resource for breeding wheat varieties with improved salt tolerance using molecular breeding technology.
P. tenebrifer (PT) belongs to the Diptera order and Stratiomyidae family. Recently, insect industry have been focused as food, animal feed and environmental advantages. γ-aminobutyric acid (GABA) and melatonin have been associated with regulating sleep and depression. GABA is the primary inhibitory neurotransmitter and is synthesized via biotransformation of monosodium glutamate (MSG) to GABA by lactic acid bacteria. In this study, we first used a GABA-enhanced PT extract, wherein GABA was enhanced by feeding MSG to PT. The underlying mechanisms preventing stress and insomnia were investigated in a corticosterone (CORT)-induced endoplasmic reticulum (ER) stress and chronic restraint stress (CRS)-exposed mouse model, as well as in pentobarbital (45 mg/kg)-induced sleep behaviors in mice. In the present study, the GABA peak was detected in high-performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) analysis and showed in Ptecticus tenebrifer water extract (PTW) but not in non-PTW extract. The results showed that PTW and Ptecticus tenebrifer with 70% ethanol extract (PTE) exerted neuroprotective effects by protecting against CORT-induced downregulation of phosphorylated extracellular signal-regulated kinase 1/2 (ERK1/2) and cAMP-response element binding protein (CREB) expression. In addition, PTW (300 mg/kg) significantly reduced CORT levels in CRS-exposed mice. Furthermore, PTW (100 and 300 mg/kg) significantly reduced sleep latency and increased total sleep duration in pentobarbital (45 mg/kg)-induced sleeping behaviors, which was related to serum melatonin levels. In conclusion, our results suggest that PTW exerts anti-stress and sleep-enhancing effects by regulating serum CORT and melatonin levels.
