Antioxidative ability of ethanol extract from the leaves of Leucaena leucocephala (Lam.) de Wit (백연수(Leucaena leucocephala (Lam.) de Wit) 잎 에탄올 추출물의 항산화능)

  • Antioxidative ability of 50% ethanol extract from leaves of Leucaena leucocephala (Lam.) de Wit was investigated. The extraction yield, total polyphenols and total flavonoids of the ethanol extract were $18.3{\pm}0.40%$, $121.3{\pm}11.04{\mu}g$ gallic acid equivalents/mg and $105.2{\pm}2.77{\mu}g$ quercetin equivalents/mg, respectively. The ethanol extract possessed the antioxidative activities such as free radical, cation radical, and nitrite scavenging activities as well as reducing power and lipid peroxidation inhibitory activity. These antioxidative activities of the ethanol extract increased in proportion to the extract concentration. $EC_{50}$ values for free radical scavenging, cation radical scavenging nitrite scavenging, and reducing power were $133.51{\mu}g/mL$, $667.22{\mu}g/mL$, 2.58 mg/mL, and $250.00{\mu}g/mL$, respectively. Polyphenols and flavonoids are suggested to be the key contributors for the antioxidative ability of the ethanol extract from Leucaena leucocephala leaves.

Antioxidant Effect and Tyrosinase Inhibition Activity of Seaweeds Ethanol Extracts (해조류 에탄올 추출물의 항산화 및 Tyrosinase 억제 활성)

  • Seaweeds, laver, sea mustard, kelp, and fusiformis, were prepared and investigated for its antioxidant and tyrosinase inhibition activities. The extracts yield, color, total phenolic contents, antioxidative activity, and tyrosinase inhibition activity of the extract samples were measured. Hunter Lightness values of laver, sea mustard, kelp, and fusiformis extracts were 82.88, 78.53, 83.04, and 78.11, respectively. The contents of total phenolic compounds of the seaweed extracts powder, laver, sea mustard, kelp, and fusiformis were 43.23, 11.59, 10.09, and 46.59 mg/g of sample, respectively. 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the fusiformis extract was shown to be the highest value compared with other seaweed extracts. 2,2'-azino-bis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) radical scavenging activities of laver, sea mustard, kelp, and fusiformis extracts were 258.00, 219.26, 95.77, and $1186.62{\mu}mol$ trolox equivalence per gram, respectively, at the 1,000 ppm level. TBARS value of oil emulsion, samples without extracts was higher than those of the samples prepared with laver and sea mustard extracts. The inhibition rates (%) of the mushroom tyrosinase of laver, sea mustard, kelp, and fusiformis extracts powder were 25.93, 26.32, 24.76 and 20.24% at 1,000 ppm, respectively. The results indicated that laver, sea mustard, kelp and fusiformis extracts possess biological activities such as antioxidant activity and tyrosinase inhibition effect.

Antioxidant Activities of Different Parts of Sparassis crispa Depending on Extraction Temperature (추출 온도에 따른 꽃송이버섯(Sparassis crispa)의 부위별 항산화 활성)

  • This study was carried out to investigate antioxidant compounds (total polyphenol and total flavonoid) and antioxidant activities (DPPH radical scavenging activity, ABTS radical scavenging activity, FRAP activity, and reducing power] of the mycelium and fruit body of Sparassis crispa depending on extraction temperature ($60^{\circ}C$ and $95^{\circ}C$). For total polyphenols, total flavonoids, FRAP activity, and reducing power, the mycelium of S. crispa extracted at $95^{\circ}C$ showed the highest contents and activities. The mycelium of S. crispa extracted at $60^{\circ}C$ and fruit body of S. crispa extracted at $95^{\circ}C$ showed the highest DPPH radical scavenging activity and ABTS radical scavenging activity, respectively. This study suggests that the antioxidant activities of S. crispa extracted at $95^{\circ}C$ are better than those of S. crispa extracted at $60^{\circ}C$, and the mycelium contained more antioxidant compounds than the fruit body.

In Vitro antioxidant effect of ethanol extract from Pennisetum purpureum (Napier grass (Penninsetum purpureum) 에탄올 추출물의 in vitro 항산화 효과)

  • In vitro antioxidant effect of 50% ethanol extract from Napier grass (Penninsetum purpureum) was investigated. The yield and polyphenol content of the Napier grass extract were $6.3{\pm}0.35%$ and $79.6{\pm}3.65{\mu}g$ gallic acid equivalents/mg-extract, respectively. Antioxidant ability of the Napier grass extract such as fee radical and cation radical scavenging activities, reducing power, nitrite scavenging activity, and lipid peroxidation inhibitory activity proportionally increased as concentration of the extract increased. $EC_{50}$ values of the Napier grass extract for free radical scavenging, cation radical scavenging, reducing power, and nitrite scavenging were 1,930.0, 350.0, 840.0, and $1,470.0{\mu}g/mL$, respectively. In the presence of $85.0{\mu}g/mL$ of the Napier grass extract, lipid peroxidation was potently inhibited by 74.6%.

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined in vitro their hyaluronidase inhibitory effect and radical scavenging activity. Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix and Moutan Radicis Coritex showed more than 50% hyaluronidase inhibitory effects. Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed more than 90% in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical showed significantly activity.

  • Two antioxidative compounds in Chrysanthemum boreale were isolated by a bioassay using a 2, 2-diphenyl-1-picrydrazyl(DPPH) free radical. They were identified as apigenin, linarin(acacetin-7-O-rutinoside) on the basis of $^1H\;and\;^{13}C-NMR$ and MS data. The DPPH radical scavenging activity$(RC_{50}:\;13.3{\mu}g)$ of apigenin was similar to that of ${\alpha}-tocopherol\;(12.1{\mu}g)$ and L-ascorbic acid$(13.1{\mu}g)$

Antioxidant Activity of Different Parts of Lespedeza bicolor and Isolation of Antioxidant Compound (싸리나무(Lespedeza bicolor) 부위별 추출물의 항산화 활성 및 항산화물질 분리)

  • In this study, total antioxidant properties of extracts from different parts of Lespedeza bicolor were determined using techniques of measuring 1,1-diphenyl-2-picryl hydrazyl/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)-radical scavenging activity and total phenolic contents. The total antioxidant activities of leaf, stem and root extracts from various solvents (water, 50, 70, 100% ethanol, and hot-water) indicated that 50 and 70% ethanol extracts have high radical scavenging activities and phenolic contents. A systematic approach was used to determine the total antioxidant activity of different solvent fractions of the Lespedeza bicolor extracts, partitioning with chloroform, ethyl acetate, n-butanol, and water, and the ethyl acetate fraction was found to have the strongest antioxidant activity. Antioxidant assay-guided isolation was carried out to isolate potential antioxidant compounds. The ethyl acetate fraction of the leaf extract was subjected to silica gel, LH-20 and RP-18 column chromatography successively, and afforded compound 1, which was identified as eriodictyol by NMR and MS analysis, after which its antioxidant activity was determined.

Antioxidant and pancreatic lipase inhibitory activities of Anemarrhena asphodeloides (지모 추출물의 항산화 및 pancreatic lipase 저해 활성 평가)

  • In this study, the antioxidant and pancreatic lipase inhibitory activities of aqueous methanolic (70% methanol) extract from the roots of Anemarrhena asphodeloides were investigated. The extracts of four solvent fractions (the n-hexane layer, EtOAc layer, n-BuOH layer, and $H_2O$ layer) of the 70% methanol extract were also investigated. Furthermore, the total phenolic content was quantified using a spectrophotometric method. All the tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. In particular, the pancreatic lipase inhibitory activity of the ethyl acetate soluble portion (the EtOAc layer) from the rhizomes of the A. asphodeloides was higher than that of the other solvent-soluble portions. The antioxidant property of the extracts was evaluated using radical scavenging assays with DPPH and $ABTS^+$ radicals. 1000 mg/ml of the n-BuOH layer extract showed 91.2% DPPH radical scavenging activity. The EtOAc layer extract and the n-BuOH layer extract showed $IC_{50}=20.5{\pm}1.7mg/ml$ and $IC_{50}=50.5{\pm}0.7mg/ml$ $ABTS^+$ radical scavenging activities, respectively. The anti-obesity efficacy of the A. asphodeloides extract was tested via porcine pancreatic lipase assay. A pancreatic lipase inhibitory activity ($IC_{50}$) of $31.3{\pm}0.1mg/ml$ was obtained from the EtOAc layer extract. These results suggest that A. asphodeloides can be considered a new potential source of natural antioxidant and anti-obesity agents.

Screening for DPPH Free Radical Scavenging Activities of Autogenous Seaweeds in Jeju Island Using a Electron Spin Resonance (ESR) Spectroscopy (Electron Spin Resonance을 이용한 제주 자생 해조류의 DPPH Free Radical 소거활성 검색)

  • Extracts which were prepared by four different extractions - 80% methanol extracts (ME) at high ($70^{\circ}C$) and a room temperature ($20^{\circ}C$), respectively and aqueous extracts (AE) at both temperatures with the residue after the methanol extracts - of 10 green, 19 brown and 25 red seaweeds collected in Jeju Island coast were examined for their DPPH free radical scavenging activity using a ESR (electron spin resonance) spectroscopy. A variety of the extracts showed positive scavenging effect against DPPH free radical (except the green seaweeds). Among the extracts, the brown seaweed extracts exhibited the highest scavenging activity. Especially, Sargassum spp. of the brown seaweeds have remarkable scavenging activities - both methanolic and aqueous at the both temperatures ($20^{\circ}C$ and $70^{\circ}C$). On the other hand, ME showed better scavenging activity than AE in the red seaweed extracts. These results indicate that autogenous seaweeds in Jeju will be potential natural antioxidants for functional food compounds.

  • Antioxidant capacity and acetylcholinesterase (AChE) inhibitory activity of the aqueous methanolic extract of Rhus javanica were investigated in vitro. The antioxidant properties was measured by radical scavenging assays using DPPH and $ABTS^+$ radicals. The AChE inhibitory efficacy of R. javanica was tested by Ellman's assay and the total phenolic content was determined using a spectrophotometric method. All tested samples showed a dose-dependent AChE inhibitory and radical scavenging activities. In particularly, ethyl acetate (EtOAc)-soluble portion from the methanolic extract of R. javanica showed significantly higher inhibitory activity than other organic solvent soluble-portions in an AChE and radical scavenging assay systems. These results suggest that R. javanica may be possess potential benefits which might be useful in development of antioxidant and anti-alzheimer's disease ingredient.
