Association between Economic Traits and Candidate Gene Polymorphism in Korean Native Pig and Duroc (한국 재래 돼지와 듀록의 경제형질과 후보 유전자 다형성간의 연관성 분석)

  • MC4R, PRKAG3, FABP3, and ESR have reported as important candidate genes related to some economic traits in pigs. To investigate the association between these genes and economic traits, the analysis of restriction fragment length polymorphism (RFLP) was conducted on 147 individuals (96 Durocs and 86 Korea native pigs; KNP) using single nucleotide polymorphism (SNP). Different genotype frequencies of 4 candidate genes were observed in Duroc and KNP. There were significant associations between MC4R polymorphic site and average daily gain (ADG, p<0.05) and backfat thickness (BF, p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in KNP. PRXAG3 polymorphic site were significantly .elated to BF (p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in the KNP. In FABP3, association with BF (p<0.05) in the Duroc, ADG (p<0.05) and days to 70 kg (p<0.05) in the KNP were found. ESR polymorphic site was not significantly associated to any other traits.

Effects of Breeds on Body Weight, Age, Body Conformation, Backfat Thickness and Steroid Hormone Concentrations at First Estrus and Mating of Gilts (첫 발정과 종부시 체중, 연령, 체형, 등지방 두께 및 스테로이드 호르몬 농도에 미치는 종빈돈 품종의 영향)

  • This study was carried out to get a fundamental information fur improvement of reproductive performance in gilt. We investigated the effects of breeds on body weight, age, body length, wither's height, girth and backfat thickness, and the serum concentrations of estradiol-17$\beta$, cortisol and progesterone at first estrus and mating of gilts. A total of 47 gilts of Duroc, Landrace and Yorkshire, produced at Livestock Experiment Station, Chungnam Livestock Sanitation Research Institute from 2000 through 2002, were used for this experiment. Body weight, age and girth of Duroc at first estrus and mating were higher than those of Landrace and Yorkshire. There were no differences on body length among the three breeds at first estrus and mating. Wither's height of Duroc and Yorkshire at first estrus and mating was higher than that of Landrace. Backfat thickness of Yorkshire was thinnest among the three breeds at first estrus, but there were no differences on backfat thickness among the three breeds at first mating. Serum estradiol-17$\beta$ concentration of Landrace (45.0 pg/ml) at first estrus was higher than that of Yorkshire (27.4 pg/ml) and Duroc (21.8 pg/ml), but there were no differences on estradiol-17$\beta$ concentration (from 18.5 to 31.9 pg/ml) among the three breeds at first mating. Serum cortisol concentration of Duroc at first estrus and mating was higher than that of Landrace and Yorkshire. There were no differences on serum progesterone concentration among the three breeds at first estrus and mating of gilt.

Genetic Parameter Estimates for Productive Traits in Duroc Pigs (듀록종의 산육형질에 대한 유전모수 추정)

  • The purpose of this study was to estimate genetic parameters for productive traits in Duroc breed. In this study, 40,657 records for productive traits and the pedigree data of 47,974 families were collected from 41 farms registered at the Korean Animal Improvement Association (KAIA) from 2004 to 2011. The REMLf90 program was used to analyze a multiple traits animal model with fixed effects of sex, contemporary group, parity and age at the end of the test as covariate and random effects of animal and residual error. The heritabilities of days to 90 kg (D90KG), average daily gain (ADG), backfat thickness (BF) and eye muscle areas (EMA) were estimated to be 0.334, 0.340, 0.335, and 0.200, respectively. The genetic correlation coefficients were -0.992 between D90KG and ADG, -0.142 between ADG and BF, -0.361 between ADG and EMA, and -0.243 between BF and EMA. Conversely, positive genetic correlations for D90KG with BF and EMA were 0.13 and 0.36, respectively.

Effect of Spring and Summer Influencing Semen Characteristics, Frozen-Thawed Sperm Viability and Testosterone Concentration in Duroc Boars (듀록 종모돈의 정액성상, 동결 .융해 후 정자의 생존성 및 테스토스테론의 농도에 미치는 봄과 여름의 영향)

  • This study was carried out to investigate the effects of spring (March~May) and summer (June~August) influencing semen characteristics, frozen-thawed sperm viability and serum testosterone concentration in Duroc boars. Results of this study were as follows: 1. There were no significant differences in the semen volume, pH and sperm concentration of Duroc boars between spring and summer. 2. Sperm motility and normal acrosome of raw semen in Duroc boars did not differ significantly between spring and summer. However, motility and normal acrosome of frozen-thawed sperm were higher in spring season than in summer season(P<0.05). 3. Serum testosterone concentrations in Duroc boars were 2.15 ng/$m\ell$ in spring and 0.65 ng/$m\ell$ in summer. Serum testosterone concentrations in spring were higher thin those in summer (P<0.05). 4. In conclusion, when serum testosterone concentrations in Duroc boars were higher, frozen-thawed sperm viability were higher.

  • This study was carried out to investigate the effects of semen characteristics, frozen-thawed sperm viability and serum FSH, LH, estradiol-17β and testosterone concentrations between breeds and among seasons in boars. In all seasons, Yorkshire boars produced higher semen volume compared with Duroc boars, whereas sperm concentration did not differ significantly between Duroc and Yorkshire boars. Semen volume in spring was higher compared with summer, autumn and winter in both Duroc and Yorkshire boars, but sperm concentration did not differ significantly among seasons. Sperm motility and normal acrosome rate of frozen-thawed sperm produced in spring were higher than those in summer, autumn and winter in both Duroc and Yorkshire boars. Sperm motility of frozen-thawed sperm in Yorkshire boars was higher than that in Duroc boars regardless of seasons. However, normal acrosome rate did not differ significantly between Duroc and Yorkshire boars. Serum FSH concentration in Yorkshire boars was lower than that in Duroc boars in all seasons. However, there were no significant differences on serum FSH concentration of Duroc and Yorkshire boars among seasons. Serum LH and estradiol-17β concentrations did not differ significantly between Duroc and Yorkshire boars. Also, there were no significant differences in serum LH and estradiol-17β concentrations of Duroc and Yorkshire boars among seasons. Serum testosterone concentration in Yorkshire boars was higher than that in Duroc boars in all seasons. In both breeds, serum testosterone concentrations were higher in spring than in summer, autumn and winter. In conclusion, when serum FSH concentrations were low, semen volumes were high, and when serum testosterone concentrations were high, sperm motility and normal acrosome rate of frozen-thawed sperm were high.

Characteristics of Carcass and Meat Yields of Fattening Pigs by Production Step (비육돈 생산단계에 따른 도체 및 부분육 생산 특성)

  • The characteristics of carcass and meat yields of fattening pigs by production steps were investigated with Landrace (LL, n=41), Yorkshire (YY, n=33), and Duroc (DD, n=30), $F_1$ (LY, n=25), the Crossbred of LYD (n=48). Duroc had more weight loss in carcasses weight than that of the other breed(p<0.05). Yorkshire and $F_1$ had higher production weight in retail cut than that of the other pure or crossbred. Carcass from Landrace and $F_1$ were significantly longer in length than the other breed(p<0.05). $F_1$ produced wider carcass than the other breed. Carcass thickness at aitch bone was higher for $F_1$ and Crossbred than the other breed(p<0.05). Landrace, Yorkshire and $F_1$ produced more loin and tenderloin in weight than the other breed (p<0.05). Yorkshire and $F_1$ produced more picnic shoulder when compared to the other breed. The hind legs produced from Yorkshire and $F_1$ were higher in weight and the fore legs produced from the Crossbred were higher in weight. Duroc produced the lowest weight of belly among the breeds. The acceptance level of loin were extremely low for all breed. Landrace had the highest acceptance level for tenderloin. Yorkshire had the highest acceptance level for picnic shoulder and ham when evaluated by export standard of Japan. In conclusion, The introduction of pure breed and establishment of mating steps are necessary to produce highly accepted pork with high acceptance in carcass and meat yields.

Studies on the MC1R Gene Frequencies in Landrace, Large White, Duroc and Jeju Native Black Pigs (랜드레이스, 대요크셔, 듀록 및 제주 흑돈의 Melanocortin 1 Receptor(MC1R) 유전자의 유전자형 분석)

  • This study was conducted to investigate the genotypes and frequencies of Melanocortin 1 Receptor(MC1R) genes in pigs which plays a central role in regulation of eumelanin (black/brown) and phaeomelanin(red/yellow) pigment synthesis within the mammalian melanocytes. Four different breeds of pigs(20 Landrace, 20 Yorkshire, 20 Duroc, and 93 Jeju native black pigs) were used and PCR-RFLP analysis of MC1R gene was also carried out. Two regions of MC1R genes (428bp and 405bp) were amplified using two specific primers (MERL1-EPIG2, EPIG1-EPIG3), respectively and MC1R allele were determined using 2 restriction enzymes (BspHⅠ, AccⅡ). The results of this experiment indicated that MC1R allelic type in Landrace, Large Yorkshire and Duroc were MC1R *2 (Ep), MC1R *2 (Ep), MC1R *4 (e), respectively. However, various allelic types of MC1R genes were detected in Jeju native black pigs. MC1R allelic type of Jeju black pigs was MC1R*2 type as in Meishan and Large black breeds or MC1R*3 type as in Hampshire and Berkshire breeds and the gene frequencies of ED1 and ED2 were 0.554 and 0.446 in average.

Association of Melanocortin 4 Receptor (MC4R) Genotypes with Fatty Acid Compositions in an Intercross Population between Duroc and Jeju Native Pigs (제주재래돼지와 듀록 참조축군에서 Melanocortin 4 Receptor (MC4R) 유전자형과 지방산 조성간의 관련성 분석)

  • This study was conducted to examine association between melanocortin-4-receptor (MC4R) genotypes and fatty acid (FA) composition in an F2 intercross between Duroc and Jeju (South Korea) Native pigs (JNP). Fourteen FA composition traits were measured in more than 290 F2 progeny population produced between Duroc and JNP. All experimental pigs were successfully genotyped for the MC4R c.1426A>G (p.Asp298Asn) single nucleotide polymorphism (SNP) by using Taq I PCR-RFLP methods. We detected three MC4R genotypes, AA, AG, and GG with 0.299, 0.542, and 0.159 genotype frequencies, respectively. The MC4R AA genotype animals showed higher levels in palmitic acid (C16:0, p<0.05), stearic acid (C18:0, p<0.01), eicosenoic acid (C20:1n9, p<0.05), saturated fatty acid (SFA, p<0.01) than GG homozygotes, respectively. Whereas MC4R GG genotype showed higher values in linoleic acid (C18:2n6, p<0.001), linolenic acid (C18:3n3, p<0.001), linolenic acid (C18:3n6, p<0.001), arachidonic acid (C20:4n6, p<0.001) and unsaturated fatty acid (USFA, p<0.01) than AA pigs, respectively. The MC4R GG genotype was associated with increasing USFA and decreasing SFA in the F2 progeny population produced between Duroc and JNP. Our finding suggests that the MC4R polymorphisms can be used as a valuable genetic marker for Duroc and JNP breeding programs to improve meat quality and to control FA compositions.
