Study of optimal pathogenicity condition on cordyceps (눈꽃동충하초 및 번데기동충하초균의 적정 병원력 유지조건 구명)

  • This study was carried out to investigate pathogenicity of liquid spawns on dongchunghacho growing using silkworm, optimum infective condition for mass production and infection rate on various conservation periods and temperatures. We compared with the infection rates after conservation the liquid spawns (Paecilomyces tenuipes) at 4$^{\circ}C$ and 25$^{\circ}C$ for 10 days and 20 days. In these results, infection rate of conservated liquid spawns at 4$^{\circ}C$ was 88.0 percent in the tenth day, and then again it was 5.3 percent at 25$^{\circ}C$. Infection rates at 4$^{\circ}C$ were on the whole excellent out of long-term preservation methods.

  • 최근 들어서 대기오염의 변화, 미세먼지, 황사, 각종 중금속 오염, 방사능 등의 대기오염으로 인하여 인간의 질병에 대한 면역기능이 떨어지고 있다. 또한 바이러스 침입에 의한 면역기능의 약화는 심각한 문제로 대두되고 있다. 따라서 항염, 항균, 항암, 항바이러스에 대한 관심이 매우 고조되고 있다. 이러한 문제들을 해결하고 인체의 면역기능을 활성화시키기 위해서는 정신수양과 먹거리의 환경변화가 절실히 요구되어지고 있다. 면역기능을 활성화 시키는데 가장 좋은 식품으로는 야생에서 자생하는 동충하초로 알려져 있다. 이러한 동총하초는 가격이 매우 비싸서 일반적으로 식품으로 활용하기에는 많은 어려움이 따르고 있다. 중국의 해발 3000m 이상에서 야생하는 동충하초를 실험한 결과는 다음과 같다. 동충하초를 메탄올과 에탄올에서 농축하여 실험을 하였던 결과 에탄올 처리구보다는 메탄올 처리구에서 훨씬 더 양호한 결과를 나타내었다. 이러한 현상은 총 폴리페놀 함량, 항산화, 항균, 항암활성에서 같은 반응을 보였다. 동충하초의 항균활성은 일반적으로 실험에 사용된 5종(Bacillus subtilis, Candida albicans, Escherichia coli, Corynebacterium ammoniagenes, Pseudomonas aeruginosa)의 균에서 대부분 억제활성을 나타내었다.

The Cultural Characteristics and Antibacterial Activities of Cordyceps militaris and Paecilomyces tenuipes (번데기동충하초와 눈꽃동충하초의 배양특성 및 항균활성)

  • The cultural characteristics and antibacterial activities of Cordyceps militaris and Paecilomyces tenuipes were compared. The mycelial growth was the highest on MCM (Mushroom Complete Medium) for C. militaris and on YMA (Yeast Malt Agar) for P. tenuipes. But the mycelial density on MMM (Mushroom Minimal Medium) was lower than other on media. The optimum mycelial growth was observed at $25^{\circ}C$. C. militaris was low mycelial growth when it was transferred over 5 times generation. The carbon source for the optimum mycelial growth was fructose of monosaccharide, maltose of disaccharide and dextrin of polysaccharide. The calcium nitrate of organonitrogen was found the best mycelial growth on C. militaris, while the sodium nitrate observed to be well for mycelial growth on P. tenuipes. The ammonium tartrate was observed to be the best among the inorganonitrogen used for mycelial growth. Antibacterial activities were found out just C. militaris against Bacillus cereus of Gram (+).

Inhibitory effect of mushrooms extract on TNF-α/INF-γ induced-cytokine in human keratinocytes, HaCaT (버섯류 추출물의 피부각질세포(HaCaTcell) 내 염증성 사이토카인 억제효과)

  • Mushroom is known for anti-inflammatory and anti-oxidative potential. This study provides evidence that the inhibitory effect of mushroom on the expression of pro-inflammatory cytokines in human keratinocytes, HaCaT cells. To define the underlying mechanisms of action, tumor necrosis factor${\alpha}$/$IFN{\gamma}$-activated human keratinocytes model was used. Mushroom significantly inhibited the expression of cytokines in HaCaT cells. Taken together, the results demonstrate that mushroom inhibited inflammtion, suggesting that mushroom (DW extract: Grifola frondosa Cordyceps militaris), (Ethanol extract: Ganoderma lucidum, Lentinus edodes, Cordyceps militaris, Flammulina velutipes) might be a candidate for the treatment of skin inflammation.

  • The radioprotective effects of white ginseng and Entomopathogenic Fugi Extract on liver damage induced by X-ray were investigated. To one group of ICR male mice were given white ginseng(150 mg/kg/day for 7days, orally) and Entomopathogenic Fugi (200 mg/kg/day for 7days, orally) before X-ray irradiation. To another group were irradiated by 5 Gy(1.01 Gy/min) dose of X-ray. Contrast group were given with saline(0.1 ml). The levels of reduced(GSH) and oxidized(GSSG) glutathione in liver tissue were measured. The ratio of GSSG/total GSH was significantly decreased in the white ginseng and Entomopathogenic Fugi (200 mg/kg/day)(150 mg/kg/day) groups than irradiation group.

  • This experiment was carried out to find the method for mass production by artifical cultivition and to analyze the components of Paecilomyces japonica according to several media. Time of inoculation of the Paecilomyces japonica using silkworm was on first day of five molting and infection rate was 72.0%. Optium medium for mass production of the Paecilomyces japonica was known effective for increasing dry weight and fruitbody at brown rice 80g plus pupa powder 20g. Dry weight of Paecilomyces japonica using fungus of silkworm was 1.2g including pupa and length of fruitbody was appeared 3.0cm to 3.5cm. Content of $\beta$ - glucan was very high as 40.5% at inoculation on the first day of the five molting while 16.4% at brown rice, 20.7% at pupa, 23.1% at brown rice plus pupa powder, and 28.7% at pine sawdust plus wheat bran. Mycelium was poor and pinkly conidiospore was formed on media of centipede, maggot and powder of silkworm.

Optimization for the Preparation of Jeung-Pyun, with Added Paecilomyces japonica Powder, Using a Response Surface Methodology (반응표면분석법에 의한 동충하초 첨가 증편 제조의 최적화)

  • To optimize the preparation process of Jeung-Pyun, an experiment was derived, using a central composite design, to fmd the optimal mixing conditions. The addition of Paecilomyces japonica powder, the volume of Tak-Ju and the second fermentation time were independent variables, with the pH, volume, color, sensory and texture properties of the Jeung-Pyun selected as response variables. As the Paecilomyces japonica powder and fermentation time were increased, the b values and volume after the second fermentation also increased. The color of the Jeung-Pyun became deeper with increases in the Paecilomyces japonica powder and fermentation time, and was deepest on the addition of 30% Tak-Ju. The pH increased with increasing fermentation time of the Jeung-Pyun, and on the addition of 3% Paecilomyces japonica rapidly increased more than with the other groups. The optimal mixing conditions for the best quality Jeung-Pyun, with restricted conditions above a b value of 3, a color intensity above 3, and a volume of 40mQ and a pH of 4.02-4.04 after the second fermentation, were 4.0%, 20% and 45 mins for the Paecilomyces japonica powder, Tak-Ju and second fermentation time, respectively.

Quality Characteristics of Kochujang Prepared with Paecilomyces japonica from Silkworm (누에 동충하초(Paecilomyces japonica)를 첨가하여 제조한 고추장의 품질특성 변화에 관한 연구)

  • Changes in quality properties of Kochujang prepared with Paecilomyces japonica powder and extract using different solvents were investigated during 90 days of fermentation at $20^{\circ}C$. Although moisture contents were not significantly different, pH of P. japonica-added Kochujang was lower than that of control group without P. japonica, and decreased with increasing fermentation time. Amino nitrogen content increased up to 60 days of fermentation and decreased slightly after 90 days, with that of P. japonica-added Kochujang showing highest on 30 and 60 days at 179.2 and 282.2 mg%, respectively, higher than control gruup. L, a, and b values decreased in proportion to fermentation period, with P. japonica-added Kochujang, particularly P. japonica powder-added Kochujang, lower than those of control g개up. Sensory evaluation test showed color of control group was 'clear red', whereas that of P. japonica powder-added Kochujang was 'dark reddish brown' and P. japonica extract-added Kochujang was darker than control group; consumer preference for dark color was low, Textures of all samples were 'glossy and smooth', showing high consumer preference. Salt content of P. japonica-added Kochujang was higher than that of control group, with P. japonica extract-added Kochujang higher than that made with powder Hot taste or P. japonica-added Kochujang was weaker, whereas its flavor higher, than control group, with P. japonica powder-added Kochujang showing highest flavor score. Overall preference was higher for P. japonica-added Kochujang than control group, with P. japonica water extract-added Kochujang showing the highest score.

Improvement Effect of Cordycepin-Enriched Cordyceps militaris JLM 0636 Powder against Orotic Acid-Induced Fatty Liver in Rats (Cordycepin 고함유 동충하초(Cordyceps militaris JLM 0636)의 orotic acid 유발 흰쥐의 지방간 개선효과)

  • The concentration of cordycepin produced by crossbred Cordyceps militaris JLM 0636 (CM${\alpha}$) was 7.42 mg/g dry weight, which was 7-fold higher than that of C. militaris (CM). However, cordycepin was not detected in Paecilomyces japonica (PJ). The improvement effects of CM${\alpha}$, CM and PJ on orotic acid (OA)-induced fatty liver in male Sprague-Dawley rats was investigated. Rats were randomly divided into five groups (n=6) based on five dietary categories: normal (N), OA control (C), OA plus 3% (w/w) PJ (PJ), OA plus 3% CM (CM), and OA plus 3% CM${\alpha}$ (CM${\alpha}$). OA treatment induced the retardation of body weight gain and enlargement of the liver. The concentration of hepatic triglyceride was markedly increased by OA-feeding, whereas this rise was significantly reduced by simultaneous feeding of OA, PJ, CM, and CM${\alpha}$, and this phenomenon was more pronounced by cordycepin-enriched CM${\alpha}$. The contents of total lipid, triglyceride, and free fatty acid in the serum were significantly or slightly lower in the OA control group than those of the N group, but there were no significant differences among the OA treatment groups. The hepatocytes in the OA-induced fatty liver contained numerous large lipid droplets, but PJ, CM, and CM${\alpha}$ feeding prevented the OA-induced lipid droplet accumulation in the hepatocytes. This effect was more pronounced in cordycepin-enriched CM${\alpha}$ than in PJ or CM in OA treatment rats. Accordingly, cordycepin-enriched CM${\alpha}$ could be an ideal candidate material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.
