Production of T-2 Toxin and Its Metabolites by Fusarium sporotrichioides Isolates from the Corn Producing Area in Korea (우리나라 옥수수산지에서 분리한 Fusarium sporotrichioides 균주들에 의한 T-2 독소 및 관련 대사물의 생성)

  • Four isolates of Fusarium sporotrichioides obtained from the corn producing area were tested for their toxicities by feeding the crude cultures to rats. Three out of four isolates were highly toxic and killed all rats within 3-4 days after feeding. The chemical analyses of toxic cultures by thin layer chromatography and gas chromatography-mass spectrometry revealed that two isolates from Jeongsun district produced T-2 toxin and its related trichothecenes. This is the first report that F. sporotrichioides isolates produce T-2 toxin in Korea.

  • 인삼의 효능 및 유효성분에 관한 수많은 문헌 및 연구사례가 보고 되어 있음에도 불구하고 인삼의 주성분인 ginsenosides의 합성경로나 그와 관련된 유전인자에 대하여서는 거의 밝혀져 있지 않다. 인삼의 생리적 특성규명 및 ginsenosides의 생합성 경로를 밝히기 위한 한 방법으로써 프로테옴 분석이 시도되었다. 본지에서는 고려인삼 (Panax ginseng C.A. Meyer)을 대상으로 이루어진 최근의 프로테옴 분석 및 이차 대사산물의 생산과 연관된 유전인자를 찾기 위하여 이루어진 인삼의 기능유전체 분석을 소개하고자 한다.

Estrogenic Potentials of Organochlorine Pesticides which Detected in Korean Adipose Tissue (한국인의 지방조직에서 검출되는 유기염소제 농약의 에스트로겐 활성)

  • 유기염소계 농약은 화학적으로 안정하고, 지용성이 크며 체내 대사에 대해 저항성을 가지고 있어서 인체 및 생태계의 생물체 중 축적성이 매우 크다. 또한 대부분 내분비계 장애물질로 분류되어 있어 관심의 대상이 되고 있다. 유기염소계 농약은 잔류성으로 인하여 사용이 금지되었음에도 불구하고 인체에서 검출되고 있으며 한국인의 지방조직을 분석한 결과 9종의 유기염소계 농약이 주로 축적되어 있음이 보고된 바 있다. 본 연구에서는 이들 유기염소계 농약의 에스트로겐 활성을 MCF-7 BUS cell을 이용한 E-screen assay competitive binding assay 및 pS$_2$ gene experession assay에 의해 조사하였다. o,p'-DDT,p, p'-DDT,p,p'-DDD,p,p'-DDE등 4종의 유기염소계 농약은 에스트로겐 수용체에 대한 ligand의존적인 작용기전에 의해 에스트로겐 활성을 가지며, $\alpha$-, $\beta$-, ${\gamma}$-, $\delta$-BHC, dieldrin등 5종의 유기염소계 농약은 lingand-비의존적 작용기전에 의해 에스트로겐 활성을 보였다. 또한 이들 유기염소계 농약을 혼합 투여하여 에스트로겐 활성을 관찰한 결과 DDT류의 경우에는 단독투여시 보다 그 대사체와 혼합 투여할 때 에스트로겐 활성에 상승적 효과가 나타났으며 o,p'-와 p,p'-DDT의 두 이성질체를 혼합 투여할 경우가 단독 투여시 보다 상승적 효과가 나타났다. 따라서 지방조직에서 검출되는 유기염소계농약은 상호작용에 의해 개별 물질이 나타내는 내분비계장애작용 보다 실제로는 강한 효과를 나타낼 것으로 추정되었다.

시상하부의 심혈관 조절기전: 후시상하부의 세포외액의 monoamine 농도에 대한 전신 혈압변화의 영향

  • 본 연구에서는 생체 뇌 국소부위의 세포외액을 대상으로 실험할 수 있는 뇌 미세투석법을 이용하여 후시상하부에서 유리되는 monoamine의 세포외액 농도에 대한 전신 혈압변화의 영향을 관찰함으로써, 심혈관 조절에 대한 후시상하부의 monoamine성 뉴론의 생리적 역할을 규명하고자 하였다. 마취한 흰쥐의 머리를 뇌정위 고정장치에 고정시키고 미세투석관 (microdialysis probe)을 후시상하부에 위치시킨 후 링거액으로 관류하였다. 미세투석액내에 존재하는 monoamine성 신경 전달물질들과 그 대사체들의 정량분석은 고속액체크로마토그라피와 전기화학검출기를 이용하여 실시하였다. 전신 혈압을 40분 동안 약 40mmHg 상승 혹은 30mmHg 감소시키기 위하여 L-phenylephrine hydrochloride (800ng/100 g/min) 혹은 nitroprusside dihydrate (500 ng/100 g/min)를 대퇴정맥을 통하여 주사하였다. 후시상하부로 부터 20분 간격으로 얻은 투석액에서 신정화학물질들의 농도는 미세투석관 삽입후 2시간에 안정되었다. 관류액의 $K^{+}$ 농도를 90 mM로 증가시켰을 때 후시상하부의 투석액에서 norepinephrine (NE) 과 serotonin (5-HT)의 농도는 각각 기준치의 176.5 $\pm$ 14.8%, 149.1 $\pm$ 2.3%로 증가하였다. Phenylephrine (i.v.)으로 유발된 전신 혈압상승에 의하여 NE과 5-HT의 양은 각각 기준치의 79.3 $\pm$ 4.4%, 61.4 $\pm$ 10.3%로 유의하게 감소하였다. 또한 nitroprusside (i.v.)에 의하여 전신혈압이 감소하였을 때 투석액내 5-HT의 양은 기준치의 195.0 $\pm$ 23.0% 로 유의하게 증가하였다. 후시상하부의 투석액내 monoamine성 대사체들의 경우에는 전신 혈압의 변화에 대한 유의한 반응을 나타내지 않았다. 이상을 종합하면 전신 혈압이 증가되었을 때는 말초 압수용체의 흥분에서 기시한 신경충동이 후시상하부에 전달되어 신경말단에서 5-HT 과 NE 의 유리가 감소되고, 5-HT의 경우에는 전신 혈압이 감소되었을때 그 반대 현상이 일어난다는 가설을 제시할 수 있다. 본 연구의 결과는 생체상태에서 혈압변화에 대한 후시상하부의 monoamine성 조절양상을 규명한 것으로서 특히 후시상하부의 serotonin성 신경계를 통한 심혈관 중추조절의 가능성을 처음으로 제시한 것이다.

Metabolome Analysis and Aroma Characteristics of Fermented Fruit Vinegar (발효 과일식초의 대사체 분석 및 향기 특성)

  • Vinegar was prepared from the fruits produced in Gangwon province, and major metabolite and aroma components were investigated for acetic acid fermentation. In the case of Meoru-Bokbunja vinegar, the ${\text\tiny{L}}$-alanine content was greatly changed by acetic acid fermentation. Acetic acid had the highest content (43%) of total aromatic components, and the contents of ester compounds, such as ethyl acetate and isoamyl acetate, were significantly increased after fermentation. Omija-Makgeolli vinegar produced linalool and hexanoic acid by fermentation, and terpenoid compound was prevalent (41.5%). ${\text\tiny{L}}$-alanine was also increased in Omija-Makgeolli vinegar, similar to that of Meoru-Bokbunja vinegar. Terpene compounds, such as terpinel-4-ol and ${\alpha}$-terpineol in Omija-Makgeolli vinegar, and ethyl acetate in Meoru-Bokbunja vinegar, were identified as major components in each aromatic formulation.

Effects of Walking Activity Monitoring and Feedback Using Wearable Device on Body Composition and Metabolic Syndrome Risk Factors in Middle-aged Women

  • The purpose of this study is to analyze the effect of monitoring and feedback on walking activity using wearable devices on changes in body composition and metabolic syndrome factors in middle-aged woman. The subjects use a mobile interlocking wearable device for 6 month, feedback is provided through mobile app automatically transmitted step counter and walking activity. As a results, significantly difference shows on body weight, skeletal muscle mass, and body fat in statistically(p<.001). In changes of metabolic syndrome factor, systolic(p<.05), fasting flucose(p<.01), abdominal circumference(p<.001), TC(p<.001), and LDL-C(p<.05) are significantly difference. In conclusion, wearable devices that can be conveniently applied is an effective tool to increase walking activity and prevent metabolic syndrome of people with a sedentary lifestyle.

Identification of Key Metabolites Involved in Quantitative Growth of Pinus koraiensis trees (II) (잣나무 생장과 관련이 있는 주요 대사물질 인자(II))

  • A metabolomic study using GC/MS analysis was conducted to identify key metabolic components regulating the growth of open-pollinated Pinus koraiensis families, which were grown for 29 years at three different locations. Among 110 individual metabolites identified, the contents of 62 metabolites were higher in the superior than in the inferior families (p<0.05), together with 22 metabolites, such as phosphoric acid, alanine, glycine, malic acid, and sucrose, being accumulated 1.5-fold higher in the superior families. In addition, 15 metabolites including alanine, malic acid, sucrose, d-turanose, and succinic acid showed positive correlation with the growth (p<0.01). Furthermore, the metabolites, of which contents were correlated with the growth but not significantly changed at different locations, were acetic acid, succinic acid, butanoic acid, glutamic acid, and inositol. Therefore we suggest that several metabolites selected in this study may be used as metabolic markers for quantitative growth trait in P. koraiensis.

Analysis of toxicity using bio-digital contents (바이오 디지털 콘텐츠를 이용한 독성의 분석)

  • Numerous bio-digital contents have been produced by new technology using biochip and others for analyzing early chemical-induced genes. These contents have little meaning by themselves, and so they should be modified and extracted after consideration of biological meaning. These include genomics, transcriptomics, protenomics, metabolomics, which combined into omics. Omics tools could be applied into toxicology, forming a new field of toxicogenomics. It is possible that approach of toxicogenomics can estimate toxicity more quickly and accurately by analyzing gene/protein/metabolite profiles. These approaches should help not only to discover highly sensitive and predictive biomarkers but also to understand molecular mechanism(s) of toxicity, based on the development of analysing technology. Furthermore, it is important that bio-digital contents should be obtained from specific cells having biological events more than from whole cells. Taken together, many bio-digital contents should be analyzed by careful calculating algorism under well-designed experimental protocols, network analysis using computational algorism and related profound databases.

Transciptomic Analysis of Larval Fat Body of Plutella xylostella under Low Temperature (저온조건에서 배추좀나방(Plutella xylostella) 지방체 유전자 발현 변화)

  • BACKGROUND: Temperature is known to be the main factor affecting development, growth and reproduction of organisms and also a physical factor directly related to insect survival. Insects as ectothermal species should be responsive to climate changes for their survival and develop various survival strategies under the unfavorable temperature such as low temperature. The purpose of this study is to identify genes contributing to adaptation of low temperature. METHODS AND RESULTS: To identify genes contributing to adaptation of low temperature, the transcriptomic data were obtained from fat body in Plutella xyostella larvae via next generation sequencing. We identified structural proteins, heat shock proteins, antioxidant enzymes, detoxification proteins, and cryoprotectant mobilization and biosynthesis-related proteins. Genes encoding chitinase, cuticular protein, Hsp23, chytochrome protein, Glutathione S transferase, and phospholipase 2 were up-regulated under low temperature. Proteins related to energy metabolism such as UDP-glycosy ltransferase, trehalase and trehalose transporter were down-regulated. CONCLUSION: When insect pests were exposed to low temperature, changes in gene expression of fat body could provide some hints for understanding temperature adaptation strategies.

Determination of Pyribencarb and Its Metabolite KIE-9749 in Agricultural Commodities by High-Performance Liquid Chromatography (액체크로마토그래피를 이용한 농산물중 pyribencarb와 대사물 KIE-9749의 잔류 분석법)

  • The aim of this study was to develop an analytical method for the determination of pyribencarb and its metabolite KIE-9749 in agricultural commodities. The experiment was performed with a range of concentrations $0.05{\sim}2.5{\mu}g/g$ in apple, green pepper, potato, hulled rice, soybean, pear, peach, grape and cucumber. Each samples were extracted with acetone and cleaned by dichloromethane/saline water partition and purified with Florisil solid phase extraction (SPE) cartridge and aminopropyl SPE cartridge. Pyribencarb and KIE-9749 were separated and quantified by HPLC/UVD at 265nm using acetonitrile and water as mobile phase. The recoveries of pyribencarb and KIE-9749 were within 78.3~108.4% and 73.9~113.7% with RSD below 12.2% and 15.0%, respectively. The limits of quantification (LOQ) were both $0.05{\mu}g/g$. LC/ESI-MS/MS was optimized for confirmation of residue identity.
