Five Genes Regulated by Oryctes rhinoceros nudivirus Infection in the Intestinal Tube of Allomyrina dichotoma (장수풍뎅이(Allomyrina dichotoma)에 Oryctes rhinoceros nudivirus 감염에 의해서 유전자 발현이 조절되는 5개의 유전자)

  • Korean rhinoceros beetles (Allomyrina dichotoma), which can be found in broad-leaved forests in mountainous habitats and lives for around one year in wild. This beetle is currently popular as a pet and traditionally regarded as a medicine for liver-related diseases in Korea. It is reported that the economic losses in the mass-rearing facilities by virus infection have been increased since the 2010s in Korea. The causing virus for the A. dichotoma was firstly reported as an Oryctes rhinoceros nudivirus (OrNV) in 2015. We, here, observes that serious morphological changes in the intestinal tube from the OrNV-infected beetles, and report five genes, which are regulated by OrNV infection in the intestine; Krueppel-like factor 15 (Klf15), Endoplasmic reticulum aminopeptidase 2 (ERAP2), U5 small nuclear ribonucleoprotein 200 kDa helicase (Snrnp200), Muscleblind-like protein 2a (mbnl2a), and MIMI_L93. The results may provide a clue to the early diagnosis and disease treatment during the mass-rearing facilities of the A. dichotoma.

  • The white-spotted flower chafer (Protaetia brevitarsis) larva is one of the insects widely-used for edible and medicinal purposes in Eastern Asia. As a result of increasing demand for P. brevitarsis, massrearing systems in domestic farms have become necessary. However, the mass-rearing of larvae under confined rearing conditions could provide conditions unsuitable for preventing entomopathogenic diseases. Metarhizium anisopliae is the strongest fatal entomopathogenic fungus against P. brevitarsis. For inhibition of M. anisopliae, we used a Rhus verniciflua Stokes extract that has antifungal components. We investigated the inhibitory effect of the R. verniciflua extract at 1%, 5%, and 10% concentrations. The results showed that a 1% R. verniciflua extract added to sawdust produced a significantly low P. brevitarsis mortality rate. Moreover, extract-treated groups were heavier and had a shorter larval period than those of the untreated group. Consequently, we suggest that using an R. verniciflua extract can reduce the P. brevitarsis fatality rate from entomopathogenic fungi (e.g. M. anisopliae), resulting in more effective mass-rearing systems for P. brevitarsis.

  • White spot disease by Cryptocaryon irritans broke out among cultured river puffer (Takifugu obscurus) cultured in sea water in Oct. 2001 and May 2004. Many white spots were easily observed on the gills with the naked eyes. The parasites attached on the gills at the base of the gill lamellae. The trophonts penetrated the interlamellar epithelium. Epithelial cell proliferation in interlamellar spaces occurred. PAS- positive mucous cells were prominent in the proliferated epithelium.

Environmental Factors Related to Mass Moralities of Young Red Seabream (Pagrus major) in the Artificial Seed Production (일본산 참돔, Pagrus major 종묘생산과정중 사육수 변화로 인한 대량폐사)

  • As seed production program is growing prosperously in various fishes in southern Korea, disease problems in larval and juvenile stages have emerged as a new research object. The following results were obtained from investigation about environmental factors related to mass mortalities of young Kinki red seabream, Pagrus major in process of artificial seedling production. Total length of red seabream larvae hatched was 2.93mm, and became 18.83~20.12mm at day 40. The first noticeable mortality of red seabream larvae (7.98~9.37mm) occurred in 25~30 day-old fish with the survival rates of 59.8~60.3%. Thereafter the mortality of larvae decreased, survival rate was 20.5~25.45% in day 40. After 20~30 days, the quality of pond water was bellow II class. During the experimental period COD, $PO_4$-P, $NO_2$-N, $NO_3$-N and $NH_$-N increased up to 3, 7, 34, 6 and 8 times, respectively, compared to initial ones. The number of viable bacteria in pond water and seabream larvae were $6.3{\times}10^6$~$2.3{\times}10^7$ cfu/ml, 4.3~$7.4{\times}10^6$ cfu/g in day 25, respectively. Among the isolated bacteria from the diseased red seabream in day 25, Vibrio spp. was considered to be the causative organism. External symptoms of this disease were floated, spined near the surface and inflated abdomen. When the isolated strain of the Vibrio was bathed to seabream larvae, $LD_50$ of seabream larvae was over $10^6$ cfu/ml of Vibrio spp.

Biological Characteristics of Tetrastichus sp. reared on Artificial Host (인공숙주에서 증식된 Tetrastichus sp.의 생물학적 특성)

  • A gregarious pupal endoparasitoid Tetrastichsus sp. (Hymenoptera: Eulophidae) was reared in vitro from oviposition to adult emergence on an artificial host. The host pupal case was made using 0.02 mm-thick polypropylene film, and was filled with a diet consisting of powders of Antheraea pernyi pupa, chicken yolk, infant formula, royal jelly, and Neisenheimer's salt solution. Female parasitoids reared in the artificial host produced smaller sized progeny than those reared in in vivo, but the adults reproduced fertile offsprings. Furthermore in vitro second-generation (G$_2$) females showed more improved biological characteristics, compared with their parents. The fecundity (mean no. adult progeny), oviposition period (days), and longevity (days) of G$_2$ female were evaluated as 45.7, 7.8, and 13.8, respectively Female biased sex ratio was obtained with 76.9% female progeny. The results demonstrated that Tetrastichus sp. is a promising parasitoid for in vitro mass production.

Development of Artificial Diets for Green Lacewing, Chrysopa pallens (Rambur), by Addition of Natural Products (천연첨가물을 이용한 칠성풀잠자리붙이 인공사료 개발)

  • The green lacewing, Chrysopa pallens (Rambur), was reared on artificial diets supplementedwith natural products such as lyophilized beef liver, silk worm pupae powder and drone honey beepowder. The performance of the green lacewing was improved siginificantly on only the diet to whichadded 1 % of silk worm pupae powder (wt/v). Larval developement was completed in 15.6 f 0.99 days,cocoon weight was 16.4f2.94 mg, and suvival to adult was 89.3% at 27$^{\circ}$C. And another diet made bymixing 4 part of lyophilized beef liver powder, 4 part of lyophilized beef powder, 3 part of lyophilizedchicken egg, 4 part of sucrose, and 50 mg each of vitamin C and Vanderzant's vitamin B mixture waspromising. Larval development of the green lacewing on this diet was completed in 12.7 f 1.45 days at27"C, pupal weight was 18.1 f 2.24mg, suvival to adult was loo%, and 633f70.4 eggs were laid in 20days after beginning of oviposition.ginning of oviposition.

Prevalence of viral nervous necrosis (VNN) in sevenband grouper Epinephelus septemfasciatus farms (능성어 양식장에서의 viral nervous necrosis (VNN) 발생양상)

  • Prevalence of viral nervous necrosis (VNN) in sevenband grouper Epinephelus septemfasciatus farms was investigated during the period of 2006-2008. The outbreak of the VNN was observed from August (water temperature: $24-26^{\circ}C$) to September or October ($20-25^{\circ}C$). The viral infection resulted in acute or chronic mortality of the host, where the mortality rate of juvenile fish was higher than that of the adult fish. Phylogenetic analysis based on partial RNA2 coat protein gene nucleotide sequences revealed that all the isolates from sevenband grouper were classified into the genotype redspotted grouper nervous necrosis virus (RGNNV). In conclusion, VNN of juvenile and adult sevenband groupers during the summer season (July to October, water temperature: about $24^{\circ}C$) was caused by virus belonging to the genotype RGNNV.

  • This experiment was carried out to develop an artificial diet and a mass rearing system, which is essentIal in the pheromone research for Ostrmia furnacalis. Component deletion and addit.ion methods were applied t.o define the nut.ritional Importance of some components, especially yeast, Wesson's salt mixture and vitamin complex. Secondly we have developed a proper rearmg syst.em for O. furnacalis. and also tried to find out the factors affecting rapid sclerotization (dark-canning) of pupae. AddItion of yeast raIsed the pupal weight. and pupation rate. However Wesson's salt mixture got the female pupae lighter. Vitamin complex as a substitue for yeast showed a good survivahty in an early larval stage buL pupal weight was lower than that of the individuals reared on the yeast dIet. Also corrugated cardboard was found to be very good for pupation. And the rate of dark~tannmg in pupae was higher as the period of larval development was shorter.

  • We investigated the growth and survival in seedling production, and growth performance was compared with the different rearing tanks and protein source of formulated feed for juvenile Pacific bluefin tuna Thunnus orientalis (PBT). The survival rate at the end of nursery culture at 30 days after hatching was $0.69{\pm}0.40%$, and total length and mean body weight were $49.83{\pm}2.52\;mm$ and $1.03{\pm}0.09\;g$, respectively. Growth performance has no significant difference in fish reared by different tanks forms for 10 days. In order to develop an artificial diet, we evaluated the dietary utility of enzyme treated fish meal (Bio-CP, BIO) for juvenile PBT. Only diet BIO sustained similar growth and higher feed efficiency, and final carcass lipid content as compared to those of Sand lance (SL) These results revealed that BIO-CP a suitable dietary protein source, could sustain growth of PBT.

  • The superworm, as known the larva of Zophobas morio, has been officially imported from 2011 and bred commercially in Korea. But it is named as the corrected scientific name, Zophobas atratus by junior synonym throughout traditional taxonomy in this study and newly designated Korean name as 'a-me-ri-ca-wang-geo-jeo-ri' in terms of resource management. Z. atratus was compared with wild native tenebrionids, Promethis valgipes and a commercial reared Tenebrio molitor on the basis of DNA barcode analysis. As the results, the average genetic divergence was 21.4% between Z. atratus and P. valgipes, and 20.9% between Z. atratus and T. molitor. These large divergences imply these tenebrionids species can be easily identified by DNA barcodes. The results of genetic divergences within species also suggest that Korean populations of Z. atratus, having the same haplotype, might be introduced from the same area of foreign country. On the other hand, a population of T. molitor was separated into two distinct intra-specific groups with DNA barcoding gaps ranged from 1.17- 2.19%. We suppose that domestic breeding entities of T. molitor might be introduced and mixed from two different local groups. Through this study, we expect that classification for two tenebrionid introduced from foreign countries can be used for the management of insect resources in Korea.
