Monoclonal antibodies against structural proteins of bovine viral diarrhea virus (소 설사병 바이러스 구조단백에 대한 단크론항체 성상에 대한 연구)

  • Monoclonal antibodies against structural proteins of bovine viral diarrhea virus(BVDV) were derived by classical hybridoma techniques. These antibodies were characterized by serum neutralization, immunoblotting and immunoprecipitation. The neutralizing monoclonal antibody reacted with the 56kd to 54kd(M.W.) viral protein in western blotting and immunoprecipitation analysis. Although there was no neutralizing activity, another monoclanal antibody reacted with the 45kd protein by immunoprecipitation and with both the 45kd and 36kd proteins in immunoblotting analysis. respectively. Densitometer scanning of purified BVDV and the immunopreipitation of whole virus particles with neutralizing monoclonal antibody revealed the presence of more than twelve viral polypeptides. Although no possible precursor form of protein was identified with the neutralizing monoclonal antibody. the presence of intact virion was detected in the infected cell supernatant immediatelty after pulse labeling, indicating rapid translational processing as well as packaging of the virus. The partial peptide mapping of 45kd and 36kd proteins with Staphylococcus aureus V 8 protease showed that these two proteins are related.

Study on the effect of glucose upon the digestibility in the perilla frutescens leave (들깨엽(葉) 단백질(蛋白質)의 소화(消化)에 있어서 glucose의 영향(影響)에 관(關)한 연구(硏究))

  • The testing materials which kept perilla frutescens' leaves frezen are divided into four parts, 1. freezing green leaves, 2. glucose added to the leaves dried in the sun, 3. glucose unadded to the leaves dried in the sun and 4. the leaves dried in the immediately after collecting sample. The perilla frutescens' leaves are treated with the artificial digestion test to investigate the effects of the digestibility of ingredients and of protein. The results obtained were as follows ; 1. The digestibility of crude protein of sample using the common leaves dried in the sun immediately after collecting sample was presented highest at 83.15%, the freezing green leaves at 68.35%, glucose added to the leaves dried in the sun at 64.25% and glucose unadded to the leaves dried in the sun at 62.12%. The digestibility of perilla frutescens' by freezing green leaves, glucose added or glucose unadded to the leaves dried in the sun is on the decrease without difference. 2. It was suggested that glucose and reductive sugars to perilla frutescens' leaves is not affected by the decreased digestibility of protein, dince the digestibility of glucose added to the leaves dried in the sun and glucose unadded to the leaves dried in the sun almost never makes a difference. 3. The digestibility of freezing the green leaves for six months was quite different from the leaves that were dried in the sun immediately after collecting sample, in that the leaves that were frozen for six months were decreased 1/5 quantity of the shole crude protein.

Effect of Godulbaegi(Korean Lettuce, Ixeris sonchifolia H.) Kimchi on the in vitro Digestibility of Proteins (고들빼기 김치가 단백질 소화율에 미치는 영향)

  • Extent of reduction in protein digestibility by Godulbaegi(lxeris sonchifolia H.) kimchi powder appeared to be related to the kimchi weight-to-protein weight ratio, the kind of protein sources, part of plants and fementation period. In vitro digestibilities of protein were significantly(p<0.05) reduced as the freeze dried kimchi powder weight-to-protein ration increased from 0.5 : 1.0 to 1.0 : 1.0 for all Godulbaegi samples and protein sources. Overall digestibility of protein sources in the presence of kimchi powder, the reduction ranked in the following order : soybean>casein>beef>squid. Lower(p<0.005) reduction occurred for each protein source when raw plant was exchanged for kimchi products. Some greater reduction of digestibility was noted in young plants and leaf samples than ripe or root samples. Trypsin inhibitor, which expressed as soybena trypsin inhibitor, was inversely related(r=0.8437) to in vitro protein digestibility of casein in the presence of Godulbaegi kimchi powder. More than three times of total polyphenols contained in leaves than in roots. Young leaves had 30% more total polyphenols(37.64mg/g sample) than that in ripe ones. Soaking in 5% NaCl solution for 24 hrs was markedly reduced in total polyphenols as 73% for leaves and 33% for roots. Remarkable reduction in total polyphenols was not checked during fermentation followed after soaking. Trypsin inhibitor content correlated well(r=0.8873) with total polyphenols in all of Godulbaegi samples.

  • The study of pancreatin extraction was investigated by supercritical fluid process. Using supercritical carbon dioxide extraction with entrainer the purification of pancreatin was possible to remove lipids from Hog pancreas. To observe the optimum conditions different experimental variables were changed as pressure, temperature, flow rate of solvent and 0.25 mm of sample size were evaluated for effective removal of lipids. Ethanol and n-hexane were used as an entrainer with 5 mL/min. Increasing pressure at constant temperature the efficiency of the lipid removal in Hog pancreas was improved and the protein was concentrated without denaturalization, compared that of the control Hog pancreas. The most efficient conditions of lipid elimination were 17 MPa of pressure and 35$^{\circ}C$ of temperature and 0.25 mm of sample size.

  • Three experiment were designed to determine the effects of dietary(n-3) fatty acids on the growth-suppressive effects of the inflammatory response and anti-oxidant enzyme activity and balances of nitrogen and energy. Day-old chicks were fed diets containing 1.0 % soybean oil in experiment 1, or diets containing 1.0 % of fish oil in experiment 2, and then on 7-d of age chicks were fed diets containing 1.0 % of either soybean oil or fish oil. In experiment 3, day-old chicks were fed on diets containing 1.0 % of either soybean oil or fish oil. In chicks of half pens within each dietary treatments of each experiments, the acute phase response were induced by injecting with Salmonella typhymurium lipopolysaccaride(LPS) i.p. 3 times at 8, 10 and 12-d of age. During acute phase response, chicks fed dietary fish oil from 1-d of age enhanced energy balances and did not give negative effect on nitrogen balance and uric acid excretion, but elevated MnSOD and Cu/ZnSOD activity In liver and erythrocyte cytosols compared with those fed dietary soybean oil. The results indicated that enhanced daily gain and metabolizable energy value of diet in birds fed fish oil from one-d of age seems due to change on the antioxidants systems during acute phase response.

Electron Microscopic Observations on the tissues infected with Nuclear Polyhedosis Virus of Bombyx mori during eary stage (가잠 핵다각체병 바이러스 초기감염에 관한 전자현미경적 관찰)

  • To investigate the pathway of Nuclear Polyhedrosis Virus(NPV) of Bombyx mori in early stage infection of 2nd instar larva, the larval tissues were observed under electron microscope at interval of 6, 12, 24, and 48 hours after virus inoculation. The results are as follows. 1. The intact and enveloped nucleocapsids released from the polyhedra protein in the gut lumen apparently entered with the microvilli. 2. Virus progenies were observed in columnar cell nuclei 24 hours after inoculation, but polyhedra was not seen. The enveloped virus was observed in some of the intercellular spaces between mid-gut cells. 3. Many enveloped virus particles appeared in the basement membrane. These enveloped virus particles passed the basement membrane and gathered at blood cells in heomocoeal. 4. The NPV muliplicates in nuclei of the blood cells and the tracheal cells normally.

Pheromone Biosynthesis Activating Neuropeptide (PBAN) in Insects (곤충의 페로몬 생합성 활성화 신경펩타이드(PBAN))

  • Neuropeptides produced in neurosecretory cells are the largest group of insect hormones. They regulate various physiological functions, such as fat body homeostasis, feeding, digestion, excretion, circulation, reproduction, metamorphosis, and behavior throughout all life stages. The PRXamide peptide family (X, a variable amino acid) is a well-characterized neuropeptide component with a common amino acid sequence, PRXamide (NH2), at the C-terminal end conserved across Insecta. The PRXamide peptides are classified into three subfamilies, each having diverse biological roles in insects: (1) pyrokinin (PK) includes the pheromone biosynthesis activating neuropeptide (PBAN) and the diapause hormone (DH), (2) the capability (CAPA) peptides, and (3) the ecdysis-triggering hormone (ETH). PBAN as a member of PK subfamily was first identified to stimulate pheromone biosynthesis in moths three decades ago. Since then, PBAN peptides have been extensively studied by various research groups from a broad spectrum of arthropods. In this paper, we briefly review insect PBAN molecules with emphasis on gene structure and expression, signal transduction, physiological mechanism in sex pheromone biosynthesis, and application for pest management.

Effects of Dietary Ptecticu tenebrifer Powder and Canned Mixtures on Protein Digestibility by Different Breeds of Companion Dogs (반려견 품종별 동애등에 분말과 통조림 혼합사료 급여가 단백질 소화율에 미치는 영향)

  • This study investigated the effects of dietary Ptecticu tenebrifer powder and canned mixtures on protein digestibility by different breeds of companion dogs (15 Bichons, 15 Malteses, 15 Chihuahuas and 15 Poodles). The mixtures were divided into Diet A, Diet B, Diet C, Diet D, and Diet E, which were supplied from five farms. Twenty-five grams each was mixed with 100 g of each canned food, and a total of 125 g was measured for each breed of dog. The result of component analysis of the mixtures showed the highest protein contents rather than dry matter, crude ash or crude fat. There were statistical significances (p<0.05) in all mixed feeds fed to bichon, maltese, chihuahua and poodle dog. Overall, protein digestibility by the breeds of dog ranged from 87.44% to 97.18%. Result of breed of dog comparison revealed that Diet E by poodle dog had the highest protein digestibility, and the lowest protein digestibility was observed in Diet C by Maltese. In conclusion, the use of dietary Ptecticu tenebrifer powder and canned mixtures did not only increased protein digestibility by different breeds of dog but also maintained normal manure properties.

Effect of Mixing Mealworm Pellets with Canine Diets on Protein Digestibility and Fecal Properties in Companion Canine (밀웜 펠렛과 반려견 사료의 혼합급여가 반려견의 단백질 소화율과 분변성상에 미치는 영향)

  • This study aimed to investigate the effects of feeding mealworm pellets on protein digestibility and fecal composition of companion canines. The canine breeds used in the specification test were Maltese (average weight 2.0 kg) and Poodle (average weight 2.5 kg), with a total of six canines, three of each breed, without discriminating between males or females. The control feed for the two canines was based on 100 g of commercially available general feed, to which the dogs were allowed to adapt for approximately 3 d. Ony on Day 0 of the experiments, dogs were fed twice, in the morning and afternoon. One day later, the treated group was 80 g of regular feed with 20 g of mealworm pellets added, and the dog was fed for 1 to 7 d in the same manner as the control group. The addition of mealworm pellets increased the protein digestibility in both canine breeds, however, the differences were no significances (p>0.05). Based on the normal stool score of 3, no remarkable changes were observed in stool score from Days 1 to 6, showing that the canines had adapted to the mealworm pellet feed. In particular, on Day 7, feeding mealworm pellet appeared to affect the fecal properties differently between the two breeds. This difference can be owing to differences in the intestinal environment and digestion and absorption ability between the two breeds. Consequently, using mealworm pellets improved the protein digestibility of canines as well as maintained normal fecal properties.

Influence of Dietary Protein and Feeding Pattern on the Weight Gain, Metabolism and Body Composition of Rats (식이단백질과 급식형태가 흰쥐의 성장, 대사 및 체조성에 미치는 영향)

  • A series of growing and digestion trials was conducted using Sprague- Dawley weanling male rats in order to determine the effects of two protein sources (casein and ISP (isolated soyprotein) ), three protein levels (10, 20 and 30%), and two feeding patterns (ad libitum and meal feeding) on the growth, protein and energy metabolism, and body composition of rats. The total energy level of experimental diets was kept constant in 3,600 kcal ME /kg diet. The results were as follows : 1) The amount of food intake and the weight gain of meal-fed group were lower than those of ad libitum group. Though the intake of meal-fed group on 20 and 30% casein diet was only 85% of ad libitum group, it was able to gain as much as ad libitum group. 2) There were no significant differences in the food efficiency ratio (FER) and the energy efficiency (weight gain per 100 kcal GE intake) between ad libitum and meal feeding group. The FER and the energy efficiency of 20 and 30% casein diets of meal-fed group were greater than those of ad libitum group. 3) Though the gross energy intake (GE ), the digestible energy (DE) and the metabolizable energy (ME) tended to be lower at meal-fed group, the DE/GE and the ME/GE ratios for meal-fed group were the same as those for ad libitum. 4) Though meal- fed group fed less amount of nitrogen than ad libitum group, there were no differences in nitrogen balance and the retention of rats among the treatments. Actually meal-fed group retained more nitrogen than ad libitum group at the levels of 20 and 30% dietary protein. 5) After growing and digestion trials, the body composition of rats was constant among all treatments. Significantly high negative correlation coefficient (r = -0.77) was found between the body fat content and the body moisture content. Consequently, this study suggests that meal- fed group on 20 and 30% casein diets has shown more effective utilization of the ingested food and energy than ad libitum group, and increasing tendency of weight gain and the body fat deposition. Those influences of meal feeding pattern in rats were more effective on the casein diet than on the ISP diet.
