Correlation between physicochemical properties and biological half-life of triazole fungicides in perilla leaf (들깻잎에서 Triazole계 살균제의 생물학적 반감기와 물리화학적 특성과의 상관관계)

  • The biological half-life of pesticides applied on crops is the key indicator for ensuring the safety of agricultural products. The biological half-life is affected by the several factors like growing conditions of the crop, climate, application method, and physicochemical properties of pesticides. In this study, the biological half-life was calculated and the degradation rates of six triazole fungicides sprayed on perilla leaves were evaluated. Moreover, the statistical analysis confirmed the correlation between the biological half-life and physicochemical properties of six triazole pesticides. The recoveries of the six pesticides were between 84.8-104.9%, which satisfied the residual pesticide analysis criteria. The biological half-life of six pesticides sprayed on perilla leaves, calculated using the first-order kinetics model, ranged between 6.4-15.1 days. When the biological half-life and the physicochemical properties were correlated using the principal component analysis: pKa and Log P, the biological half-life was found to be affected by PC1. The correlation coefficient between biological half-life and physicochemical properties (pKa), calculated by Spearman rank-order correlation, was R2 = -0.928 (p <0.01). Biological half-life has been shown to correlate with pKa. In conclusion, it can be used as a database for the relationship between biological half-life and physicochemical properties and will contribute to ensure safe supply of agricultural products.

Residue Studies of Difenoconazole and Thiamethoxam during Cultivation of Sweet Persimmon for Export (수출용 단감에 대한 Difenoconazole과 Thiamethoxam의 잔류특성 연구)

  • BACKGROUND: In order to elucidate residual characteristics of difenoconazole and thiamethoxam by treatment to sweet persimmons for one year and to generate the data for the maximum residue limit (MRL) establishment for those pesticides in or on sweet persimmon. METHODS AND RESULTS: Systemic fungicide difenoconazole WP (10% a.i.) and systemic insecticide thiamethoxam WG (10% a.i.) were sprayed onto 12~25-years-old sweet persimmons according to its preharvest interval (PHI), respectively, and then fresh sweet persimmons were harvested at 0, 1, 3, 7, 14, 21 days after treatment from pesticide-sprayed plots at each 3 sites. The analytical methods were evaluated to limit of quantification, linearity, specificity, reproducibility and recoveries. The crop samples were extracted with acetone and performed dichloromethane partition process. The extracted samples of difenoconazole were analyzed by GC-ECD and the thiamethoxam extracted samples were analyzed by HPLC with good sensitivity and selectivity of the method. The average recoveries of difenoconazole ranged from 87.5 to 99.5% with the percentage of coefficient variation in the range 4.1~7.6% at three different spiking levels(0.02, 0.2 and 2.0 mg/kg). And the average recoveries of thiamethoxam and clothianidin ranged from 88.8 to 98.9% and 83.2 to 96.6% with the percentage of coefficient variation in the range 3.6~5.0% and 3.8~9.4% at three different spiking levels(0.02, 0.2 and 2.0 mg/kg), respectively. The residue amounts ranges of difenoconazole were 0.2~0.56 mg/kg and the residue amount was decreased below the MRL level, 1.0 mg/kg, after 1 day harvest. The residue amounts ranges of thiamethoxam were 0.08~0.28 mg/kg and the residue amount was decreased below the MRL level, 0.5 mg/kg, after 1 day harvest. And the residue amount of clothianidin was below then 0.03 mg/kg for only one test site of 14 and 28 day samples. CONCLUSION: As a result, the residual amounts of difenoconazole and thiamethoxam were not exceeded the MRL of established criteria for sweet persimmon. The biological half-lives of difenoconazole and thiamethoxam were 13.6, 19.4, 16.3 and 10.0, 15.3, 14.0 days at each three test sites, respectively.

Herbicidal Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Substance (생분해성 광활성 물질 5-aminolevulinic acid의 제초활성)

  • Laboratory and greenhouse experiments were conducted to determine the herbicidal effect of two types of ${\delta}$-aminolevulinic acid (ALA), microbiologically-produced ALA (Bio-ALA) and synthetically produced ALA (Synthetic-ALA), on plant growth and chlorophyll content of Chinese cabbage. ALA effect on early plant growth was greatly concentration dependant, showing significant inhibition at higher concentrations. Both pre- and post-emergence application of ALA exhibited significant degree of photodynamic phytotoxicity. Older plants with many leaves were more tolerant to ALA than younger plants, showing less injury. No significant difference in herbicidal activity of two types of ALA, Bio-ALA and Synthetic-ALA, on plant height and chlorophyll content of Chinese cabbage was observed. However, residual biological activity and physico-chemical properties of Synthetic-ALA were more stable than those of Bio-ALA. Our results suggest that ALA had herbicidal potential with both pre- and post-emergence application, and that the chemical may be a valuable mean of eco-friendly weed control based on natural microbial substance.

Control of Phythophthora capsici and residual characteristics by drenching of pesticides on tomato in hydroponic culture system (약제 관주처리에 의한 양액재배 토마토의 역병 방제 및 농약잔류 특성)

  • To establish effective and safe control method against Phytophthora root rot caused by Phytophthora capsici on tomato in hydroponic culture, three pesticides, oxadixyl copper hydroxide 8% WP, metalaxyl copper oxychloride 15% WP, and dimethomorph. dithianon 38% WP at 4 concentration levels were tested on potato dextrose agar medium inoculated with Phytophthora capsici. All pesticides inhibited mycelial growth, but two pesticides of them, metalaxyl copper oxychloride WP and dimethomorph. dithianon WP, were selected as effective pesticides for the efficacy test in a hydroponic culture. Forty days after transplanting of tomato seedlings, 4 ml of sporangia of P. capsici (about 25 sporangi/ml) per plot was inoculated around tomato plant root, and then 5 days after inoculation, the pesticides diluted at 5,000 times were drenched 1, 2 or 3 times per plot on the culture cube at 15 days interval. Fifteen days after drenching, tomato fruits and hydroponic culture solution were sampled for the analysis of pesticide residues. Dimethomorph was detected 0.001 and 0.003 mg/kg in tomato of the plots sprayed 2 and 3 times with dimethomorph dithianon WP of which detection levels were far below compared with 1.0 mg/kg of the Korean MRL of dimethomorph on tomato. Incidences of Phytophthora root rot were $30.5{\sim}50%$ in the plots drenched at 1 or 2 times with metalaxyl.copper oxychloride WP, and $16.7{\sim}25%$ in the plots treated with dimethomorph dithianon WP. However, there was no incidence of Phytophthora root rot in the plots treated at 3 times with both of pesticides, showing no phytotoxic effect. Based on the results, the drenching of these pesticides on the culture cube could be recommended as a very safe and effective control method for Phytophthora root rot in tomato.

Pesticides, heavy metals, radioactive substances, and ash of Poria cocos distributed in Korea (국내 유통 복령(Poria cocos)의 농약, 중금속, 방사선물질 및 회분 함량 조사 연구)

  • The purpose of this study is to provide basic data on the safety of Poria cocos, a major Korean herbal medicine, and to establish tolerance levels for hazardous substances. Residual pesticides (321 types), heavy metals (7 types), radioactivity (3 types), and ash content were analyzed in 10 Poria cocos products distributed in Korea (5 domestic and 5 in China). Of the residual pesticides, cypermethrin was detected at 0.03 mg/kg in one domestic sample and was below the standard value. Of the heavy metals, the levels of Zn, Pb, Ni, Cu, Hg, and Cr were below the standard values in all samples. Hg was not detected in any sample. Cd was detected at 0.011 mg/kg in one Chinese sample. The radioactivity concentration was also below the MDA value in all P. cocos samples. The ash content was less than the standard value of 1.0% in all P. cocos samples. The findings indicate that P. cocos distributed in Korea is safe. However, because P. cocos is always exposed to harmful substances because of the nature of its cultivation in the soil, it seems necessary to establish systematic management standards for the entire production stage, such as cultivation, storage, and distribution.

Development and Validation of a Simultaneous Analytical Method for 5 Residual Pesticides in Agricultural Products using GC-MS/MS (GC-MS/MS를 이용한 농산물 중 잔류농약 5종 동시시험법 개발 및 검증)

  • The aim of this research was to develop a rapid and easy multi-residue method for determining dimethipin, omethoate, dimethipin, chlorfenvinphos and azinphos-methyl in agricultural products (hulled rice, potato, soybean, mandarin and green pepper). Samples were prepared using QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) and analyzed using gas chromatography-tandem mass spectrometry (GC-MS/MS). Residual pesticides were extracted with 1% acetic acid in acetonitrile followed by addition of anhydrous magnesium sulfate (MgSO4) and anhydrous sodium acetate. The extracts were cleaned up using MgSO4, primary secondary amine (PSA) and octadecyl (C18). The linearity of the calibration curves, which waas excellent by matrix-matched standards, ranged from 0.005 mg/kg to 0.3 mg/kg and yielded the coefficients of determination (R2) ≥ 0.9934 for all analytes. Average recoveries spiked at three levels (0.01, 0.1, 0.5 mg/kg) and were in the range of 74.2-119.3%, while standard deviation values were less than 14.6%, which is below the Codex guideline (CODEX CAC/GL 40).

Monitoring of Polycyclic Aromatic Hydrocarbon Residues in Environmental Samples in Korea (국내 PAHs 오염 우려지역의 환경 시료 중 PAHs 잔류량 모니터링)

  • The aim of this study was to determine the residual amounts of PAHs in environmental samples such as crop, soil and water collected from paddy, upland fields and forestlands near industrial zone and/or a thermal power plant in South Korea. All of the samples were analyzed by GC-mass spectrometer. The average contents of total PAHs in soil samples were 140.2 ${\mu}g\;kg^{-1}$ and the range was from 4.3 to $662.9{\mu}g\;kg^{-1}$. The detection of benzo(a)anthracene, chrysene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)pyrene and dibenzo(a,h) anthracene which have strong carcinogenecity was ranged from 14.2 to 167.8 ${\mu}g\;kg^{-1}$. The residual amounts and detection frequency of PAHs in soil samples from the iron and heavy industrial areas near Pohang and Busan were 3-folds more than those of the other areas. Amounts of PAHs in upland soil samples was 1.5 folds higher than those of paddy soil samples, suggesting that it may be related to the content of organic matter in soil. The average contents of total PAHs in crop samples were 9.7 ${\mu}g\;kg^{-1}$ which ranged from 4.5 to 52.2 ${\mu}g\;kg^{-1}$. However, the residual amounts of PAHs in water samples were not detected. These results showed that soils and crops were slightly contaminated with PAHs. Therefore, the investigation should be continued for evaluating a safety or risk assessment through expansion of regions and crops.

Improvement of an Analytical Method for Fluoroimide Residue in Agricultural Products Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 Fluoroimide의 잔류농약 분석법 개선)

  • Fluoroimide is a fungicide and is also used as a pesticide for persimmons and potatoes. The established fluoroimide pesticide analysis method takes a long time to perform and uses benzene, a carcinogen. In addition, a lower limit of quantification is required due to enforcement of the Positive List System. Therefore, this study aimed to improve the analysis method for residual fluoroimide to resolve the problems associated with the current method. The analytical method was improved with reference to the increased stability of fluoroimide under acidic conditions. Fluoroimide was extracted under acidic conditions by hydrogen chloride (4 N) and acetic acid. MgSO4 and NaCl were used with acetonitrile. C18 (octadecylsilane) 500 mg and graphitized carbon black 40 mg were used in the purification process. The experiment was conducted with agricultural products (hulled rice, potato, soybean, mandarin, green pepper), and liquid chromatograph-tandem mass spectrometry was used for the instrumental analysis. Recovery of fluoroimide was 85.7-106.9% with relative standard deviations (RSDs) of less than 15.6%. This study reports an improved method for the analysis of fluoroimide that might contribute to safety by substituting the use of benzene, a harmful solvent. Furthermore, the use of QuEChERS increased the efficiency of the improved method. Finally, this research confirmed the precise limit of quantification and these results could be used to improve the analysis of other residual pesticides in agricultural products.

Residual Characteristics of Bistrifluron and Chlorantraniliprole in Strawberry (Fragaria ananassa Duch.) for Establishing Pre-Harvest Residue Limit (생산단계 잔류허용기준 설정을 위한 딸기 중 bistrifluron과 chlorantraniliprole의 잔류 특성 연구)

  • BACKGROUND: Pesticide residue analysis is essentially required for safety evaluation of agricultural products. Bistrifluron and chlorantraniliprole have been currently considered as potentials to deeply evaluate their residues in agricultural products because they are frequently found in strawberry. This work was performed to investigate the residual patterns of bistrifluron and chlorantraniliprole in strawberry after harvest. METHODS AND RESULTS: Strawberry was treated with bistrifluron and chlorantraniliprole 0, 1, 2, 3, 5, 7 and 10 days before harvest under greenhouse conditions. The strawberry samples were subjected to solvent and solid phase extractions followed by LC-MS/MS analysis. There covery percentages of bistrifluron and chlorantraniliprole for tified in the control samples ranged from approximately 82 to 103% with the method limit of 0.005 mg/kg. The concentrations of bistrifluron and chlorantraniliprole in strawberry samples decreased significantly in 10 days after treatment, giving the safety levels of 0.04 to 0.06 mg/kg at 10 days after application, as considered maximum residue limit. The half-lives of bistrifluron and chlorantraniliprole based on first order kinetics were determined to 6.3 days and 6.4 days, respectively. CONCLUSION: Bistrifluron and chlorantraniliprole are suggested to use in strawberry 10 days before harvest to reach residual safety levels.

Development and Validation of the Analytical Method for Oxytetracycline in Agricultural Products using QuEChERS and LC-MS/MS (QuEChERS법 및 LC-MS/MS를 이용한 농산물 중 Oxytetracycline의 잔류시험법 개발 및 검증)

  • An analytical method was developed for the determination of oxytetracycline in agricultural products using the QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) method by liquid chromatography-tandem mass spectrometry (LC-MS/MS). After the samples were extracted with methanol, the extracts were adjusted to pH 4 by formic acid and sodium chloride was added to remove water. Dispersive solid phase extraction (d-SPE) cleanup was carried out using $MgSO_4$ (anhydrous magnesium sulfate), PSA (primary secondary amine), $C_{18}$ (octadecyl) and GCB (graphitized carbon black). The analytes were quantified and confirmed with LC-MS/MS using ESI (electrospray ionization) in positive ion MRM (multiple reaction monitoring) mode. The matrix-matched calibration curves were constructed using six levels ($0.001{\sim}0.25{\mu}g/mL$) and coefficient of determination ($r^2$) was above 0.99. Recovery results at three concentrations (LOQ, $10{\times}LOQ$, and $50{\times}LOQ$, n=5) were from 80.0 to 108.2% with relative standard deviations (RSDs) less than of 11.4%. For inter-laboratory validation, the average recovery was in the range of 83.5~103.2% and the coefficient of variation (CV) was below 14.1%. All results satisfied the criteria ranges requested in the Codex guidelines (CAC/GL 40-1993, 2003) and the Food Safety Evaluation Department guidelines (2016). The proposed analytical method was accurate, effective and sensitive for oxytetracycline determination in agricultural commodities. This study could be useful for safety management of oxytetracycline residues in agricultural products.
