The Effect of Combined Estrogen/calcium Therapy on Bone Metabolism in Ovariectomized Rats II. A Study on Metabolism of calcium, Phosphorus and Nitrogen in Ovariectomized Rats (에스트로겐/칼슘 혼합요법이 난소절제 쥐의 골대사에 미치는 영향 II. 칼슘, 인 및 질소대사에 관한 연구)

  • This study was carried out to investigate the effects of dietary calcium-salt, estrogen-treatment, and estrogen/calcium treatment on calcium, phosphorus and nitrogen metabolism. Female Sprague-Dawley rats with a body weight of 250~280g were underwent ovariectomy or sham-operation. The ovariectomized rats were divided into 9 different experimental groups including the saline-treated group, the estrogen-treated group, the high calcium salt-treated group, and the estrogen/calcium treated groups and fed for 6 weeks. Each group daily intake and excretion of calcium, phosphorus and nitrogen were measured and apparent digestibility and balance were also studied. The results were as follows: The excretion level of winn calcium was significantly higher the ovariectomized rats than the sham-operation group(p<0.01) and reduced with estrogen treatment but this difference was not statistically significant. Fecal loss of calcium was higher the ovariectomized rats than the sham-operation group(p<0.001). When the estrogen was treated, fecal loss was decreased and then apparent digestibility of calcium was increased. Calcium balance was significantly higher the high calcium treated groups than the control diet groups. The excretion level of urinary Phosphorus was higher the ovariectomized rats than sham-operation group, while these showed to be decreased in the calcium salt, the estrogen and the estrogen/calcium treated groups(p<0.01). Fecal loss of phosphrous was higher in the ovariectomized rats. When the estrogen was treated, the fecal loss was decreased in the avariectomized rat than that of other groups. The excretion level of urinary nitrogen was higher the ovariectomized rats than the sham-operation, while these showed to be decreased in the estrogen, the estrogen/calcium, and the estrogen gradually reduction/calcium intensification. Fecal loss of nitrogen was decreased in tile estrogen treated group. The results in this study showed that high calcium, estrogen/calcium and estrogen gradually reduction/calcium intensification in the ovariectomized rats enhanced calcium balance compared to the ovariectomized rats without calcium treatment, but little effects on the phosphorus and nitrogen balance.

Anti-obesity Effects of Genistein in Female Ovariectomy-induced Obese Mice (난소절제로 비만이 유도된 암컷 쥐에서 제니스테인의 항비만 효과)

  • To investigate whether genistein regulates menopause-induced obesity, it was studied the effects of genistein on anti-obesity effects in female ovariectomized (OVX) mice, an animal model of postmenopausal women. 7-week-old female mice (C57BL/6J) were randomly divided into three groups. All the animals received a high fat diet or a high fat diet supplemented with genistein for 8 weeks and variables and determinants of obesity were measured. The OVX mice had significantly higher body weight and adipose tissue mass than sham mice. However, genistein supplementation reduced body weight, adipose tissue mass, and adipocyte size of OVX mice. The OVX mice treated with genistein had significantly lower levels of serum triglycerides and total cholesterol than the vehicle-treated OVX mice. Lipid accumulation in liver was also markedly decreased by genistein in OVX mice. The results suggest that genistein can effectively prevent adiposity, adipocyte phertrophy, and llipid disorders caused by ovariectomy. Moreover, this study may contribute to the alleviation of metabolic syndrome, including obesity and hyerlipidemia in postmenopausal women.

Effect of Ledebouriella seseloides Extracts on Lipid Parameters in Ovariectomized Rats (방풍나물 추출물이 난소절제 흰쥐의 혈중 지질 변화에 미치는 영향)

  • This study was investigated the improvement effects of Ledebouriella seseloides (LS) ethanol extracts on lipid parameters in an ovariectomized animal model. Sixty, nine-week old female Sprague Dawley rats were randomly assigned to four groups as follows: sham-operated rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with LS ethanol extracts (50 mg/kg/day and 200 mg/kg/day, respectively). The diets were fed to the rats for six weeks after their operation. The total-cholesterol and triglyceride contents on serum increased in the OVX-CON group compared to the SHAM group, but supplementation with the LS extract caused these factors to decrease. Notably, the serum LDL-cholesterol concentration in the supplemented 200 mg/kg/day LS ethanol extract group was significantly more reduced than the OVX-CON group. In addition, the platelet aggregation ability was lower in groups treated with LS than in the OVX-CON group. The alkaline phosphatase (ALP) activity was lower in the LS extract group compared to the OVX-CON group. Collagen content, in bone and cartilage, were reduced by ovariectomy, but the supplemented LS extract groups exhibited higher concentrations in their bones. According to these results, the improvement effects of LS extract on serum lipid parameters and osteogenesis in ovariectomized rats were illuminated.

The Effects of Ca and Mg Supplementation on Serum and Liver Lipid Parameters in Ovariectomized and Ca Deficiency Rats (난소절제 칼슘결핍 흰쥐에서 칼슘과 마그네슘의 보충이 혈액과 간의 지질함량에 미치는 영향)

  • This study was carried out to investigate the Ca and Mg supplementation on the serum and liver lipid parameters in ovariectomized and Ca deficiency rats. Total 50 Sprague Dawley female rats (6 weeks) were divided into 5 groups and bred for 12 weeks: sham operated control group (SNCa), OVX Ca deficiency poop (OLCa) with Ca deficiency diet (0.1% Ca modified AIN-93N diet), OVX Ca deficiency & Mg supplement group (OLCaMg), OVX adequate Ca group (OACa; 0.5% Ca AIN-93N diet) and OVX adequate Ca & Mg supplement group (OACaMg). There were no significant difference among the five groups in serum total cholesterol, triglyceride and HDL-cholesterol levels. LDL-cholesterol of OVX groups was significantly higher than that of SNCa group (p < 0.01). AI (Atherogenic index), TPH (Total cholesterol/HDL-C) and LPH (LDL-C/HDL-C) of OACa group were significantly lower than those of OLCa groups. OACaMg group had significantly lower levels LDL, AI and TPH than OLCa group. There was no significant difference in lever cholesterol level. However, liver total fat content of OACa was significantly lower than that of OLCa. From the above the results, it is concluded that the accumulation level of calcium shows how the supplement of magnesium affects hyperlipidemia. Therefore, in order to prevent women#s hyperlipidemia after menopause, and to keep healthy, it is encourage able to consider how the supplement of magnesium relates calcium intake.

Effects of Dietary Calcium and Soy Isoflavones Supplementation on Bone Metabolism in the Ovariectomized Rats (난소절제 흰쥐의 골격대사에 대한 식이 칼슘과 대두 이소플라본의 섭취효과)

  • This study was conducted to demonstrate whether dietary Ca and the soy isoflavones supplementation could reduce the bone loss associated with estrogen deficiency. Nine week-old female rats (SD) were ovariectomized and then fed on diet of low $(0.1\%)$ or normal $(0.5\%)$ Ca supplemented with soy isoflavones (80 and 160 ppm) for 6 weeks. The ovariectomized and sham-operated rats showed normal serum Ca and P levels, and dietary Ca and soy isoflavones did not changed them. The serum alkaline phosphatase activity was increased in all ovariectomized rats, especially in the rats fed low Ca diet regardless of isoflavone supplementation. The serum tartrate-resistance acid phosphatase was not significantly different among the ovariectomized rats and were not changed by dietary Ca and isoflavones. Breaking force of femur was higher in the rats fed the nomral Ca diets and not statistically changed by soy isoflavone supplementation. Femoral and lumbar Ca and P contents decreased in the ovariectomized rats and the soy isoflavones-80 ppm supplementation significantly enhanced bone minerals, but the soy isoflavones-160 ppm supplementation did not. Dietary Ca increased lumbar Ca and P contents. The results of this study have suggested that the soy isoflavones 80 ppm supplementation could be sufficient to prevent bone loss in ovariectomized rats and normal Ca supplementation could enhance the effect of soy isoflavones on bone protection.

The Effect of Ecklonia cava Extracts on Bone Turnover Markers in Ovariectomized Rats (갱년기 유도 흰쥐에서 감태 추출물이 골 대사 지표물질의 변화에 미치는 영향)

  • Menopause is often associated with the incidence of several chronic diseases including osteoporosis, cardiovascular disease, and obesity. Hormone replacement therapy (HRT) is an effective regimen that has been found to prevent these diseases in postmenopausal women. However, HRT is accompanied by an increased risk of unfavorable outcomes. Therefore, this study was conducted to evaluate the effects of Ecklonia cava, a kind of seaweed, extract on bone turnover markers in symptomatic menopausal women. For this study, the following four groups of 9-week-old Sprague-Dawley rats were evaluated over 6 weeks: normal rats (SHAM), ovariectomized rats (OVX-CON) and ovariectomized rats that were treated with Ecklonia cava extracts. The optimum extraction temperature and solvent of Ecklonia cava were found to be $80^{\circ}C$ and 80% ethanol. We measured the osteocalcin and CTx content, enzyme ALP activity in serum and collagen content in the cartilage, bone, skin and lungs. We found that the levels of indicators of bone metabolism such as ALP, osteocalcin and CTx were lower in rats in the Ecklonia cava extract group than the OVX-CON group. In addition, the collagen contents in the bone, cartilage, skin and lungs decreased in response to ovariectomy, but the levels of collagen were greater in the bone of rats that were treated with Ecklonia cava extract than in the bone of rats in the OVX-CON group. According to these results, we were able to know the effects of Ecklonia cava extract on bone aging in ovariectomized rats. Consequently, we expect Ecklonia cava extract to have an effect on bone aging in postmenopausal women.

The Effect of Sea Tangle Extract on Serum Lipid Level in Ovariectomized Rats (다시마 추출물이 난소를 절제한 흰 쥐의 혈 중 지질 함량에 미치는 영향)

  • This study was performed to investigate the effects of sea tangle (ST) extracts on serum lipid concentration in ovariectomized rats. Weight-matched female Sprague-Dawley strain rats were assigned to four groups. Three groups were surgically ovariectomized (OVX). The fourth group was sham operated. Rats were randomly assigned to the following groups: sham-operated rats (Sham), ovariectomized control rats (OVX-control), ovariectomized rats supplemented with ST at 50 mg/kg body wt (OVX-ST50) and ovariectomized rats supplemented with ST at 200 mg/kg body wt (OVX-ST200). The activities of serum GOT and GPT were increased by ovariectomized. The serum GOT levels in OVX-ST50 and OVX-ST200 groups were decreased than OVX-control group. But GPT level was only decreased in the serum of OVX-ST200 group. Serum total cholesterol and triglyceride contents decreased in sham group compared with OVX-control group by ovariectomy. Six weeks feeding of ST extract resulted in a significant lowering of serum triglyceride and lowering tendency of total cholesterol Level. The serum HDL-cholesterol was higher in ST groups than OVX-control group. These results suggest that the beneficial effect of ST extract may be used to possibly improve on the lipid metabolic syndrome of menopausal women.

Effects of Soy Isoflavone Intake on Urinary and Fecal Excretion of Daidzein and Genistein in Ovariectomized Rats (대두 이소플라본 섭취수준이 난소절제한 흰쥐에서 Daidzein과 Genistein의 뇨와 변 중 배설에 미치는 영향)

  • This study was performed to determine the effect of three different concentrations of soy-isoflavones on excretions through urine and feces in either sham-operated or ovariectomized female rats. Seventy-two 16-week old Sprague-Dawley rats underwent sham operation or bilateral ovariectomy. They were provided diets containing different levels of soy isoflavones for 6 weeks: 50 ppm (low isoflavone intake; LI), 250 ppm (medium isoflavone intake; MI) and 500 ppm (high isoflavone intake; HI). The subsequent fecal and urinary excretions of daidzein and genistein were then measured. In the sham operated rats, body weight gains and food efficiency ratio of the MI and HI groups were significantly lower than control group, while food intake was not different. However, there was no significant difference in ovariectomized rats. The fecal excretion of daidzein was significantly higher in the HI group than the LI and MI groups, and that of genistein increased as dietary isoflavone intakes increased in both the sham operation and ovariectomy. The reverse tendency of fecal recovery was shown with dietary isoflavone dose only in genistein among ovariectomized rats. When dietary isoflavones were increased from 50 ppm to 500 ppm, the amounts of daidzein and genistein in the urine increased dose-dependently. The higher intakes of isoflavones leaded to lower recovery rates of daidzein and genistein in the urine in the sham-operated rats but not in the ovariectomized rats. The urinary recovery was significantly higher in the LI group than the MI and HI groups in the sham-operated rats. The excreted amounts and recovery of the two isoflavones in the urine were higher in the sham operated groups than in the ovariectomized groups, which implied an increased bioavailability of isoflavones by ovariectomy. Therefore, the results suggest that a more efficient use of soy isoflavones in ovariectomized conditions may occur, which indicates that a higher dose of soy isoflavones is necessary for the postmenopausal states.

Effect of Isoflavone Supplementation on Bone Metabolism in Ovariectomized Rats at Different Ages (이소플라본 보충이 다른 나이에 난소를 절제한 흰쥐의 골대사에 미치는 영향)

  • Osteoporosis that is associated with estrogen deficiency in menopause is by far the most common cause of age-related bone loss. Since isoflavone had been reported as a natural substance that minimizes bone loss, we have begun this study to examine the effect of the substance on bone metabolism in ovariectomized rats. Five week-old (n=22) and 25 week-old (n=22) Sprage-Dawley female rats were classified into young (Y) and adult (A) groups. Each group consisted of three subgroups : sham operated group (SH), ovariectomized group (OVX), and isoflavone supplemented group (OVX+ISO 80 mg/kg B.W.). They were fed chow for 9 weeks. The result showed that body weight gain was increased in YOVX in comparison to YSH group, (p<0.05) serum osteocalcin concentration and urinal deoxypyridinoline (DPD) excretion had significantly increased in YOVX more than in YSH group, and significantly decrease in OVX+ISO than in YOVX group (p<0.05). We concluded that soy isoflavones may decrease bone turnover in young rats. However, isoflavone supplement didn't show significant influence on bone metabolism of adult rats.

Preventive Effects of Drynariae Rhizoma on Experimental Bone Disorder in Rats (실험적으로 유발된 골손실에 미치는 골쇄보의 예방 효과)

  • 흰쥐의 난소 절제로 야기된 골손실에 미치는 골쇄보의 영향을 관찰하였다. 난소 절제 후 흰쥐의 체중은 증가하고 자궁의 무게는 감소하였는데, 골쇄보는 특별한 영향을 주지 않았다. 그러나, 난소 절제 후 흰쥐의 tibia에서 측정한 골밀도는 대조군에 비하여 22% 감소되었으나, 골쇄보와 17be1a-estradiol를 투여한 경우는 골밀도의 감소가 현저히 억제되었다. 난소를 절제한 실험군에 골쇄보와 17be1a-estradiol를 투여하고 전자 현미경으로 관찰한 결과, trabecular bone의 미세분자 표면이 보호되는 것으로 관찰되었다. 이러한 결과들을 볼 때, 골쇄보가 17be1a-estradiol 만큼 난소 절제술 후 진행되는 골손실을 예방하는데 효과적임을 강하게 나타내고 있다고 사료된다.
