Fabrication of the poly (methyl methacrylate)/clay (modified with fluorinated surfactant) nanocomposites using supercritical fluid process (초임계 공정을 이용한 poly(methyl methacrylate)/클레이 나노복합체 제조)

  • The supercritical fluids (SCFs) have been widely used for material synthesis and processing due to their remarkable properties including low viscosity, high diffusivity and low surface tension. Carbon dioxide is one of the suitable solvents in SCFs processes in terms of its advantages such as easy processibility (with low critical temperature and pressure), inexpensive, nonflammable, nontoxic, and readily available. However, it has generally low solubility for high molecular weight polymers with the exception of fluoropolymers and siloxane polymers. Therefore, hydrocarbon solvents and hydrochlorofluorocarbons have been used for various SCFs process by its high solubility for high molecular weight polymers. In this report, a PMMA/clay nanocomposites were fabricated by using supercritical fluid process. The $Na^+$-MMT(montmorillonites)was modified by a fluorinated surfactant which is able to enhance compatibility with the chlorodifluoromethane(HCFC-22) and thus, improve dispersability of the clay in the polymer matrix. The PMMA/fluorinated surfactant modified clay nanocomposite shows enhanced mechanical and thermal properties which characterized by X-raydiffraction(XRD), Thermo gravimetric analysis(TGA), Dynamic mechanical analysis (DMA) and Transmission electron microscopy (TEM).

  • Alkaline fuel cells using liquid fuels such as hydrazine and ammonia are gaining great attention as a clean and renewable energy solution possibly owing to advantages such as excellent energy density, simple structure, compact size in fuel container, and ease of storage and transportation. However, common shortcomings including cathode flooding, fuel crossover, side yield reactions, and fuel security and toxicity are still challenging issues. Real time monitoring of fuel concentrations integrated into a fuel cell device can help improving fuel cell performance via predicting any loss of fuels used at a cathode for efficient energy production. There have been extensive research efforts made on developing real-time sensing platforms for hydrazine and ammonia. Among these, recent advancements in electrochemical sensors offering high sensitivity and selectivity, easy fabrication, and fast monitoring capability for analysis of hydrazine and ammonia concentrations will be introduced. In particular, research trend on the integration of metallic and metal oxide nanoparticles and also their hybrids with carbon-based nanomaterials into electrochemical sensing platforms for improvement in sensitivity and selectivity will be highlighted.

Pre-validation of Colony Forming Efficiency Assay for Assessing the Cytotoxicity of Nanomaterials (나노물질의 세포독성 평가법으로 Colony Forming Efficiency Assay에 대한 검증연구)

  • Objectives: The cytotoxcities of Au, Ag, SWCNT, $SiO_2$, and ZnO nanomaterials were evaluated in order to assess their potential toxicological effects in in vitro cell models using colony forming efficiency (CFE) assay. Methods: The CFE assay of the test materials was carried out on Hep G2 cells. The size distribution of nanomaterials was studied by transmission electron microscopy (TEM). Changes in cell viability after treatment with a toxicant will result in a decreased number of colonies formed in comparison to solvent. Results: The TEM images show that all the particles except SWCNT and ZnO can be considered approximately spherical. The gold and $SiO_2$ nanoparticles show no response (no toxicity) in concentration response experiments. A statistically significant toxic effect was found in Hep G2 cells treated with Ag, SWCNT and ZnO nanomaterials. Conclusion: In this study, we considered CFE assay to be a promising test for screening studies for cytotoxicity with physicochemical analysis.

Evaluating Cadmium Ion Removal in Aqueous Solutions and Cytotoxicity Evaluation of Carbon, Synthesized Layered Double Hydroxide, and Multi-wall Carbon Nanotube (활성탄, 합성 층상이중층 수산화물, 카본나노튜브를 이용한 수용액상의 카드뮴의 제거와 흡착제 독성 연구)

  • This experiment was carried out with the purpose of testing cadmium adsorption abilities of multi wall carbon nanotube (MWCNT), activated carbon, and synthesized layered double hydroxide (LDH). In the acidic condition, only MWCNT was effective for removing cadmium ion in the aqueous phase while other adsorbents rarely removed it. The MWCNT and cadmium ion adsorption reactions followed pseudo-first order kinetic. When the initial pH value was neutral (pH = 7), cadmium ion was rapidly removed by MWCNT and activated carbon in 4 hr (100% and 99.2%, respectively). Increasing adsorbent dosages affects the pH evolution and cadmium ion removal (0 to 99%). Cytotoxicity test showed that both MWCNT and LDH has low cytotoxic effects on three kind of human cells (A549, HS-294t, and HT-29).

Application of sickle red blood cells for targeted cancer therapy (항암치료를 위한 겸형적혈구의 응용)

  • Conventional drug carriers such as liposomes, nanoparticles, polymer micelles, polymeric conjugate and lipid microemulsion for cancer chemotherapy shield normal tissues from toxic drugs to treat cancer cells in tumors. However, inaccurate tumor targeting uncontrolled drug release from the carriers and unwanted accumulation in healthy sites can limit treatment efficacy with current conventional drug carriers with insufficient concentrations of drugs in the tumors and unexpected side effects as a result. In this research, we examined the use of sickle red blood cells as a new drug carrier with novel tumor targeting and controlled release properties. Sickle red blood cells show natural tumor preferential accumulation without any manipulation and controlled drug release is possible using a hemolysis method with photosensitizers.

Public and Experts Perception about Nanotechnology Hazards in Korea (대한민국에서의 나노물질 위해성에 관한 일반인/전문가 인식 설문조사)

  • Public perceptions of nanotechnology and its potential risk can be an important measure for the sustainable advances of the technology. We conducted a survey for public (N =599) and experts (N = 165). They answered the same questions and results were analyzed. 74% of the public have not heard about the potential risk of nanotechnology and 77% expected that nanotechnology/materials are not harmful to the human and nature. 74% of experts realized the potential hazards by nanomaterials. The results represented that large perception gab between public and experts has existed in Korea. Interestingly, Korean public thought that nanotechnology is more profitable and less risky than that of American. We suggest that 'now is the best time to give appropriate information on the potential risk of nanotechnology to Korean public without preconception or exaggeration.'

  • 자연계의 많은 생물들은 의사소통이나 분위기 표현, 위장 등을 하기위해 자신의 색을 바꾸는 것으로 알려져 있으며, 현재 많은 연구자들이 이러한 자연 현상으로부터 영감을 받아 생체모방 구조와 메커니즘을 이용한 바이오센서를 개발하고 있다. 하지만 기존의 컬러센서는 수용체 개발에 있어 복잡한 디자인, 어려운 합성 방법 및 낮은 감도와 저선택성 등의 한계점을 가지고 있다. 이에 본 연구에서 우리는 바이러스(M13-박테리오파지)를 기반으로 한 신개념 고감도 고선택성 컬러센서를 개발하고자 한다. 우리가 개발하고자 하는 컬러센서는 자가 조립방법으로 만들어진 나노 구조체로 형성되어 있으며, 다양한 종류의 화학물질이나 오염물질을 감지할 수 있다. 이 컬러센서는 아주 낮은 농도의 휘발성 유기화합물(volatile organic compounds)을 감지해 색변화를 보였으며, 다양한 독성 물질이나 방향족을 가진 화학 물질을 감지할 수 있었다. 따라서 우리가 개발한 컬러 센서는 국가의 안보나 국민의 건강을 증진시키기에 아주 유용할 것으로 보인다.

  • Existing organic UV protection materials seem to be problematic due to their penetration and irritation to skin. Inorganic UV protection materials are also at issue for safety of their nano-type transformation. Therefore, the recent studies of UV protection materials have been focused not only on the effectiveness but also on their safety. One of the UV protection materials in study which have higher safety is the organic-inorganic conjugation type UV protection material. Previously, we have reported the manufacturing process, physical property and UV protection efficiency of methoxychinnamidoprophy poloysilsesquixan as a new cross-linked polymer type UV protection material. In this study, we have evaluated the effect of the methoxychinnamidoprophy poloysilsesquixan on embryo-fetal development in SD rats. This study is expected to show some definite information related to the effect on pregnancy or embryo-fetal abnormality in case of the clinical exposure of the methoxychinnamidoprophy poloysilsesquixan.

Polymeric Micelle Using Poly((R)-3-hydroxybutyric acid)/Poly(ethylene glycol) Amphiphilic Block Copolymer for Drug Delivery System (Poly((R)-3-hydroxybutyric acid)/Poly(ethylene glycol) 양친성 블록 공중합체를 이용한 약물전달체용 고분자 미셀)

  • A biodegradable polymer poly((R) -3-hydroxybutyric acid) (PHB) was conjugated with a hydrophilic polymer poly(ethylene glycol) (PEG) by the ttansesterification reaction to form the amphiphilic block copolymer. PHB with low molecular weight ($3000{\sim}30000$) was appropriated for the drug delivery materials. High molecular weight PHB was hydrolyzed by an acid-catalyst to produce the low molecular weight one. Amphiphilic block copolymer was formed the self-assembled polymeric micelle system in the aqueous solution that the hydrophillic PEG was wraped the hydrophobic PHB. Generally, polymeric micelle forms the small particle between $10{\sim}200nm$. These polymeric micelle systems have been widely used for the drug delivery systems because they were biodegradable, biocompatible, non-toxic and patient compliant. The hydroxyl group of PEG was substituted with carboxyl group which has the reactivity to the ester group of PHB. Amphiphilic block copolymer was conjugated between PHB, and modified PEG at $176^{\circ}C$ which was higher than the melting point of PHB. Transesterification reaction was verified with DSC, FTIR, $^1H-NMR$. In the aqueous solution, critical micelle concentration (CMC) of the mPEG-co-PHB copolymer measured by the fluororescence scanning spectrometer was $5{\times}10^{-5}g/L$. The shape and size of the nanoparticle was taken by dynamic light scattering and atomic force microscopy. The size of the nanoparticle was about 130 nm and the shape was spherical. Our polymeric micelle system can be used as the passive targeting drug delivery system.

Synthesis and Characterization of Porous Poly(ε-caprolactone)/Silica Nanocomposites (다공성 폴리(ε-카프로락톤)/실리카 복합체의 제조 및 특성평가)

  • Poly(${\varepsilon}$-caprolactone) (PCL) nanofibers and PCL/silica membranes were synthesized by sol-gel derived electrospinning and casting, respectively. Smooth PCL nanofibers were obtained from the precursor containing N,N-dimethylformamide (DMF). PCL/silica membranes were prepared by varying the tetraethyl orthosilicate (TEOS) contents from 0 to 40 vol% to investigate the effect of silica addition on mechanical properties and cytotoxicity of the membranes. Although the strength of the membranes decreased from 12 to 8 MPa with increasing the silica content, the strength remained almost constant 7 weeks after dipping in phosphate buffered saline solution (PBS). The strength reduction was attributed to the presence of a patterned surface pores and micro-pores present in the walls between pores. The crystal structure of the membranes was orthorhombic and the crystallite size decreased from 57 to 18 nm with increasing the silica content. From the agar overlay test, the PCL/silica membranes exhibited neither deformation and discoloration nor lysis of L-929 fibroblast cells.
