행인(杏仁)과 길경(桔梗)이 Asthma model 내(內)의 Cytokine IL-4, IL-5, IL-6에 미치는 영향(影響) (The Effects of Armeniacae Arnarum Semen and Platycodi Radix on IL-4, IL-5, IL-6 in Asthma Model)

  • Background : Nowadays asthma is considered to be the inflammatory disease characterized by airway hyperresponsiveness and pulmonary eosinophilia, and mediated by Th lymphocytes expressing the Th2 cytokine pattern. In many recent studies, molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of asthma. Objective : We aimed to identify the effect of Armeniacae Arnarum Semen and Platycodi Radix on the transcriptional activities of cytokine IL-4, IL-5 and IL-6 involved in asthma model. Materials and Methods : RBL-2H3 cell lines were used. Cells were stimulated with calcium inophore for maximal gene expression. After 24 hours of Armeniacae Arnarum Semen and Platycodi Radix-treatment, total cellular RNAs were collected using Trizol solution method. Then transcriptional activities of IL-4, IL-S and IL-6 were measured by RT-PCR with electrophoresis. Results : In IL-4 study, Armeniacae Arnarum Semen treated group showed 48.4% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 45.4% of transcriptional activities compared to the control group. In IL-5 study, Armeniacae Arnarum Semen treated group showed 52.7%of transcriptional activities compared to the control group and Platycodi Radix treated group showed 60.2% of transcriptional activities compared to the control group. In IL-6 study, Armeniacae Amarum Semen treated group showed 42.3% of transcriptional activities compared to the control group and Platycodi Radix treated group showed 69.1% of transcriptional activities compared to the control group. Conclusion : This study shows that Armeniacae Arnarum Semen and Platycodi Radix have the inhibitory effect on the transcription of IL-4, IL-5 and IL-6 gene expression in RBL-2H3 cell lines. Advanced studies are required to investigate the mechanisms of inhibition by herbal medicine in asthma model.

태음인(太陰人) 이열병(裡熱病)의 병증(病證) 약리(藥理)에 대한 연구(硏究) (A Study on the medical and pharmacological theory of Interior-Overheated-Disease of Taeumin)

  • 1. 연구배경 및 목적 이제마가 태음인(太陰人) 이열병(裡熱病)을 인식하고 태음인(太陰人) 처방(處方)을 계발한 과정을 파악함으로써 태음인의 병증(病證) 약리(藥理)에 대한 이해를 높이고자 한다. 2. 방 법 동의수세보원(東醫壽世保元) 병증론(病證論)에 나타난 인용문과 처방의 출전을 시대별로 고찰함으로써 병증(病證)과 약리(藥理)에 대한 인식의 변화를 관찰하고 분석하였다. 3. 결 과 이제마(李濟馬) 이전에는 태음인(太陰人) 간열증(肝熱證)이 소양인(少陽人) 위열증(胃熱證)과 다르다는 것은 알았으나, 병리에 있어서는 둘을 구분하지 못하고 똑같이 열결(熱結)로만 이해하였고, 그러면서도 처방은 어느 정도 서로 다르게 쓰여지는 경향이 있었다. 문헌 고찰 결과 위열이 상하로 치성(熾盛)하여 번조(煩燥), 두통(頭痛) 소편불이(小便不利), 편비(便秘)를 거의 대부분 동반하는 소양인 위열증과 '호산지기(呼散之氣) 부족흡취지기(不足吸聚之氣) 태과(太過)로 인해 내부에 조와 열(熱)이 발생함으로써 목동(目疼), 비건(鼻乾), 인통(咽痛), 음일수이 등의 특징적 증상을 보이는 태음인 간열증을 구분할 수 있었다. 한편 약재에 있어서도 갈근, 승마(升麻), 고본(藁本), 백지(白芷), 대황(大黃)은 태음인 간열증의 치방에 군약으로 쓰이는 점에서, 황금(黃芩)과 길경(桔梗)은 폐에 작용하는 약임이 분명하므로, 그리고 라복자(蘿卜子)와 급각은 각각 치담약(治痰藥)과 개열락(開悅樂)이면서 천식(喘息), 해수(咳嗽) 등 폐병증을 함께 치료하는 점에서 각각 태음인 약으로 알 수 있었다.

길경 수용액 추출물에 의한 인체 폐암세포의 성장억제 기전 연구 (Cell Cycle Arrest of Human Lung Carcinoma A549 Cells by an Aqueous Extract from the Roots of Platycodon grandiflorum)

  • Platycodi Radix, the root of Platycodon grandiflorum, commonly known as Doraji, is used as a traditional oriental medicine. Extracts from the roots of P. grandiflorum have been reported to have wide ranging health benefits. We investigated the effects of an aqueous extract from the roots of P. grandiflorum (AEPG) on the cell proliferation of human lung carcinoma A549 cells in order to understand its anti-proliferative mechanism. AEPG treatment resulted in the inhibition of cell proliferation in a concentration-dependent manner. This anti-proliferative effect of A549 cells by AEPG treatment was associated with morphological changes such as membrane shrinking, cell rounding up and inhibition of cell migration. DNA flow cytometric histograms showed that populations of both Sand G2/M phase of the cell cycle were increased by AEPG treatment in a concentration-dependent manner. AEPG treatment induced a marked accumulation of tumor suppressor p53 and a concomitant induction of cyclin-dependent kinase (Cdk) inhibitor p21 and p27. In addition, SSS treatment resulted in down-regulation of Cdk2 and Cdk4 expression. The present results indicated that AEPG-induced inhibition of lung cancer cell proliferation is associated with the blockage of S to G2/M phase progression the induction of apoptosis. Taken together, these findings suggest that P. grandiflorum has strong potential for development as an agent for prevention against human lung cancer.

길경 투여가 고지방, 고탄수화물 식이로 유발된 비만형 제2형 당뇨병 동물모델에 미치는 영향 (The Effects of Platycodi Radix on Obese Type 2 Diabetes Mouse Model Induced by High Fat, High Carbohydrate Diet)

  • Objectives: This study was designed to investigate the anti-obesity, anti-diabetic and anti-inflammatory effects of Platycodi radix on obese type 2 diabetes mouse model. Methods: Obese type 2 diabetes mouse model was induced by Surwit's high fat, high sucrose diet for 8 weeks. Models were divided into 4 groups of normal diet (ND, n=10), high fat and high sucrose diet (HFD, n=10), high fat and high sucrose diet with Platycodi radix (PR, n=10), and high fat and high sucrose diet with Metformin (Met, n=10). Body weights were measured every week. After 7 weeks fasting, blood sugar and oral glucose tolerance tests were conducted. After 8 weeks blood samples were taken from mouse hearts and analyzed biochemically. Lipid profile, fructosamine, leptin and weight of epididymal fat pad and liver were measured. Adipose tissue macrophage percentage was analyzed by fluorescence-activated cell sorting (FACS). Results: Compared with the HFD group, body weight, glucose level, fructosamine, weight of epididymal fat pad and adipose tissue macrophage percentage decreased in the PR group. Conclusions: These results suggest that Platycodi Radix has anti-obesity, anti-diabetic, and anti-inflammatory effects on obese type 2 diabetes mouse model.

  • This study was conducted to investigate the contents of dietary fiber (DF), compositions of fatty acids in lipid fraction and amino acids in salt-soluble protein from the functional food source such as Platycodi radix, perilla seed, evening primrose seed and aloe vera. The contents of dietary fiber, neutral detergent fiber (N.D.F) , acid detergent fiber( A.D.F) . kignin, hemicellulose an dcellulose in evening primorse seed were higher than those of other samples, except the content of cellulose .The ration of polyunsaturated /saturated (P/S) fatty acid in total lipids was 6.31 in perilla seed, which was higher than those of other samples. The content of linolenic acid (n-3) in perilla seed was 55.47%. The content of linoleic acid (n-6) in evening primrose seed was 71.88% , which was higher than those of other samples. The fatty acid composition in neutral lipids were the same as those of total lipids. The PUFA contents of fatty acid in glycolipids were 61.76% in perilla seed. And also, the ratio of n-6/n-3 in evening primrose seed was 15.19. The fatty acid compositions in phospholipids were the sameas those of glycolipids. The contents of PUFA in Platycodi radix were 62.96% . The essential amino acid contents of salt-soluble protein were 47 mole % in Platycodi radix , which was slightly higher than those other samples. The ration of essential amino acid /nonessential amino acid (E/N) was 0.9 and 0.66 in Platycodi radix and aloe vera, respectively.

길경 재배년수, 수확시기 및 건조방법에 따른 조사포닌 함량 (Crude Saponin Contents of Platycodon grandiflorum(Jacq.) A.DC.)

  • 길경의 품질평가 기준으로 활용하고 있는 crude saponin 함량 변화를 밝히고 고품질 약재생산 및 가공을 위한 기초자료를 얻고자 생육년수, 부위, 수확시기, 및 건조방법별로 조사포닌 함량을 분석한 결과는 다음과 같다. Crude saponin 함량은 생육년수가 증가함에 따라 점점 감소하였다. 부위별로는 미부에서 2.74%로 두부 1.65%보다 1.6배 높았으며, 외피가 내심보다 1.8배 높았다. 수확시기별로는 3월과 12월에 수확했을 때가 각각 2.82, 2.74%이었으며 6월과 9월은 각각 2.12, 2.09%이었다. 건조방법별로는 증건했을 때 가 1.65%로 가장 낮았고 양건, 음건, 열풍건 및 원적외선 건조에서는 차이가 없었다.

길경(桔經)의 적응면역계(適應免疫界) 증강(增强) 효과(效果) (Enhancing the Immunogenicity of Platycodon Grandiflorum on Adaptive Immune System)

  • Purpose: This study was designed to investigate enhancing the immunogenicity effects of Platycodon grandiflorum(PG) on adaptive immune system. Methods: To investigate the effect of PG as an adjuvant, we used the ovalbumin (OVA) as an antigen at first. The proliferation of lymphocytes, the antibody titer, the subisotypes of antibodies and the production of cytokines were measured. Results: The proliferation of lymphocytes and the antibody titer were increased after PG treatment. The increased subisotypes of antibodies were IgG2 and IgG3 induced from T1-helper cells. However IgE induced from T2-helper cells was decreased. The production of cytokines derived from T1-helper cells was increased but that from T2-helper cells was decreased. Conclusion: It is supposed that PG has an immunogenicity effect as an adjuvant on adaptive immune system.

HPLC-ELSD법에 의한 길경의 platycodin D 정량분석 (Quantitative Analysis of Platycodin D from Platycodon grandiflorum by HPLC-ELSD)

  • 길경의 부탄올 분획으로부터 지표성분을 분리, 정제하기 위해 $SiO_2$ 컬럼크마토그래피를 실시한 결과 1종(화합물 1)의 단일 물질을 순수 분리하였으며 화합물 1의 구조는 $^1H$, $^{13}C$ NMR 등 분광학적 관측 결과 platycodin D로 확인되었다. Platycodin D의 HPLC 정량법을 검토한 결과, 검출기는 증기화광산란검출기(ELSD)를, 컬럼은 역상컬럼을 사용하여 길경 MeOH 추출액으로부터 효율적인 지표성분 정량이 가능하였다. 추출조건을 재검토한 결과 환류추출보다는 초음파 추출이 더 효율적이었으며 추출용매로는 80% MeOH이 다른 용매조건보다 추출효율이 우수하였다. 건조조건의 검토에서는 platycodin D의 함량이 양건이나 양건과 건조기 혼용건조 보다 건조기 단독 건조시 더 높았으며 특히 $60^{\circ}C$ 열풍건조에서 그함량이 0.083%로서 가장 높았다. 묽은 에탄올 엑스 함량도 건조기만을 이용한 열풍건조가 양건과 건조기 열풍건조를 혼용한 방법보다 4% 정도 더 높았다.

길경탕의 치밀결합 강화 및 MMPs의 활성 억제를 통한 인체방광암세포의 이동성 및 침윤성의 억제 (Gilgyung-tang Inhibits the Migration and Invasion of Human Bladder Cancer 5637 Cells through the Tightening of Tight Junctions and Inhibition of Matrix Metalloproteinase Activity)

  • Objectives: Gilgyung-tang (GGT) has been used as one of the main multi-herb formulas to treat “Peo-ong” (lung abscess). In this study, we investigated the inhibitory effects of water extracts of GGT on cell migration and invasion, two critical cellular processes that are often deregulated during metastasis, in human bladder cancer 5637 cells.Methods: Effects on cell viability were quantified using an MTT assay. To analyze the anti-metastatic effects, we conducted a wound healing migration assay, an in vitro invasiveness assay, and a measurement of the transepithelial electrical resistance (TER). The expression of protein and mRNA were measured by Western blotting and real-time polymerase chain reaction (RT-PCR), respectively.Results: GGT markedly inhibited the cell motility and invasiveness of 5637 cells within the concentration range that was not cytotoxic. The inhibitory effects of GGT on cell invasiveness were associated with tightening of the tight junctions (TJs), which was demonstrated by an increase in the TER. The RT-PCR and Western blotting results indicated that GGT decreased the levels of claudin proteins. GGT also inhibited the activity and expression of matrix metalloproteinase (MMP)-2 and -9 and simultaneously increased the levels of tissue inhibitor of metalloproteinase-1 and -2.Conclusions: Our findings suggest that GGT reduces both the migration and the invasion of 5637 cells by modulating the activity of TJs and MMPs.

길경배지 유산균이 마우스 대식세포의 NO 및 TNF-${\alpha}$에 미치는 영향 (Effects of Lactobacillus Plantarum Cultured in Platycodi Radix Decoction on the Eexpression of NO and TNF-${\alpha}$ in Mouse Macrophage RAW264.7 Cell Line)

  • This study examined the effects of Lactobacillus plantarum(LP) cultured in Platycodi Radix decoction(LPPR) on the expressions of NO and TNF-${\alpha}$ in mouse macrophage RAW264.7 cell line. Cells were stimulated with LP or LPPR (0.1, 1, and 10 bacteria/cell) in all assays. More NO was induced in LPPR than LPM at 0.1 and 1 of a LP: cell ratio. The iNOS mRNA expression was also enhanced in LPPR stimulated cells. TNF-${\alpha}$ was increased in LPPR stimulated cells at the protein and mRNA level compared with LPM. In conclusion, LP fermented in Platycodi Radix decoction induced stronger activity in NO and TNF in mouse macrophages than LPM. These results suggest that fermentation by Platycodi Radix can be useful in enhancing the immunostimulatory activity of LP.
