Influence of Temperature, Salinity and Hypoxia on Survival and Metabolic Rate in the Ark Shell, Scapharca broughtonii (피조개의 생존과 대사에 미치는 수온, 염분 및 저산소의 영향)

  • The ark shell, Scapharca broughtonii were collected from the Jinhae bay in Kyungnam, Korea from April 2006 to October 2007. To assess how much they have tolerance against hypoxia under the compound condition of temperature and salinity, survivorship, oxygen consumption rate and external features of Scapharca broughtonii were measured, The 7 days-$LC_{50}$ of dissolved oxygen (DO) for Scapharca broughtonii at $15^{\circ}C$-ordinary sea water and $26{\pm}1$ psu was 1.98mg/L (confidence limit: 1,45-2.39 mg/L) and 2.63mg/L (confidence limit: 1.76-3.37mg/L) respectively. Whereas 3 days-$LC_{50}$ at $25^{\circ}C$-ordinary sea water was 3.89mg/L (confidence limit: 3.36-4.55 mg/L) and 4 days-$LC_{50}$ at $25^{\circ}C-26{\pm}1$ psu was 3.74mg/L (confidence limit: 3.19-4,43 mg/L). Oxygen consumption rate with each experimental group was decreased during the period exposed to hypoxia. To recovery, they were replaced to ordinary and oxygen consumption rate was increased a little, but all died, The color of the umbo of shell in Scapharca broughtonii was changed dark during hypoxia. These data will provide important fundamental information for examining the causes of mass mortality of shellfish in the summer.

  • Streptomycetes is a group of Gram-positive soil bacteria that growas a branching vegetative mycelium leading to the formation of spores, and display a physiological differenti-ation related to the synthesis of many secondary metabolites including antibiotics. Their complex life cycle and multicellular differentiation require various levels of regulation and types of signal transduction systems including eukaryotic-type serine/threonine protein kinases and prokaryotic-type histidine/aspartic acid protein kinases. Akt kinase that was found in cells is a sorine/threonine kinase controlling signal pathway for multi-tude of important cellular events. The activation or inactivation of Akt kinase in the cell is one of the critical regulatory points to deliver cell proliferation, differentiation, survival or apoptosis signal. To find the regula-tory protein homologous to Akt in Streptomyces, the fluorescien-labeled synthetic peptide (FITC-TRRSR-TESIT) was designed from the consensus sequence of target proteins for Akt kinase. From the difference of the mobility between the nonphosphorylated and phosphorylated synthetic peptides on Agarose gel electro-phoresis, the Akt-phosphorylating activity was monitored. The cell-free extract prepared from Streptomyces griseus IFO 13350 and the Akt homologous protein was purified by ammonium sulfate fractionation and many steps of column chromatographies such as, DEAE-Sepharose, Mono Q, Resource Phenyl-Soporose and Gel permeation column chromatographies. As a result, the protein phosphorylating the fluorescien-labeled Akt substrate was identified and it's molecular weight was estimated as 39 kDa on SDS-PAGE.

Study on the Relationship between Skin Dryness and Amino Acids in Stratum Corneum (아미노산 동시분석을 통한 피부보습능과 각질 중 아미노산 함량과의 상관관계 연구)

  • Natural moisturizing factors (NMFs) are hydrophilic and water-soluble components in stratum corneum of the skin. NMFs absorb water from outer environment and enhance the water-holding capacity of stratum corneum and thereby, prevent the dryness and increase flexibility and plasticity of skin. NMFs are mainly composed of amino acids and their metabolites that are produced from the degradation of filaggrin. Here we established a simultaneous quantification method for 22 amino acids in tape-stripped stratum corneum samples using UPLC-PDA. With this method, we analyzed amino acid contents from tape-stripped stratum corneum samples of forearm and forehead regions from 15 healthy volunteers. Amino acid contents of inner (or upper) region were higher than outer (or lower) region of stratum corneum. Amino acid contents of stratum corneum of forearm were higher by 1.5 fold than forehead region. Of note, total amino acid contents were significantly and inversely correlated with trans-epidermal water loss (TEWL), an index for skin barrier function. Especially, Ser, Glu, Gly, Ala and Thr were determined to positively affect skin mositurizing activities. In conclusion, we could demonstrate that amino acid contents of stratum corneum are closely linked with skin barrier and moisturizing function, providing an important and fundamental methodology for the study of amino acids in skin physiology.

Construction and Analysis of a DNA Microarray for the Screening of Biosynthetic Genes of Secondary-Metabolites formation in Streptomyces (방선균 유래 이차대사 생합성 유전자 분석용 DNA Microarray 제작 및 해석)

  • Streptomyces produces many kinds of secondary-metabolites including antibiotics. Screening of a new compound and elucidation of a biosynthetic pathway for the secondary metabolites are very important fields of biology, however, there is a main problem that most of the identified compounds are already researched compounds. To solve these problems, a microarray system that is based on the data related to the biosynthetic genes for secondary-metabolites was designed. For the main contents of DNA microarray, the important genes for the bio-synthesis of aminoglycosides, polyenes group, enediyne group, alpha-glucosidase inhibitors, glycopeptide group, and orthosomycin group were chosen. A DNA microarray with 69 genes that were involved in the bio-synthesis for the antibiotics mentioned above was prepared. The usability of the DNA microarray was confirmed with the chromosomal DNA and total RNA extracted from S. coelicolor whose genomic sequence had already been reported.

Assessment of Basal Energy Expenditure in Normal Healthy Volunteers and Patients Receiving TPN by Bioelectrical Impedance Analysis (Bioelectrical Impedance Analysis를 통한 건강한 성인과 TPN환자의 기초대사량 산출)

  • Adequate nutrition is important in maintaining optimal health. Malnutrition can expose individual to increased risks of morbidity and mortality. The purposes of this study were to determine the basal energy expenditure (BEE) of Korean healthy subjects and TPN patients using Bioelectrical Impedance Analysis (BIA) method and to compare these values with those predicted by Harris-Benedict equation (H-B). BEE values measured by BIA were compared with predicted BEE values by the H-B formula in 59 clinically stable TPN patients and 65 healthy volunteers. In healthy volunteers and TPN patients, statistically significant differences were not shown between the BEE values measured by BIA (1392.5 Kcal and 1325.9 Kcal) and those predicted by H-B formula (1384.1 Kcal and 1270.1 Kcal). In male volunteers, statistically significant differences were not shown between BEE values measured by BIA (1670.7 Kcal) and the H-B formula (1550.9 Kcal), but in female volunteers, statistically significant differences were shown between BEE values measured by BIA (1194.8 Kcal) and the H-B formula (1265.6 Kcal). In male TPN patients, statistically significant differences were shown between BEE values measured by BIA (1453.5 Kcal) and the H-B formula (1335.9 Kcal), but in female TPN patients, statistically significant differences were not shown between BEE values measured by BIA (1126.4 Kcal) and the H-B formula (1167.2 Kcal). In normal healthy volunteers, $90.8\%$ of BEE values measured by BIA and in TPN patients $89.8\%$ of BEE values measured by BIA were within $15\%$ of BEE values predicted by the H-B formula in non-obese subjects. In conclusion, BEE values predicted by H-B formula or measured by BIA can be applied to non-obese Koreans. However, these values should be confirmed with Indirect calorimetry for Koreans.

Evaluation of metabolic tumor volume using different image reconstruction on 18F-FDG PET/CT fusion image (18F-FDG PET/CT 융합영상에서 영상 재구성 차이에 의한 MTV (Metabolic tumor volume) 평가)

  • Recently, MTV(metabolic tumor volume) has been used as indices of the whole tumor FDG uptake on FDG PET image but it is influenced by image reconstruction. The purpose of this study was to evaluate the correlation between actual volume and metabolic tumor volume applying different SUVmax threshold for different reconstruction algorithm on phantom study. Measurement were performed on a Siemens Biograph mCT40 using a NEMA IEC body phantom containing different size six spheres filled with F18-FDG applying four SBRs (4:1, 8:1, 10:1, 20:1). Images reconstructed four algorithms (OSEM3D, OSEM3D+PSF, OSEM3D +TOF, OSEM3D+TOF+PSF) and MTV were measured with different SUVmax threshold. Overall, the use of increasing thresholds result in decreasing MTV. and increasing the signal to background ratio decreased MTV by applying same SUVmax threshold. The 40% SUVmax threshold gave the best concordance between measured and actual volume in PSF and PSF+TOF reconstruction image. and the 45% threshold had the best correlation between the volume measured and actual volume in OSEM3D and TOF reconstruction image. we believe that this study will be used when the measurement of MTV applying various reconstruction image.

Functional characterization of a CCCH type zinc-finger protein gene OsZF2 by ectopic overexpression of the gene in rice (과발현 형질전환벼에서 CCCH type zinc-finger protein 유전자 OsZF2 기능 분석)

  • We have previously isolated a CCCH type zinc-finger protein gene, OsZF2 (Oryza sativa Zinc Finger 2), from the cold-treated rice cDNA library. To investigate the potential role of OsZF2, transgenic rice lines over-expressing OsZF2 under the control of CaMV 35S promoter have been developed through Agrobacterium-mediated transformation. Elevated level of OsZF2 transcripts was confirmed by RNA gel blot analysis in transgenic rice. Under the 100 mM NaCl condition, the transgenic rice showed significantly enhanced growth rate in terms of shoot length and fresh weight, implicating that OsZF2 is likely to be involved in salt response of rice. In the field condition, however, the transgenic rice showed a dwarf phenotype and flowering time was delayed. Genome expression profiling analysis of transgenic plants using the 20K NSF rice oligonucleotide array revealed many up-regulated genes related to stress responses and signaling pathways such as chaperone protein dnaJ 72, salt stress-induced protein, PR protein, disease resistance proteins RPM1 and Cf2/Cf5 disease resistance protein, carbohydrate/ sugar transporter, OsWAK kinase, brassinosteroid LRR receptor kinase, and jasmonate O-methyltransferase. These data suggest that the CCCH type zinc-finger protein OsZF2 is a upstream transcriptional factor regulating growth and stress responsiveness of rice.

Histone H3 Lysine Methylation in Adipogenesis (Adipogenesis에서 히스톤 H3 lysine methylation)

  • Adipogenesis as a model system is needed to understand the molecular mechanisms of human adipocyte biology and the pathogenesis of obesity, diabetes, and other metabolic syndromes. Many relevant studies have been conducted with a focus on gene expression regulation and intracellular signaling relating to Peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT/enhancer-binding protein alpha (C/EBPα), which are master adipogenic transcription factors. However, epigenome regulation of adipogenesis by epigenomic modifiers or histone mutations is not fully understood. Histone methylation is one of the major epigenetic modifications on gene expression in mammals, and histone H3 lysine methylation (H3Kme) in particular implicates cell differentiation during various tissue and organ development. During adipogenesis, cell type-specific enhancers are marked by histone H3K4me1 with the active enhancer mark H3K27ac. Mixed-lineage leukemia 4 (MLL4) is a major H3K4 mono-methyltransferase on the adipogenic enhancers of PPARγ and C/EBPα loci. Thus, MLL4 is an important epigenomic modifier for adipogenesis. The repressive mark H3K27me3 is mediated by the enzymatic subunit Enhancer zeste homolog 2 (EZH2) of the polycomb repressive complex 2. EZH2-mediated H3K27 tri-methylation on the Wnt gene increases adipogenesis because WNT signaling is a negative regulator of adipogenesis. This review summarizes current knowledge about the epigenomic regulation of adipogenesis by histone H3 lysine methylation which fundamentally regulates gene expression.

Histological Observation on the Seasonal Changes of Distribution of Muscle Components in Hibernant Fish - 1.   • This study was carried out to obtain fundamental data on the metabolism of hibernant fish loach, Misgurnus mizolepis. Main focus was on the compositon of muscle components and its changes in fresh - water loach before and after spawning season and before and after hibernation. Distributional changes of carbohydrate, protein and lipid in the muscle tissues were also investigated. Change patterns of miosture and crude protein, and moisture and crude lipid were in inversely proportional, i.e. : moisture amount showed the lowest value after spawnig season, the highest just after hibernation, but crude protein and crude lipid were the highest values after spawning season, and the lowest just after hibernation. Carbohydrate showed the highest value just before hibernation and tended to decrease thereafter. Muco layer of epidermis and muscle cell of hypodermis layer in loach were remarkable in its PAS dyeing degree after sapwning season, and it was presumed to include high percentage of protein or carbohydrate. Dermis layer became thinner before spawning hibernation. Lipid component in female tended to distribute relatively widely in the muscle cell layer before spawning season, but in case of male mainly in muco layer and epidermis layer. It appeared that lipid was spreaded mainly in epidermis and hypodermis tissue after spawning season, while it prevailed in almost all tissues but tended to decrease after hibernation.

Vegetation Structure around Abeliophyllum distichum Habitats (미선나무 자생지 주변의 식생구조)

  • This study was conducted to establish the effective and systematic counter plan of conservation by analyzing vegetation on Korean endemic plants and rare and endangered plants, Abeliophyllum distichum, habitats and to present the raw data and direction on counter plan of restoration. The dominant species of tree layers were Platycarya strobilacea(Goesan-gun Chujeom-ri I), Pinus densiflora(Goesan-gun Songdeok-ri II and Yulji-ri III), Robinia pseudo-acacia(Yeongdong-eup Maechon-ri IV), Quercus variabilis(Jincheon-gun Yongjeong-ri V) and Pinus densiflora$.$Quercus variabilis(Buan-gun Junggye-ri Ⅵ). Subtree layers were dominated by Quercus dentata(1st site), Quercus serratu(2nd site), Quercus acutissima(3rd site), Robinia pseudo-acacia(4th site), Zelkova serrata(5th site) and Quercus variabilis(6th site). And the dominant species of shrub layers were Lindera obtusiloba(1st and 5th site), Lespedeza bicolor(2nd site), Stephunandra incisa(3rd site), Quercus aliena(4th site) and Rhododendron schlippen-bachii(6th site). The dominant species of herb layers were surveyed Humulus japonicus$.$Pueraria thunbergiana(1st site), Oplismenus undulatifolius(2nd and 4th site), Carex siderosticta$.$Pueraria thunbergiana(3rd site), Streptolirion cordifolium$.$Humulus japonicus(5th site) and Sasa borealis(6th site). Among the surveyed sites, the Goesan-gun Yulji-ri supremely abounded in species and individuals, and there was surveyed each 21 species and 116 individuals. As well, this site showed the highest H'(1.123) and H'$_{max}$(1.322). Therefore, Goesan-gun Yulji-ri formed diverse vegetation and Yeongdong-eup Maechon-ri and Buan-gun Junggye-ri showed relatively poor vegetation.ation.
