Changes in Coagulase Serotype of Staphylococcus aureus Isolates in Busan, 1994-2005 (부산지역에서 분리된 Staphylococcus aureus의 Coagulase 혈청형 변화)

  • We analyzed the phenotypic changes in coagulase serotype of S. aureus isolated from clinical sources and nasal cavities of healthy persons, $1994{\sim}2005$. A total of 715 isolates, 408 methicillin resistant S. aureus (MRSA) from clinical sources and 307 methicillin-susceptible S. aureus (MSSA), were classified into eight coagulase sero-types, I to VIII. The most prevalent serotype in MRSA was type II (54.3%, 222/408) and followed IV (24.7%), III (10.9%), and V (5.2%), whereas the majorities in MSSA were type VII (30.9%, 95/307), IV (22.2%), V (22.2%) and II (7.1%). Among the isolates collected periods, significant changes of coagulase serotypes in both strains were observed. In MRSA strains, the serotype V was not detected until 1997, but rapidly increased to 18.5% (20/108) in 2005, and the serotypes III decreased from 27% (31/115) in 1994 to 0.9% (1/108) in 2005. A similar trend in MSSA strains was observed but serotype II strain was not detected in 2005. The antigenic shift and changes in the coagulase of S. aureus were confirmed.

Isolation and Charactrization of Lactic Acid Bacteria and Yeast from Traditional Andong sikhe (전통 안동식혜로부터 젖산균 및 효모의 분리 및 그 특성)

  • We isolated seventy two lactic acid bacteria and two hundred yeast from traditional Andong sikhe. Microorganisms were isolate from 4 kinds of traditional Andong sikhe. The optimum temperature and pH for growth of sikhe yeast, No. SCS 5, was $30^{\circ}C$ and 4.5, respectively. SCS 5 produced $CO_2$ gas and 1.5% of alcohol in malt extract broth. SCS 5 was identified as S. cersvisiae from the observation of shape of vegetative reproduction, morphological and cultural chararteristics, fermentation and assimilation of carbon sources, and physiological characteristics.

  • This study was conducted to investigate the fermentation forms of goat milk, and the changes in the flavor of goat milk fermented using single and mixed strains of commercial Lactobacilli during storage. The mixed strains reached a lower pH more quickly than the single strains. The mixed strains also had higher rates of Lactobacillus proliferation. The tastes detected in sensory tests can differ depending on the skill levels of the panel, making it difficult to obtain reproducible and objective data when numerous samples are analyzed. Therefore, we measured changes in flavor during storage using taste sensors. The taste sensors measured diverse flavor changes in goat milk fermented using single strains and mixed strains. Notably, this study is the first in our country to measure changes in the taste and composition of fermented milk during cold storage using taste sensors. This work could have great value for the maintenance and monitoring of dairy products within their expiration dates.

Variation of Disease Severity by Mixed Inoculation of Compatible and Incompatible Races of Bacterial Blight in Rice (비친화적 및 친화적 레이스의 혼합접종에 따른 벼흰잎마름병 발병도의 변화)

  • Compatible and incompatible interactions of near-isogenic lines containing one of Xa1, Xa3, and Xa7 resistance genes with Japanese bacterial blight isolates (T7174, T7147, and T7133) were examined in order to determine the variation of bacterial blight resistance and the stability of resistance gene. IRBB 101 line having a Xal gene was compatible (host susceptible) with T7147 and T7133 isolates but incompatible (host resistant) with T7174 isolate at all the tested rice growth stages. IRBB 103 line having a Xa3 gene was susceptible or moderately resistant to the three isolates at seedling and maximum tillering stage but resistant at heading stage. IRBB 101 line having a Xa7 gene was semi-compatible with the three isolates at seedling stage but incompatible at the other growth stages. Overall there were clear differences between compatible and incompatible interactions of rice with Xanthomonas oryzae pv. oryzae races. In the mixed inoculations of compatible and incompatible isolates, the lesion length from near-isogenic lines decreased as the ratios of incompatible races increased. When the distinction between compatible and incompatible isolates was unclear, there was almost no variation of lesion length regardless of mixed ratios. The pathogenicity of the mixed races in the incompatible Interactions increased rather than the individual inoculation whereas the lesion length of compatible interactions was similar to that of the individual inoculation. These data indicate the incompatible races inhibit the virulence of a compatible race but compatible races increase the disease occurrence due to incompatible races. Furthermore, IRBB 107 line that showed resistance to all the isolates at all the tested growth stages was considered as a good parent f3r breeding of resistant variety.

The influence on cultivation characteristics of fruiting body of winter mushroom by growing humidity (재배사의 습도가 팽이버섯 재배에 미치는 영향)

  • To elucidate the effect of humidity to the characteristics related to mushroom cultivation, five white strains and four brown strains of Flammulina velutipes were cultivated and investigated on their characters. The periods for fruiting initiation, growth and harvest were a little decreased when humidity increased after peak at 75%. Brown strains showed fast fruiting than white ones. ASI 4103 was the fastest and ASI 4166 and ASI 4153 were the latest. The productivity of ASI 4166 and ASI 4149 were the best in all conditions. In general, white strains showed high yields at 75% and brown ones did at 95%. Individual mushroom weight decreased and water content of mushroom increased in most strains when humidity increased.

Serological Analysis of the Antigenicity during Cultivation of Streptomyces Strains (Streptomyces 속 균주들의 생장에 따른 외피 항원성 변화의 혈청학적 분석)

  • The changes in antigenicity during cultivation of streptomycetes were determined by immunodiffusion assay and indirect ELISA. New precipitin lines in immunodiffusion assay began to appear in the growth period of the soluble pigment production and became thickened thereafter. The increase in the antigenicity was also confirmed by ELISA. The antigenic development was relatively weak for S. lavendulae and S. viridochromogenes while that was strong for S. lavendulae and S. viridochromogenes. The results indicated that Streptomyces strains, even though not proved for some strains, changed the compositions of cell envelope during submerged growth and this could be estimated quantitatively by serological method.

  • Prophage cured strain derivatives from Luctobacillirs araei YIT 9018 were isolated from thermoinducible mutant of the parent lysogenic strain. Two thermoinducible mutants were isolated from L. casei YIT 9018 strain treated with N-methyl-N'-nitro-N-nitrosoguanidine. Prophage cured strains were selected after heat induction of thermoinducible strains at $42^{\circ}C$ for 30 min in MRT medium containing anti- 4 FSV serum. The prophage cured strains, L. casei HYM 1213 and L. casei HYM 4024, could be used an indicator strain for temperate phage $\phi$ FSW. The growth, lactic acid producing ability and carbohydrates fermentation of L. casei HYM 1213 were similar to the parent L. cmei YIT 9018 strain, but A. casei HYM 4024 was not. One of the prophage cured strain, L. cmei HYM 1213, could be used industrially .to produce lactic acid beverages because this strah could not induce the virulent phage$\phi$FSV. The physiological characterization of L. casei HYM 1213 strain was similar to the parent L. casei YIT 9018 strain.

The Preventive Effect of Bacillus polyfermenticus KJS-2 and Bacillus mojavensis KJS-3 on Triton WR-1339-induced Hyperlipidemia (Triton WR-1339로 유도된 고지혈증에 대한 Bacillus polyfermenticus KJS-2와 Bacillus mojavensis KJS-3의 예방효과)

  • The purpose of this study was to evaluate the possibility that administration of Bacillus polyfermenticus KJS-2 (BP2), Bacillus mojavensis KJS-3 (Moja3), and their mixtures could control serum lipid levels. We observed changes in the blood cell level, metabolic function evaluation, and blood lipid levels after two weeks of oral administration of these microbial strains to hyperlipidemia-induced rats. Measurements of major cell changes in the white blood cells (WBC) indicated no significant effects due to the administration of the microbial strains. Platelet (PLT) levels decreased by 18.4% in the Triton WR-1339-treated group (NCON) and recovered to the control (CON) group levels in the positive control (PCON) group and the microbial strain-administered groups (p<0.05). No functional changes were observed in red blood cells (RBC) by Triton WR-1339-induced hyperlipidemia. The blood AST, ALT, BUN, and creatinine levels did not indicated effects on liver and kidney function, and all rats administered the microbial mixture recovered. The blood lipid levels in the microbe-treated groups indicated reduced levels of triglyceride (TG) and total cholesterol (TC), and increased levels of serum HDL cholesterol. The HMG-CoA inhibition rate of 7-O-succinyl macrolactin A (SMA) produced by BP2 showed similar activity at a concentration of 1,000 times lower than that achieved with atorvastatin. The administration of the microbial strains to the Triton WR-1339-induced rat model of hyperlipidemia resulted in reduced weight gain without affecting the food and water intake. Thus, blood circulation can be improved by controlling serum lipid levels by the combined administration of the BP2 and Moja3 microbial strains.

  • 5'-Xanthylic acid 생성균주인 Brevibacterium ammoniagenes ATCC 21263 R에 XMP에서 GMP로의 전환효소인 GMP synthetase활성을 부여하기 위해 동종간 세포융합을 시도하여 융합균주들을 얻었다. 이들 우량 융합균주들과 융합모균의 GMP synthetase 활성을 측정하여 상호 비교하였으며, pH 변화에 따른 GMP synthetase 활성과 GMP 생성량과의 관계를 검토하였다. 또한 최적 pH에서 균성장에 따른 당소모량과 GMP 생성량을 비교하였다.
