Selection and Antagonistic Mechanism of Pseudomonas fluorescens 4059 Against Phytophthora Blight Disease (고추역병과 시들음병을 방제하는 토착길항세균 Pseudomonas fluorescens 4059의 선발과 길항기작)

  • In oder to select the powerful rhizophere-dorminatable biocontrol agent, we had isolated an indigenous antagonistic bacterium which produced antibiotic and siderophore from a disease suppressive local field soil of Gyungsan, Korea. And we could select the Pseudomosp. 4059 which can strongly antagonize against Fusarium oxysporum and Phytophthora capsici by two kinds of antifungal mechanism that can be caused by the antibiotic of Phenazin, a siderophore and a auxin like subThe selected strain was identified as Pseudomonas fluorescens (biotype A) 4059 by biochemical tests, API $\textregistered$ test, MicroLog TM system and 16S rDNA analysis. The selected antagonistic microorganism, Pseudomosp. 4059 had an antifungal mechanism of antifungal antibiotic and sidrophore. And we were confirmed the antagonistic activity of P fluorescens 4059 with in vitro antifungal test against Phytophthora capsici and in vivo by red-pepper.

The Use of Glufosinate as a Selective Marker for the Transformation of Cucumber (Cucumis sativus L.) (오이의 형질전환을 위반 선발마커로서 Glufosinate의 이용)

  • Agrobacterium tumefaciens-mediated cotyledonary explants transformation was used to produce transgenic cucumber. Cotyledonary explants of cucumber (c.v., Eunchim) were co-cultivated with strains Agrobaderium (LBA4404, GV3101, EHA101) containing the binary vector (pPTN289) carrying with CaMV 355 promoter-gus gene as reporter and NOS promoter-bar gene conferring resistance to glufosinate (herbicide Basta) as selectable marker. There was a significant difference in the transformation frequency depending Agrobacterium strains. The EHA101 of bacterial strains employed gave the maximum frequency (0.35%) for cucumber transformation. Histochemical gus and leaf painting assay showed that 15 individual lines were transgenic with the gus and bar gene. Southern blot analysis also revealed that the gus gene was successfully integrated into each genome of transgenic cucumber.

Antagonistic Assay of Bacillus spp. for Eco-friendly Biological Control of Melon Powdery Mildew (멜론 흰가루병 친환경 생물적 방제를 위한 Bacillus속 균의 길항력 평가)

  • Melon powdery mildew, caused by Podosphaera fusca, is one of the serious diseases of melon plant in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on the inhibition of mycelial growth of various plant pathogens, and control of melon powdery mildew. Based on the 16S rDNA and gyrA gene sequences, the selected antagonistic bacteria, M09, M70, and M99-1, were identified as Bacillus velezensis. These bacteria not only inhibited the mycelial growth of 47~69% in various plant pathogens, but also significantly reduced the incidence of powdery mildew. The three strains selected in this study could be used as potential biological control agents for various plant diseases as well as melon powdery mildew.

A Efficient Selection of Hybrids Following Intergeneric Transfer of Nuclei from Trichoderma harzianum into Gliocladium virens Protoplasts (Gliocladium virens와 Trichoderma harzianum의 속간(屬間) 핵(核) 전이체(轉移體)의 효율적(效率的) 선발(選拔))

  • To obtain hybrids producing antagonisms and plant growth promoting effects by intergeneric nuclei transfer, the nuclei were isolated from the protoplasts of Trichoderma harzianum T95 and treated with colchicine. The nuclei were tranferred into protoplast of multi-auxotrophic Gliocladium virens G88 which cannot grow in minimal medium. The nuclei tranferred into protoplasts of G. virens G88 were selected on the regeneration minimal medium containing chloroneb as a haploid inducer. Low transfer frequency of 0.08% was observed with three chemical treatment, however no segregants were found in the intergeneric nuclei transfer. The various types of hybrids with different morphology were detected when different concentration of chloroneb were treated. These morphologies were classified as parental, recombinant and petite type.

Comparison of the fermented property and isolation of acetic-acid bacteria from traditional Korean vinegar (재래 식초에서 초산균의 분리와 발효특성 신속 비교)

  • For the selection of a starter for vinegar, six strains of acetic-acid bacteria were isolated from traditional Korean vinegar fermented through the static method. These strains were investigated for their acetic-acid fermentation and identification characteristics. The 16S rRNA sequences of six strains were identified as Acetobacter pasteurianus, A. malorum, Gluconacetobacter entanii, Ga. intermedius, and Ga. xylinus respectively. The overoxidation of acetic acid, acetic-acid and pH tolerances, and acetic-acid production of these strains were investigated. None seemed to have been overoxidized. The Gluconacetobacter genus showed acetic-acid tolerance. Among the acetic-acid bacteria, A. malorum V5-7 exhibited the highest pH tolerance. The Ga. intermedius V11-5 and Ga. xylinus V8-1 strains produced colloids that exopolysaccharides of fiber. The acetic-acid production by isolated acetic-acid bacteria and type strain was a achieved at a shaking culture at $30^{\circ}C$ for 5 days. A. malorum V5-7, A. pasteurianus Gam2, and Ga. intermedius V11-5 exhibited the highest acetic acid production. The study results indicate that appropriate strains of acetic-acid bacteria improved the thraditional Korean vinegar fermented through the static method.

Selection of Beneficial Microbial Agents for Control of Fungal Diseases in the Phyllosphere of Cucumber Plant (오이 지상부의 주요 곰팡이 병해의 생물적 방제용 유용미생물의 선발)

  • Bacillus subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 obtained from phyllosphere of cucumber plants were selected for biological control of fungal air-borne diseases. For the downy mildew, diseased area of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 showed 0.5%, 20.2% and 42.0%, but that of control was 82.0% respectively, in cucumber seedling test. Incidence of powdery mildew by once application of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 was 2.8%, 3.6% and 12.3%, respectively, whereas that of control was 65.6%. On the gray mold, diseased area of B. subtilis B29, B. subtilis M10 and Streptomyces sp. CC19 was 8.0%, 30.8% and 5.2%, respectively, compared to 81.2% for the control. Therefore, B. subtilis B29 could be a prospective antagonist for biological control of powdery mildew, downy mildew and gray mold of cucumber plant.

${\beta}$-1,4-Xylosidase Activity of Leuconostoc Lactic Acid Bacteria Isolated from Kimchi (김치에서 분리된 Leuconostoc 속 젖산균의 ${\beta}$-1,4-xylosidase 효소생산 특성)

  • The ${\beta}$-xylosidase (EC 3.2.1.37) production capabilities of lactic acid bacteria in the genus Leuconostoc, isolated from a variety of kimchi (fermented vegetables), were examined. The intracellular levels of ${\beta}$-xylosidase were similar to the extracellular levels, when most Leuconostoc lactic acid bacteria were grown in a medium containing xylose as the carbon source. Intracellular ${\beta}$-xylosidase with a maximum activity of $1.2{\pm}0.1units/mL$ (mean${\pm}$standard error) was obtained from Leuconostoc lactis KCTC 13344, which was isolated from fermented Chinese cabbage. The optimum reaction conditions for Leu. lactis KCTC 13344 ${\beta}$-xylosidase activity were pH 6.0 and $30^{\circ}C$, and the addition of most divalent cations, except zinc, to the reaction mixture resulted in a slight increase in enzyme activity. Compared with a media containing other carbon sources, the ${\beta}$-xylosidase activity was 5 times higher when Leu. lactis KCTC 13344 was grown in a medium containing xylose as carbon source. Zymographic analysis indicated that the synthesis of Leu. lactis KCTC 13344 ${\beta}$-xylosidase (approximate size, 64 kDa) is induced by xylose. A maximum intracellular ${\beta}$-xylosidase activity of $7.1{\pm}0.3units/mL$ was obtained in a batch cultivation in an MRS medium containing 30 g/L xylose.

Study on the Contamination of Pseudomonas tolaasii in Oyster Mushroom (느타리버섯에서의 Pseudomonas tolaasii오염 연구)

  • One hundred twenty five bacterial isolates were obtained from the brown blotch-diseased oyster mushrooms collected from markets. Among them, 45 were determined as pathogenic bacteria and white line forming organisms(WLFO) were 6 strains and white line reaction organisms (WLRO) were 6 strains. All of the white line forming isolates were identified as Pseudomonas tolaasii which is a known pathogen of brown blotch disease of oyster mushroom by GC-MIS(Gas chromatography-microbial identification system). Six of the white line reacting organisms were identified as P. chlomraphis, P. fluorescens biotype A and type C. The rest of them were P gingeri, P. agarici, P. fluorescens biotype B, P. chloroyaphis, non-pathogenic P. tolaasii, P. putida biotype A and B etc. For spectrum of activity of tolaasin, culture filtrates from pathogenic isolates were examined by browning of mushroom tissue and pitting of mushroom caps. The weak pathogenic bacteria didn't induce browning or pitting of mushroom tissue. On the other hand, strong pathogenic isolates showed browning and pitting reaction on mushroom. An extracellular toxin produced by P. tolaasii, was investigated. The hemolysis activity test of 6 strains identified as P. tolaasii were 0.8∼0.9 at 600 nm and 3 strains of WLRO were 0.9∼1.0 and Pseudomonas app. were 1.0∼1.2. Observation of fresh mushroom tissue using confocal laser scanning microscopy was carried out for images of optical sectioning and vertical sectioning. Also images of brown blotch diseased oyster mushroom tissue after contamination P. tolaasii was obtained by CLSM.

Effect of Feeding Yogurt Using Enterococcus faecium KHM-11 on the Growth in Piglet (Enterococcus faecium KHM-11를 이용한 요구르트 급여가 자돈의 성장에 미치는 영향)

  • A total of 70 colonies were isolated from the Korean human milk samples on the BCP plate count agar. These LAB isolates were subcultured in 10% reconstituted skim milk, and two strain thereof were finally selected for their highest acid productions. These strains were identified as Enterococcusfaecium based on 16S rDNA sequencing data, named as Enterococcus faecium KHM-11. Sugar utilization of E. faecium KHM-11 was investigated by API 50CH kit, and 19 different sugars including D-arabinose, L-arabinose, galactose, D-glucose, D-fructose, and D-mannose were utilized. For fermentation profiles, a yogurt inoculated by E. faecium KHM-11 after 15 hour, reached at pH 4.09, titratable acidity at 1.10% and average viable counts $1.30{\times}10^9\;CFU/mL$. Effects of the administration of yogurt 0.5% of piglet diet to piglets were investigated for growth rate, analysis of blood and incidence of diarrhea. 24 heads of piglets were divided into two groups: the experimental and the control of 12 animals each. The average growth rate in the yogurt-fed group was higher for 21.67%, compared with control (p<0.05). There were no differences in the concentrations of blood glucose, cholesterol, albumin and globulin between the two treatments. Incidence of diarrhea was no in pigs fed yogurt as compare to control.

Isolation and characterization of antifungal violacein producing bacterium Collimonas sp. DEC-B5 (항진균활성 violacein 색소를 생산하는 Collimonas sp. DEC-B5 균주의 분리 및 특성)

  • Forty-nine pigments were extracted from the collections of 106 pigment producing bacteria from the plant rhizosphere soil. Antibacterial activity test was performed in the subjects of the extracted pigments with plant pathogenic bacteria including Xanthomonas axonopodis and Xanthomonas campestris, and with plant pathogenic fungi including Botrytis cinerea, Colletotrichum acutatum, and Fusarium oxysporum. The yellow pigment by Chryseobacterium sp. RBR9 and the red pigment by of Methylobacterium sp. RI13 showed the antibacterial activities against Xanthomonas axonopodis and Xanthomonas campestris. The violet pigment by Collimonas sp. DEC-B5 showed the antibacterial activity as well as the antifungal activities against Botrytis cinerea and Fusarium oxysporum. Especially, the violet pigment inhibited the growth of Botrytis cinerea more than 65% at MIC $20{\mu}M$. Upon the HPLC analysis result for the isolation of pigment with antifungal activity, violacein (91.6%) and deoxyviolacein (8.4%) were isolated for the pigment by Collimonas sp. DEC-B5. The production amount of the pigment was increased more than 10 times higher when D-mannitol 1.5% and yeast extract 0.2% were added as the nitrogen source to SCB medium. This study suggests that produced violacein by Collimonas sp. DEC-B5 will be effective to control strawberry gray-mold rot fungi by its preventive activity.
