This study was conducted to select rhizobia from hairy vetch (Vicia villosa Roth) with nodulation and excellent nitrogen-fixing ability. Hairy vetch root was collected from 7 of cultivation region of all over the country, rhizobia were isolated from the root nodules. Isolates were re-inoculated into a hairy vetch separately and studied nodulation and nitrogen-fixing ability. As a result, total of 52 Rhizobium isolates were isolated from the hairy vetch root nodules, among these, 16 isolates were Rhizobium which show good growth at more than 0.5% NaCl concentration. These 16 isolates were re-inoculated separately, 8 weeks after, good root nodule formation was observed from Rhizobium sp. RH1, RH3, RH81, RH82, RH84, and RH93 strain treated samples. Six isolates were positive for nitrogen fixing ability, the highest acetylene reduction activity was shown by Rhizobium sp. RH84. Results suggest that the Rhizobium sp. RH84 could be used as the possibility of its application as a green manure crop of hairy vetches in nonuniform salt distribution reclaimed land.
Actinomycetes produce diverse secondary metabolites which have the primary importance in medicine, agriculture and food production, and key to this is their ability to interact with other organisms in natural habitats. In this study, we have investigated the taxonomical and functional diversity of actinomycetes in fecal sample of rhinoceros beetle larvae (Allomyrina dichotoma L.) by using culture-dependent and -independent approaches. For the culture-independent approach, the community DNA was extracted from the sample and 16S rRNA genes of actinomycetes were amplified using actinomycetes-specific PCR primers. Thirty-seven clones were classified into 15 genera and 24 species of actinomycetes. For the culture-dependent approach, 53 strains were isolated from larval feces, of which 27 isolates were selected based on morphological characteristics. The isolates were classified into 4 genera and 14 species, and 24 isolates (89%) were identified as the genus Streptomyces. Many of the representative isolates had antimicrobial activities against plant pathogenic fungi and Gram-positive bacteria. In addition, most of the isolates (78%) showed biochemical properties to hydrolyze cellulose and casein. The results demonstrated that diverse and valuable actinomycetes could be isolated from insect fecal samples, indicating that insect guts can be rich sources for novel bioactive compounds.

Lactic acid bacteria were selected on the basis of lactic acid producing ability from kimchi, a traditional Korean fermented food. Among the initial screening of over 150 strains selected from the sample, 27 strains were selected as lactic acid producing bacteria, and 4 strains were finally selected based on their ability to produce relatively high levels of lactic acid. The four strains were identified as Lactobacillus (L.) plantarum Gk04, Pediococcus pentosaceus Gk07, L. brevis Gk35 and L. curvatus Gk36 by the conventional morphological, cultural, physiological and biochemical characteristics, as well as by 16S rRNA sequence analysis. Among the identified lactic acid bacteria, L. curvatus Gk36 was used for soymilk fermentation. The viable cell counts and acidity values measured for the L. curvatus Gk36 were comparable to the commmercial L. acidopillus. Thus, the L. curvatus Gk36 is a potential probiotic strain to prepare fermented soy products, such as kephir, yogurt, tempeh and soy sauce.
A bacterium, which was named to be Bacillus sp. E64-2, capable of degrading endosulfan was isolated from the environmental sample using enrichment culture technique. The Bacillus sp. E64-2 was able to degrade 99% of 10 mg/L endosulfan in the culture media within 7 days at $30^{\circ}C$. Endosulfan diol was the only intermediate by the endosulfan degrading bacterial culture and the pH value of the culture media was significantly increased to pH 8.4 from pH 7.0 after 7 days of incubation. When the endosulfan and the crude extract of the strain were incubated, endosulfan diol was a major metabolite. Both the enzymatic reaction and the pH-increasing effect contribute to the degradation of endosulfan by the bacterial culture.
This study was investigated the occurrence of sclerotinia rot caused by Sclerotinia sclerotiorum at the major perilla cultivating area, Gangdong-dong, Gangseo-gu, Busan in 1998. The incidence of this disease ranged from 8.1 to 28.3% at Gangdong-dong area during the growing seasons. Symptoms of the disease initially appeared damping-off of infected stems and soft-rot on the leaves of perilla. Under the relatively high humidity, abundant white mycelia of the pathogen formed on the lesion developed into black sclerotia later and the infected leaves were finally fell down. Sixteen isolates, Sl-S16, isolated from diseased lesions showing typical symptoms, and pathogenicity was tested using mycerlial disks. Among them, S2 isolate showing the most strong pathogenicity was selected and identified as Sclerotinia sclerotiorum on the basis of morphological and cultural characteristics. For biological control, an antagonistic bacteria, N4 isolate which effectively inhibited not only mycelial growth of S2 isolate but also suppress sclerotinia rot on the pot assay, was selected and identified as Bacillus megaterium according to Bergey's manual and API system., Wettable powder type, N4 formulation using B. megaterium N4 isolate was developed and estimated its control effect on perilla crops in a plastic house. As a results, N4 formulation which applied before 3 days inoculation of pathogen was effectually controlled Sclerotinia rot as the control value of 98.0%, was more effective than chemical fungicide, benomyl showing the control value of 78.0%. This is the first report of wettable powder formulation as a biocontrol agent using B. megaterium N4 against Sclerotinia rot caused by S. sclerotiorum on perilla.
Two hundred and sixty-eight bacterial strains were isolated from pine mushroom (Tricholoma matsutake) basal soil. In the course of screening for antifungal activity against seven plant pathogenic fungi (Alternaria panax, Botrytis cinerea, Colletotrichum gloeosprioides, Fusarium oxysporum, Phytopthora capsici, Pythium ultimum, Rizoctonia solani) of isolates, strain KM1-15 showed strong antibiotic activity against Alternaria panax and Colletotrichum gloeosprioides. In determining its relationship on the basis of 16S rDNA sequence, KM1-15 strain was most closely related to Bacillus $koguryoae^T$ (AY904033) (99.62%). When assayed with the API 50CHE Kit, unlike Bacillus koguryoae, it is positive for utilization of L-arabinose, cellobiose, inulin, and D-turanose. Results of cellular fatty acid analysis showed that major cellular fatty acids were 15:0 anteiso (35.78%) and 17:0 anteiso (17.97%). In particular, hydroxyl fatty acids such as 13:0 iso 3-OH, 14:0 iso 3-OH, 15:0 iso 3-OH, and 17:0 iso 3-OH were only restricted to strain KM1-15. DNA G+C content was 43.7 mol% and quinone system was MK-7 (100%) in strain KM1-15.
This study was examined to identify the race differentiation and distribution of mating type on Phytophthora capsici population in Korea. One hundred forty three isolates of P. capsici were collected from several locations of Korea in 2005-2007. In 2005, 20 isolates of P. capsici were collected and surveyed as A1 mating type of 75% and A2 mating type of 25%. In 2006, a total of 91 isolates were collected and separated as A1 mating type of 49.0%, A2 mating type of 42.9% and S type (sterile) of 3.3%. Isolates obtained in 2007 were similar to 2006 results. Totally, ratio of mating type of 153 isolates was confirmed that A1 type was 56.6%, A2 type was 39.2%, and S type was 4.2%. Thirteen pepper cultivars with different pathogenic response to 3 typical isolates having different mating were screened among 50 pepper cultivars and determined as race differential cultivars for investigation. The 11 races of P. capsici were found by using 13-race differential cultivars. These results indicated that at least 11 races of P. capsici are existed and confirmed race differentiation of P. capsici in pepper.
This study was carried out to isolate the soil bacteria degrading herbicide napropamide [N,N-diethyl-2-(1-naphthoxy)-propionamide] from the clayey loam soil tested and to clarify the characteristics of the napropamide-degrading bacteria. Twenty strains of the gram-positive and the gram-negative bacteria were isolated and identified from the clayey loam soil tested. Most of them were vigorously proliferated at 100ppm of napropamide and two strains of Staphylococcus spp., Corynebacterium spp. II and Other spp. II were very tolerated to napropamide even at the concentration of 1500ppm. Staphylococcus spp. II and Actinobacillus spp. II of the isolated bacteria degraded more than 20% of the treated napropamide. These two strains could not utilize napropamide as sole nitrogen sources, but could use this compound as sole carbon sources. Napropamide was rapidly decomposed by Staphylococcus spp. II at one-time application and at three-time application of napropamide, but wasn't at two-time application of napropamide.

Nine microbial strains were isolated from the byproduct of ginseng processing and field of ginseng cultivation. Two strains among them were confirmed. Phylogenetic analysis of these 𝛽-Glucosidase strains confirmed that strain GYP-1 belongs to the Rhodotorula and strain GYP-3-3 belong to genus Brachybacterium. Rhodotorula sp. GYP-1 was finally selected due to its high biomass production. The 𝛽-Glucosidase activity of Rhodotorula sp. GPY-1 was assessed at 30 ℃, and Higher than 70% of the enzyme activity was maintained at the temperature range of 20-40℃. Although the optimum pH for the highest enzyme activity was pH 5.0, the enzyme was stable throughout the pH range of 5.0-8.0. In addition, Rhodotorula sp. demonstrated antifungal activity against the ginseng root rot disease caused by Botrytis.
Rice bakanae disease caused by Fusarium fujikuroi is one of the most serious rice diseases in Korea. From 2006 to 2009, 118 F. fujikuroi isolates were collected from various regions of rice fields in Korea. Resistance assay of 118 F. fujikuroi isolates to prochloraz, tebuconazole, and benomyl, were performed using agar dilution method. To investigate inhibitory effects of the fungicides, minimum inhibitory concentration of mycelial growth (MIC) and effective concentration inhibiting mycelial growth by 50% ($EC_{50}$) for 118 isolates were calculated using Sigmaplot 8.02 (Antro, SPSS UK, Ltd). Based on the means of $EC_{50}$ values, baseline resistance values were determined as $0.5{\mu}g{\cdot}mL^{-1}$ for prochloraz, $5.0{\mu}g{\cdot}mL^{-1}$ for tebuconazole and $2.5{\mu}g{\cdot}mL^{-1}$ for benomyl. Number of resistant isolates to each fungicide was 17, 19 and 43 for prochloraz, tebuconazole and benomyl, respectively. Furthermore, 4 isolates showed the double resistance to both prochloraz and tebuconazole, 6 isolates to prochloraz and benomyl, and 11 isolates to tebuconazole and benomyl. Isolates CF366 and LF335 isolated from Gyeongbuk province were resistant to the three fungicides tested, prochloraz, tebuconazole and benomyl.
