Anti-inflammatory effects of fruit and leaf extracts of Lycium barbarum in lipopolysaccharide-stimulated RAW264.7 cells and animal model (염증유도 RAW264.7 세포와 동물모델에서 구기자와 구기엽의 항염 효능)

  • Purpose: Medicinal herbs have recently attracted attention as health beneficial foods and source materials for drug development. Recent studies have demonstrated that extracts of Lycium's fruits and roots have a range of physiologically active substances. The extract of Lycium's leaves has been reported to have excellent anti-oxidant and anti-microbial activity, but its anti-inflammatory efficacy is not known. The chlorophyll present in the leaves can act as an anti-oxidant or pro-oxidant depending on the presence of light. Therefore, this study analyzed the anti-inflammatory effects of Lycium's fruit extract (LFE), leaf extract (LLE), and leaf extract with chlorophyll removal (LLE with CR). Methods: This study examined the inhibitory effects of LFE, LLE, and LLE with CR on pro-inflammatory mediator production as well as on the expression of iNOS and COX-2 in lipopolysaccharide (LPS)-stimulated RAW264.7 cells and BALB/c mice. Results: LFE, LLE, and LLE with CR inhibited the production of pro-inflammatory mediators (NO, $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$) and the expression of iNOS and COX-2 in LPS-stimulated RAW 264.7 cells in a dose-dependent manner. Furthermore, the administration of LLE and LLE with CR inhibited the serum pro-inflammatory cytokine levels and suppressed DNA damage in BALB/c mice. In particular, LLE with CR exhibited the highest anti-inflammatory activity. Conclusion: These results suggest that the fruit and leaves of Lycium are potential therapeutic agents against inflammation.

A Study on Exit Delay of Dominant Technology in Technology Competition: Focused on Sailing Ship Effect (기술경쟁에서 구기술의 퇴출 지연 현상에 관한 연구: 범선 효과를 중심으로)

  • Technology competition is becoming a core element to get a competitive advantage in the market economy and its importance has been growing rapidly. Therefore, lots of studies on technology competition have been conducted and diversified. We focused on 'Sailing ship effect' which accelerates innovation in the old technology in response to the threat of the new technology. We found that previous studies had suggested the factors causing 'Sailing ship effect' from the viewpoint of either market or competitor. However, there are a lot of limitations to analyse the status and establish strategies. Hence, we hereby suggest a new technology competition analysis model considering market, technology and especially competitors at once by classifying the results of technology competition: 1) Replacement, 2) Status Quo, 3) Co-existence, and 4) Sailing ship effect. In this paper, we apply three cases to the suggested model in order to judge whether it is reasonable or not and confirm that it explains many cases related to 'exit delay' of a dominant technology. We expect this study will contribute to forecasting the results of technology competition and establish a response strategy if it enters into general use.

Free Radicals Scavenging Activity of Bulro Kugi (Lycium chinense Mill) Fruit, Leaf and Root (불로 구기 부위별 자유라디칼 소거효과에 관한 연구)

  • Free radical-scavenging activities of Korean Bulro Kugi (Lycium chinense Mill) fruit, leaf, and root were evaluated. Total phenolic contents of fruit, leaf, and root were $1,078.4{\pm}61.0$, $939.9{\pm}19.8$, and $3,792.2{\pm}106.6mg/L$, and their flavonoids were $396.7{\pm}15.2$, $1,952.9{\pm}21.3$, and $425.3{\pm}13.5mg/L$, respectively (p<0.001). HPLC revealed main polyphenolic compounds in fruit were p-coumaric and syringic acids in fruit, p-coumaric, syringic, and procatechuic acids in leaf, and p-coumaric, syringic, caffeic, and procatechuic acids in root. Highest radical-scavenging activities of superoxide anion and hydroxyl were found in leaf and root, respectively (p<0.001).

Fermentation Properties of Yogurt Added by Lycii fructus, Lycii folium and Lycii cortex (구기자, 구기엽 및 지골피를 첨가한 요구르트의 발효 특성)

  • This experiment was carried out to examine the fermentation properties of yogurt with Lycii fructus, Lycii folium and Lycii cortex powder, and extract additives at concentrations of 0.5, 1.0, 2.0, 4.0, and 6.0%. Lactic acid bacteria was used in a mixed starter culture of Streptococcus salivarius ssp. thermophilus(ST36) and Lactobacillus delbrueckii ssp. bulgaricus(LB12). When the boxthorn was added with extract types, the changes of pH, acidity and lactic acid bacteria counts of yogurt during the fermenation of 3 hours were pH 5.64, titratable acidity 0.85%, 5.80xl0$\^$6/cfu/ml of viable cell counts for control yogurt, whereas those were pH 4.10∼5.06, titratable acidity 0.98∼1.27%, 1.80∼9.60x10$\^$7/ cfu/ml of viable cell counts for Lycii fructus extract yogurt. The lactose hydrolysis ratio was better for 1.0% Lycii fructus extract yogurt(42.00%) and 1.0% Lycii folium extract yogurt(41.46%) than for control yogurt(28.40%). Also, content of lactic acid of 1.0% Lycii fructus(11.9 times) and 1.0% Lycii folium extract yogurt(10.6 times) produced more than control yogurt(7.3 times). The viscosity of yogurt was better for boxthorn extract yogurt(1,027∼1,382 cps) than for control yogurt(975cps). The sensory scores of color, taste and overall acceptability of yogurt with 0.5, and 1.0% Lycii fructus extract additive were better than other groups. The yogurts made with increased Lycii fructus extract concentration(0.5∼6.0%), showed the increase of lactic acid, titratable acidity, number of lactic acid bacteria, viscosity and lactose hydrolysis rate compared to the treatments of 0.5, 1.0, 2.0, and 4.0% Lycii folium and Lycii cortex extract and powder yogurt. We gained excellent results from the yogurt to which Lycii fructus extract was added with 0.51.0% concentration.

구기자(Lycium chinence Miller) 첨가 요구르트의 항산화 활성과 ACE 저해 및 ${\alpha}-glucosidase$ 저해 효과

  • 구기자, 구기엽, 지골피 추출물을 이용한 요구르트를 제조하여 항산화효과, ACE (angiotensin converting enzyme) 저해 효과 및 ${\alpha}-glucosidase$ 저해 활성을 시험하였다. 구기자 첨가 발효유의 Antioxidants activity는 물 추출물보다 methanol 추출물에서 더 많은 항산화 활성능을 보였으며 특히 구기엽 요구르트에서 83.85%의 높은 활성능을 보였다. 구기자와 지골피 요구르트에서는 각각 47%와 54%내외의 활성능을 나타냈다. 구기자 첨가 발효유의 ACE(angiotensin converting enzyme) 저해 효과는 원재료에서는 물 추출물이 angiotensin converting enzyme 저해능이 높았고, 구기자 요구르트의 경우 물 추출물과 methanol 추출물 모두가 angiotensin converting enzyme 저해능이 높은 것으로 나타났다. 구기자 첨가 발효유의 ${\alpha}-glucosidase$ 저해 활성은 구기자, 구기엽 및 지골피 4% 첨가 요구르트에서 methanol 추출물의 경우 각각 8.2%, 5.6%, 7.3%의 ${\alpha}-glucosidase$ 저해능을 나타냈는데 이들 모두 원재료 보다는 활성 저해능이 큰 것으로 나타났으나 물 추출물의 경우에는 활성 저해능이 거의 나타나지 않았다.

Effects of Antioxidant Activity and Changes in Vitamin C during Storage of Lycii folium Extracts Prepared by Different Cooking Methods (조리방법을 달리한 구기엽의 저장기간별 항산화 효과 및 비타민 C 함량의 변화)

  • Contents in polyphenol and vitamin C and their antioxidant activities during storage of Lycii folium prepared by different treatment methods were investigated. The antioxidant activity of extracts prepared from 80% MeOH was evaluated using DPPH radical scavenging and SOD-liked activities. Total phenolic acid contents were 1.34 mg/mL when dry, but extracts during 15-day storage after NaCl 0% treatment decreased. DPPH radical scavenging activity of extracts was 65.48% when dry and decreased during 15-day storage after dry and NaCl 0% treatment. SOD-liked activity was 98.63% in NaCl 0% during 15-day storage after dry. Contents of vitamin C decreased during 15-day storage after dry and NaCl 0% treatment. Consequently, our results indicate that Lycii folium prepared from dry treatment could be a promising biomaterial for the production of functional food for total phenolic acid and vitamin C.

Effects of the Ethanol Extract from Lycii folium Leaves on Obesity and Blood Biochemical Indices in High-fat Diet Induced Obese Rats (구기엽 에탄올 추출물이 고지방 식이 유도 비만쥐의 비만과 혈중 생화학적 지표 조절에 미치는 영향)

  • This study was conducted to investigate the effects of ethanol extract from Lycii folium (L. folium) leaves on obesity index, serum lipids, homocysteine, leptin, ghrelin, and glucose in obese rats. Sprague-Dawley rats were administrated high-fat diets to induce obesity. Then, obese rats were divided into three diet groups: a basal-diet obese group (BO group), high-fat diet obese group (FO group), and high-fat diet plus L. folium extract obese group (FLEO group). Three groups were each fed experimental diet for 8 weeks. There were no significant differences in body weight and FER among the groups. However, obesity index such as Ro?hrer index, Lee index, and T.M. index of FLEO group was significantly decreased as compared to FO group. While serum triglyceride of BO group was significantly decreased as compared to FO group, there were no significant differences in serum lipids, homocysteine, leptin, ghrelin and glucose between FLEO group and FO group. In conclusion, these results indicated that ethanol extract from L. folium leaves might be beneficial with anti-obese effect by reduction of obesity indices in obese rats.
